Dog park planning items and needs assessment in consideration of Korean pet culture (애견문화를 고려한 '도그파크' 계획항목과 요구도)

  • Rapid urbanization and social development have brought about changes in family structure. In urbanized areas, recently the number of dogs that are a majority of companion animals has increased significantly in Korea. Consequently, a lot of friction between the general public who are not fond of dogs and dog owners in public places such as parks has been happening. A lot of countries that are taking a lead role in animal welfare have built dog parks that the general public and dogs can enjoy together. This research starts with the realization that the concept of 'dog park' needs to be introduced in Korea as more and more Koreans own dogs and other pets. This research aims to provide guidance for dog park planning by drawing planning items and analyzing the needs of each item. Total 56 planning items including dog zone, dog playground, and dog training facility, are drawn in this research. There are differences in the needs according to sex, age and the amount of time they spent with their dogs, however overall there is the highest needs in separated area for dogs in parks.

  • Korea's pet-related society has been fast growing without the base and preparations of socio-cultural animal welfare. For the most part, Korea's current public parks, in particular, have a space structure in which a lot of friction between the general public who are not fond of dogs and dog owners in public places has been happening unavoidably. A lot of countries that are taking a lead role in animal welfare have built dog parte that the general public and dogs can enjoy together. This research starts with the realization that the concept of 'dog park' needs to be introduced in Korea as more and more Koreans own dogs and other pets. This research aims to provide guidance for dog park planning by drawing a dog park planning model and to promote basics for co-existence of dogs and people in a park and set in place animal welfare early and raise the level of awareness. Total 7 types of dog put 56 planning items including dog zone, dog playground, and agility, and a selective matrix by which it is possible to select essential recommendatory optional items by each type of dog park were drawn in this research. Overall there were the highest needs in separated area for dogs in park. A dog park planning model is designed as follows. 1) To select a dog park type that was introduced. 2) To set up facilities applicable in common and planning items desired in advance by each type. 3) To zone dog park areas by the priority order of planning items that were introduced and to position necessary facilities. 4) To plan program that should be were introduced. A dog park was expected to take some positive roles such as providing opportunities to socialize dogs, activating social communities, more effective park management by participating local community members and so on.

A Study on Perception and Needs of Urban Park Users on Off-Leash Recreation Area (도시 공원이용자들의 애견전용공간 계획에 대한 인식 및 요구도 연구)

  • The recent increase in the number of dogs in high-density urban environments causes conflicts between dog owners and non-dog owners and the access of dogs to urban parks has been controversial among park users. A lack of exercise and socialization, however, can cause canine behavior problems such as aggression and hyperactivity which are potential dangers to people and there has increased demand for a place where people and their dogs can interact and exercise together. In many developed countries, off-leash recreation areas (OLRA) are considered as safe and effective places that dogs can exercise, play, and socialize with other dogs while reducing conflicts between dog owners and other park users. This study aimed to review the issue of dogs' access to public parks and to examine the perception of park users on OLRA to provide insights and guidelines in establishing OLRA in Korea. The survey respondents, even non-dog owners, perceived that it is necessary to provide a separate space for dogs from other park users by providing OLRAs in urban parks and showed the willingness to use. Most of dog owners consider their dogs as companions or family members and feel the lack of places to exercise their dogs. As local government plays a major role in urban animal management which emerge as a social issue at present, more active interventions of municipalities are necessary to support various needs and activities of park users. An experimental OLRAs based on community participations would be a meaningful starting point to effective urban animal management.

Correction of the Proximal Intertarsal Instability in a Dog

  • A 22-month-old female mongrel dog weighing 4.8 kg was referred with chief complaint of bite wounds and lameness. The patient showed non-weight bearing lameness on left hindlimb and lateral instability of intertarsal joint. On radiograph, proximal intertarsal luxation between calcaneus and fourth tarsal bone was diagnosed. To imitate the long collateral and calcaneoquartal ligament, bone tunnels were drilled in the distal calcaneus and proximal fifth metatarsal bone. The figure-eight suture was placed with 0.5 mm cerclage wire through the predrilled holes. At 2 weeks after surgery, the lameness of hindlimb was still present. However, the patient had weight bearing lameness for 4 weeks after operation and was able to walk and run without lameness at 5 weeks after surgery. This case report explains the successful correction of intertarsal luxation with a single figure of eight wire suture in case of luxation to the intertarsal joint.

