  • The purpose of this paper is to examine the effects on reliability of equipment or product which spends a great deal of its time in the dormant condition. Many systems experienced periods of dormancy throughout their life cycle, such as periods of operational storage where the system waits, ready for use. The design of such systems must account for how these period of dormant effects system performance. The methodology for predict and analysis was developed to support the evaluation of dormant modes of operation of systems and subsystems. For proper handling of the dormant environment, issues relating to dormant failures need to be taken into consideration from design stage of the lifecycle. Furthermore, the relevant environmental concerns that need to be taken into consideration depend on the environmental factors associated with each different target environment. This paper will look at dormant reliability, the possible dormant reliability models and the methodology on life cycle reliability which has different dormant phase.

Relationship between reactive oxygen species and autophagy in dormant mouse blastocysts during delayed implantation

  • Objective: Under estrogen deficiency, blastocysts cannot initiate implantation and enter dormancy. Dormant blastocysts live longer in utero than normal blastocysts, and autophagy has been suggested as a mechanism underlying the sustained survival of dormant blastocysts during delayed implantation. Autophagy is a cellular degradation pathway and a central component of the integrated stress response. Reactive oxygen species (ROS) are produced within cells during normal metabolism, but their levels increase dramatically under stressful conditions. We investigated whether heightened autophagy in dormant blastocysts is associated with the increased oxidative stress under the unfavorable condition of delayed implantation. Methods: To visualize ROS production, day 8 (short-term dormancy) and day 20 (long-term dormancy) dormant blastocysts were loaded with $1-{\mu}M$ 5-(and-6)-chloromethyl-2', 7'-dichlorodihydrofluorescein diacetate, acetyl ester (CM-$H_2DCFDA$). To block autophagic activation, 3-methyladenine (3-MA) and wortmannin were used in vivo and in vitro, respectively. Results: We observed that ROS production was not significantly affected by the status of dormancy; in other words, both dormant and activated blastocysts showed high levels of ROS. However, ROS production was higher in the dormant blastocysts of the long-term dormancy group than in those of the short-term group. The addition of wortmannin to dormant blastocysts in vitro and 3-MA injection in vivo significantly increased ROS production in the short-term dormant blastocysts. In the long-term dormant blastocysts, ROS levels were not significantly affected by the treatment of the autophagy inhibitor. Conclusion: During delayed implantation, heightened autophagy in dormant blastocysts may be operative as a potential mechanism to reduce oxidative stress. Further, ROS may be one of the potential causes of compromised developmental competence of long-term dormant blastocysts after implantation.

Ornamented Resting Spores of a Green Alga, Chlorella sp., Collected from the Stone Standing Buddha Statue at Jungwon Miruksazi in Korea

  • The growth of subaerial microalgae on historic buildings or various cultural properties causes discoloration and physico-chemical deterioration of the surfaces. We collected a subaerial chlorophyte, Chlorella sp., from the stone Standing Buddha statue at Jungwon Miruksazi, which is a national treasure of Korea, and found dormant, thickwalled spores with regular pentagonal ornamentation along with the vegetative Chlorella cells. The morphology of Chlorella resting spores was compared to that of the other green algal resting cells. The ornamented spores and smooth-walled vegetative cells revived in 2 weeks in a liquid freshwater medium and started reproduction by autospores. To our knowledge, the ability of Chlorella to form ornamented dormant spores in drought condition was not previously recorded. The ornamentation of spores would supplement taxonomic characteristics of this genus.

Genotype Difference of Plant Regeneration from Dormant Bud Culture in Colocasia esculenta Schott.

  • This study was conducted to obtain the basic breeding information of Colocasia esculenta Schott. Effect of supplemental plant growth regulators and genotype difference were investigated on dormant bud tissue for proliferation. The plant regeneration ratio, plant height and root length were the best upon mixed treatment of 0.8mg/L IAA and 2.0mg/L zeatin. Both leaf weight and root weight were heavy upon culture in a dark condition. The leaf and root weights were heaviest in 6Pie sucrose concentrations. In several collected area the heaviest one was Binnangxin and then in the order of Suwon, Wanju and Puan. Genotype differences of tuber diameter and tuber weight were found in Suwon. Tuber weight was found in the order of Suwon (862mg) >Wanju(723mg) >Puan(649mg) >Binnangxin (424mg).

Cryopreservation of Mulberry Germplasm Core Collection and Assessment of Genetic Stability through ISSR Markers

  • A simple and reliable cryo technique using desiccation and slow freezing of winter dormant buds was employed for 238 core collection of mulberry germplasm collected from diverse geographical regions and maintained under tropical conditions in the ex situ field gene bank to develop long-term biodiversity conservation for ensuring sustainable utilization of these valuable resources. Desiccation and freezing tolerance of bud grafts and excised shoot apices in the axillary buds of different Morus species under in vivo and in vitro condition indicated species-specific variation and most of the wild Morus species were found sensitive. In vitro regeneration and cryopreservation($-196^{\circ}C$) protocols using differentiated bud meristem like axillary winter dormant buds were worked out for a wide range of Morus species, land races, wild and cultivated varieties. Successful cryopreservation of mulberry winter dormant buds of different accessions belonging to M. indica, M. alba, M. latifolia, M. cathayana, M. laevigata, M. nigra, M. australis, M. bombycis, M. sinensis, M multicaulis and M. rotundiloba was achieved. Among wild species Morus tiliaefolia, and M. serrata showed moderate recovery after cryopreservation. Survival rates did not alter after three years of cryopreservation of different Morus species. ISSR markers were used to ascertain the genetic stability of cryopreserved mulberry, which showed no difference detected among the plantlets regenerated from frozen apices in comparison to the non-frozen material.

