Functional Characterization and Regional Expression of Dopamine Transporter (도파민 수송체의 기능적 특성 및 발현에 관한 연구)

  • Brain dopamine systems play a central role in the control of movement, hormone release, and many complex behavior. The action of dopamine at its synapse is terminated predominately by high affinity reuptake into presynaptic terminals by dopamine transporter (DAT). The dopamine transporter(DAT) is membrane protein localized to dopamine-containing nerve terminals and closely related with cocaine abuse, Parkinsonism, and schizophrenia. In present study, the recombinant plasmid pRc/CMV-DAT, constructed by subcloning of a cDNA encoding a bovine DAT into eukaryotic expression vector pRc/CMV, was stably transfected into CV-1 cells(monkey kidney cell line). The DAT activities in the cell lines selected by Geneticin$^{R}$ were determined by measuring the uptake of $[^3H]$-dopamine. The transfected cell lines showed 30-50 fold higher activities than untransfected CV-1 cell line, and this result implies that DAT is well expressed and localized in transfected cells. The transfected cells accumulated $[^3H]$-dopamine in a dose-dependent manner with a $K_{m}$ of 991.6nM. Even though high doses of norepinephrine, epinephrine, serotonin, and choline neurotransmitters inhibited the uptake of $[^3H]$-dopamine, DAT in transfected cell line was proven to be much more specific to dopamine. The psychotropic drugs such as GBR12909, CFT, normifensine, clomipramine, desipramine, and imipramine inhibited significantly the dopamine uptake in tissue culture cells stably transfected with DAT cDNA. Radioactive in situ hybridization was done to map the cellular localization of DAT mRNA-containing cells in the adult rat central nervous system. The strong hybridization signals were detected only in the substantia nigra pars compacta and ventral tegmental area. The restricted anatomical localization of DAT mRNA-containing cells confirms the DAT as a presynaptic marker of dopamine-containing cells in the rat brain.

  • Biogenic monoamines are divided into three categories; catecholamines(dopamine, norepinephrine, and epinephrine), indoleamine(serotonin and melatonin) and histamine. Among them, serotonin has been intensively studied by many researchers with a broad spectrum of biomedical interests. A concise overview of serotonin-related topics such as biosynthetic pathway, receptor subtypes, and roles in reproduction will be provided. In particular, serotonergic efffect on the regulation of hypothalamus-pituitary-gonad hormonal axis and sexual behaviors will be emphasized. Though our Knowledge on the biological roles and its clinical applications are still limited, these topics are quite promising subjects which will be helpful for improving our 'quality of life' in near future.

The Effects of Self-administration of Methamphetamine on Serotonin Receptors Level in Rat Brain (메스암페타민 자발섭취가 흰쥐 뇌조직 중 세로토닌 수용체에 미치는 영향)

  • (+)-Methamphetamine (METH) is a psychostimulant, which has been the most popular abused drug in Korea. The rewarding mechanism in METH abuse has been reported to be mediated by dopaminergic system. Recently, it has been reported that dopamine releaser (phentermine) plays a dominant role in the discriminative stimulus effects of METH, whereas 5-HT releaser (fenfluramine) can strongly modify METH self-administration. The present study is designed to assess the behavioral changes and the changes of the serotonin receptors in the brains of rats administered repeated of self-administered METH. The repeated administration of 1.0 mg/kg/day METH for 12 days increased locomotor activities, and there was no difference between i.v. and i.p. treatment. Rats had actively acquired METH self-administration for 3 weeks at 0.1 or 0.2 mg/kg/injection. Whereas, it was taken few days to acquire sucrose pellet self-administration. The binding of [$^3$H]-8-hydroxy-DPAT (5-H $T_{1A}$ receptors) and [$^3$H]-5-carboxytryptamine (5-H $T_{1B}$ receptors) to brain sections was examined. Both passive administration and self-administration of METH did not change significantly the serotonin receptors levels in hippocampus, striatum and nucleus accumbens. These results suggest that serotonin receptors may not change in the acquisition period of METH self-administration, and we are trying to investigate the serotonin receptors levels of brain in rats maintained of METH self-administration.n.n.

