pKT230 벡터를 이용한 Pseudomonas sp. P20의 2,3-Dihydroxybiphenyl Dioxygenase 유전자의 클로닝

  • Pseudomonas sp. P20 isolated from the polluted environment is capable of degrading biphenyl and 4-chlorobiphenyl. The pcbABCD genes responsible for degradation of biphenyl and 4-chlorobiphenyl were cloned using pBluescript SK(+) from the chromosomal DNA of Pseudomonas sp. P20 to construct pCK1 and pCK102, harbouring pcbABCD and pcbCD, respectively. The 2, 3-DHBP dioxygenase gene, pcbC, was cloned again from pCK102 by using pKT230 which is known as a shuttle vector and pKK1 hybrid plasmid was constructed. The E. coli KK1 transformant obtained by transforming the pKK1 into E. coli XL1-Blue showed 2, 3-DHBP dioxygenase activity. The specific 2, 3-DHBP dioxygenase activity of E. coli KK1 was similar to that of the E. coli CK102, but much higher than those of the natural isolates, Pseudomonas sp. DJ-12 and Pseudomonas sp. P20.

  • 著者는 한국산 八放珊瑚類의 계통분류학적 연구를 하기 위하여 1965년부터 1978susRK지 동해의 울릉도 (苧洞, 道洞, 沙洞)와 남해의 尾浦, 海金剛, 鴻島, 황천도, 濟州, 爲美里, 林島, 地歸島, 西歸浦, 鳥島, 門島, 虛島등 15개 지역으로부터 채집된 標本들을 同定分類한 결과 다음과 같은 全軸類에 속하는 3科 7屬 15種을 얻었다. 이들은 모두 韓國 未記錄種이었으며 다음과 같다. Anthoplexaura dimorpha, Euplexaura anastomosans, E. abietina, E. crassa, E. recta, Ellisella maculata, E. rubra, E. limbaughi, Verrucella umbraculum, V. stellata, Primnoa reseda pacifica, Callogorgia pseudoflabellum, Plumarella spinosa, P. rigida 및 P. adharens.

창원시 노인의 영양지수에 영향을 미치는 요인 (Factors Affecting Nutrition Quotient for Elderly (NQ-E) for the Elderly in Changwon City)

  • The purpose of this study was to identify nutrition quotient for elderly (NQ-E) and to investigate factors affecting NQ-E of the elderly using welfare center and senior citizen's center in Changwon city. A self-administered questionnaire was conducted between June 2021 and early August 2021 for 320 elderly (≥65 years, male, n=52, female, n=268). As a result of the survey, the mean NQ-E score was 61.12, which was within the medium-high grade. The scores of balance, moderation, and dietary behavior factors were within the medium-high grade, while diversity factor was within the medium-low grade. According to the results of the Pearson's correlation coefficient analysis, NQ-E showed positive correlations with the 'with family' (p<0.01), 'education level' (p<0.01), 'health functional foods consumption' (p<0.01), 'monthly household income' (p<0.05), 'nutrition education experience' (p<0.05), 'reading nutrition labeling of health functional foods' (p<0.01), 'perception of the efficacy of health functional foods' (p<0.01), and 'the number of times of leisure activities per week' (p<0.01) and negative correlations with the 'gender' (p<0.05), 'age' (p<0.01), 'smoking' (p<0.05), and 'social frailty' (p<0.01). As a result of multiple regression analysis, 'gender' (p<0.05), 'perception of the efficacy of health functional foods' (p<0.05), 'the number of times of leisure activities per week' (p<0.05), and 'social frailty' (p<0.05) were found to be factors affecting NQ-E. Based on the results of this study, customized services by characteristic, nutrition education, and counseling for the elderly should be implemented and the development of various programs and continuous support of the community are necessary so that the elderly can carry out social exchange.

  • ApoE 유전자의 다형성은 인체 혈중 콜레스테롤 농도의 정상분포를 설명하는 중요한 인자이다. 특히, E2 와 E4 allele들은 고지혈증환자의 혈중에 현저히 나타나며, 각 다른 인종간의 이틀의 상대빈도수가 관상 동맥 질환 유발 위험도와의 상관관계가 아주 높다. Apo E 유전자의 다형성이 혈중 지질 대사변화 에 미치는 영향에 관한 실험은 105명의 건강한 한국여성을 대상으로 시행되었다. 난소를 제거한 생쥐 자궁조직에서의 ADAM-8, 9, 10, 12, 15, 17 그리고 ADAMTS-1의 유전자 발현 (Gene Expression of ADAM-8, 9, 10, 12, 15, 17 and ADAMTS-1 in Ovariectomized Mice Uteri)

