Complete genome sequence of Paenibacillus swuensis DY6T, a bacterium isolated from gamma-ray irradiated soil (감마선 조사된 토양에서 분리된 박테리아 Paenibacillus swuensis DY6T의 완전한 게놈 서열)

  • Several bacterial species have been reported to be surviving after the ionizing radiation treatment due to the presence of sophisticated enzymes systems and some endospores producing bacterial strains can also resist, due to the presence of thick spore coat. In this study, we report the complete genome sequence of a bacterium Paenibacillus swuensis $DY6^T$, isolated from an irradiated soil sample. The genome comprised of 5,012,599 bp with the G+C content of 49.93%, the genome included 4,463 protein coding genes and 133 RNA genes.

  • Cells of Deinococcus puniceus $DY1^T$ are Gram-positive, coccus-shaped, and crimson color-pigmented. Strain $DY1^T$ was isolated from soil irradiated with 5 kGy gamma radiation and showed resistance to UVC and gamma radiation. In this study, we report the complete genome sequence of a bacterium Deinococcus puniceus $DY1^T$ is consist of circular chromosome comprised of 2,971,983 bp, with the G + C content of 62.5%. The complete genome sequence was obtained using the PacBio RS II platform, it included 2,617 coding sequences (CDs), 2,762 genes, and 88 pseudogene.

Separation and Adsorption-Desorption Characteristics of Heavy Rare Earth Elements (Gd, Tb, Dy) using P507 Resin (P507 추출수지를 이용한 중희토류 원소(Gd, Tb, Dy)의 흡탈착 분리특성에 관한 연구)

  • This study was conducted to establish the adsorption-desorption mechanism and the optimum condition of chromatographic operation for separations of heavy rare earth elements (Gd, Tb, Dy) using a p507-containing resin. By employing Langmuir and Freundlich isotherm together with pseudo first and second order kinetics, absorption-desorption reaction mechanism was investigated. Langmuir and Freundlich isotherm was applied under assumption that adsorption reaction occurs in form of monolayer, and because the result was identical to the assumption, now we know adsorption of heavy rare earth elements occurs in form of monolayer. Concerning the pseudo first and second order kinetic, the pseudo second order seemed to be more suitable to represent heavy rare earth element adsorption mechanism. By using the extraction chromatography to separate heavy rare earth elements, ${\alpha}^{Tb}_{Gd}=1.24$, and ${\alpha}^{Dy}_{Tb}=1.03$ were confirmed in eluent HCl 0.25 M which indicates almost perfect separations of three elements. Furthermore, as concentrations of eluent became higher, the resolution value decreased and the elution area got shortened.

Behavior of Rare Earth Elements in Synthetic Fluorapatites Revealed by Rietveld Structure Refinement Data (리트벨트법에 의한 합성인회석 내의 희토류 원소의 거동에 관한 연구)

  • 이성분 희토류 원소로 치환된 4종류의 합성불화인회석(synthetic fluorapatite) (Ap49: La+Gd, Ap50:Ce+Dy, Ap51: Pr+Er, Ap54: Eu+Lu; $Ca{10-x-2y}$ $Na_{y}$ $REE_{x+y}$($P_{1-x}$ //$Si_{x}$ $_{4}$)$_{6}$ $F_{2}$, x=0.13~0.12, y=0.26~0.42)을 대상으로 X-선 회절분석을 통해 얻어진 자료를 이용하여 리트벨트 구조분석을 실시하고 치환된 희토류원소의 거동을 단결정법으로 구해진 결과와 비교.분석하였다. 리트벨트 구조분석결과 합성불화인회석은 공간군 $P6_3$/m, 단위포는 평균하여 a=9.3906(1) $\AA$, c=6.8924(1) $\AA$, V=527.36 $\AA^3$의 값을 갖는다. 구조의 정밀도를 나타내는 R 지수를 보면 $R_{B}$ / 값은 17.29~18.80이고 S(GofF)값은 1.44~1.68로 계산되었다. 불화인회석은 9개의 산소를 배위하는 Ca1자리와 6개의 산소와 하나의 불소가 배위하는 Ca2자리가 있으며 Ca1-O의 평균거리는 2.563 $\AA$이고 Ca2-O의 평균거리는 $2.460 \AA$으로 Ca1자리가 Ca2자리보다 다소 크다. 구조자리 치환식에서는 $Ca^{2+}$ / 자리를 치환하는 $REE^{3+ }$ 로 인하여 전하균형을 맞춰주기 위해서 인과규소가 함께 참여하였다. ($REE^{3}$+Si$^{ 4+}$ $2Ca^{2+}$ : Ca1) 계산된 희토류원소의 자리점유율(REE-Ca2/REE-Cal)은 원자번호가 증가함에 따라 일정하게 감소하는 경향을 보여주며 이는 희토류원소의 거동이 LREE는 크기가 상대적으로 작은 Ca2 자리에 우선 치환되고 HREE는 크기가 큰 Ca1자리에 우선 치환되는 경향을 지시한다.

