This study was conducted to establish a Habitat Suitability index (HSI) based on literature research and field surveys on ecology and habitat of 'Hynobius leechii'. And this study will be used as basic data for qualitative evaluation of habitat environment. The survey sites were divided into natural habitats close to the prototype habitat and artificial restoration areas where Hynobius leechii was monitored. So the types of habitats were diversified. Hynobius leechii is a vulnerable species to climate change because it is affected by the microhabitat and has low mobility. HSI variables of Hynobius leechii were extracted through domestic and overseas literature, and standards were extracted from literature research and field survey. The standards were presented as a value of the physical allowable category in consideration of realization. To verify the study, an in-depth consultation was conducted by amphibians experts. HSI variables of Hynobius leechii were included 9 variables such as Overstory canopy cover(%), Understory cover(%), Water-pH, Soil-pH, Soil relative humidity(%), Leaf litter depth(cm), Rock substrates (%), Type of Coarse woody, Distance from Street or Pollutant(m).

For this survey, PCR (polymerase chain reaction) testing was conducted using 14 species-specific primers to monitor the labeling of allergy-causing substances in various foods. Sixty samples from stationary stores near elementary schools and imported confectionery shops were tested, including snacks, candies, and chocolate. Allergens of milk, wheat, eggs, tomatoes, almonds and peanuts were detected in 30 cases (50.0%). In addition, many products were detected as either containing unlabeled substances or not showing allergen-related information and labeling in Korean. In order to ensure that consumers are able to purchase products safely and securely, a system for thorough guidance and monitoring of allergen-related labeling by domestic manufacturing and processing companies and import-related companies is required.

This research was conducted to investigate the genetic diversity and select useful accession with agronomic characteristics of garlic (Allium sativum L.). germplasms at Garlic Research Institute in Chungbuk Agricultural Research and Extension Service. Morphological diversity and relationships among 160 germplasms collected from 26 countries were assessed by methods of clustering and principal component analysis. Among 11 types of leaves and bulbs characteristics, emergence days of leaf showed the highest variation with coefficient of variation of 84.8%, and the bulb weight and the number of scales showed higher variability with 24.3%. Correlation analysis based on 11 quantitative traits showed that bulb weight and bulb length have very high positive correlation with bulb quantity. Plant height, leaf length, and number of leaves showed positive correlation with bulb weight as collections with better performance in growth produced large bulb with higher quality. The cluster analysis based on 5 principal components generated 6 clusters with an average distance of 1.6 among clusters. Domestic genetic resources were the largest with 36 species (22.5%) in group II.
Arsenic (As) oxidation followed by precipitation from a high-As(III)-containing leaching solution derived from a sulfidic ore was investigated in this study to remove aqueous As from the solution using activated carbon (AC) with air injection as an oxidant. To obtain the initial leaching solution, a domestic sulfidic ore was leached in a sulfuric acid solution at pH 1 and 50℃ for 95 h, and approximately 7 g/L of Fe and 3 g/L of As were leached out. To determine the effect of the oxidative reaction utilizing AC with air injection, the leaching solution was tested under the following five oxidative conditions at an initial pH of 1 and 90℃ for 72 h: air-only injection; air injection with 1, 5, and 10 w/v% of AC addition; and H2O2 addition. The tests in the presence of both air and AC revealed that the oxidation kinetics and As removal were improved by the reaction between the metallic species and the surface group formed on the AC surface. In addition, the greater the amount of AC added, the better was the reaction efficiency, removing 93-94% of As with more than 5 w/v% of AC addition. Finally, X-ray diffraction analysis confirmed that the precipitate formed from the oxidative reaction was scorodite (FeAsO4·2H2O).