Physical Dormancy in Seeds of Chinese Milk Vetch (Astragalus sinicus L.) from Korea

  • Freshly harvested seed of Chinese milk vetch (Astragalus sinicus L.; CMV) was strongly dormant because of hardseedness. Seeds of freshly harvested germinated only 8% while clipping the seed coat completely overcome the innate dormancy, which indicates inhibition of germination of the seed is mainly due to seed coat (87%). The dormant (intact) hard seeds did not imbibe water whereas the non-dormant (clipped) seeds took up rapidly. In natural environment condition, the hard seed coat dormancy was broken only after 5 months after seed harvest. To break such a strong seed coat dormancy, the chemical and heat treatments were effective. Concentrated sulfuric acid was more effective than dry heat and hot water treatments. Hot water treatment improved germination but the germination percentage was less than 41%. Treatments increased germination due to its effect on the seed coat integrity. A scanning electron microscope reveled that disruption of seed coat layers and subsequent development of numerous crack in the hilum region of the seed and on the seed coat surface of concentrated sulfuric acid treatment and formation of cracks in the dry heat treatments, respectively, were observed in the seed coat surface, which served as water entry points.

Growth and Yield of Introduced Alfalfa(Medicago sativa L.) Cultivars in Korea (도입 Alfalfa(Medicago sativa L.) 품종의 생육 및 수량)

  • This study was conducted to select the best adapted alfalfa cultivars among 32 introduced alfalfa cultivars in the Korean environmental condition. Plant height and dry matter yield per cultivar were determined. On the point of plant height, seven high ranked cultivars among the 32 alfalfa cultivars in high order were Diamond> Milkmaker> Arrow>Chalienger>Sapphire > Dekalb brand 135> LS-1920. and seven low ranked cultivars in low order were Pioneer brand 5929> Maxidor> Dekalb brand 167> Pierce> Blazer> Raidor> Pioneer brand 532. Seven high ranked cultivars from 32 alfalfa cultivars on the dry matter yield in high order were Pike>LS-1920>Challenger>Salute>Diamond>Decathlon>Pioneer brand 581, and seven low ranked cultivars in low order were Maxidor>Pioneer brand 5929>Pioneer brand 532>Florida 77> Pierce> Magnum> Blazer. Dry matter yield per cultivar was also greater when alfalfa cultivar was appeared higher plant height compared with those which were lower plant height. Leading cultivars for plant growth and yield under the Korean environmental condition were Pike, LS-1920, Challenger, Salute, Diamond among the 32 alfalfa cultivars. All those cultivars except Challenger cultivar are Semi-dormant which can be well grown under moderate temperatures.

  • In order to produce young leaf of butterbur(Petasites japoflicus MAX.) in early spring, the planting date and relationship between abscisic acid(ABA) content and dormancy were investigated. Under open field condition, the dormancy of rootstock was initiated in the beginning of October, was the deepest in the middle of November and was completely broken in the end of December. When those periods were converted by the low accumulation hour below 5$^{\circ}C$, 900 hours were required approximately. This means that the rootstock needs for dormant breaking necessitates under the low temperature. In relationship between growing period and ABA content, the ABA in root-stock did not exist during maximum growing period, from April to September. This means that the ABA together with other substances in rootstock can be transferred to the shoot part with sprouting. While shoot part withered by decreacing the open field temperature since October, the ABA intiated to exist in rootstock. This means that the ABA in the shoot part can be transferred to the rootstock part. Therefore, it was concluded that the ABA which has been known that inhibiting growth and inducing dormancy was closely related with dormancy of rootstock.

  • [ $IP^2$ ] (IP-based IMT Network Platform) is a ubiquitous platform supporting mobility using two step If address IPha (IP host address) and IPra (IP routing address) - in a backbone network. MN (Mobile Node) in $IP^2$ maintains either Active or Dormant state, which is transferred to Active state through Paging process when communication is required. In this paper, we proposed a Paging method using proxy to resolve the problem of the conventional Paging method which transmits the Paging messages to all cells in LA (Location Area) resulting in the excessive use of network resources. Performanceevaluation of the proposed method using NS-2 showed that the usage of network resources becomes more efficient by reducing paging loads, especially under the condition of increased nodes.

  • Introduced were two biological investigations in which information biology played a significant role. In the first case independent findings in cancer research over a long period were united and organized by information biology and led to the outcome that was subject to a Nobel Prize. The outcome has revealed that the cause of human cancer is located in the genome in a dormant condition. The second case shows how to elucidate the function of an unknown DNA sequence or ORF in prokaryotes by a large - scale computer homology search and analyses. For the elucidation the International DNA Databases and a large - scale computer were two key factors.
