New Triterpenoids from the Fruits of Schisandra wilsoniana and Their Biological Activities

  • Investigation of an organic extract of the fruits Schisandra wilsoniana led to the isolation of two new highly oxygenated nortriterpenoids, named schilancidilactones V-W (1-2). Their structures were elucidated by spectroscopic evidence. Compounds 1-2 feature a double bond between C-7 and C-8 compared with related known nortriterpenoids isolated from the genus Schisandra. Compounds 1 and 2 were tested for their anti-HIV-1 activities and cytotoxicity. The results revealed that compounds 1 and 2 showed moderate anti-HIV-1 activities with $EC_{50}$ 3.05 and 2.87 ${\mu}g/mL$, respectively, and compound 1 showed high cytotoxicity against KB and MDA-MB-231 cell with $IC_{50}$ values of 3.18 and 5.22 ${\mu}M$, respectively.

나노리포좀을 이용한 astaxanthin의 안정화 (Stabilization of Astaxanthin Using Nanoliposome)

  • 본 연구에서는 이중결합을 가진 불포화화합물로 제조나 저장 시 열과 산화 (빛)에 의해 쉽게 파괴되어 활성이 감소하여 응용범위에 한계가 있는 아스타잔틴의 안정성 향상을 위하여 나노리포좀 제형기술을 이용하였다. 제형안정성 평가를 통하여 리포좀 제조조건 및 조성비를 확립하였고, 포접나노리포좀을 제조하여 포접전의 아스타잔틴과 안정성을 비교 검토하였다. 아스타잔틴을 포접하여 포접나노리포좀 제조 시포접전의 아스타잔틴에 비해 열에 대한 안정성이 2배 정도 향상되고, UV 안정성 또한 3배 향상된 것을 확인하였다. 이상의 결과로부터 아스타잔틴을 포접한 포접나노리포좀은 안정성이 우수한 기능성 원료로서 산업적으로 용도를 넓히고 부가가치를 더욱 향상시킬 수 있을 것으로 기대된다.

Flavonoids의 약리작용(I) -Flavonoids 구조와 과민반응 억제작용과의 상관성- (Pharmacological Activities of Flavonoids (I) -Relationships of Chemical Structure of Flavonoids and their Inhibitory Activity of Hypersensitivities-)

  • The activities of twenty-one flavonoids and their related compounds on the hypersensitivity reaction against various antigens were studied in vitro and in vivo. 1. Generally flavonoids inhibited significantly the homologous passive cutaneous anaphylaxis (PCA) induced by reaginic antibody as compared as anaphylaxis by compound 48/80-induced mast cell degranulation, and so more strongly active in the IgE-mediated anaphylaxis than non-IgE-mediated anaphylaxis. 2. Flavonids inhibited remarkably Arths reaction, hemolysin titer, delayed hypersensitivity, haemagglutinin titer, rosette forming cells and plague forming cells against sheep red blood cells, and so it exhibited that flavonoids inhibited type 2, 3 and 4 hypersensitivity. 3. Quercetin, kaempferol, hesperetin, disodium cromoglycate, malvin and baicalein were active dose-dependently in the all types of hypersensitivity. Fisetin, daidzein, morin, narigin, flavone, catechin, rutin, hesperidin, neophsperidin, apigenin and chrysin were significantly active in the various types of hypersensitivity, but apigenin, rutin and catechin were less active in the delayed hypersensitivity. Taxifolin was significantly active in PCA and histamine-induced anaphylaxis except other types of hypersensitivity. Rotenone and cyanin also inhibited all types of hypersensitivity, but they are toxic. 4. Based on these results from hypersensitivity, the following flavonoid structure-activity relationships became apparent. 1) Flavonoids with $C_{2-3}$ double bond in C-ring were more active than that of $C_{2-3}$ saturation. 2) Flavonoids with $C_4$ ketone group in C-ring were more active than abscence of them except catechin and malvin. 3) Flavonoids with benzene ring at positions 2 or 3 in C-ring exhibited same activities. 4) Flavonoids with opening of the C-ring does not abolish their activities. 5) The glycosylated flavonoids in position 3 or 7 was less active than their aglycone. 6) Flavonoids with the more hydroxy group in A and B-ring were more active. 7) Flavonoids with or without $C_3-OH$ did not change their activities.