  • The history of pediatric psychopharmacology is very short and the research on safety, efficacy and side effects is preliminary and long-term effect on growth and maturation is not well known yet. Clinical findings have shown that the responses to antidepressants, antipsychotics, CNS stimulants and steroids in children and adolescents might be different from adult populations. Based on these findings, this paper reviewed three issues, Firstly, in developmental pharmacokinetics. the author discussed the developmental factors affecting drug absorption, distribution, protein-binding, metabolism and excretion. Secondly, in developmental pharmacodynamics, developmental characteristics of dopamine, serotonin, norepinephrine receptors and their clinical implications were reviewed. Lastly, in pharamcogenetic part, the clinical utility of pharmacogenetics, pharmacokinetic aspects of pharmacogenetics, the pharmacodynamic aspects of pharmacogenetics, the association studies of dopamine-related alleles in neuropsychiatric disorders such as attention-deficit hyperactivity disorders or Tourette’s disorders, pharmacogenetic studies dopamine-related alleles and the pharmacogenetic studies of serotonin-related alleles. Based on these preliminary research, future pharmacogenetic applications in childhood and adolescent psychiatry were also discussed.

  • Objectives : This study aimed to evaluate the anti-stress effects of Ansymondam-tang and Gamiondam-tang on the contents of monoamines in the regional brain of mice forced cold swimming stress in $4^{\circ}C$ water. Methods : The experimental animals were forced cold swimming stress for 3 minutes, and administered of Ansymondam-tang(2.2mg/10g) and Gamiondam-tang(0.83mg/10g) extract for 7 days before stress. The monoamines contents were measured by HPLC method in various part(frontal cortex, hypothalamus, corpus striatum and hippocampus) of mice brain. Results: 1. In frontal cortex, the contents of norepinephrine were increased with statistical significantly in all of the administered group compared to control group. The contents of dopamine and serotonin were increased in Gamiondam-tang administered group has a statistical significance. 2. In hypothalamus, the contents of dopamine were increased with statistical significance in all of the administered group compared to control group. 3. In corpus striatum the contents of norepinephrine were decreased and the contents of dopamine were increased with statistical significance in all of the administered group compared to control group. The contents of serotonin were increased in Gamiondam-tang administered group has a statistical significance. 4. In hippocampus the contents of norepinephrine were decreased with statistical significance in Gamiondam-tang administered group compared to control group. The contents of dopamine were increased in Ansymondam-tang administered group has a statistical significance. Conclusion : This study reaches a conclusion that Ansymondam-tang and Gamiondam-tang has significant effects .on reducing and preventing stress in mice. Especially Gamiondam-tang is more effective in the statistical significance than Ansymondam-tang on the monoamines change in the mice brain.

  • An electrochemically-modified electrode [P(EDOP-SWNTs)/GCE] was prepared by electropolymerization of 3,4-ethylenedioxy pyrrole (EDOP) single-walled carbon nanotubes (SWNTs) on the surface of a glassy carbon electrode (GCE) and characterized by SEM, CV, and DPV. This modified electrode was employed as an electrochemical biosensor for the selective determination of serotonin concentrations at pH 7.4 and exhibited a typical enhanced effect on the current response of serotonin with a lower oxidation overpotential. The linear response was in the range of $1.0{\times}10^{-7}$ to $1.0{\times}10^{-5}$ M, with a correlation coefficient of 0.998 on the anodic current. The lower detection limit was calculated as 5.0 nM. Due to the relatively low currents and difference of potentials in the electrochemical responses of uric acid (UA), ascorbic acid (AA), and dopamine (DA), the modified electrode was a useful and effective sensing device for the selective and sensitive serotonin determination in the presence of UA, AA, and DA.