The Origin of Radioactive Elements Found in Groundwater Within the Chiaksan Gneiss Complex: Focusing on the Relationship with Minerals of the Surrounding Geology (치악산 편마암 복합체에 분포하는 지하수 내 함유된 방사성 원소의 기원: 주변 지질을 구성하는 광물과의 연관성을 중심으로)

  • Petrological and mineralogical analyses were conducted to identify minerals containing radioactive elements (uranium) in the Chiaksan gneiss complex and to confirm their association with the surrounding groundwater. Fourteen minerals were identified through the microscopic and electron microscopy (SEMEDS) investigation. The principal minerals included plagioclase, biotite, quartz, alkali feldspar, chlorite, and calcite. Minor minerals were sphene, allanite, apatite, zircon, thorite, titanite, pyrite, and galena. A small amount of thorite was observed in the size of ~1 mm within macrocrystalline allanite. Allanite, which includes a large amount of rare earth elements, appeared in three distinctive patterns. The results of the EPMA analyses indicated that macrocrystalline allanite had higher elemental contents of TiO2~1.70 wt.%, Ce2O3~11.86 wt.%, FeO ~13.31 wt.%, MgO ~0.90 wt.% and ThO2 ~1.06 wt.% with the lowest average content of Al2O3 17.35 ± 2.15 wt.% (n = 7), CaO 12.13 ± 1.81 wt.% (n = 7). An allanite existing at the edge of the sphenes encompassing titanites had a higher element content of Al2O3 ~24.00 wt.%, Nd2O3 ~5.10 wt.%, Sm2O3~0.66 wt.%, Dy2O3~0.86 wt.% and Y2O3~1.38 wt.% with the lowest average content of TiO2 0.35 ± 0.21 wt.% (n = 11), Ce2O3 5.25 ± 1.03 wt.% (n = 11), FeO 9.84 ± 0.26 wt.% (n = 11), MgO 0.12 ± 0.05 wt.% (n = 11), and La2O3 1.49 ± 0.29 wt.% (n = 11). Allanites in a matrix of parental rocks exhibited intermediate values between the two elemental compositions mentioned above. None of the uranium-rich minerals were observed in the migmatitic gneiss within the study area. Consequently, the origin of uranium in the groundwater was not associated with the geology of the surrounding environment, but our investigation proved the existence of abundant allanites containing significant amounts of radioactive thorium and rare earth elements.

Effects of Radiofrequency Induced Local Hyperthermia on Normal Canine Liver (라디오 파를 이용한 국소 온열치료 : 정상 개의 간에 미치는 영향)

  • In order to assess the effects of radiofrequency-induced local hyperthermia on the normal liver, histopathologic findings and biochemical changes after localized hyperthermia in canine liver were studied. Hyperthermia was externally adminsitered using the Thermotron RF-8 (Yamamoto Vinyter Co., Japan; Capacitive type heating machine) with parallel opposed electrodes. Thirteen dogs were used and allocated into one control group (N=3) and two treatment groups according to the treatment temperature. Group I (N=5) was heated with $42.5\pm0.5^{\circ}C$ 30 minutes, and Group II (N=5) was heated with $45\pm0.5^{\circ}C$ for 15-30 minutes. Samples of liver tissue were obtained through a needle biopsy immediately after hyperthermia and T,14, and 28 days after treatment. Blood samples were obtained before treatment and W, 3,5, 7,14 and 28 days after treatment and examined for SGOT, SGPT and alkaline phosphatase. Although SGOT and SGPT were elevated after hyperthermia in both groups (three of five in each group), there was no liver cell necrosis or hyperthermia related mortality in Group 1. A hydropic swelling of hepatocytes was prominent histologic finding. Hyperthermia with $45^{\circ}C$ for 30 minutes was fatal and showed extensive liver cell necrosis. In conclusion, liverdamage dy heat of $42.5\pm0.5^{\circ}C$ for 30 minutes is reversible, and liver damage by heat of $45\pm0.5^{\circ}C$ for 30 minutes can be fatal or irreversible. However, these results cannot be applied directly to human trial. Therefore, in erder to apply hyperthermic treatment on human liver tumor safely, close obsewation of temperature with proper thermometry is mandatory. Hyperthermic treatment should be confined to the tumor area while sparing a normal liver as much as possible.

The Change in Patterns and Conditions of Algal Blooms Resulting from Construction of Weirs in the Youngsan River: Long-term Data Analysis (보 건설에 따른 영산강의 조류 발생 및 환경 변화: 수질측정망 장기 자료 분석)

  • The effect of weir construction (2009~2011) was investigated on algal bloom dynamics and surrounding conditions in the Youngsan River by analyzing the long-term (2001~2014) data provided by the Water Information System, Ministry of Environment. The data include chlorophyll a and water properties such as total suspended solids (TSS), ammonium ($NH_4{^+}$), nitrate ($NO_3{^-}$), orthophosphate ($PO{_4}^{3-}$), total nitrogen (TN), total phosphorus (TP) and DIN/DIP molar ratio collected from 12 stations along the channel of the river. Temporal variations were examined using data collected monthly from 2001~2014 and Box-Whisker plot was used to examine the difference in algal bloom dynamics between before (2006~2008) and after (2012~2014) the weir construction. Pearson's correlation analysis was also used to analyze the correlation of parameters. The results showed that TSS affecting water turbidity increased during the construction but decreased especially at the stations located in the upper and middle regions of the river after the construction. Ammonium concentrations increased whereas the concentrations of other nutrients decreased after the construction inducing an increase in N:P molar ratio. Chlorophyll a decreased suddenly during the construction but increased clearly after the construction at the stations where TSS decreased. This indicates that algal blooms can develop in the Youngsan River due to a decrease in turbidity that increases light penetration in water column although the concentrations of nutrients such as orthophosphate were reduced after the weir construction.
