국내 박물관의 체험활동프로그램 조사연구 (A Study on the Experience Program of Domestic Museums)

  • Museums have conducted the role of conserving, collecting, and displaying cultural property generally but main museum function transforms the role of social educational place accomplishing various instruction programs .recently through social perception's changing. Educational function of museum having experience program is more effect than before that. Museums need exhibition for preservation of property in regional community and history, as well as certainly require the concept to importing experience program. Through analysing experience program patterns and varieties in domestic museum. I will propose basic dater to supply preliminary and intermediate study.

도입 작약의 생육특성과 페오니플로린 함량 (Growth Characteristics and Paeoniflorin Content in Introduced Peony Varieties)

  • 도입한 작약 92계통중 12계통은 조기 고사하거나 생육이 불량하였고 80계통 중 65계통의 개화하였다. 조기개화계통으로는 9616, 9646, 9671등이 있었고 만기개화계통으로는 9632, 9638, 9658등이 있었다. 9651, 9655, 9662, 9677등은 절화용으로 좋았다. 9645, 9653은 semi-double형으로 독특하였고 9635는 백색과 황색의 겹꽃으로 아름다웠다. 9637, 9645, 9670은 키 큰 화단용으로 좋을 뿐 아니라 뿌리수량이 매우 높아 다수성 계통으로 평가되었다. 9613, 9628, 9635, 9642, 9655, 9680은 paeoniflorin 고함량 계통이었다. 이런 다수성 계통 및 Paeoniflorin 고함량 계통은 약용작약품종의 육종자원으로도 이용될 수 있을 것이며 9637, 9645, 9676, 9677등은 수량과 약효성분이 적절하여 관상용과 약용을 겸한 겸용품종으로 이용 가능성을 보였다.

엿기름의 효소활성과 관련한 보리의 품질특성 (Quality Characteristics of Barley Varieties Related to Enzymatic Activity in Malt)

  • 엿기름의 품질 요소인 당화력(DP)은 보리품종에 따라 큰 차이를 나타내 $139{\sim}220^{\circ}L$의 범위에 있었다. 당화력은 ${\alpha}-amylase$ 보다 ${\beta}-amylase$와 높은 상관관계가 있어 엿기름의 ${\beta}-amylase$ 활성이 매우 중요한 인자였으며, 엿기름 첨가에 따른 amylograph 전분기질 점도 감소는 ${\alpha}-amylase$와 관련이 있는 것으로 확인되었다. 보리품종들의 품질요소들을 분석하여 엿기름 당화력과의 상관관계를 조사한 결과 엿기름의 당화력은 원맥의 품질인자와 상관관계가 별로 높지 않았으나 중량이 낮은 품종이나 덜 풍만한 품종에서 당화력이 높은 경향을 보여주었다. 보리원맥이 지니고 있는 ${\beta}-amylase$ 활성은 엿기름의 당화력과 상관관계가 있는 것으로 평가되었으며 엿기름의 당화력을 예측할 수 있는 잠재적인 당화력으로서 엿기름 제조에 매우 유용한 품질인자인 것으로 판단되었다.

내외국산 소맥의 주요품종별 제분성 비교연구 (Studies on the milling quality of major varieties of domestic and foreign produced wheat)

  • 한국산 소맥 5품종과 미국산소맥 3품종을 밀과 밀가루의 성분ㆍ회분ㆍ조단백질함량 및 제분율을 조사하였는데 그 성적을 요약하면 다음과 같다. 1. 수분은 한국산밀이 미국산밀보다 품종과 시료에 따른 차가 컸다. 2. 밀의 회분함량이 미국산 밀에서는 경질과 연질에 따라 차가 현저했는데 한국산밀에서는 그렇지 않았다. 3. 제분율은 한국밀중에서는 육성 003 한 품종만이 비교적 높고 기타는 미국밀에 비하여 현저히 낮았다. 4. 밀가루의 회분함량은 품종간의 차를 인정할수 없었다. 5. 밀가루의 조단백질함량은 한국밀보다 미국밀중에 현저히 높은 것이 있었다. 6. 밀의 조단백질함량과 밀가루의 조단백질함량과의 상관은 한국밀에서 보다 미국밀에서 현저하게 높았다.