Objective: Efficient gene editing technology is critical for successful knock-in in domestic animals. RAD51 recombinase (RAD51) gene plays an important role in strand invasion during homologous recombination (HR) in mammals, and is regulated by checkpoint kinase 1 (CHK1) and CHK2 genes, which are upstream elements of RAD51 recombinase (RAD51). In addition, mismatch repair (MMR) system is inextricably linked to HR-related pathways and regulates HR via heteroduplex rejection. Thus, the aim of this study was to investigate whether clustered regularly interspaced short palindromic repeats/CRISPR-associated 9 (CRISPR/Cas9)-mediated knock-in efficiency of human lactoferrin (hLF) knock-in vector in the bovine β-casein gene locus can be increased by suppressing DNA MMR-related genes (MSH2, MSH3, MSH6, MLH1, and PMS2) and overexpressing DNA double-strand break (DSB) repair-related genes (RAD51, CHK1, CHK2). Methods: Bovine mammary epithelial (MAC-T) cells were transfected with a knock-in vector, RAD51, CHK1, or CHK2 overexpression vector and CRISPR/sgRNA expression vector to target the bovine β-casein gene locus, followed by treatment of the cells with CdCl2 for 24 hours. After 3 days of CdCl2 treatment, the knock-in efficiency was confirmed by polymerase chain reaction (PCR). The mRNA expression levels of DNA MMR-related and DNA DSB repair-related genes were assessed by quantitative real-time PCR (RT-qPCR). Results: Treatment with CdCl2 decreased the mRNA expression of RAD51 and MMRrelated genes but did not increase the knock-in efficiency in MAC-T cells. Also, the overexpression of DNA DSB repair-related genes in MAC-T cells did not significantly affect the mRNA expression of MMR-related genes and failed to increase the knock-in efficiency. Conclusion: Treatment with CdCl2 inhibited the mRNA levels of RAD51 and DNA MMR-related genes in MAC-T cells. However, the function of MMR pathway in relation to HR may differ in various cell types or species.
Animals exhibit certain behaviors and movement patterns as they react to their internal needs, external stimuli, and surrounding environments. They have a bounded range in which they mostly spend their time, and it is referred to as a home range. Based on the fact that the home range is a critical area for the survival and preservation of species, there has been a growing body of research on developing more precise home range estimation methods to use the estimated ranges as a ground for establishing an effective conservation policy since the early 1940s. Recent rapid advancements in telemetry technology that resulted in the presence of autocorrelation between locations with short time intervals revealed the limitations of the existing estimators. Many novel estimators have been developed to compensate for it by incorporating autocorrelation in calculating home ranges. However, studies on the animal home range are still in their early stage in Korea, and newly developed methodologies have not yet been adopted. Therefore, this study aims to introduce the foreign home range estimation methods and foster domestic research activities on home ranges. Firstly, we compared and contemplated seven estimators by categorizing them into geometrical and statistical methodologies and then divided them into estimators that assume independent observations and those that consider autocorrelation in each category. After that, the home ranges of black-tailed gulls (Larus crassirostris) were calculated using GPS tracking data for the month of June and derived home range estimators by applying the methodology introduced in this study. We analyzed and compared the results to discuss the strengths and weaknesses of each method. Lastly, we proposed a guideline that can help researchers choose an appropriate estimator for home range calculation based on the animal location data characteristics and analysis purpose.
Two kinds of chemicals (RNF-3 and RNF-4) with high anti-mold effectiveness against stored fungi causing fungal discoloration was selected from among the six kinds of fluoride-based chemicals in the preceding study. In this study, the anti-mold test using 16 species fungi isolated from the softwood lumbers which were fungal discolored and field test in the sawmill was carried out to prove the feasibility for practical using of selected chemicals.For the isolated fungi, the RNF-3 consist of F and Cu showed high mycelial growth control in the PDA medium and fungicidal effectiveness in the japanese red pine (Pinus densiflora), korean pine (Pinus koraiensis) and radiata pine (Pinus radiata) when the concentration was 2% or more. However the RNF-4 consist of F only was not effective compared with RNF-3 because of mycelial growth in the PDA medium and wood treated with 2% or more chemical solution. The RNF-3 also showed a strong anti-mold effectiveness because there was no fungal discoloration for the radiata pine boards treated by 2% and 10 min. soaking in the field test. These results mean that RNF-3 can be used as domestic anti-stain chemicals for prevention of fungal discoloration of the softwood lumber

The black cutworm, Agrotis ipsilon (Hufnagel), is an important crop pest worldwide that feeds more than 80 plant species including cabbage, potato, maize, wheat and bean, and this moth is a typical pest attacking underground parts of crops. It has been known in farm booklets that the larvae of A. ipsilon overwinter in the soil in Korea, but no definitive data exist yet. This study was conducted to evaluate that the specific appearance time of A. ipsilon observed actually in the field could be explained when we assumed that this pest overwinters in a form of larvae or pupae. Degree day-based phenology models were applied for tracking forward or backward to find the predicted developmental stage which developed at a specific stage found in the field. As a result of the analysis, it was confirmed that an initial population could be established in a group that does not overwinter as larvae or pupae in Korea. In other words, the appearance of adults in early March to April could not be explained by the presence of domestic overwintering populations. Populations that overwinter as larvae or pupae in Korea were able to emerge as adults in June to July at the earliest. Therefore, the group of adults appearing in early spring is highly likely to be a population that migrated from outside Korea. Taken together, it was estimated that the colony of A. ipsilon in Korea would be formed by a mixture of a migrant population through long-distance migration and a overwintering population.
