Quantification of Karanjin, Tannin and Trypsin Inhibitors in Raw and Detoxified Expeller and Solvent Extracted Karanj (Pongamia glabra) Cake

  • Despite being a rich source of protein (28-34%), karanj (Pongamia glabra) cake is found to be bitter in taste and toxic in nature owing to the presence of flavonoid (karanjin), tannin and trypsin inhibitor, thereby restricting its safe inclusion in poultry rations. Feeding of karanj cake at higher levels (>10%) adversely affected the growth performance of poultry due to the presence of these toxic factors. Therefore, efforts were made to detoxify karanj cake by various physico-chemical methods such as dry heat, water washing, pressure cooking, alkali and acid treatments and microbiological treatment with Sacchraromyces cerevisiae (strain S-49). The level of residual karanjin in raw and variously processed cake was quantified by high performance liquid chromatography and tannin and trypsin inhibitor was quantified by titrametric and colorimetric methods, respectively. The karanjin, tannin and trypsin inhibitor levels in such solvent and expeller pressed karanj cake were 0.132, 3.766 and 6.550 and 0.324, 3.172 and 8.513%, respectively. Pressure-cooking of solvent extracted karanj cake (SKC) substantially reduced the karanjin content at a cake:water ratio of 1:0.5 with 30-minute cooking. Among chemical methods, 1.5% (w/w) NaOH was very effective in reducing the karanjin content. $Ca(OH)_2$ treatment was also equally effective in karanjin reduction, but at a higher concentration of 3.0% (w/w). A similar trend was noticed with respect to treatment of expeller pressed karanj cake (EKC). Pressure cooking of EKC was effective in reducing the karanjin level of the cake. Among chemical methods alkali treatment [2% (w/w) NaOH] substantially reduced the karanjin levels of the cake. Other methods such as water washing, dry heat, HCl, glacial acetic acid, urea-ammoniation, combined acid and alkali, and microbiological treatments marginally reduced the karanjin concentration of SKC and EKC. Treatment of both SKC and EKC with 1.5% and 2.0% NaOH (w/w) was the most effective method in reducing the tannin content. Among the various methods of detoxification, dry heat, pressure cooking and microbiological treatment with Saccharomyces cerevisiae were substantially effective in reducing the trypsin inhibitor activity in both SKC and EKC. Based on reduction in karanjin, in addition to tannin and trypsin inhibitor activity, detoxification of SKC with either 1.5% NaOH or 3% $Ca(OH)_2$, w/w) and with 2% NaOH were more effective. Despite the effectiveness of pressure cooking in reducing the karanjin content, it could not be recommended for detoxification because of the practical difficulties in adopting the technology as well as for economic considerations.

Efficacy of Amaranth(Amaranthus spp. L.) Plant as a Natural Dye Resource: Focused on Wool Dyeing (아마란스 식물의 천연염재로서의 유효성 연구: 모직물 염색을 중심으로)

  • In this study, the efficacy of Amaranth(Amaranthus spp. L.) as a natural dye resource was investigated for wool fabrics. It is known that a large amount of flavonoid and anthocyanin colorant are contained in leaves and stems, as well as red flowers. The optimum condition of dyeing was 1.3% of dye concentration(o.w.b.) at 100℃ for 60 minutes, resulting the K/S value, 23.43 and R Munsell color on the wool fabrics. Al, Fe, Zinc and Titanium were used as a mordant. The mordant improved the dye uptake, regardless of the mordant type and mordant method. The pre-mordanting method was more effective than the post-mordanting method. Al pre-mordanted fabric showed the highest K/S, 30.02. Light fastness and washing fastness were high in grades 4-5 and 5, and rubbing fastness was good in grades 4 and 4-5 in dry condition, but low in grades 2-3 and 3 in wet condition. The dry cleaning fastness was excellent in all 5 grades. However, the alkaline perspiration fastness ratings were low in grades 2-3 and 3. The results show Amaranthus spp. L. colorant can be used as a functional natural dye for wool fabrics.

Natural Dyeing of Cotton Fabrics with Rumex crispus L. Root (소루쟁이뿌리를 이용한 면직물 천연염색)

  • The natural dyeing of cotton fabrics with Rumex crispus L. root extract was investigated. The dyeability of Rumex crispus L. root extract was evaluated with conditions of concentration, temperature, time, repeat-numbers, pH, mordants variables, methods of mordanting, color fastness and antibacterial activity, etc. The maximum V-visible spectrum possessed absorption band of Rumex crispus L. extract appeared at 274nm and 336nm. The amount of dyes extracted was increased with extracting concentration, temperature and time. The K/S value increased with increasing dyeing concentration and repeat-numbers. The K/S value increased with increasing dyeing temperature and time, the exhaustion was saturated in $90^{\circ}C\;and\;80min$, respectively. Surface colors of fabrics dyed with pH 3, 7, 11 extract were RP-R-YR-Y range. The light fastness and washing fastness showed good results in Fe-mordanted. The dry leaning fastness appeared more than 4 grade. Rubbing fastness was better in dry methods han that in wet methods. In the result of antibacterial activity, the decrease rate was 9.9% to Staphylococcus aureus with the dyed fabric of cotton.

