  • This study aims to present a model of digital archiving based on E-ARK. It analyzed the international standards and technological specifications designed for digital archiving. The analysis employed in study explored the common specifications including core processes, information packages, and metadata structure needed for digital archiving. Based on the analysis and reviews, this study developed a model for digital archiving, in order to achieve interoperability of information packages throughout the process.

Changes in Antioxidant Enzyme Activity and Physiological Responses to Cadmium and Tributyltin Exposure in the Ark Shell, Scapharca Broughtonii

  • Cadmium (Cd) and tributyltin (TBT) are common contaminants of marine and freshwater ecosystems, and can induce the formation of reactive oxygen species (ROS). These ROS can, in turn, cause oxidative stress. In the present study, we investigated time-related effects of Cd (0.05 and 0.1 ppm) and TBT (5 and 10 ppb) treatment on antioxidant enzyme activity, i.e., the activity of superoxide dismutase (SOD) and catalase (CAT) in the gills and digestive glands of the ark shell, Scapharca broughtonii. In addition, hydrogen peroxide ($H_2O_2$) concentrations, lysozyme activity, and glutamate oxaloacetate transaminase (GOT) and glutamate pyruvate transaminase (GPT) levels were measured in the hemolymph. We found that Cd and TBT treatment significantly increased antioxidant enzyme mRNA expression and activity in the digestive glands and gills in a time-dependent manner. In response to the Cd and TBT treatments, antioxidant enzymes mRNA expression and activity increased up to day 5 in the digestive glands and then decreased by day 7. In the gills, antioxidant enzymes mRNA expression and activity increased up to day 3 and then decreased by day 5. Likewise, $H_2O_2$ concentrations significantly increased up to day 5 and then decreased by day 7. Finally, lysozyme activity decreased during the experimental period, whereas GOT and GPT levels were significantly increased in a time-dependent manner. These results suggest that antioxidant enzymes play an important role in decreasing ROS levels and oxidative stress in ark shells exposed to Cd and TBT.

Assessment of Bacteriological Safety of the Seawater and Ark shell (Scapharca subcrenata) in Yeoja Bay, Korea (전남 여자만 해수 및 새꼬막(Scapharca subcrenata)의 세균학적 위생 평가)

  • This study evaluated the sanitary state of seawater and shellfish in Yeoja bay from January 2014 to June 2016. The sampling stations for sanitary survey in Yeoja Bay was composed of 50 stations for seawater and 4 stations for ark shell and the samples have been collected monthly at a stated date. The total and fecal coliforms in seawater and Escherichia coli (E. coli) in shellfish were investigated as sanitary indicator bacteria, respectively. The geometric mean and the estimated 90th percentile ranges of total and fecal coliforms for seawater were <1.8-3.9, <1.8-2.6 and <1.8-29.7, <1.8-9.3 MPN/100 mL, respectively. The range of E. coli levels for ark shell were <20~330 MPN/100 g. The sanitary state in Yeoja bay was evaluated clean area according Korea criteria and conditionally approved area according US criteria and Class B area according EU criteria.

Treatment of Acid dye Using Microbial Immobilization (미생물 고정화를 이용한 산성염료의 처리)

  • Strains degrading and decolorizing acid dyes, Nylosan red E-BL 150%. were isolated from natural system, was named as ARK3. The optimal culture conditions of temperature and pH were $35^\circ{C}$, 7.0, respectively. Growth rate of cells in conditions of aerobic shaking more than standing culture conspicuously increased, and optical density of those to strain ARK3 were found as 1.38 and 0.25 after 42 hrs. Decolorization efficiency in batch culture which used as immobilization media to natural zeolite was 15% after 6 hrs, while suspension culture was 5%, also its of immobilization and suspension culture were 90% and 85% after 48 hrs, respectively. Decolorization efficiency of air-lift bioreactor was more than 90% to a dilution rate of $0.038hr^{-1}$, but that was decreased as 70%, when the dilution rate was $0.05hr^{-1}$. Even though at maximum dilution rate of this study, there was not appeared "wash out" phenomienon of biomass. Decolorization efficiency was 97.7% at a dilution rate of $0.025hr^{-1}$, when influent dye concentration was $100mg/\ell$. But if influent dye concentration increased as $150mg/\ell$, even though MLVSS increased, that of treatment water decreased as 93%. Also, when influent dye concentration increased as $200mg/\ell$ and $300mg/\ell$, decolorization efficiencies of treatment water abruptly decreased as 85% and 63%, respectively. Decolorization efficiency was more than 92% to the limit volumetric loading rate of $3.75mg/\ell\cdot{hr}$hr, without regard to variation of influent dye concentration or hydraulic retention time. if volumetric loading rate was more than $3.80mg/\ell\cdot{hr}$, at same condition, decolorization efficiency was lower decrease of retention time than increase of influent dye concentration.entration.

