A Novel Esterase from Paenibacillus sp. PBS-2 Is a New Member of the ${\beta}$-Lactamase Belonging to the Family VIII Lipases/Esterases

  • Screening of a gene library from Paenibacillus sp. PBS-2 generated in Escherichia coli led to the identification of a clone with lipolytic activity. Sequence analysis showed an open reading frame encoding a polypeptide of 378 amino acid residues with a predicted molecular mass of 42 kDa. The esterase displayed 69% and 42% identity with the putative ${\beta}$-lactamases from Paenibacillus sp. JDR-2 and Clostridium sp. BNL1100, respectively. The esterase contained a Ser-x-x-Lys motif that is conserved among all ${\beta}$-lactamases found to date. The protein PBS-2 was produced in both soluble and insoluble forms when E. coli cells harboring the gene were cultured at $18^{\circ}C$. The enzyme is a serine protein and was active against p-nitrophenyl esters of $C_2$, $C_4$, $C_8$, and $C_{10}$. The optimum pH and temperature for enzyme activity were pH 9.0 and $30^{\circ}C$, respectively. Relative activity of 55% remained at up to $5^{\circ}C$ with an activation energy of 5.84 kcal/mol, which indicates that the enzyme is cold-adapted. Enzyme activity was inhibited by $Cd^{2+}$, $Cu^{2+}$, and $Hg^{2+}$ ions. As expected for a serine esterase, activity was inhibited by phenylmethylsulfonyl fluoride. The enzyme was remarkably active and stable in the presence of commercial detergents and organic solvents. This cold-adapted esterase has potential as a biocatalyst and detergent additive for use at low temperatures.

  • 정보의 자원화를 국가 발전의 기본적인 원동력으로 간주하고 있는 일본의 정책 현 황을 일본학술회의, 과학기술청, 통상산업성, 문부성을 중심으로 분석하였다. 정책이 효과적 으로 실행되기 위하여서는 최고 통치권자를 자문하는 협의 기관이 반드시 필요하며 이를 능 독적으로 활동시키기 위한 제도 또한 중요하다. 법률에 의한 명백한 임무의 부여와 관 에리 트와 지식 인간의 협력 또는 마찰의 정도가 정보 정책을 시행하는데 결정적인 요인이 됨을 현재 진행되고 있는 여러 시책 사항을 통하여 지적하였다. 문부성에서 의욕적인 정책으로 추진하여 소기의 성과를 거두고 있는 일본학술정보센터의 정책적 효과를 온라인 네트워크를 편성하는 기본 조직으로 주의 깊게 관찰할 대상임을 강조하였으며 과학기술청이 주관하는 일본 과학기술정보센터의 기능 또한 중요한 기관으로 간주됨을 분석하였다. 이들 기관 이외 에 광범위한 활동을 전개하고 있는 각급 사회 단체 연구소등 반관반민의 조직 또는 민간 레 벨의 조직은 본론에서는 제외되었다.

Cloning of Mouse AQP-CD Gene

  • Water transport in highly-permeable membranes is facilitated by some specialized pathways, which are called aquaporins (AQP). AQP1 (AQP-CHIP) is the first recognized aquaporin identified from red cells and renal proximal tubules. Up until now 4 other aquaporin homologs have been reported. Each aquaporin has its unique tissue distribution and regulatory mechanims. To elucidate molecular mechanisms for their transcription regulation and tissue-specific expression isolation of aquaporin genes is required. To clone promoters of the AQP family mouse genomic library was screened by the 1st exon-specific probe of AQP4, and 5 different plaques were positively hybridized. Phage DNAs were purified and characterized by restriction mapping and sequencing. One of them is the mouse AQP-CD gene. The gene was consisted of 4 exons and the exon-intron boundaries of mouse AQP-CD gene were identified at identical positions in other related genes. The 5'-flanking region of AQP-CD gene contains one classic TATA box, a GATA consensus sequence, an E-box and a cyclic AMP-responsive element. The cloning of the mouse AQP-CD gene, of which product is expressed in the collecting duct and is responsible for antidiuresis by vasopressin, will contribute to understand the molecular mechanisms of tissue-specific expression and regulation of AQP-CD gene under various conditions.

