진전(震顫)을 주소로 내원한 환자 31례에 대한 임상고찰 -체질의학(體質醫學)적 치료를 중심으로 (Clinical Study for Tremor in 31 Outpatient Cases)

  • The clinical study was carried out the 31 patients with tremor who were treated in Daejeon University Oriental Hospital from 2 March 2002 to 29 June 2002. The results were summarized as follows. 1. In classification of Four Human coporeal constitution the number of patients So-Yang-In(少陽人) was the most, in the ratio of sex, female was higher than man and in the age distribution the aged over 60 were higher frequence. 2. The 31 patients did not have especially past history and inducing factors. 3. Tremor frequently appeared in extremities but in the case of So-Euem-In(少陰人) tremor frequently appeared in the face. Tremor appeared to be accompanied with dizziness, palpitation, especially in So-Euem-In(少陰人) tremor appeared to be accompanied with anorexia, abdomen pain, weight loss e.t.c. 4. In the case of Tae-Eum-In(太陰人) CHUNGSIMYUNJATANG(淸心蓮子湯), CHUNGPESAGANTANG(淸肺寫肝湯) were used to be busy, in the case of So-Yang-In(少陽人) YANGJUKSANHWATANG(凉膈散火湯) were used to be busy but in the most case of So-Eum-In(少陰人) drug was not used. 5. The rate of treatment was collectively improved. The rate of treatment was higher in the female than in the male and the rate of treatment was higher in short period of the clinical history than long period. But in the age the rate of treatment was not worth special mention.

흰쥐에서 Cyclosporine의 약동학적 지표에 대한 Nicardipine의 영향 (Effect of Nicardipine on the Pharmacokinetic Parameters of Cyclosporine in Rat)

  • Cyclosporine (CsA) is a major immunosuppressive drug used widely to prevent organ allograft rejection. fits potential organotoxicity by prolonged use is known to cause both direct tissue damage and indirect pharmacokinetic interactions with other drugs. This study was performed to determine the effect of nicardipine (NCP) on the pharmacokinetic parameters of CsA in Sprague-Dawley rats. Each rat was administered with CsA in saline-treated group or in NCP-treated group which was pretreated with NCP (5 mg/kg/12 hours, i.p.) for 6 days. The plasma CsA concentration were analyzed by reversed HPLC: UV system at 0.5, 1, 2, 4, 6, and 8 hours after bolus injection of CsA (10 mg/kg). Pharmacokinetic parameters (mean$\pm$ SD, n=7) such as initial plasma concentration (C(0)), mean residence time (MRT), steady-state volume of distribution (Vdss), terminal half-life (t$\frac{1}{2}$($\beta$)) and plasma clearance (CLp) of CsA in each groups (saline-group vs NCP-group) were determined as follows: C(0) (5.66$\pm$ 1.98 vs 17.98$\pm$2.36, p<0.01); Vdss (2.68$\pm$ 1.6 vs 0.94 $\pm$ 0.25, p<0.01); CLp (0.53 $\pm$0.18 vs 0.21 $\pm$0.06, p<0.01). Therefore, Our results indicate that nicardipine significantly affects the pharmacokinetic parameters of cyclosporme, especially C(0), Vdss, and CLp in NCP-treated group. We suggest that the significant pharmacokinetic interaction between cyclosporine and nicardipine should be considered and cyclosporine level should be closely monitored and dosage reduction made as necessary in clinical situation that was coadministered with CsA and NCP.