Regional Difference in Fatty Acid Content of Korean Shellfish

  • Regional variation in the fatty acid content of shellfish was investigated on 5 species of Korean shellfish including murex shell, ark shell, jack-knife clam, orient hard clam, and little neck clam that were originated from 2 geographically different regions in Korea (Region 1: South coast, $34-35^{\circ}N$, $127-129^{\circ}E$; Region 2: West coast, $36-38^{\circ}N$, $126-127^{\circ}E$). Significant regional difference in total fatty acids content was observed in murex shell and little neck clam (p<0.01), but not in the other species of shellfish. The contents of saturated fatty acids, monounsaturated fatty acids, and polyunsaturated fatty acids including n-3 and n-6 fatty acids were appreciably higher in murex shell from Region 2 and in little neck clam from Region 1 than the shellfish originated from their counterpart areas (p<0.05). Nevertheless, relative percentages of the fatty acids remained constant within same species regardless of geographic regions or species. Considering the facts of that the fish/shellfish are unique sources of n-3 fatty acids and a little neck clam is the most-consumed shellfish in Korea, n-3 fatty acids intake might vary with the habitat of the shellfish that Koreans consume.

Antimicrobial Resistance in Escherichia coli Isolated from the Shellfish Farms in the Southern Coast of Korea (남해안 패류양식장에서 분리된 대장균(Escherichia coli)의 항균제 내성)

  • This study evaluated the impact of antimicrobial resistant bacteria produced by inland pollution sources on coastal areas, and investigates antimicrobial resistance profiles of bacteria isolated from marine filter feeders. A total of 107 shellfish (short-necked clams Ruditapes philippinarum, ark shells Scapharca broughtonii, and oysters Crassostrea gigas) were collected from the southern coast of Korea, from which 204 Escherichia coli strains were isolated. The antimicrobial susceptibility patterns of isolated strains were analyzed for 15 antimicrobial agents used in Korea for clinical or veterinary therapy. Antimicrobial resistance was present in 44.6% of E. coli isolates against at least one antimicrobial agent. The rate of antimicrobial resistance in the Narodo area was higher than isolates from the Gangjinman area and Kamak Bay. E. coli isolates had a higher rate of resistance against: tetracycline (29.9%), streptomycin (25.5%), and trimethoprim (14.2%). Of 204 isolates, 29 (14.2%) were resistant to multiple antimicrobial agents.

THE FOOD AND GROWTH OF THE LARVAE OF THE ARK SHELL ANADARA BROUGHTONI SCHRENCK (피조개의 먹이와 성장)

  • The larvae of the ark shell Anadare broughtoni(Schrenck) were grown at room temporature (approximately $20.4^{\circ}C$), and fed laboratory-cultured Cyclotella nana. The egg of the ark shell produced in the laboratory measured about $54.9\mu$ in diameter. The embryos gradually developed into larvae up to $110.8\mu$ shell length, $83.9\mu$ shell height and with shell breadth of $58.2\mu$ even in the absence of the algal food. Beyond this sire, however, the growth of the larvae was considerably retarded. The larvae showed better growth rate when they were fed the algal food two days after spawning, i. e., early straight-hinge stage. Daily rate of food consumption varies according to the larval sizes. But the rate increases considerably when the larvae begin to form umbos. In general the rate Is indicated by the following formula: $Y=0.0025161\;X^{2.76459}$. The growth experiments of the larvae indicate that the efficiency of food conversion was higher when fed centrifuged food. Regarding to the difference in the slopes of growth curve, centrifuged food showed better growth rate as compared to those grown with the non-centrifuged food. The smaller the larval size, the greater will be the difference in growth. The larvae began settling when they reathed 261.7 to $289.6\;{\mu}$ in shell length, 199.2 to $221.7\mu$ in shell height and 147.6 to $170.8\mu$ in shell breadth. The time which elapsed from spawning to the larval settlement was about 28 days. The mean growth of the larvae is indicated with regression line and exponential curve equations as follows. Inhibitory effect of broccoli leaf extract on PGE2 production by NF-κB inhibition (NF-κB 저해를 통한 브로콜리 잎 추출물의 PGE2 저해효과)

  • Objective : Broccoli is edible green plant that has a wide variety of health benefits including cancer prevention and cholesterol reduction. However, leaves of broccoli are not eaten and are mostly left as waste. This study was conducted to evaluate the effects of the broccoli leaf extract (BLE) on prostaglandin E2 (PGE2) production related to nuclear factor kappa B (NF-κB) signaling in lipopolysaccharide (LPS)-activated macrophages. Methods : BLE was prepared by extracting dried leaf with ethanol. Cell viability was determined by 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. PGE2 and inflammatory cytokines were detected by enzyme-linked immunosorbent assay (ELISA). Expression level of each protein was monitored by Western blot analysis. Results : In LPS-activated Raw264.7 cells, PGE2 release into culture medium was dramatically enhanced compared to control cells. However, increased PGE2 was attenuated dose-dependently by treatment with BLE. Inhibition of PGE2 production by BLE was due to the suppression of cyclooxygenase-2 (COX-2) expression determined by Western blot analysis. BLE also inhibited the production of inflammatory cytokines such as interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α). Inhibition at PGE2 and cytokine was mediated from inhibition of nuclear translocation of NF-κB due to the repression of inhibitory kappa B alpha (IκBα) phosphorylation and degradation. Conclusion : This study showed that BLE exerted inhibitory activities against PGE2, which is critical for the initiation and resolution of inflammatory responses, and that inhibition of PGE2 was mediated by suppression of NF-κB signaling. These results suggest that the waste broccoli leaves could be used for controlling inflammation.