고부가가치 맞춤형 상추품종 개발을 통한 국내 상추유통 제고 전략 (Strategies to Increase Domestic Lettuce Circulations through Improving Valuable End-User Traits)

  • Purpose - Lettuce (Lactuca sativ L.) is one of the economically important vegetable crops, which worldwide market value is over 100 billion U.S. dollar. In Korea, about 89.7 kilo ton of lettuce was produced in 3400ha in 2016, recoded as No. 1 vegetable crop in domestic green house production. However, recently, domestic lettuce production and cultivation areas are all getting decreased. Thus, novel approaches are needed to be implemented to revive the production. Research design, data and methodology - In this review paper, we first prioritized the end-user traits which are imperative to positively stimulate the domestic lettuce market and discussed relevant genomics strategies. Especially, we assessed a possibility whether school meal program would be a potential niche market. Results - The genomics technologies, which become widely applied in the crop biotechnology since 2008 when next generation sequencing method was developed, may be a good solution in the crop improvement, efficiently gathering valuable information of agriculturally useful traits. Significantly, in lettuce, the high quality whole genome sequence, based on Lactuca sativa cv. Salinas, is publically available and this genomics platform, thus, would be implemented in lettuce breeding program to innovate relevant end-user traits both for the farmers and customers, including the disease resistance to the Fusarium wilt, productivity under hot weather conditions, various nutritional qualities and so forth. These improvements will boost domestic lettuce industries in the near future. Conclusions - Due to the nutritional distinctions comparing to the western style lettuces, domestic leaf lettuces could be one of the important vegetables in the school meal programs. To make it happen, we would better devise diverse recipes to make a salad with it, instead of only using as a wrap vegetable. Meanwhile, novel lettuce varieties need to be developed, which are favorable to the students and also easy to be handled with while processing. Overall, to achieve international competence in the lettuce industries, we need to create elite lettuce varieties that satisfies domestic farmers as well as customers, suitable to various niche markets, such as school meal program. Thus, efficient breeding programs using genomics approaches should be established in advance and careful monitoring on the preference of the related customers for a niche market be continued persistently.

  • 1973년(年) 7월(月) 서울특별시내(特別市內) Smoke통닭 공장(工場)에서 얻은 이병닭 폐(肺)에서 Aspergilli를 분리(分離)할 수 있었고 또 C-source 및 N-source의 배양조건(培養條件)을 실험(實驗)한 결과(結果) 다음과 같은 결론(結論)을 얻었다. 1. Aspergillus fumigatus group에 속(屬)한 1 strain을 분리(分離) 하였다. 2. 분리용(分離用) 배지(培地)로서는 Czapek agar에 누에, 메주를 4g씩 첨가(添加)한 것이 과거 Austuwick(1962)가 보고한 2% malt extract agar medium보다 양호하였다. 3. 배양(培養)에 있어서 C-source는 종류(種類)와 양(量)에 따라 생장율과 sporulation의 차(差)가 많았고 20%에서 생장율이 떨어겼다. N-source에서는 종류(種類)에 따라 생장율의 차가 생기나 양(量)에 따라서는 생장율의 차(差)가 거의없고 다만 mycelium과 sporulation에서 차(差)가 생겼다.

우리나라 양곡가공공장의 현황분석 (The Profile of Milling Plants in Korea)

