In Vitro Antibacterial Effects of Yeonkyokeumpae-jeon against Escherichia coli (연교김패전(連翹金貝煎)의 Escherichia coli에 대한시험관내 항균력 평가)

  • Objectives: The object of this study was to observe the in vitro antibacterial effects of Yeonkyokeumpae-jeon (YKKPJ) have been used for treating various gynecological diseases including mastitis in Korea, and individual six kinds of herbal composition aqueous extracts - Forsythiae Fructus (FF), Millettiae Caulis (MC), Lonicerae Flos (LF), Fritillaria Thunbergii Bulb (FT), Taraxci Herba (TH) and Prunellae Spica (PS) against E. coli. Methods: Antibacterial activities against E. coli of YKKPJ, FF, MC, LF, FT, TH and PS aqueous extracts were detected using standard agar microdilution methods. In addition, the effects on the bacterial growth curve were also monitored at MIC and $MIC{\times}2$ levels. The effects on the intracellular killing and bacterial invasion of individual test materials were also observed using Raw 264.7 and MCF-7. The results were compared with ciprofloxacin, a second generation of quinolone antibiotics in the present study. Results: MIC of YKKPJ, FF, MC, LF, FT, TH, PS aqueous extracts against E. coli were detected as $0.039{\pm}0.013mg/ml$, $0.064{\pm}0.033mg/ml$, $0.108{\pm}0.053mg/ml$, $0.078{\pm}0.027mg/ml$, $16.250{\pm}8.385mg/ml$, $15.625{\pm}9.375mg/ml$, $0.254{\pm}0.131mg/ml$, repectively. YKKPJ, FF, MC, LF, FT, TH, PS aqueous extracts showed antibacterial effects against to E. coli, except for FT and TH, which were showed negligible antibacterial effects, respectively. In addition, ciprofloxacin with YKKPJ, FF, MC, LF and PS aqueous extracts also showed marked dosage-dependent inhibition of bacterial growth, and favorable inhibitory effects on the both bacterial invasion and intracellular killing assays using MCF-7 and Raw 264.7 cells were detected in this experiment. Conclusions: The results obtained in this study suggest that traditional polyherbal formula YKKPJ aqueous extracts showed more favorable antibacterial activities as compared to individual six kinds of herbal composition aqueous extracts. The antibacterial effects of YKKPJ against E. coli considered as results of complicated synergic effects of their six kinds of herbal components rather than simple antibacterial effects of single herbal components. It means, YKKPJ aqueous extracts may show potent anti-infectious effects against E. coil for mastitis.

Growth Inhibition Effects of Ethanol and Sodium Chloride on Bacillus cereus (Ethanol과 NaCl에 의한 Bacillus cereus 생육저해 영향)

  • Agricultural foodstuffs, usually consumed without sterilization, are frequently contaminated with foodborne pathogen B. cereus. Ethanol and sodium chloride were used to inhibit this pathogen for its effective control. Though five minutes of exposure to 20% ethanol did not inhibit the growth of B. cereus, inhibition was detected to 30% ethanol solution. As exposed longer, B. cereus was more effectively inhibited than E. coli and S. typhimurium. B. cereus, E. coli and S. typhimurium were not inhibited when exposed in a 10% sodium chloride solution for five minutes. However, these bacteria were inhibited with a combination of 10% sodium chloride and 20% ethanol solution for five minutes. Much synergistic growth inhibition on B. cereus was found at the treatment. Its viable count was reduced from $10^8cfu/ml$ to $10^2cfu/ml$ after five minutes and showed no count after ten minutes. This trend was also confirmed for the wild types of B. cereus. This method may be applied for the effective pre-treatment of many agricultural foodstuffs, especially uncooked foodstuffs, without the hazards that accompany special sanitizers and the nutritional loss from harsh sterilization.

Cloning and mutational analysis of pyrroquinoline quinone(PQQ) genes from a phosphate - solubilizing biocontrol bacterium Enterobacter intermedium.