Simultaneous Determination of Doxifluridine and 5-FU in Liver and Intestine Tissue Using LC/MS/MS (LC/MS/MS를 이용한 원숭이 및 비글견의 간 및 장관 조직에서의 Doxifluridine과 대사체 5-FU 동시분석법 개발)

  • A liquid chromatographic method with tandom spectrometric detection (LC/MS/MS) for the simultaneous determination of doxifluridine and its active metabolite, 5-fluorouracil (5-FU) was developed over the concentration range of $5{\sim}2000$ ng/ml, respectively. Doxifluridine, 5-FU and internal standard, 5-chlorouracil (5-CU), were extracted from liver and intestine tissue via protein precipitation. Acetonitrile was used as the extraction solvent and the supernatant was evaporated and reconstructed in mobile phase. Optimum chromatographic separation was achieved on a Agilent Zorbax $C_{18}$ ($100\;mm{\times}2.1\;mm$, $3.5\;{\mu}m$) column with mobile phase run in isocratic with methanol : water (20 : 80, v/v). The flow rate was 0.2 ml/min with total cycle time of 5 min. The lower limit of quantification was validated at 5.0 ng/ml of liver and intestine tissue, for both doxifluridine and 5-FU, respectively. The intra-day and inter-day precision and accuracy of quality control (QC) samples were <11% coefficient of variation and <7% relative error from theoretical concentration for both analytes. In addition, the special designed stability study was performed, because the metabolism of doxifluridine occurs spontaneously even in ice bath for monkey liver. The stability of doxifluridine in liver and intestine of monkey and beagle dog was compared. It was found that bioanalytical validation could not be performed for the monkey liver; however, beagle dog's liver has relatively low speed of metabolism compared to monkey liver and instead of monkey liver, beagle dog's liver could be used for the validation. Bioanalytical validation could be performed in monkey intestine. Eventually, this developed method for liver and intestine will be useful in support of the toxicokinetic and pharmacokinetic studies of doxifluridine and 5-FU.

A Case of Osteotomy of the Distal Radius and Ulna with Panarthrodesis of Carpus for Reform of (성장판 조기페쇄에 따른 요척골 외전과 완관절 아탈구 교정들 위한 요척골 절단술 및 광범위 완관절 고술의 일례)

  • A 23.5 kg179, 8 months olds non-spaycd female Rottweiler dog was submitted to the veterinary teaching animal hospitals Chonbuk: national university, for chronic bitten trauma on right foreleg with pain, lameness, and pronation. The patient fought with a neighboring dog about five months ago and had mild anorexia, depression, bolt normal walking at that time. CBC and blood chemical examination were in normal range. Physical examination resulted in the right foreleg with edema and interval rotation of carpal joint. Radiograph of the right carpus showed varus and subluxation. We finally diagnosed the patient as the growth deformity of distal radius and ulna caused by medial premature closure of distal radial physis. Osteotomy was performed fur reforming of the varus of the distal radius and ulna with 6-hole straight plate and six 3.5 mm screws. Panarthrodcsis of carpus was preformed for correcting subluxation using 6-hole T-shaped plate and four 3.5 mm screws with cortical onlay autograft and strengthened by two 3.5 mm screws and tension wire band in lateral aspect of the carpus. Follow-up radiographs after 16 weeds of the surgery showed complete coaptation of osteotomy sire of the radius and uIna. After 6 months of the procedurc, talc plate, screws and the wire were removed except 7-shaped plate and four screws fur arthrodesis. The patient was seen in normal forelimb and could walk and run without lameness after 7 months of the procedure.

Early rehabilitation treatment helpful in a case of pectus excavatum of a dog (개에서 발생한 누두흉의 초기 재활치료의 효과)

  • A 6-week-old female cocker spaniel, with a history of abasia astasia, was referred to the Department of Laboratory Animal Medicine, Medical Research Center, Yonsei University College of Medicine on February 23, 2000. The ribs inclined downwards with a sharp slope on both lateral sides of the thorax. The animal was diagnosed as pectus excavatum. She showed no signs of dyspnea, dyschezia, or dysuria. There was no heart murmur or sign of neural disease. For rehabilitation treatment, she was given physical exercise therapy 2 to 3 times a day by bending and stretching each articulation of the hind limbs and pressing the costochondral junction in and upward with both hands placed on each lateral side. After a month of therapy, she was able to stand up and walk. The therapy continued and resulted in the slight reformation of a round thoracic wall. The flattened rib cartilages grew more round by palpation and the thoracic cavity distended. Rehabilitation therapy yielded good results, enabling the subject to walk and run normally, as the thoracic cavity was distended by the rounded rib junction.

