The Hippo-YAP signaling pathway is critical for cell proliferation, survival, and self-renewal in both Drosophila and mammals. Disorder of Hippo-YAP pathway leads to tumor development, progression and poor prognosis in various cancers. YAP/TAZ are the key downstream effectors of the Hippo pathway and they can be inhibited through LATS1/2, core kinases in the Hippo pathway, mediated phosphorylation. In this study, we investigated the effect of Angelica gigas Nakai extract (AGNE) on Hippo-YAP/TAZ pathway. First, ANGE induced YAP/TAZ phosphorylation and dissociation of the YAP/TAZ-TEAD transcription complex. By qRT-PCR, we found that ANGE inhibits the expression of YAP/TAZ-TEAD target gene, CTGF and CYR61. In addition, the transcriptional activity of YAP/TAZ was not suppressed significantly in LATS1/2 double-knockout (DKO) cells by ANGE compared to LATS1/2 wild-type (WT) cells, which means AGNE inhibits YAP/TAZ signaling through direct action on LATS1/2. Further, it was confirmed that AGNE-induced activation of LATS1/2 inhibited the migration potential of the vector-expressing cells by suppressing YAP/TAZ activity. The reduced migration potential was restored in active YAP-TEAD expressing cells. Taken together, the results of this study indicate that ANGE downregulates YAP/TAZ signaling in cells through the activation of LATS1/2.
Background: Efficient gene editing technology is needed for successful knock-in. Homologous recombination (HR) is a major double-strand break repair pathway that can be utilized for accurately inserting foreign genes into the genome. HR occurs during the S/G2 phase, and the DNA mismatch repair (MMR) pathway is inextricably linked to HR to maintain HR fidelity. This study was conducted to investigate the effect of inhibiting MMR-related genes using CdCl2, an MMR-related gene inhibitor, on HR efficiency in HC11 cells. Methods: The mRNA and protein expression levels of MMR-related genes (Msh2, Msh3, Msh6, Mlh1, Pms2), the HR-related gene Rad51, and the NHEJ-related gene DNA Ligase IV were assessed in HC11 cells treated with 10 μM of CdCl2 for 48 hours. In addition, HC11 cells were transfected with a CRISPR/sgRNA expression vector and a knock-in vector targeting Exon3 of the mouse-beta casein locus, and treated with 10 μM cadmium for 48 hours. The knock-in efficiency was monitored through PCR. Results: The treatment of HC11 cells with a high-dose of CdCl2 decreased the mRNA expression of the HR-related gene Rad51 in HC11 cells. In addition, the inhibition of MMR-related genes through CdCl2 treatment did not lead to an increase in knock-in efficiency. Conclusions: The inhibition of MMR-related gene expression through high-dose CdCl2 treatment reduces the expression of the HR-related gene Rad51, which is active during recombination. Therefore, it was determined that CdCl2 is an inappropriate compound for improving HR efficiency.
Objective: To investigate the effect of high expression of XAF1 in vivo or in vitro on lung cancer cell growth and apoptosis. Methods: 1. The A549 human lung cancer cell line was transfected with Ad5/F35 - XAF1, or Ad5/F35 - Null at the same multiplicity of infection (MOI); (hereinafter referred to as transient transfected cell strain); XAF1 gene mRNA and protein expression was detected by reverse transcription polymerase chain reaction (RT-PCR) and Western blotting respectively. 2. Methyl thiazolyl tetrazolium (MTT) and annexin V-FITC/PI double staining were used to detect cell proliferation and apoptosis before and after infection of Ad5/F35 - XAF1 with Western blotting for apoptosis related proteins, caspase 3, caspase - 8 and PARP. 3. After the XAF1 gene was transfected into lung cancer A549 cells by lentiviral vectors, and selected by screening with Blasticidin, reverse transcription polymerase chain reaction (RT-PCR) and Western blotting were applied to detect mRNA and protein expression, to establish a line with a stable high expression of XAF1 (hereinafter referred to as stable expression cell strain). Twenty nude mice were randomly divided into groups A and B, 10 in each group: A549/XAF1 stable expression cell strain was subcutaneously injected in group A, and A549/Ctrl stable cell line stable expression cell strain in group B (control group), to observe transplanted tumor growth in nude mice. Results: The mRNA and protein expression of XAF1 in A549 cells transfected by Ad5/F35 - XAF1 was significantly higher than in the control group. XAF1 mediated by adenovirus vector demonstrated a dose dependent inhibition of lung cancer cell proliferation and induction of apoptosis. This was accompanied by cleavage of caspase -3, -8, -9 and PARP, suggesting activation of intrinsic or extrinsic apoptotic pathways. A cell strain of lung cancer highly expressing XAF1 was established, and this demonstrated delayed tumor growth after transplantation in vivo. Conclusion: Adenovirus mediated XAF1 gene expression could inhibit proliferation and induce apoptosis in lung cancer cells in vitro; highly stable expression of XAF1 could also significantly inhibit the growth of transplanted tumors in nude mouse, with no obvious adverse reactions observed. Therefore, the XAF1 gene could become a new target for lung cancer treatment.