  • 난소가 제거된 생쥐를 이용하여 자궁조직에서의 ADAM-8, 9, 10, 12, 15, 17, 그리고 ADAMTS-1의 유전자의 발현이 생식호르몬에 의하여 조절되는 지를 알아보았다. 암컷 생쥐의 난소를 제거하고, 2주후에 sesame oil, 17 ${\beta}$-estradiol ($E_2$), progesterone ($P_4$ 혹은 이 둘 혼합액 ($E_2+P_4$)을 피하 주사하였다. RT-PCR 방법을 이용하여 유전자 전사체의 발현을 조사한 결과 ADAM-8, 12, 그리고 17은 oil을 주사하거나 $P_4$만을 주사한 군보다 $E_2$를 주사한 군에서 자궁조직에서의 mRNA의 양이 현저하게 증가하였다. 반면 ADAM-9, 10, 15, 그리고 ADAMTS-1은 oil을 주사하거나 $E_2$만을 주사한 군보다 $P_4$를 주사한 군에서 mRNA의 양이 현저하게 증가하였다. 또한 단백질의 발현양상의 결과도 RT-PCR의 결과와 동일하게 관찰되었다. 이러한 결과로 미루어 ADAM-8, 12, 그리고 17은 17 ${\beta}$-estradiol에 의하여, ADAM-9, 10, 15, 그리고 ADAMTS-1은 progesterone에 의하여 유전자의 발현이 upregulation 되는 것으로 생각되어진다.

  • 폴리에틸렌필름(P.E)멀칭이 제초제(除草劑) alachlor, pendimethalin, diphenamid의 토양(土壤) 및 수확물중(收穫物中)의 잔류성(殘留性)에 미치는 영향(影響)에 대(對)하여 고추, 땅콩, 참깨포장(圃場)에서 로지조건(露地條件)을 대조(對照)로 하여 조사(調査)하였다. 토양중(土壤中)에 있어서 3종제초제(種除草劑)의 로지조건(露地條件)에서의 반감기(半減期)는 alachlor의 경우 땅콩과 참깨포장(圃場)에서 $3{\sim}4$ 일(日), pendimethalin은 고추포장(圃場)에서 30일(日), diphenamid는 고추, 땅콩, 참깨포장(圃場)에서 $24{\sim}46$ 일정도(日程度)였다. 분해(分解)가 빨랐던 alachlor의 반감기(半減期)는 로지조건(露地條件)과 P.E.피복조건(被覆條件)에서 거의 차(差)가 없었다. 분해(分解)가 느렸던 pendimethalin과 diphenamid는 로지조건(露地條件)에서보다 P.E.피복조건(被覆條件)에서 각각(各各) 30일(日)과 $20{\sim}90$ 일간(日間) 더 길었다. P.E.피복(被覆)의 유무(有無)에 관계(關係)없이 pendimethalin은 최종적(最終的)으로 수확(收穫)된 고추에서 검출(檢出)되지 않았으며 alachlor도 땅콩과 참깨에서 검출(檢出)되지 않았다. diphenamid의 땅콩중(中) 잔류량(殘留量)은 0.147ppm(투명(透明) P.E. 피복조건(被覆條件))${\sim}0.071ppm$(로지조건(露地條件))이었고, 참깨중(中) 잔류량(殘留量)은 0.022ppm(투명(透明) P.E. 피복조건(被覆條件))${\sim}0.129ppm$(로지조건(露地條件))이었는 바, 이는 양조건(兩條件)에서 잔류허용량(殘留許容量)(0.2ppm) 이하(以下)였다.

  • Recently, much attention has been paid to problems which is concerned with voltage instability phenomena and much works on these phenomena have been made. In this paper, by substituting d $P_{k}$ d $e_{k}$ ( $v^{\rarw}$= e +j f) for $P_{k}$ in conventional load flow, direct method for finging the limit of voltage stability is proposed. Here, by using the fact that taylor se- ries expansion in .DELTA. $P_{k}$ and .DELTA. $Q_{k}$ is terminated at the second-order terms, constraint equation (d $P_{k}$ d $e_{k}$ =0) and power flow equations are formulated with new variables .DSLTA. e and .DELTA.f, so partial differentiations for constraint equation are precisely calculated. The fact that iteratively calculated equations are reformulated with new variables .DELTA.e and .DELTA.f means that limit of voltage stability can be traced precisely through recalculation of jacobian matrix at e+.DELTA.e and f+.DELTA.f state. Then, during iterative process divergence may be avoid. Also, as elements of Hessian mat rix are constant, its computations are required only once during iterative process. 일부 전문대 응급구조학생의 응급구조전문직관의 변화에 대한 연구 (A study on the change of the views on E.M.T. profession of the E.M.T. college students)

