Background: Interspecific ginseng hybrid, Panax ginseng ${\times}$ Panax quenquifolius (Pgq) has vigorous growth and produces larger roots than its parents. However, F1 progenies are complete male sterile. Plant tissue culture technology can circumvent the issue and propagate the hybrid. Methods: Murashige and Skoog (MS) medium with different concentrations (0, 2, 4, and 6 mg/L) of 2,4-dichlorophenoxyacetic acid (2,4-D) was used for callus induction and somatic embryogenesis (SE). The embryos, after culturing on $GA_3$ supplemented medium, were transferred to hormone free 1/2 Schenk and Hildebrandt (SH) medium. The developed taproots with dormant buds were treated with $GA_3$ to break the bud dormancy, and transferred to soil. Hybrid Pgq plants were verified by random amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR) analyses and by LC-IT-TOF-MS. Results: We conducted a comparative study of somatic embryogenesis (SE) in Pgq and its parents, and attempted to establish the soil transfer of in vitro propagated Pgq tap roots. The Pgq explants showed higher rate of embryogenesis (~56% at 2 mg/L 2,4-D concentration) as well as higher number of embryos per explants (~7 at the same 2,4-D concentration) compared to its either parents. The germinated embryos, after culturing on $GA_3$ supplemented medium, were transferred to hormone free 1/2 SH medium to support the continued growth and kept until nutrient depletion induced senescence (NuDIS) of leaf defoliation occurred (4 months). By that time, thickened tap roots with well-developed lateral roots and dormant buds were obtained. All Pgq tap roots pretreated with 20 mg/L $GA_3$ for at least a week produced new shoots after soil transfer. We selected the discriminatory RAPD and ISSR markers to find the interspecific ginseng hybrid among its parents. The $F_1$ hybrid (Pgq) contained species specific 2 ginsenosides (ginsenoside Rf in P. ginseng and pseudoginsenosides $F_{11}$ in P. quinquefolius), and higher amount of other ginsenosides than its parents. Conclusion: Micropropagation of interspecific hybrid ginseng can give an opportunity for continuous production of plants.
The seeds, scions and plants of chestnut tree (Castanea. crenata) and Chinese chestnut tree (C. bungeana) were irradiated by gamma ray in order to know their biological effects on germination, plant growth and mutation at several accumulative doses and dose rates. The results of this study could be summarized as follows: 1. In general, the radio-sensitivity of scions was more sensitive to irradiation, showing 50% reduction dose of the control for grafting percentage at 3.1 kR as compared with 4.6 kR for the above reduction dose in germination rates of seeds. 2. The seeds treated by 5 kR dose resulted in some albino mutants at a rate of 0.84%. There was a general tendency that the seedling height reduced significantly as the irradiation dose increased. 3. The scions treated by an acute irradiation showed their 50% reduction dose in grafting at 3.2 kR for Chinese chestnut and at 3.1 kR for chestnut, respectively, while their irradiation doses increased three times, having 10.2 kR for the 50% reduction dose in the case of semi-acute irradiation. 4. When Chinese chestnut trees were irradiated during their dormant period with a total dose of 7.5 kR to 4.9 kR at a dose rate of 150 R to 98 R per day, there were induced giant leaf bud-sports at a frequency of 16.6%. The averag leaf area of the giant leaf bud-sports were measured at 96.36 square centimeters, while the area of normal leaf was only 26.28 square centimeters.
This studies were conducted to get the basic information on the growth habits of Scirpus planiculmis F. Schmidt for control in poldered paddy field. The distribution of corm was about 16% in 0-5cm, 66% in 5-8 cm. 18% in 8-10cm depth of underground. The mean depth of total corm distribution was about 6.51cm. Number of rhizomes and dormant buds per corm were one to four, respectively. Number of rhizomes. dormant buds and total buds(rhizome+dormant bud) were 2.34, 2.85 and 5.20, respectively. The phyllotaxy of S. planiculmis was 1/3 and bracteal leaf were showed from first to 4th leaves. The period of propagation to 100 plants from emergence was 58.52 and 48 days for corm seeded on March 25th, b4ay 7th and June 6th respectively. Days to emergence of S. planiculmis from emergence of previous plant was gradually shorted. as order of propagation from corm seeded progressed. The other side, length of rhizome, plant height and diameter of culm was increased. The plant height of S. planiculmis was higher than rice plant from early stage in rice direct seeding culture, and from 30 dats after transplanting in rice transplanting culture. Speed of propagation was faster in rice direct seeding culture than in rice trasplanting culture.
The population ecology (Ecological characteristics) of Squalidus chankaensis tsuchigae were investigated at Geum River from March to November, 2013. This species inhabited midstream of the river where was mostly covered with cobble and sand. The depth of the habitat was 30~60 cm and velocity of stream was 2.27~4.88 cm/sec. The sex ratio of female to male was 1 : 0.86. They were sexually mature when they were over two years of age. The spawning period was from June to July with water temperatures from $25^{\circ}C$ to $25^{\circ}C$. Sexually matured individuals appeared over the two year of age. Frequency analysis of total length indicated that the group with 55~70 mm in total length was one year old, the 71~90 mm was two years old, and the group over 91 mm was three years old. The average number of eggs in the ovaries was estimated as 2,219 per matured female. The number of matured eggs size in ovaries was 0.64~0.98 (mean: 0.85) mm. Calulation length-weight relation of S. chankaensis tsuchigae show the value of contant a as 0.0002 and b as 2.81, and condition factor (K) was 1.06 on average. The stomach contents were Cyanophyta, Chlorophyta, Bacillariophyta, Crustaceae, Copepoda, Nematota, Actithocephala, and Ephemeroptera, Hemiptera, Diptera, and Trichoptera that belong to Aquatic insect, and dormant bud of Pectinatella that belongs to Phylactolaemata. The diet of S. chankaensis tsuchigae was omnivorous, mainly vegetable feed on attached algae (filament form), and mainly animality feeded on benthic invertebrates such as Chironomidae larvae.