니켈촉매에 의한 크로톤 알데히드의 액상 수소첨가반응 (Liquid Phase Hydrogenation of Croton Aldehyde with Nickel Catalysts)

  • 에탄올의 제조시 불순물로서 미량 생성되는 크로톤 알데히드를 수소와 반응시켜 n-부틸알콜로 전환하여 불포화 탄화수소를 제거하는 공정에 액상수소 첨가반응을 이용하고자 하며, 기존의 기상 수소첨가반응보다 월등한 에너지 절약 효과가 있다. 반응촉매는 내구성 및 가격 등 경제적인 측면을 고려하여 니켈촉매를 선택하였으며, 반응전화율의 측정은 PMT(permanganate time) test 방법을 적용하였다. PMT는 에탄올에 미량으로 함유되어 있는 크로톤알데히드의 초기농도 증가에 따라 급격히 감소하였으며, 크로톤 알데히드로부터 n-부틸알콜로의 수첨반응은 탄소-탄소 이중결합의 환원 후, 알데히드의 환원 과정이 연속적으로 일어나고, 각 반응단계는 0차 반응속도 상수를 가진다. 실험조건 범위 내에서는 반응 온도가 높을수록, LHSV가 느릴수록 PMT는 길어지고, 반응압력은 PMT와 거의 무관함을 보였다.

황산으로 개질된 TiO2 촉매의 특성 (Characterization of TiO2 Catalyst Modified with H2SO4)

  • $TiCl_4-HCl$ 혼합용액으로부터 침전법으로 $TiO_2$를 만들고 이를 황산으로 개질하여 $TiO_2/SO_4{^{2-}}$ 촉매를 제조하였으며 제조된 촉매의 특성을 IR, XPS, XRD 및 DT-TGA로 조사하였다. 개질된 촉매의 산의 세기는 적어도 $H_0{\leq}-14.52$로 초강산 성질을 나타내었으며 이 성질은 $TiO_2$와 황산이온에 의하여 형성된 착물의 S=O 결합성질에 기인하였다. $TiO_2/SO_4{^{2-}}$의 IR spectra를 조사한 결과 황산이온은 $TiO_2$ 표면에 2자리 ligand로 배위하였다. $TiO_2/SO_4{^{2-}}$는 황산으로 개질하지 아니한 순수한 $TiO_2$에 비하여 표면적이 상당히 컸으며 또한 무정형에서 anatase상으로 되는 전이온도도 높았다.

  • 식물 세포의 미토콘드리아와 엽록체는 광 증감제로 작용할 가능성이 있는 여러 가지 색소들을 함유하고 있다. 이들 색소들은 대부분이 막에 결합되어있으며 청색광 영역에서 강한 흡수대를 가지므로 청색광하에서 노출된 이들 소기관에서는 활성산소가 발생되어 막의 구조적, 기능적 피해를 유발하는 요인이 된다. 활성산소의 광발생에 따른 막구조의 변화는 일차적으로 막지질 지방산성분의 산화적 파괴에 기인할 것이다. 본 연구에서는 식물이 고광도의 가시광선(특히 청색광선)하에서 일어나는 막지질 지방산의 산화적 파괴에 지방산 대신 상대적으로 둔감한 포화 지방산의 조성비를 높일 것이라고 가정하고, 이를 뒷받침할만한 실험결과를 얻었다. 즉, 광질이 서로 다른 가시광선 처리조건하에서 생장중인 식물 및 그 소기관들을 대상으로 막지질의 과산화, 지방산조성 및 불포화도, 막결합 단백질의 활성 등을 측정하고 이를 제시하였다.

Microcrystalline cellulose에 자유 라디칼을 이용한 methyl methacrylate의 그래프팅 반응과 이를 이용한 천연복합필름의 제조 (Grafting of MMA onto MCC through free radical method and its application to all natural cellulose composite film preparation)

  • Methyl methacrylate(MMA) was grafted onto microcrystalline cellulose(MCC) with ceric ammonium nitrate(CAN) as a redox initiator at the various conditions. The cellulose triacetate(CTA) composite films added MCC and MMA-grafted MCC powders were prepared on a glass plate. The graft yield(GY) and graft efficiency(GE) of the grafted MCC were calculated with the simple equations by the weight balance method. The double bond of C=O on the grafted MCC surfaces was confirmed by the fourier transform infrared spectroscopy with attenuated total reflection(FT-IT ATR) spectrophotometer. After grafting, the degree of crystallinity of cellulose powders was decresed by judging from x-ray diffraction(XRD) data. Scanning electron microscope(SEM) photos showed the only solvent and CAN solution could change the roughness of MCC powders and the effect of powder dispersions in composite matrix. The tensile strength of MCC/CTA composite films was decreased with increase of MCC powder contents. When 5% grafted MCC was added, the tensile strength of grafted MCC/CTA composite films was increased from 82.3 MPa to 97.2 MPa. The thermal property of powders was also analyzed by the thermogravimetric analysis(TGA).