국내 식품 냉장.냉동 창고 위생관리 수준 분석 (Sanitary Conditions for Cold and Frozen Food Storage Warehouses in Korea)

  • 전국 17곳의 식품 냉장 냉동 창고를 대상으로 입고, 교차오염, 보관, 온도 및 기록관리 등 5개 대분류와 20개 세부평가항목으로 분류하여 위생관리 수준을 분석하였다. 5개 대분류 평가항목별 수준에 대한 빈도분석결과, 기록관 리가 가장 높은 수준으로 나타났고, 이어서 온도 및 보관 관리 항목으로 나타났으며, 교차오염관리가 가장 낮은 수준으로 나타나 이 항목이 냉장 냉동창고 위생관리 중 현장에서 가장 잘 수행되지 못하는 항목으로 추정되었다. 총점과의 상관분석결과 교차오염과 보관관리가 높게 나타나 이들 2항목을 잘 관리하는 업소 일수록 전체적으로 냉장 냉동창고 위생관리를 잘 수행하는 것으로 판단되었으며, 20개의 세부평가항목 중 "교차오염과 관련한 발송시 교차 오염 방지를 위한 포장 여부"가 평균 $2.67{\pm}1.80$로 가장 낮게 나타났다.

Histone Deacetylase Inhibitor Trichostatin A Enhances Antitumor Effects of Docetaxel or Erlotinib in A549 Cell Line

  • Background and Objective: Histone deacetylase (HDAC) inhibitors represent a promising class of potential anticancer agents for treatment of human malignancies. In this study, we investigated the effect of trichostatin A (TSA), one such HDAC inhibitor, in combination with docetaxel (TXT), a cytotoxic chemotherapy agent or erlotinib, a novel molecular target therapy drug, on lung cancer A549 cells. Methods: A549 cells were treated with TXT, erlotinib alone or in combination with TSA, respectively. Cell viability, apoptosis, and cell cycle distribution were evaluated using MTT (3- (4, 5-dimethylthiazol-2-yl) -2, 5-diphenyltetrazolium bromide) assay, Hochst33258 staining and flow cytometry. Moreover, immunofluorescent staining and Western blot analysis were employed to examine alterations of ${\alpha}$-tubulin, heat shock protein 90 (hsp90), epidermal growth factor receptor (EGFR), and caspase-3 in response to the different exogenous stimuli. Results: Compared with single-agent treatment, co-treatment of A549 cells with TSA/TXT or TSA/erlotinib synergistically inhibited cell proliferation, induced apoptosis, and caused cell cycle delay at the $G_2/M$ transition. Treatment with TSA/TXT or TSA/erlotinib led to a significant increase of cleaved caspase-3 expression, also resulting in elevated acetylation of ${\alpha}$-tubulin or hsp90 and decreased expression of EGFR, which was negatively associated with the level of acetylated hsp90. Conclusions: Synergistic anti-tumor effects are observed between TXT or erlotinib and TSA on lung cancer cells. Such combinations may provide a more effective strategy for treating human lung cancer.

Molecular Genetic Studies on 167 Pediatric ALL Patients from Different Areas of Pakistan Confirm a Low Frequency of the Favorable Prognosis Fusion Oncogene TEL-AML1 (t 12; 21) in Underdeveloped Countries of the Region

  • TEL-AML1 fusion oncogene (t 12; 21) is the most common chromosomal abnormality in childhood acute lymphoblastic leukemia (ALL). This translocation is associated with a good prognosis and rarely shows chemotherapeutic resistance to 3-drug based remission induction phase of treatment as well as overall treatment. Thus, the higher the frequency of this fusion oncogene, the easier to manage childhood ALL in a given region with less intensive chemotherapy. Although global frequency of TEL-AML1 has been reported to be 20-30%, a very low frequency has been found in some geographical regions, including one study from Lahore, Punjab, Pakistan and others from India. The objective of present study was to investigate if this low frequency of TEL-AML1 in pediatric ALL is only in Lahore region or similar situation exists at other representative oncology centers of Pakistan. A total of 167 pediatric ALL patients were recruited from major pediatric oncology centers situated in Lahore, Faisalabad, Peshawar and Islamabad. Patients were tested for TEL-AML1 using nested reverse transcription polymerase chain reaction (RT-PCR). Only 17 out of 167 (10.2%) patients were found to be TEL-AML1 positive. TEL-AML1+ALL patients had favorable prognosis, most of them (82.4%, 14/17) showing early remission and good overall survival. Thus, our findings indicate an overall low frequency of TEL-AML1 in Pakistan pediatric ALL patients, in accordance with lower representation of this prognostically important genetic abnormality in other less developed countries, specifically in south Asia, thus associating it with poor living standards in these ethnic groups. It also indicates ethnic and geographical differences in the distribution of this prognostically important genetic abnormality among childhood ALL patients, which may have a significant bearing on ALL management strategies in different parts of the world.