Treadmill exercise ameliorates post-traumatic stress disorder-induced memory impairment in Sprague-Dawley rats

  • Post-traumatic stress disorder (PTSD) is a stress-related mental disorder induced by severe external stressors such as assault, disaster or severe accident. We investigated the effects of treadmill exercise on short-term memory in relation to apoptosis and cell proliferation in the hippocampus following PTSD. Stress to the pregnant rats was induced by exposure of maternal rats to the hunting dog in an enclosed room. Exposure time was 10 min, repeated three times per day, with 1 hour interval. Exposure of maternal rats to the hunting dog was continued 7 days after pregnancy until delivery. The pregnant rats in the exercise groups were forced to run on a treadmill for 30 min once a day for the same duration of stress exposure. Step-down avoidance task for short-term memory, western blot for Bcl-2, Bax, and immunohistochemistry for caspase-3, 5-bromo-2'-deoxyuridine (BrdU), and Ki-67 were conducted. Maternal rats exposed to stress during pregnancy showed short-term memory impairment. Expressions of Bax, Bcl-2, ratio of Bax to Bcl-2, and caspase-3 in the hippocampus were increased in the PTSD rats. Cell proliferation in the hippocampal dentate gyrus was decreased in the PTSD rats. Treadmill exercise alleviated short-term memory impairment and suppressed expressions of Bax, the ratio of Bax to Bcl-2, and caspase-3. Treadmill exercise also increased cell proliferation. The present results indicate that treadmill exercise alleviated PTSD-induced short-term memory impairment by suppressing apoptotic cell death and enhancing cell proliferation in the hippocampus.

Determination of urinary cortisol : creatinine ratios by sequential thin layer chromatography and ELISA in dogs (개에서 thin layer chromatography 박층크로마토그라피 및 ELISA를 이용한 요중의 cortisol : creatinine비 측정)

  • This study was conducted to evaluate the ELISA kit for measuring the level of cortisol in the urine. The CV of within-run variation and day to day variation were 0.4~2.8 and 1.8~5.7, respectively. The minimum limitation of measurement was 1ng/ml. The cross reaction was high ($CR_{50}(%)=11.4{\sim}43.2$) in prednisolone, 11-deoxycortisol, 21-deoxycortisol and predinosone. There was low and no cross reaction in other steroid. To develop the ELISA kit we measured the cortisol level in diluted urine with PBS (procedure I), extracted urine with methylene chloride (procedure II) and extracted methylene chloride-extracted urine from thin-layer chromatography (procedure III). The CV value of procedure I, II, III was 9.4~28.3%, 7.2~8.9% and 2.5~5.7%, respectively. There was significant difference between procedure I with II, and pro-cedure I with III(p < 0.01), but no difference between procedure II with III significantly(p < 0.01). The mean UCCR of urine collected through am 8 to 10 was $9.5{\pm}7.6$(0.14~28.0) in 12-month-old dog(n = 47). In this study we can measure the cortisol level in extracted urine with methylene chloride and sequential thin-layer chromatography accurately using ELISA kit.

Development of ELISA for cortisol and it's application to clinical use (ELISA를 이용한 cortisol 측정법의 정립 및 임상적 응용)

  • ELISA kit for cortisol was developed and then evaluated. Polyclonal antihydrocortisone-3-(o-carboxymethyl)oxime BSA rabbit serum was used to coat the 96-well microplates. The minimum detection limit of the kit was 250pg of cortisol per milliliter. The within-run variation and the day to day variation of the ELISA system were 2.0 and 5.9 at maximum, respectively. The kit was used to determine whether salivary cortisol concentration could replace blood cortisol concentration in dexamathasone suppression test of dogs. Changes of cortisol concentration were measured in serum or saliva after intravenous administration of 0.01mg of dexamethasone per kilogram of body weight. Blood alone, saliva alone or both were collected at 0, 30, 60, 120, 240, and 360 minutes after injection of dexamethasone. The change in blood cortisol concentration was found to be suitable in dexamathasone suppression test of dogs, but the change in salivary cortisol concentration was not. The kit was also used to determine whether salivary cortisol concentration could be a stress index as well as blood cortisol concentration in dogs. Two types of trial were performed to estimate the stress either by blood or salivary cortisol concentration. The first trial was stress experiment by intravenous injection of 0.2IU of PZI-insulin per kilogram body weight. Either blood alone or saliva alone was collected at 0, 30, 60, and 90 minutes after insulin administration. Both blood and salivary cortisol concentration were found to be suitable index in estimating stress from hypoglycemia by injection of insulin. The second trial was stress experiment by electrical irritation. The dogs were irritated with anti-bark device for 10 seconds. Blood was collected before and at 2 and 5 minutes after electrical irritation. Saliva was collected before and at 3 and 6 minutes after electrical irritation. The blood cortisol concentration, but not the salivary cortisol concentration was found to be suitable index in estimating stress from electrical irritation. Cushing syndrome in a dog was also successfully diagnosed with this kit.