Simultaneous Comparison of Efficacy and Adverse Events of Interventions for Patients with Esophageal Cancer: Protocol for a Systematic Review and Bayesian Network Meta-analysis

  • Background: Esophageal cancer is one of the most serious malignancies. Due to the aggressive nature of this cancer, the prognosis is poor. A network meta-analysis with simultaneous comparison of multiple treatments can help determine better treatment options that have higher effects on overall survival of patients with lower adverse events. The aim of this review is to simultaneously compare efficacy and adverse events of treatment interventions for esophageal cancer. Materials and Methods: In this review, only randomized control trials (RCT) will be considered for network meta-analysis. All international electronic databases including Medline, Web of Sciences, Scopus, Cochran's library, EMBASE and Cancerlit will be searched to find randomized control trials which compared two or more treatment interventions for esophageal cancer. A network plot will be drawn for visual representation of all available treatment interventions. Bayesian approach will be used to combine the direct and indirect evidence. Treatment effects (e.g. hazard ratio for time to event outcomes, risk ratio for binary outcomes, and rate ratio for count outcomes with 95% credible interval) will be reported. Moreover, cumulative probability of the treatment ranks will be reported using the surface under the cumulative ranking (SUCRA) graphs. Consistency assumption will be assessed by the loop-specific and design-by-treatment interaction approaches. Conclusions: The results of this study may be helpful for the patients, clinicians and health policy makers in selecting treatments that have the best effect on survival and lowest adverse events.

Expression and Characterization of Protein Latcripin-3, an Antioxidant and Antitumor Molecule from Lentinula edodes C91-3

  • In this study, an anti-oxidant and anti-tumor protein Latcripin-3 of Lentinula edodes C91-3 was expressed in Escherichia coli. for the first time. According to the cDNA library, the full-length gene of Latcripin-3 was cloned by the methods of 3'-full rapid amplification of cDNA Ends (RACE) and 5'-full RACE. The structural domain gene of Latcripin-3 was inserted into the pET32 a(+). The functional protein of Latcripin-3 was expressed in Rosetta-gami (DE3) E. coli, evaluated by Western blotting and mass spectrometry. DPPH testing showed that the protein Latcripin-3 can scavenge free radicals remarkably well. The activity of functional protein Latcripin-3 on A549 cells was studied with flow cytometry and the MTT method. The MTT assay results showed that there was a decreases in cell viability in a dose-dependent and time-dependent manner in protein Latcripin-3 treated groups. Flow cytometry demonstrated that Latcripin-3 can induce apoptosis and block S phase dramatically in human A549 lung cancer cells as compared to the control group. At the same time, the cell ultrastructure observed by transmission electron microscopy supported the results of flow cytometry. This research offers new insights and advantages for identifying anti-oxidant and anti-tumor proteins.

Gene Cloning, Expression, and Characterization of a New Carboxylesterase from Serratia sp. SES-01: Comparison with Escherichia coli BioHe Enzyme

  • The carboxylesterase-encoding gene(bioHs) of a newly isolated strain, Serratia sp. SES-01, was cloned from the genomic DNA library by detecting formation of transparent halo around the colony on LB-tributyrin agar plates. The amino acid sequence of BioHs was highly similar to the members of the BioH enzyme family involved in the biotin biosynthetic pathway; it showed the highest similarity(91%) with that of Serratia proteamaculans. To compare BioHs with other BioH enzymes, the relatively well-known bioHe gene of E. coli was cloned with PCR. After we achieved high-level expression of soluble BioHs and BioHe through the exploration of different culture conditions, the purified BioHs and BioHe enzymes were characterized in terms of specificity, activity, and stability. BioHe was generally more robust to a change in temperature and pH and an addition of organic solvents than BioHs. The two enzymes exhibited a strong preference for carboxylesterase rather than for thioesterase and were optimal at relatively low temperatures($20-40^{\circ}C$) and alkaline pHs(7.5-9.0). The results in this study strongly suggested that both the BioHs and BioHe enzymes would be potential candidates for use as a carboxylesterase in many industrial applications.

  • 본 연구의 목적은 외국학술지 지원센터로 시범 선정된 K대 도서관의 원문복사서비스에 대한 경제적 가치를 추정하기 위한 것이다. 신제품이나 신규 서비스의 수요 예측에 활용되는 Bass 확산모형이 본 연구에서 활용되었다. 본 연구를 위하여 2007년 9월 10일부터 9월 20일까지 11일간에 걸친 설문조사도 함께 이루어졌다. 경제적 가치의 추정결과 유용가치는 2015년에 20억원을 약간 상회하는 것으로 나타났고, 지불의사가치의 경우는 20억원에 조금 못 미치는 것으로 조사되었다. 본 연구에서 외국학술지 지원센터의 경제적 가치분석은 원문복사서비스에 한정되었고, 이를 위한 분석방법도 유용가치와 지불의사가치로 제한되었다. 유용가치와 지불의사가치의 인구통계적 분석(성별, 연령, 근무처, 학력)도 함께 이루어졌다.