Endoscopic transthoracic sympathectomy (ETS) has recently become estabilished as a successful treatment for severe palmar and axillary hyperhidrosis. Descriptions have been published of neurolytic, operative and alternative endoscopic procedures involving thermocoagulation, laser coagulation, or or nonvideo-assisted ganglionectomy using equipment not widely available, with low morbidity and excellent results. All methods have advantage and disadvantages. A 19-year-old male who suffered from severe hyperhidrosis on face, palms and axillary areas, has been initially treated with stellate ganglion block in other pain clinic. He was transfered to our pain clinic for endoscopic thoracic sympathectomy. The patient was intubated left side 34 Fr. double lumen tube and positioned left semi-lateral position for right sympathectomy. Right side pneumothorax was created by clamping the ipsilateral side of the double lumen tube and aspiration of air. 11-mm trocar was introduced through incision at the third intercostal space in anterior axillary line, and then additional two 11-mm and 5-mm trocar was introduced through second and fifth intercostal space in mid axillary line. The lung was gently retracted and the parietal pleura over the heads of the appropriate ribs excised using 5-mm sharp insulated coagulating microprocesss. The T4, T3, and T2 ganglions, as well as accompanying rami communicantes, and other branchs arising from upper thoracic nerves to the brachial plexus and surrounding tissues were carefully dissected, coagulated. During sympathectomy, skin temperature of middle was continuously monitored. Elevation of palmar skin temperature intraoperatively indicated an adequate sympathectomy with a definite therapeutic effect. A No. 28 Fr. thoracotomy tube was introduced through a troca under video guidance, placed under water seal after the lung was reinflated. the controlateral side was performed same procedure. After bilateral sympathectomy, chest tubes were removed, and then, he was discharged 2 days after operation with great satisfaction. The ETS provides a well-tolerated, cost-effective alternative to thoracic sympathectomy for primary hyperhidrosis and sympathetic mediated neuropathic pain disorder. And T2 ganglion is considered the key ganglion for the treatment of primary hyperhidrosis. The low incidence of compensatory sweating may by explained by the limited extent of the sympathectomy.
This study attempts to verify the double mediating effects of trait anxiety and burnout in the relationship between socially prescribed perfectionism and subjective well-being of employees. For this study, 300 office workers were surveyed to measure the trait anxiety, the burnout, the socially prescribed perfectionism and subjective well-being. The collected data were analyzed with SPSS PROCESS Macroin an bootstrapping method. The result of this study can be summarized as follows. First, socially prescribed perfectionism had significant positive relationship with trait anxiety and burnout. Socially prescribed perfectionism had significant negative relationship with subjective well-being. Also trait anxiety had significant positive relationship with burnout and significant negative relationship with subjective well-being. In addition, burnout had significant negative relationship with subjective well-being. Second, trait anxiety and burnout fully mediated the relationship between socially prescribed perfectionism and subjective well-being. Third, there was a dual mediation effect on trait anxiety and burnout in the relationship between socially prescribed perfectionism and subjective well-being. In conclusion, it suggests that in order to elevate subjective well-being of employees with a high level of socially prescribed perfectionism, it would be more effective to deal with trait anxiety and burnout which are proved to make them happy than to deal with perfectionism itself.
Women with children suffer from double burden of childcare and household labor. Based on the framework of social determinants of mental health, this study investigated the moderated mediation effect of income in the relationship between time poverty and mental health among women with children. The study sample included 1,505 women from the $6^{th}$ wave of Korean Longitudinal Survey of Women and Families (KLoWF). Results showed that the lower level of time poverty was associated with decreased stress and depression, and stress mediated the relationship between time poverty and depression. Income had a moderated mediation effect in the relationship between time poverty, stress, and depression. Time poor women were more vulnerable to poor mental health, and income buffered the effect of time poverty on women's mental health. This implies that women in the lower end of socioeconomic ladder are more prone to the effect of time poverty. The study speaks to the importance of social factors on mental health which has usually been considered an individual problem. Policies and services need to address women's working conditions and income security in order to increase their mental health.