Resolution of Molecular Species of the Triacylglycerol Containing Petroselinic Acid $(cis-C_{18:1{\omega}12)$ by Silver Ion-HPLC

  • On the analysis of triacylglycerol (TG) from the kernels of Acanthopanax sessiliflorus by reversed phase-HPLC, it was separated into three main fractions of PN 44, 46 and 48, according to partition number (PN). On the contrary, it could be clearly classified into seven fractions of SMM, MMM, SMD, MMD, SDD, MDD and MDT by silver ion-HPLC by the number of double bond in the acyl chains of TG species. But resolution of so-called critical pairs of TG molecular species such as molecular pairs of $P_eLL$ $[C_{18:1{\omega}12}/(C_{18:2{\omega}6)2}]$ and OLL $[C_{18:1{\omega}9}/(C_{18:2{\omega}6)2}]$ and OOL $[(C_{18:1{\omega}9)2}/C_{18:2{\omega}6]$, and $P_eP_eL$ $[(C_{18:2{\omega}12)2}/C_{18:1{\omega}6]$ was not achieved $(P_e;$ petroselinic acid, L; linoleic acid, O; oleic acid). On the other hand, TG extracted from Aralia continentalis kernels were also fractionated into seven groups of SSM, SMM, MMM, SMD, MMD, SDD and MDD (S; saturated acid, M; monoenoic acid, D; dienoic acid) by silver ion-HPLC, although it's were classified into three groups of PN 44, 46 and 48 by reversed phase-HPLC. The fractions of SMM, MMM, MMD and MDD were divided into two subfractions, respectively; the fractions of SMM, MMM, MMD and MDD were resolved into the subfraction of $PP_e/P_e$ and POO (critical pairs from each other), that of $P_e/P_e/P_e$ and OOO, that of $P_e/P_e/L$ and OOL, and that of $P_e/L/L$ and OLL.

2-메타크릴로일옥시에틸 이소시아네이트에 의한 삼나무재의 화학처리 (Chemical Modification of Japanese Cedar with 2-Methacryloyloxyethyl Isocyanate)

  • 목재는 2-메타크릴로일옥시에틸 이소시아네이트(MOI)와 $50-60^{\circ}C$에서 촉매존재하에서 매우 쉽게 반응하였다. 적외선 분광분석을 통하여, 목재가 MOI와 우레탄결합($1715cm^{-1}$)를 형성하고 있을 뿐만 아니라, 올레핀구조의 탄소간 이중결합 ($1635cm^{-1}$)이 존재하고 있음이 밝혀졌다. 도입된 메타크릴산기는 다른 비닐모노머와의 그래프트공중합시 반응시점으로 작용할 것이다. 또한 MOI처리목재는, MOI와의 반응에 의해 친수성이던 목재의 표면이 소수화되어 접촉각이 증대되는 결과를 낳았다. X선광전자분광분석(XPS)의 결과, 목재의 표면부위의 대부분은 MOI에 의해 개질되었음을 밝혀졌다.

유한요소해석을 통한 섬유보강 아스팔트의 파괴거동특성 분석 (Finite Element Analysis for Fracture Resistance of Fiber-reinforced Asphalt Concrete)

  • PURPOSES : In this study, a fracture-based finite element (FE) model is proposed to evaluate the fracture behavior of fiber-reinforced asphalt (FRA) concrete under various interface conditions. METHODS : A fracture-based FE model was developed to simulate a double-edge notched tension (DENT) test. A cohesive zone model (CZM) and linear viscoelastic model were implemented to model the fracture behavior and viscous behavior of the FRA concrete, respectively. Three models were developed to characterize the behavior of interfacial bonding between the fiber reinforcement and surrounding materials. In the first model, the fracture property of the asphalt concrete was modified to study the effect of fiber reinforcement. In the second model, spring elements were used to simulated the fiber reinforcement. In the third method, bar and spring elements, based on a nonlinear bond-slip model, were used to simulate the fiber reinforcement and interfacial bonding conditions. The performance of the FRA in resisting crack development under various interfacial conditions was evaluated. RESULTS : The elastic modulus of the fibers was not sensitive to the behavior of the FRA in the DENT test before crack initiation. After crack development, the fracture resistance of the FRA was found to have enhanced considerably as the elastic modulus of the fibers increased from 450 MPa to 900 MPa. When the adhesion between the fibers and asphalt concrete was sufficiently high, the fiber reinforcement was effective. It means that the interfacial bonding conditions affect the fracture resistance of the FRA significantly. CONCLUSIONS : The bar/spring element models were more effective in representing the local behavior of the fibers and interfacial bonding than the fracture energy approach. The reinforcement effect is more significant after crack initiation, as the fibers can be pulled out sufficiently. Both the elastic modulus of the fiber reinforcement and the interfacial bonding were significant in controlling crack development in the FRA.