에너지 소비 모니터링을 위한 재목적 인스트럭션-셋 시뮬레이터 (Retargetable Instruction-Set Simulator for Energy Consumption Monitoring)

  • 재목적성(retargetability)은 아키텍처 정보를 정형화된 형식으로 기술하여 컴파일러, 시뮬레이터와 같은 소프트웨어 개발 도구(SDK)를 생성하는데 이용된다. 시뮬레이터는 임베디드 프로세서의 설계를 하드웨어로 구현하기 전에 아키텍처의 다양한 성능 확인과 개선을 위해 소프트웨어적으로 검증할 수 있는 중요한 하드웨어 및 소프트웨어 개발 도구이다. 이러한 시뮬레이터는 시스템의 기능 검증, 성능 측정, 전력 에너지 소비 측정 결과 등을 하드웨어 설계 과정에서 중요하게 활용한다. 이 논문에서는 에너지 소비 측정이 가능한 시뮬레이터를 ADL로부터 생성하기 위해 첫째 에너지 소비 측정 및 모니터링 요소를 ADL에 표현한다. 둘째, ADL 표현으로부터 에너지 측정 및 모니터링 시뮬레이션 라이브러리를 생성한 후 시뮬레이터인 RenenrgySim 을 구축한다. 마지막으로, MiPS R4000에 대한 ADL을 표현을 작성하여 에너지 소비 측정 결과를 제시한다. 이러한 연구는 모바일 임베디드 소프트웨어 개발 분야에서 소프트웨어적인 실험을 통해 효과적인 아키텍처 개발과 신속한 SDK 생성에 활용될 수 있다.

인공지능 학습을 위한 웹 컴파일러 설계 및 구현 (Design and Implementation of Web Compiler for Learning of Artificial Intelligence)

  • 4차 산업혁명과 ICT 기술의 중요성이 증가함에 따라 소프트웨어 중심 사회가 초래되었다. 기존 소프트웨어 교육은 학습 환경구성에 제한적이었으며, 초기에 많은 비용이 발생하였다. 이를 해결하기 위하여 웹 컴파일러를 활용하는 형태의 학습 방법이 개발되었다. 웹 컴파일러는 다양한 소프트웨어 언어를 지원하며, 컴파일 결과를 사용자에게 웹을 통해 보여준다. 하지만 4차 산업혁명의 핵심기술인 인공지능에 대한 웹 컴파일러는 아직 미비한 상황이다. 본 논문에서는 구글 인공지능 라이브러리인 텐서플로우 기반의 웹 컴파일러를 설계, 구현하였다. nodeJS 기반의 서버에 텐서플로우와 텐서플로우 서빙, 파이썬 주피터를 구현하고, meteorJS 기반의 웹 서버를 구축하여 인공지능 학습을 위한 시스템을 구현하였다. 소프트웨어 중심 사회에서 인공지능 학습을 위한 도구로써의 활용이 가능할 것으로 기대된다.

Dose coefficients of mesh-type ICRP reference computational phantoms for external exposures of neutrons, protons, and helium ions

  • Recently, the International Commission on Radiological Protection (ICRP) has developed the Mesh-type Reference Computational Phantoms (MRCPs) for adult male and female to overcome the limitations of the current Voxel-type Reference Computational Phantoms (VRCPs) described in ICRP Publication 110 due to the limited voxel resolutions and the nature of voxel geometry. In our previous study, the MRCPs were used to calculate the dose coefficients (DCs) for idealized external exposures of photons and electrons. The present study is an extension of the previous study to include three additional particles (i.e., neutrons, protons, and helium ions) into the DC library by conducting Monte Carlo radiation transport simulations with the Geant4 code. The calculated MRCP DCs were compared with the reference DCs of ICRP Publication 116 which are based on the VRCPs, to appreciate the impact of the new reference phantoms on the DC values. We found that the MRCP DCs of organ/tissue doses and effective doses were generally similar to the ICRP-116 DCs for neutrons, whereas there were significant DC differences up to several orders of magnitude for protons and helium ions due mainly to the improved representation of the detailed anatomical structures in the MRCPs over the VRCPs.