  • The purpose of this study was to identify the change of perceptions related to E.M.T. profession of E.M.T. student and to provide the basic data for development of curriculum. The subjects of the survey were 216 students from 3 junior college by questionaire from November 25 to November 29, 1996 in Kwang-Ju, Cheong-Ju, and Kong-Ju. The results were analyzed using the SAS and can be summerized as follows : 1. The view on profession was a statistically significant difference inspecial knowledge & skill training (t=-1.686, P<.001), high income (t=-0.753, P<.05) 2. The view on competence as a E.M.T. was a statistically significant differncein sufficient professional knowledge on the E.M.T. (t=-1.144, P<.001), own's health (t=-0.808, P<.001), sincerity and responsibility (t=-1.429, P<.01), satisfaction and effort (t=-1.335, P<.01), boundary of affairs (t=-1.356, P<.01) 3. The view on E.M.T. profession was a statistically significant difference inprotect of pt. life & to relieve the pain (t=-2.388, P<.001) 4. The view on necessary knowledge of E.M.T. was a statistically significant difference in patient assessment (t=-2.168, P<.001), ACLS (t=1.678, P<.001), hemorrhage & shock (t=-3.252, P<.001), trauma (t=-4.284, P<.001), cardiopulmonary disturbance (t=-2.897, P<.001), acute abdomen (t=-4.284, P<.001), neonatal disease (t=-4.032, P<.001), OBGY disease(t=-4.151, P<.001), emergency delivery (t=-2.825, P<.001), infectious disease (t=-2.930, P<.001), environmental emergency (t=-3.123, P<.05), recording(t=2.22, P<.05) 5. The view on ideology of E.M.T. was a statistically significant difference in belief & faith (t=-2.344, P<.001), knowing of own (t=-2.142, P<.01), humanity (t=-2.581, P<.05), knowing of patient (t=-2.079, P<.05).

The Study of Bfa1pE438K Suggests that Bfa1 Control the MitoticExit Network in Different Mechanisms Depending on DifferentCheckpoint-activating Signals

  • During mitosis, genomic integrity is maintained by the proper coordination of anaphase entry and mitotic exit via mitotic checkpoints. In budding yeast, mitotic exit is controlled by a regulatory cascade called the mitotic exit network (MEN). The MEN is regulated by a small GTPase, Tem1p, which in turn is controlled by a two-component GAP, Bfa1p-Bub2p. Recent results suggested that phosphorylation of Bfa1p by the polorelated kinase Cdc5p is also required for triggering mitotic exit, since it decreases the GAP activity of Bfa1p-Bub2p. However, the dispensability of GEF Lte1p for mitotic exit has raised questions about regulation of the MEN by the GTPase activity of Tem1p. We isolated a Bfa1p mutant, $Bfa1p^{E438K}$, whose overexpression only partially induced anaphase arrest. The molecular and biochemical functions of $Bfa1p^{E438K}$ are similar to those of wild type Bfa1p, except for decreased GAP activity. Interestingly, in $BFA1^{E438K}$ cells, the MEN could be regulated with nearly wild type kinetics at physiological temperature, as well as in response to various checkpoint-activating signals, but the cells were more sensitive to spindle damage than wild type. These results suggest that the GAP activity of Bfa1p-Bub2p is responsible for the mitotic arrest caused by spindle damage and Bfa1p overproduction. In addition, the viability of cdc5-2 ${\Delta}bfa1 $ cells was not reduced by $BFA1^{E438K}$, suggesting that Cdc5p also regulates Bfa1p to activate mitotic exit by other mechanism(s), besides phosphorylation.

P-e 곡선의 타원 특성을 이용한 전력계통 최대허용부하의 예측 (Estimation of Maximum Loadability in Power Systems By Using Elliptic Properties of P-e Curve)

  • This paper presents an efficient algorithm to estimate the maximum load level for heavily loaded power systems with the load-generation vector obtained by ELD (Economic Load Dispach) and/or short term load forecasting while utilizing the elliptic pattern of the P-e curve. It is well known the power flow equation in the rectangular corrdinate is jully quadratic. However, the coupling between e and f makes it difficult to take advantage of this quadratic characteristic. In this paper, the elliptic characteristics of P-e curve are illustrated and a simple technique is proposed to reflect the e-f coupling effects on the estimation of maximum loadability with theoretical analysis. An efficient estimation algorithm has been developed with the use of the elliptic properties of the P-e curve. The proposed algorithm is tested on IEEE 14 bus system, New England 39 bus system and IEEE 118 bus system, which shows that the maximum load level can be efficiently estimated with remarkable improvement in accuracy.