  • This study was conducted to obtain a basic information necessary to assess present rice milling technology in Korea The profiles for milling plants was analyzed by survey work.For the private custom-work mills, which process about 80 percent of domestic rice consumption ,their actual milling test for the identical samples as used for filed mills was conducted. Two rice varieties Japonica and Tongil-type were associated with the experiments. The results are summarized as follows: 1. Analyses for private custom-work mills showed their general aspects as; about 91 percent of the mills belonged to an individual owner ship ; more than 93 percent of the mills was established earlier than 1950 ; about 80 percent of the mills was powered with electric motor; mills having less than two employees were about 75 percent; about 45 percent of the mills provided for warehouse in storing customers cereal grains. 2. The polishers installed in 1,255mills within the surveyed area (7 counties) have been supplied by 44 different domestic manufacturers ;in but about 60 percent of which was supplied by 6major manufacturers. The polishers could be classified into two categories in terms of principles of their polishing actions ;jet-pearler and friction types. About 51 percent of the mills was equipped with the former which has been recognized as giving greater milling recovery than the friction types. 3. Reason for owners of private mills to supplement new machines was due mainly to pgrading their mills to meet the requirements that established by the Government. However, about 60 percent of the mill owners intended to replace with new pearler by their own needs to meet with new high yielding varieties. 4. Processing systems of each previate rice mills surveyed could be classified into three categories, depending upon whether the systems posessed such components as precleaner and paddy separator or not. Only 36.7 percent of mills was installed with both precleanr ad paddy seperrator, 5.0 percent of mills did have neither percleaner nor paddy seperator, and rest of them equipped only one of the two. Hence,it is needed for about 63% of rice miils to be supplemented with these basic facilities to meet with the requirements for the standaized system. 5. Actual milling capacity measured at each field rice mills was shown a wide variation, having range from about 190 to 1,210 kg/hr. The percentages of mills classified according to daily milling capacity based on this hourly capacity were 24.3% for the capacity less than 3 M/T a day; 20.0% for 3-4 M/T; 15.6% for 4-5 M/T; 6.7% for 5-6 M/T; 22.3% for 6-7 M/T; and 11.0% for more than 7 M/T a day. 6. Actual amount of rice processed was about 310 M/T a year in average. About 42% of total milled rice was processed during October to Decembear, which formed a peak demand period for rice mills. The amount of rice milled during January to May was relatively small, but it had still a large amount compared to that during June to September. 7. Utilization rate of milling facility, i. e., percentage of the actual amount of milled rice to the capacity of rice mills, was about 18% on the year round average, about 41% in the peak demand season, and about 10% during June to September. Average number of operating days for mills surveyed was about 250 days a year, and about 21 days a month. 8. Moisture contents of paddy at the time of field mill tests were ranged 14.5% to 19.5% for both Japonica and Tong-i] varieties, majority of paddy grains having moisture level much higher than 1530. To aviod potential reduction of milling recovery while milling and deterioration of milled rice while storage due to these high grain mJisture contents, it may be very important for farmers holding rice to dry by an artificial drying method. 9. Milling recovery of JapJnica varieties in rice mills was 75.0% in average and it was widely ranged from 69.0% to 78.0 % according to mills. Potential increase in milJing recovery of Japonica variety with improvement of mill facilities was estimated to about 1.9%. On the other hand, milling recovery of Tong-il varieties in the field mill tests was 69.8% in average and it ranged from 62% to 77 %, which is much wider than that of Japonica varieties. It is noticed that the average milling recovery of Tong-il variety of 69.8% was much less than that of the Japonica-type. It was estimated th3.t up to about 5.0% of milling recovery for Tong-il variety could be improved by improving the present lo'.ver graded milling technology. 10. Head rice recoveries, as a factor of representing the quality of commercial goods, of Japonica and Tong-il varieties were 65.9% and 53.8% in average, and they were widely ranged from 52% to 73% and from 44% to 65% , respectively. It was assessed that head rice recovery of Japonica varieties can be improved up 3.3% and that of Tong-il varieties by 7.0% by improving mill components and systems.

흰색 팽이버섯 신품종 '백운' 육성 (Breeding a new variety of the white winter mushroom, Flammulina velutipes 'Baekwoon')

  • 본 연구는 농촌진흥청에서 육성한 백승과 국외 수집자원(KMCC02260)을 모본으로 하여 단핵균주를 분리 후 mon-mon교잡을 통해 '백운' 품종을 육성하였다. 균사배양 특성은 25℃에서 두 품종의 균사생장이 가장 좋았으며 15, 25℃에서 백운의 균사생장 속도가 대조품종보다 더 빨랐으며 배지별 시험에서도 모든 배지에서 백운의 균사생장이 우수하였다. 톱밥 병에 접종 후 균사 배양기간은 백운이 2일, 초발이일수도 1일 더 빨라 전체적인 생육기간을 단축시켰으며 수량은 228.0±10.9 g/병(850 ml)으로 대조품종 대비 8.3% 증수하였다. 백운의 형태적인 특성은 갓이 크고 대가 굵으며 갓이 순백색이었다. 대치배양과 유전적 다형성 검정 결과, 백운이 두 모본과 대조품종과 유전적으로 다르다는 것을 확인하였다.