The purpose of this study was to examine the effect of out-of-school adolescents' social stigma on smart-phone addiction through career disorders and depression. In the process that social stigma of adolescents outside of school affects smart-phone addiction, the mediating effect of career barriers and depression was verified. The subjects of this study were 318 subjects who participated in the 5th year survey of out-of-school youth panel by the Korea Youth Policy Institute. For data analysis, the correlations and mediating effects between major variables were verified. As a result of the study, first, all of the major variables were found to have statistically significant correlations. Second, the social stigma of out-of-school adolescents was found to have an effect on smart-phone addiction through career disorders and depression, confirming a double mediation. In addition, in the process in which out-of-school adolescents' social stigma affects smart-phone addiction, it was confirmed that career disorder and depression were respectively mediated, but social stigma did not have a significant effect on smart-phone addiction. Based on these results, prevention of smart-phone addiction among out-of-school adolescents and intervention methods and implications for youth coaching were presented.
The purpose of this study was to examine the effects of the job characteristics of the PR practitioner on flourishing and to identify the mediating effect of the meaning of work and job engagement. This study used a survey of 353 PR practitioners from PR firms and the hypothesis was verified by gathering the data of the job characteristics, the meaning of work, the job engagement and the flourishing by conducting hierarchical regression analysis and SPSS Process Macro bootstrapping analysis. The results showed that the job characteristics of the PR practitioner were found to influence the flourishing with the double-mediation effect, the meaning of the work and job engagement. The more PR practitioners regard their work affects their surroundings, the higher the degree of autonomy is, and the more they get feedbacks they engaged more, as they valued their job more thereby experience flourish more. Moreover, when PR practitioners valued their job more, they more engaged in their work and experience flourish. The meaning of work revealed to be the important factor to affect flourish regardless of the job characteristics and the job engagement so that gained the results that PR firms' effort to elevate the meaning of work of PR practitioners has is important. The study findings suggest that PR practitioners' flourishing is manageable in the organization by paying attention to oneself in the aspect of the organization, not leaving in individual areas. Limitaions and implications for future studies were discussed.

This study focused on whether employees' perceived leader's creativity has a significant effect on employees' job satisfaction and innovative behavior. In addition, this study analyzed the mediating effect of positive psychological capital and creative self-efficacy in the relationship between perceived leader creativity and employee satisfaction with the job or innovation behavior, and analyzed the moderated mediating effect of team cohesion. This study conducted confirmatory factor analysis and path analysis for each factor using the measurement tool verification method using the SmartPLS 3.0 structural equation model, and empirically analyzed the research model. As a result, it was found that the creativity of employees' perceived leaders did not directly significantly affect employees' job satisfaction and innovation behavior. However, it was found that positive psychological capital significantly mediated the relationship between perceived leader creativity and job satisfaction or innovative behavior. In addition, positive psychological capital and creative self-efficacy have a complete double-mediation relationship between the employees' perceived leader's creativity and employees' innovative behavior. However, positive psychological capital and creative self-efficacy don't have a double-mediation relationship between the employee's perceived leader's creativity and employees' job satisfaction. Finally, Team cohesion was found to moderate the relationship between perceived leader creativity, positive psychological capital (or creative self-efficacy). However, the research model that the moderated mediating effect of team cohesion on the relationship between leader's creativity, the mediating variables(positive psychological capital and creative self-efficacy), and job satisfaction (or innovative behavior) was significant only for groups with high team cohesiveness. This study examined the needs of the times for the importance of creativity of perceived leaders and analyzed the relationship between factors influencing job satisfaction and innovation behavior of organizational members. The study is meaningful in that it suggested a plan to establish a specific organizational strategy to enhance positive psychological capital and creative self-efficacy.
This study is an empirical research based on data analysis obtained by surveying 655 adult men and women. The notable findings are as follow: First, overall, individuals' perception of extramarital affair is negative, but about 16.4% respondents have a positive perception of it. Second, the respondents have a more critical perception of their spouse's extramarital affairs compared to their own. Thirdly, the less critical perception of extramarital affair, the more acceptance of my extramarital relationship, the less tolerance of my spouse's extramarital relationship regardless marital status. Forth, while the less tolerance of spouse's extramarital relationship, the less openness to extramarital sex among unmarried group, the more acceptance of my own extramarital relationship, the more openness to extramarital sex among married group. Lastly, individuals' perception of extramarital relationship does not directly affect the openness to extramarital sex only among unmarried group, however, the correlation between the perception of extramarital affairs and the openness to extramarital sex is fully mediated by the effect of unacceptance of spouse's extramarital affairs. The moderating effect of marital status is proved.