Small-sized persimmons produced by high crop load are better accepted in the export markets. However, maintaining high crop load frequently results in weakness of tree vigor, deterioration of fruit quality, and increase of the risks for alternate bearing. This experiment was conducted to determine the combined effects of fertilization rate and leaf-fruit (L/F) ratio on container-grown 3-year-old 'Fuyu' persimmon trees. Application of 3.6-g N, 2.1-g $P_2O_5$, 2.7-g $K_2O$, 2.7-g CaO, and 0.6-g MgO was for the control fertilization rate (CF) and that of a 3-fold CF was for the high fertilization rate (HF). Commercial fertilizers were surface-applied to a container on July 6, July 17, and August 10 in three equal aliquots. Single tree for each fertilization rate was assigned for 12 L/F ratios (5, 6.3, 7.7, 9, 10.4, 13, 15.5, 18, 21, 24, 27, and 33) mostly by fruit thinning or rarely by defoliation on July 1. HF did not affect the yield, weight and soluble solids of the fruits but decreased skin color. As L/F ratio increased, yield decreased but average weight, skin color, and soluble solids of fruits increased. With HF, N and K concentrations in leaves, fruits, and shoots increased to some extent but soluble sugars in dormant shoots decreased. Many shoots were cold-injured with low L/F ratio especially at the HF. HF did not increase number of flower buds the next spring either on a shoot or on a tree basis but increased shoot length, compared with the CF. Increasing L/F ratio markedly increased number of flower buds and shoot growth the following year at both fertilization rates. Therefore, an appropriate combination of fertilization rate and L/F ratio should be necessary to maintain stable fruit production and tree vigor at high crop load.
This study aimed to know difference in freezing resistance among different clonal seedlings or different seed source seedlings of Cryptomeria japonica which has been selected where extreme cold prevails in Korea and Japan. The freezing resistance of three 12-50 year old trees was also measured in the experiment. The freezing resistance was measured in different tissue parts: mainly leaf, cambiam and xylem, at three different collection dates in two different collection places during the winter of 1977-1978. The following results and discussions were made: 1. The clonal difference in freezing resistance of Cryptomeria japonica was $9^{\circ}$ to $15^{\circ}C$ in maximum according to the collection place. However, the clonal difference in freezing resistance was not related to the difference in climatic conditions where the parent tree have been growing. This impiled that the natural selection of cold resistant genes in Cryptomeria japonica has not reached its evolutional equilibrium yet since most of the Cryptomeria forest has been established by artificial regeneration. 2. The difference in freezing resistance among leaf, cambium and xylem was not apparent except that leaf of several clones showed higher freezing resistance than cambium or xylem when they collected at mid-winter. The least freezing resistant tissue part, thought its freezing resistance was not measure in all clones and all temperatures were appeared in the apical buds. The new shoot growth was observed in the next spring with being replaced by its dormant or adventitious bud growth when the apical bud was injured dy cold during winter. 3. The freezing resistance of leaf, cambium and xylem was shown high enough so that freezing resistance Cryptomeria clones in this experiment were supposed to be able to survive in cold winter conditions at the middle part of Korea. However, it was reported that the most susceptible tissue part to winter injury was the basal stem, but of which freezing resistance was not-measured in this experiment. Several silvicultural methods for prevention of Cryptomeria seedlings from cold damage were discussed in literature.
The objectives of this study were to obtain standardized basic data on cambial electrical resistance (CER) of ornamental trees to be used for estimation of tree vigor and to compare CER between shrubs, deciduous, and coniferous trees in relation to bud opening, flowering, and growth cessation. Eighty healthy trees with four trees each for twenty ornamental tree species growing at an university campus in Suwon were selected and their CER was measured using a Shigometer every week from March to May, once a month from June to October and every two weeks in November, 1996. The CER of all tree species was high in March, started to decrease in April, maintained minimum in May through August, began to increase in September, and returned in November to the similar high level to March. Among the 20 species, Metasequoia glyptostroboides showed the lowest($5.5k{\Omega}$) annual average CER, while Cercis chinensis and Ligustrum obtusifolium had the highest($22.7k{\Omega}$, $22.9k{\Omega}$) annual average CER. The lowest CER($2.4k{\Omega}$) in summer(June through August) was obtained from Wisteria floribunda, second lowest($2.5k{\Omega}$) from Metasequoia glyptostroboides, the highest CER($46.8k{\Omega}$) during dormant season from Euonymus japonica, and second highest($45.0k{\Omega}$) obtained from Ligustrum obtusifolium. The CER of most species, particularly shrub species, started to decrease with bud opening, and many species, particularly shrubs and deciduous trees with large amount of flowers, showed sharp decrease with flowering. When CER was compared between shrubs and trees, shrubs showed higher average CER than trees, and seasonal difference in CER of evergreen species was smaller than that of deciduous species. It is interesting to note that the deciduous Metasequoia glyptostroboides had the lowest annual average CER.