Recently, sporadic cases of human infection by genetic reassortants of H7Nx influenza A viruses have been reported; such viruses have also been continuously isolated from avian species. In this study, A/wild bird/South Korea/sw-anu/2023, a novel reassortant of the H7N1 avian influenza virus, was analyzed using full-genome sequencing and molecular characterization. Phylogenetic analysis showed that A/wild bird/South Korea/sw-anu/2023 belonged to the Eurasian lineage of H7Nx viruses. The polymerase basic (PB)2, PB1, polymerase acidic (PA), and nucleoprotein (NP) genes of these viruses were found to be closely related to those of avian influenza viruses isolated from wild birds, while the hemagglutinin (HA), neuraminidase (NA), matrix (M), and nonstructural (NS) genes were similar to those of avian influenza viruses isolated from domestic ducks. In addition, A/wild bird/South Korea/sw-anu/2023 also had a high binding preference for avian-specific glycans in the solid-phase direct binding assay. These results suggest the presence of a new generation of H7N1 avian influenza viruses in wild birds and highlight the reassortment of avian influenza viruses found along the East Asian-Australasian flyway. Overall, H7Nx viruses circulate worldwide, and mutated H7N1 avian viruses may infect humans, which emphasizes the requirement for continued surveillance of the H7N1 avian influenza virus in wild birds and poultry.
Purpose - This study empirically analyzes the consumer value of risk management associated with illegal, unreported, and unregulated (IUU) fishing of fishery products imported to Korea. The global regulatory paradigm for IUU fishing has shifted from production-centered to market-centered. As a result, changes in the international fisheries trade environment emphasizing "transparency" and "legality" of the production process are accelerating. Therefore, changes in the management systems of fishery products entering the country are also needed. Accordingly, this study estimated the consumer value for risk management of IUU fishing, targeting major fish species imported to Korea, and derived the feasibility of introducing related policies. Design/methodology - This study used the choice experiment as an analysis model to estimate consumers' willingness to pay for the "possibility to check for IUU fishing." The choice experiment assumes that the value of a good or service is composed of separable attributes and that the sum of the part-worth of these individual attributes becomes the total value. In this study, respondents were presented with profiles comprising three attributes (country of origin, price, and possibility of checking IUU fishing) and the levels of frozen poulp squid, the subject of the analysis. The participants were asked to select their preferred profile. The marginal willingness to pay for each attribute was derived from the results of the respondents' choices using conditional logit model estimates. Findings - There is a marked difference in utility based on the preference of the country of origin of fishery products among consumers. In addition, the utility of fishery products that have undergone IUU fishing verification was observed to be higher, with the utility marked to be higher for lower prices. Originality/value - Estimating the policy value of the risk management in IUU fishing of imported fisheries products in this study is a novel attempt that has never been conducted before. Several studies have been conducted to assess the risk of IUU fishing associated with the import of fishery products internationally. However, such studies are yet to be conducted in Korea. Instead, policies and studies have focused on issues related to complying with trading partners' legal and transparent standards for exporting fishery products. This study should be the beginning of more in-depth empirical and theoretical explorations to establish order in the domestic seafood market and respond to changes in international regulations on IUU fishing.