국내산 품종의 녹두껍질 추출물의 산화방지활성에 미치는 추출방법의 영향 (The Antioxidant Activities of the Korean Variety Mung Bean Hull Extracts as Dependent on the Different Extraction Methods)

  • In comparison of the antioxidant activities by the different extraction methods of the domestic mung bean varieties, 'Geumsung', 'Dahyeon', 'Sohyeon', and 'Eohul', were soaked and dehulled, with the extracts obtained by using 80% ethanol at room temperature or heated. The mung bean hull was freeze-dried, ground, and passed through 100 mesh sieve. The moisture content of the hull powder ranged from 1.70 to 3.15%. The total dietary fiber content ranged from 84.42 to 88.47%, with the Sohyeon hull showing the highest value. The L value of Geumsung hull (62.93) was the highest, and the a value of Sohyeon hull (-1.69) was the lowest (p<0.05). The color difference was the highest in Dahyeon hull (42.52) and lowest in Geumsung hull (38.52). The Eohul hull's chlorophyll a and b contents were the highest with the 80% ethanol extract by heating (10.55 and 6.12 ${\mu}g/mL$, respectively), but the Sohyeon hull showed the highest in 80% ethanol extract at room temperature (7.63 and 3.93 ${\mu}g/mL$, respectively). The total phenolic and total flavonoid contents of the ethanol extracts were significantly different between the varieties regardless of the extraction temperature. The hull extract from the heating extraction was higher in Eohul than in other varieties, and was the lowest in the Dahyeon variety. The extract from Sohyeon hull at room temperature showed the highest contents of total phenolic and flavonoid contents. In the mung bean hull extract by heating, DPPH and ABTs radical scavenging activity ranged from 13.49-16.75% and 22.93-39.42% respectively and those at room temperature ranged from 39.86-41.88% and 49.71-70.92%. The reducing powers of extracts by heating ranged from 7.29 to 9.36 mg BHT/g hull, and at room temperature, they ranged from 19.90 to 20.97 mg BHT/g hull. Our results indicate that different extraction methods influenced antioxidant activities in the domestic mung bean varieties.

몇 가지 벼 품종으로부터 분리한 4-HPPD저해 제초제에 감수성인 HIS1 유전자 특성 (Characteristics of Sensitive HIS1 Genes to the 4-HPPD Inhibiting Rice Herbicides Isolated from Several Rice Cultivars)

  • 4-HPPD저해 제초제에 의한 주요 벼품종의 약해반응과 약제반응에 관여하는 유전자(HIS1) 특성을 구명하기 위해 본시험을 수행하였다. Benzobicyclon 액상수화제 처리에 Japonica 품종인 상주벼에는 백화증상의 약해증상이 발생하지 않았으며, Japonica 품종임에도 불구하고 삼백벼에서는 기준량에서는 2-3, 배량에서는 3-4의 백화증상의 약해가 발생하였으며, 산들진미, 금영 품종에서는 3-4 및 4-5의 약해증상을 보였다. 또한 Indica 품종인 IR-8 품종에서는 기준량에서 3~5, 배량에서는 4~6의 가증 심한 약해증상을 보였다. 4-HPPD저해 제초제애 대해서 약제반응에 관여하는 HIS1 유전자 보유여부를 확인하기 위해 일반계 품종인 상주벼와, 삼백벼, 산들진미, 금영과 통일형 품종인 IR-8품종으로부터 저항성 표적유전자와 동일 HIS1 유전자를 분리하였다. 각 품종별 HIS1유전자를 PCR등을 통해 염기서열을 확인한 결과 HIS1 유전자는 생태형에 상관없이 시험한 5품종 모두에서 HIS1유전자를 보유하고 있는 것으로 확인되어, 본 유전자로 HPPD 약해 발생 유무를 검증할 수 없음이 확인되었다.