  • E. intermedium 60-2G possessing a strong ability to solubilize insoluble phosphate, has plant growth-promoting activity, induced systemic resistance activity against scab pathogen in cucumber, and antifungal activity against various phytopathogenic fungi. The phosphate solubilizing activity of 60-2G may be mainly accomplished by production of gluconic acid through a direct extracellular oxidation of glucose by glucose dehydrogenase that required a PQQ cofactor for its activation. A pqq gene cluster conferred Phosphate-solubilizing activity in E. coli DH5${\alpha}$ was cloned and sequenced. The 6,783 bP pqq sequence had six open reading frames (from A to F) and showed 50-95% homology to pqq genes from other bacteria. The E. coli strain expressing the pqq genes solubilized phosphate from hydroxyapatite after a pH drop to 4.0, which paralleled in time the secretion of gluconic acid. To study the role of PQQ in biocontrol traits of E. intermedium, PQQ mutants of 60-2G were constructed by marker exchangee mutagenesis. The PQQ mutants of E. intermedium were lost activities of solubilizing phosphate, growth inhibition of phytopathogenic fungi, and plant growth promotion. These findings suggest that PQQ plays an important role, possibly activation of certain enzymes, in several beneficial bacterial traits of E. intermedium by as yet an unknown mechanism.

  • GlcNAc 2-epimerase gene from human was cloned. However GIcNAc 2-epimerase was expressed in E. coli as inclusion body formation. Several approaches were tried such as expression in low temperature and low concentration of IPTG. With these treatments production of active form of human GIcNAc 2-epimerase ι ,vas enhanced. For the direct synthesis of NeuAc from GlcNAc and pyruvate, NALase and GlcNAc 2-epimerase were characterized in terms of temperature effect on activity. equilibrium and stability, inhibition by pyruvate etc. For cheap and ease preparation of both the NALase and GlcNAc 2-epimerase, pEN24ma vector was made. which express both the NALasc and GIcNAc 2-epimerase simultaneously. In addition, E. coli BL21(DE3) harboring two plasmids was also made. Of the two systems, the latter was better for the expression of both enzymes.

Effect of Pyrimidylsalicylate on the Valine Sensitive Acetolactate Synthase Purified from Serroatia marcescens

  • The inhibitory effect of herbicides such as sulfonylurea derivatives, imidazolinones and pyrimidylsalicylate has been examined on the purified valine sensitive acetolactate synthase (ALS) from Serratia marcescens. The concentration of sulfometuron methyl which inhibits 50% of the ALS activity was 2.5 mM. The required concentrations of triasulfuron, primisulfuron methyl and imazaquin for the 50% inhibition of the ALS activity were 1 mM. The resistance of Serratia ALS to sulfometuron methyl, imazapyr and imazaquin is similar to that of E. coli ALS 1. However, pyrimidylsalicylate showed a potent inhibitory effect on the Serratia ALS almost 13 times more potent than on E. coli ALS II, which is known as herbicide-sensitive isozyme. The inhibitory mode was competitive against pyruvate. 150 value was determined to be $17{\mu}M$ in an assay mixture containing 20 mM pyruvate, and the $K_1$, value was calculated to be $0.4{\mu}m$ from the modified double reciprocal plot of 1/V versus $1/S^2$.

Microbial Inhibition by an Isolate of Pediococcus from Kimchi (김치에서 분리한 Pediococcus 의 미생물 생육 저해)

  • Lactic acid bacteria isolated from Kimchi were tested for inhibitory activity against E. coli, Streptococcus faecalis and Lactobacillus bulgaricus. The inhibitory strains appeared most frequently around the middle stage of fermentation during two weeks of observation, and the majority of them were identified as Pediococcus cerevisiae. The agent(s) responsible for the inhibitory activity of P. cerevisiae was inactivated by heat treatment at moderate temperature, but resistant to proteolytic enzymes. The production of the inhibitory compound(s) decreased when the pH of the medium was maintained at about 7 with phosphate buffer.

Molecular Cloning and Characterization of a Gene Encoding Thermostable Pectinase from Thermotoga maritima

  • A gene encoding thermostable pectinase (TmPec) was isolated from hyperthermophilic microorganism, Thermotoga maritima. The open reading frame (ORF) of TmPec gene is 1,104 bp long and encodes 367 amino acid residues with a molecular weight of 40,605 Da. To analyze the enzymatic activity and biochemical properties, the ORF of TmPec gene excluding putative signal sequence of 27 amino acids was introduced into the E. coli expression vector, pRSET-B, and overexpressed in E. coli BL21. Protein concentration of purified recombinant TmPec was 1.1 mg/mL with specific activity of 56 U/mg protein on pectin. The recombinant TmPec showed the highest activity at around $85-95^{\circ}C$, and at around pH 6.5. It was stable at temperature below $85^{\circ}C$. In the presence of $Ca^{2+}$, the activity of recombinant TmPec was increased to 146.3% of normal level. In contrast, $Ba^{2+}$ and Mn2+ showed strong inhibition to the recombinant TmPec.