  • Premenstrual syndrome (PMS) is a common disorder affecting the emotional and physical health of women during certain periods of the menstrual cycle. Many researchers who have previously studied PMS have believed that PMS is associated with changes in sex hormones and serotonin levels at the beginning of the menstrual cycle. However, recent studies suggest that progesterone/estrogen imbalance and elevation of prolactin-induced by dopamine low-secretion play a crucial role in increasing PMS symptoms. Because of this, we have focused on mitigating PMS symptoms through the mechanism of prolactin secretion inhibition by dopamine receptor activation. The inhibition of prolactin secretion by 61-kinds of medicinal herb extracts was investigated in GH3 pituitary cells. Among them, Inula heleniun L. root extract (IHE) showed excellent prolactin secretion inhibitory effect. IHEs were prepared using 30, 50, and 70% ethanol. And the yield, cytotoxicity, dopamine receptor activity and inhibition of prolactin secretion of each extract were measured. Through a series of experiments, we found that prolactin secretion was significantly reduced (P<0.01) by the components present in IHE and that dopamine receptor regulation was possible (P<0.05). Considering yield and safety, we suggest the use of 30% ethanol IHE in the development of PMS symptom relief products.

  • Effect of the standardized ginseng extract(G115) on the central monoaminergic systems were investigated in comparison with that of halcperidol in rats. Immediately after sacrificed by decapitation, the strlata and frontal cortex were removerl. Concentations of the monoamines dopamine and serctorLin and their metabolites were deterinintd by HPLC-EC. G115 increased the concentration of 5-HIAA and DOPAC/UA ratio in striatum. However, dopaminrrgic neuronal activities were not affected by G115 that decreased the concentratio,Is of 5-HT and 5-HIAA in frontal cortex. G115 in combination with apomorphine significantly irlcreased the concentration of DA and S-HT but decreased the DO PAC/DA ratio and 5-HIAA/5-HT ratio only in frontal cortex. These results suggest that G115 like HPD inhibits the activity of nigrostriatal dopamine neuron in striatum. However, unlike HPD it activates central monoaminergic neuron activity in frontal cortex.

Effects of Gypenosides on Acute Stress in Mice

  • The effects of gypenosides (GPS) on electric footshock (EF)-induced acute stress in mice were investigated. Mice were treated orally with GPS (30-400 mg/kg) once a day for 5 days. After 2 days of GPS treatment, mice were exposed to EF stimuli (intensity, 2 mA; interval, 10 s; duration, 3 min) for acute stress for 3 days. Spontaneous locomotor activity was increased by acute EF stress, which was decreased by treatment with GPS (100 and 400 mg/kg). In addition, the increased levels of dopamine and serotonin by acute EF stress in the brain were reduced by treatment with GPS (100 and 400 mg/kg). The serum levels of corticosterone increased by acute EF stress were also reduced by GPS (100 and 400 mg/kg). These results suggest that GPS shows the ameliorating effects on acute EF stress by modulating the activity of dopaminergic and serotonergic neurons, and the serum levels of corticosterone. Clinical trials of GPS need to be conducted further so as to develop promising anti-stress agents.

Plasma Serotonin Level of Vietnam War Veterans with Post-Traumatic Stress Disorder and Symptom Severity (혈장 세로토닌과 외상후 스트레스 장애 : 월남전 참전 재향군인을 대상으로)

  • Objective : The objective of this study was to examine the relationship between plasma serotonin concentration and posttraumatic stress disorder (PTSD) symptoms in chronic PTSD patients who have been taking medication. Methods : Plasma serotonin level of 14 PTSD patients and a control group of 28 Vietnam War veterans was measured by HPLC (high performance liquid chromatography). The Combat Exposure Scale (CES), Mississippi Scale for Combat-Related Posttraumatic Stress Disorder (M-PTSD), Clinician Administered PTSD Scale (CAPS), Hamilton Rating Scale for Depression (HRSD), and Hamiltion Anxiety Scale (HAS) were used to evaluate PTSD symptom severity. Results : Serotonin level was significantly higher in the PTSD group than in the control group (p=0.036, p=0.006, respectively). M-PTSD (p<0.001), CAPS (p<0.001), HRSD (p<0.001), and HAS (p<0.001) scale scores were significantly higher in the PTSD group than in the control group; however, the CES score failed to show a significant improvement (p=0.964). There were no significant differences between plasma serotonin and PTSD symptoms. Conclusion : In chronic PTSD patients who have been taking medications, we can not predict treatment effect and symptom severity by measuring only plasma serotonin levels. PTSD is a complicated disorder which may likely be related to a variety of neurotransmitter systems. Therefore, further research which investigate relationships with norepinephrine, dopamine, and other neurotransmitters as well as serotonin is needed to improve the treatment of PTSD.