Genomic Screening for Targets Regulated by Berberine in Breast Cancer Cells

  • Berberine, a common isoquinoline alkaloid, has been shown to possess anti-cancer activities. However, the underlying molecular mechanisms are still not completely understood. In the current study, we investigated the effects of berberine on cell growth, colony formation, cell cycle distribution, and whether it improved the anticancer efficiency of cisplatin and doxorubicin in human breast cancer estrogen receptor positive (ER+) MCF-7 cells and estrogen receptor negative (ER-) MDA-MB-231 cells. Notably, berberine treatment significantly inhibited cell growth and colony formation in the two cell lines, berberine in combination with cisplatin exerting synergistic growth inhibitory effects. Accompanied by decreased growth, berberine induced G1 phase arrest in MCF-7 but not MDA-MB-231 cells. To provide a more detailed understanding of the mechanisms of action of berberine, we performed genome-wide expression profiling of berberine-treated cells using cDNA microarrays. This revealed that there were 3,397 and 2,706 genes regulated by berberine in MCF-7 and MDA-MB-231 cells, respectively. Fene oncology (GO) analysis identified that many of the target genes were involved in regulation of the cell cycle, cell migration, apoptosis, and drug responses. To confirm the microarray data, qPCR analysis was conducted for 10 selected genes based on previously reported associations with breast cancer and GO analysis. In conclusion, berberine exhibits inhibitory effects on breast cancer cells proliferation, which is likely mediated by alteration of gene expression profiles.

Effects of Chlorpromazine·HCl on the Structural Parameters of Bovine Brain Membranes

  • Fluorescence probes located in different membrane regions were used to evaluate the effects of chlorpromazine HCl on structural parameters (transbilayer lateral mobility, annular lipid fluidity, protein distribution, and lipid bilayer thickness) of synaptosomal plasma membrane vesicles (SPMVs) isolated from bovine cerebral cortex. The experimental procedure was based on the selective quenching of 1,3-di(1-pyrenyl)propane (Py-3-Py) by trinitrophenyl groups, radiationless energy transfer from the tryptophan of membrane proteins to Py-3-Py, and energy transfer from Py-3-Py monomers to 1-anilinonaphthalene-8-sulfonic acid (ANS). In this study, chlorpromazine HCl decreased the lateral mobility of Py-3-Py in a concentration dependent-manner, showed a greater ordering effect on the inner monolayer than on the outer monolayer, decreased annular lipid fluidity in a dose dependent-manner, and contracted the membrane lipid bilayer. Furthermore, the drug was found to have a clustering effect on membrane proteins.

No Association between the CCR5Δ32 Polymorphism and Sporadic Esophageal Cancer in Punjab, North-West India