Inhibition of adhesion and biofilm formation in Escherichia coli O157:H7 by diosmin (다이오스민(diosmin)에 의한 병원성 대장균 세포부착 및 생물막 형성 억제)

  • Escherichia coli O157:H7 is one of the most common foodborne pathogens responsible for outbreaks of hemorrhagic colitis, which can lead to the life-threatening hemolytic-uremic syndrome. In this study, we identified phytochemicals that specifically inhibit the expression of LEE operon in E. coli O157:H7. Among phytochemicals, diosmin decreased the adherence of E. coli O157:H7 towards Caco-2 cells in vitro (p<0.01) and its biofilm formation activity (p<0.05). Quantitative RT-PCR analysis revealed that the transcripts of Ler-regulated genes and genes related to curli production were significantly reduced in the presence of diosmin. However, diosmin does not affect bacterial viability, indicating that the resistance rate to diosmin was remarkably low. Overall, these results provide significant insights into the development of a novel anti-infective agent that is different from conventional antibiotics.

Expression of a Glutathione Reductase from Brassica rapa subsp. pekinensis Enhanced Cellular Redox Homeostasis by Modulating Antioxidant Proteins in Escherichia coli

  • Glutathione reductase (GR) is an enzyme that recycles a key cellular antioxidant molecule glutathione (GSH) from its oxidized form (GSSG) thus maintaining cellular redox homeostasis. A recombinant plasmid to overexpress a GR of Brassica rapa subsp. pekinensis (BrGR) in E. coli BL21 (DE3) was constructed using an expression vector pKM260. Expression of the introduced gene was confirmed by semi-quantitative RT-PCR, immunoblotting and enzyme assays. Purification of the BrGR protein was performed by IMAC method and indicated that the BrGR was a dimmer. The BrGR required NADPH as a cofactor and specific activity was approximately 458 U. The BrGR-expressing E. coli cells showed increased GR activity and tolerance to $H_2O_2$, menadione, and heavy metal ($CdCl_2$, $ZnCl_2$ and $AlCl_2$)-mediated growth inhibition. The ectopic expression of BrGR provoked the co-regulation of a variety of antioxidant enzymes including catalase, superoxide dismutase, glutathione peroxidase, and glucose-6-phosphate dehydrogenase. Consequently, the transformed cells showed decreased hydroperoxide levels when exposed to stressful conditions. A proteomic analysis demonstrated the higher level of induction of proteins involved in glycolysis, detoxification/oxidative stress response, protein folding, transport/binding proteins, cell envelope/porins, and protein translation and modification when exposed to $H_2O_2$ stress. Taken together, these results indicate that the plant GR protein is functional in a cooperative way in the E. coli system to protect cells against oxidative stress.

Effects of Spice Addition on the Inhibition of Bacterial Growth in Ground Chicken Meat (향신료 첨가에 의한 닭고기 분쇄육에서의 미생물 증식 억제 효과)

  • Addition of spice for inhibition of bacterial growth in ground chicken meat was investigated. The ground chicken meat approximately contained 72.98±0.15% moisture, 23.37±0.46% crude protein, 1.00±0.03% crude fat, and 1.90±0.03% ashes. Addition of rosemary showed the maximum bacterial inhibition, followed by garlic and mustard. The inhibitory effect increased with the addition of a greater quantity of spices. The optimal added concentration of spices for inhibition of total viable cell and proliferation of Escherichia coli in ground chicken meat was 2%, 4%, and 1.2% for rosemary, garlic, and mustard, respectively. The growth inhibition of total viable cells and E. coli differed during storage period for MixA (97.4%) > rosemary (96.9%) > MixB (96.3%) > garlic (53.7%) > mustard (33.3%). The addition of sterilized garlic to ground chicken meat showed that the total viable cells was low at 2.6-3.0 log CFU/g on the 0-day and 2.4-3.2 log CFU/g on the 9-day, and the number decreased as the storage lengthened. Non-sterilized garlic treatment showed a higher number of total viable cells than the control group, and this increased with elapse of storage time. The number of E. coli, was low at 0.4-1.0 log CFU/g on the 0-day and 0.5-1.5 log CFU/g on the 9-day for the sterilized group, and the change during the storage showed a similar trend for the total viable cells. In conclusion, the microbial safety of ground chicken meat products was improved by various mixed applications of rosemary, garlic, and mustard.