Sanitation Assessment and Microbiological Analysis of the Domestic Kitchen Utensils (가정 내 주방용품 및 기구의 위생실태 및 미생물학적 분석)

  • This study was conducted to evaluate the sanitation level and use of dish cloths, chopping board, and drawer in the refrigerator in 50 university students' houses located in Seoul and Kyonggido. The survey questionnaire evaluated general information, frequency of the washing, cleaning method, drying method and the associated factors. The highest percentage of the cleaning and drying methods of dish cloths were “boiling(76.0%)”, and “dry in the kitchen or on the sink(78.0%)”, respectively. For the chopping board and drawer in the refrigerator, the highest percentage of cleaning method was “use detergent” and drying method was “dry in the kitchen”. The mean of coliforms for dish cloths, chopping board, and drawer in the refrigerator was 4.46${\pm}$1.95 log CFU/100 $cm^2$, 3.92${\pm}$1.64 log CFU/100 $cm^2$, 3.48${\pm}$2.01 log CFU/ 100 $cm^2$, respectively. Salmonella spp. were not detected from all samples. But E. coli were detected from dish cloths(4.0%) and S. aureus were detected from dish cloths(16.0%), chopping board(12.0%), and drawer in the refrigerator(8.0%). The results of microbiological test of kitchen utensils indicated that the sanitary conditions of dish cloths, chopping board, drawer in the refrigerator should be improve promptly.

The Effects of the Pre-treatments with Proteins on Dyeing of Silk Fabric with Caesalpinia sappan (견직물의 소목염색에서 단백질 전처리 효과)

  • This study investigated the effects of the working mechanisms of proteins, mordants, and dyes, as well as the mordanting order, on dye uptake by silk fabric pre-treated with proteins and dyed with freeze-dried sappan wood water extract. Soybean protein and sodium caseinate were used as the proteins. 1. When Al mordants were not used, the dyeability of the fabrics increased upon protein pre-treatment as compared to the case without treatment. 2. Dyeing with protein pre-treatment, followed by mordanting, led to the highest dye uptake, and the optimal protein concentration was 5%. 3. The K/S values slightly decreased with an increase in the dyeing temperature, and the fabric turned dark red in color when dyeing was carried out at increasing temperature. Fabrics showed the highest dye uptake at $40^{\circ}C$. 4. Regarding the effect of time, the K/S values of the fabrics with and without protein treatment showed almost no increase after the initial dyeing time of 10min; further, there was hardly any difference in the cases with and without protein pre-treatment. 5. In case of protein pre-treatment fabrics, the washing fastness was level 2. The dry cleaning fastness showed very excellent result with level 4-5. The rubbing fastness was better in dry rubbing than in wet rubbing of the fabrics. For the light fastness, all dyed fabrics showed low fastness.