  • Background: Chemokines and their receptors influence carcinogenesis and cysteine-cysteine chemokine receptor 5 (CCR5) directs spread of cancer to other tissues. A 32 base pair deletion in the coding region of CCR5 that might alter the expression or function of the protein has been implicated in a variety of immune-mediated diseases. The action of antiviral drugs being proposed as adjuvant therapy in cancer is dependent on CCR5 wild type status. In the present study, distribution of CCR5${\Delta}32$ polymorphism was assessed in North Indian esophageal cancer patients to explore the potential of using chemokine receptors antagonists as adjuvant therapy. Materials and Methods: DNA samples of 175 sporadic esophageal cancer patients (69 males and 106 females) and 175 unrelated healthy control individuals (69 males and 106 females) were screened for the CCR5${\Delta}32$ polymorphism by direct polymerase chain reaction (PCR). Results: The frequencies of wild type homozygous (CCR5/CCR5), heterozygous (CCR5/${\Delta}32$) and homozygous mutant (${\Delta}32/{\Delta}32$) genotypes were 96.0 vs 97.72%, 4.0 vs 1.71% and 0 vs 0.57% in patients and controls respectively. There was no difference in the genotype and allele frequencies of CCR5${\Delta}32$ polymorphism in esophageal cancer patients and control group. Conclusions: The CCR5${\Delta}32$ polymorphism is not associated with esophageal cancer in North Indians. As the majority of patients express the wild type allele, there is potential of using antiviral drug therapy as adjuvant therapy.

다층 배양된 암세포에서 파크리탁셀의 항증식효과 분석 (Anti-proliferative Effect of Paclitaxel in Multicellular Layers of Human Cancer Cells)

  • Human solid tumors exhibit a multicellular resistance (MCR) resulting from limited drug penetration and decreased sensitivity of tumor cells when interacting with their microenvironments. Multicellular cultures represent solid tumor condition in vivo and provide clinically relevant data. There is little data on antitumor effect of paclitaxel (PTX) in multicellular cultures although its MCR has been demonstrated. In the present study, we evaluated antiproliferative effects of PTX in multicellular layers (MCL) of DLD-1 human colorectal carcinoma cells. BrdU labeling index (LI), thickness of MCL, cell cycle distribution and cellular uptake of calcein were measured before and after exposure to PTX at 0.1 to 50 ${\mu}M$ for 24, 48 and 72 hrs. BrdU LI and thickness of MCL showed a concentration- and time-dependent decrease and the changes in both parameters were similar, i.e., 34.2% and 40.6% decrease in BrdU LI and thickness, respectively, when exposed to $50\;{\mu}M$ for 72 hr. The DLD-1 cells grown in MCL showed increase in $%G_{0}/G_{1}$ and resistance to cell cycle arrest and apoptosis compared to monolayers. Calcein uptake in MCL did not change upon PTX exposure, indicating technical problems in multicellular system. Overall, these data indicate that antitumor activity of PTX may be limited in human solid tumors (a multicellular system) and MCL may be an appropriate model to study further pharmacodynamics of PTX.

HPLC를 이용한 어류 중의 Spiramycin 분석 (Analysis of Spiramycin in Fish Using High Performance Liquid Chromatography)

  • A high performance liquid chromatography assay method for spiramycin in fish muscle was developed. The developed method was evaluated and validated by monitoring spiramycin in olive flounder (Paralichthys olivaceus), black rock fish (Sebastes schlegeli) and in live conger eel (Anguilla japonica) in fish farms and distribution centers. Using the developed method, the recovery rate was up to 82.4-88.8%, which was higher than that of conventional methods (77.6-87.1%). In particular, the proposed sample treatment protocol was suitable for use with fish samples to remove low molecular weight materials and pigments that could interfere an accurate analysis. The prepared stock solution was very stable, and it remained chemically stable for 5 weeks at $4^{\circ}C$. The performance limit of the developed method for spiramycin acid in fish muscle was 0.05 ppm. One hundred thirty-four fish samples including olive flounder, black rock fish and live conger eel were analyzed to evaluate the overall efficiency of the modified method and to monitor the actual condition of spiramycin usage in fish farms. Olive flounder and black rock fish were collected from fish farms in coastal areas of Korea, and live conger eels were purchased from a fish market in the Busan area from September 2001 to March 2002. According to the overall performance of the developed method, it was considered suitable for the monitoring of spiramycin in fish muscle. The suggested method of analysis for spiramycin in fish muscle is registered in the Korean Official Methods of Food Analysis and is currently tieing utilized for fishery products inspection by the Korea Food and Drug Administration and the National Fisheries Products Quality Inspection Service.