  • Silk textile finishing has been studied for many years by many workers in order to meet more utilities for various endusers. Such studies, however, could not be successful because any natural fibers are hardly change their natures by artificial treating methods. Textile finishing is of course to improve the mechandise qualities and the poor natures of silk so that it may be available as the best textile fiber in the world. Sometimes, famous trade marked textile plays more power than its quality in the silk market, nevertheless, this should be over line of research activities. Meantime, the silk demand has been also transferred from ladies stocking to other clothes since nylon or other synthetic fibers were developed. That is why, the extension of silk demand should be developed by various research works. Specially, silk is known as difficult textile to handle it during washing or ironing process which happened to depress down the silk usage for house wives. In order to solve such problems, the reporter has been worked for many years and now, he believes that he has developed a proper finishing method to coversuch problems. The developed finishing method may be said to eligible with economical aspect and shorten the dry duration after water washing in half against normal silk textile without harming the specific silk nature. As all of us know, silk fiber starts to denature since it was spinned by silkworm and the fiber is formed as overlapped "S" type curves during its concooning process. After it is made as raw silk or sericin silk, it shows as straight line form, but it changes in to waved form in case refining or degumming process in order return to its original spinned form. Such nature is continued during its textile form and ends with hard ironing nature than other textile fibers. Mean while, the silk fiber keeps to continue its denaturing and this is iniciated by repeat of washing and drying which takes many years to reach its final stage, The reporter has found the iniciating denature of silk by his finishing process, with out heat, decreasing the swollen nature which ended with shortening the drying duration after wash. Each washing was carried out by soaking the previously weighed sample in cold water for one hour, then pressed the sample for ten minutes to eliminate its free water component before weighing with same condition. According to this, the treated silk showed much denaturing after the finishing, but the standard silk progressed the denaturing by and by with the repeat of washing and drying, finally reached the same swollen degree of treated silk, Such treating result explains that the treated silk happened to be stebilized nature by the treating immediately. On the other that the treated silk happened to be stebilized nature by the treating immediately. On the other hand, standard silk may reach to such condition by the time of worn out clothes after repeat of washing and drying for many years while the clothes will be no more useful. The decreased swelling nature has brought about the drying period in half against standard silk after all. Not only the tests of tenacity and elongation but also crease resistance recovery, stiffness and shrinkage tests were carried out after each washing and drying which he has found better result on the treated silk textile against the standard silk. The most important thing was to keep the textile feeling of silk by such finishing work before improve any poor nature of silk. The general silk has a nature to absorb smoke or dirt from its surrounding air and reaches to dirty color shade upon such exposure, but the treated one has improved such nature because of its artificial denaturing, another word, it keeps clean longer than the normal silk. Many previous finishing works could improve some specific nature of silk, but it happened to deprave other important natures. The reporters work is, however, specialized to improve the silk to be useful as Wash and Wear Silk without harming its standard natures. So far, this work happened to be a overall innovative finishing method of silk textile.

Cadmium Uptake by Mon-viable Biomass from a Marine Brown Alga Ecklonia radiata Turn.

  • Biomass of non-viable and dried brown marine algae Ecklonia radiata Turn. was used to examine Its cadmium uptake capability. Twelve different pretreatments on the algal biomass were prepared. Among these pretreatments, the algal biomass, which treated with 0.1 M NaOH and kept in water. bath (100$\^{C}$, 18 h) followed by washing with distilled water and squeezing, showed the highest amount of cadmium uptake as 1634 $\pm$ 195 mg/g dry biomass at pH 4.0 and 50$\^{C}$. Adsorption temperatures and pH levels played some Important role In cadmium uptake. However, cadmium uptake decreased dramatically at a lower pH than 4.0. Freundlich adsorption isotherm showed potent cadmium uptake capacity of the non-viable biomass. Pretreatments on the non-viable algal biomass shown in this study nay enhance the Eadmium removal in the industrial wastewater.

The Research of New Azo Red Pigments for Textile Printing (섬유날염용 신규 아조 적색안료 연구)

  • The azo red pigments for textile printing containing amino-N-substituted benzamide derivatives as diazo components and 2-hydroxy-3-naphthoyl derivatives as coupling components were prepared. They have been printed on cotton, and the fastness, such as light, washing, dry cleaning, rubbing and heat stability was estimated. The new azo red pigments were valuable colorants in case of textile printing.

Hygienic Aspects of Campylobacter Enteritis (Campylobacter장염에 의한 식품위생학적인 고찰)

  • Campylobacter jejuni has been recognized as one of the causes of human gastroenteritis. The feces of a variety of reservoir animals contain c. jejuni as commensals in the intestinal tracts, and are fundamental source of contamination. The intestinal organisms contaminate carcasses, equipment tools hands of the processing line workers and air of the processing facility. Once the contamination happens in the slaughterhouses or the meat processing facilities, it is very difficult to keep the carcasses free from the infection of c. jejuni. Various disinfectants are effective in minimizing the number of Campylobacter infections in the processing facilities by washing contaminated carcasses, tools, and hands. Direct contact with infected animals has been incriminated in transmission of infection caused by C. jejuni. Freezing, cooling with dry air and gamma irradiation are an effective way for preserving the meat and eliminating the transmission, but broad and enforced studies are needed for the practical use.

  • The purpose of this study was to suggest the spatial structure among residential behaviors in relation to work space. To find the structure, Multi-Dimensional Scaling analysis was used. Respondents were 349 middle income housewives living in apartments in the area of Caopital region. Data were collected through the questinnaire survey. For the future residential interior design, following points were sugtested. A laundary space neess to be developed independently , being separated from both bathroom and kitchen/dining space. A new area which will give housewives some territoriality and also function as home office needs to be considered. A space configuration which satisfies the organic relationships among laundary behaviors such as washing dying, ironing and folding , needs to be systematically planned, with two adjoining separate zones of wet and dry utility . The present bathroom nees to be more incorporated to the private bedroom rather than to workspace.
