Extract of Broussometia kazinoki Induces Apoptosis Through the Mitochondria/Caspase Pathway in A549 Lung Cancer Cells (A549세포에서 닥나무 추출물의 미토콘드리아/Caspase 경로를 통한 Apoptosis 유도작용)

  • Extract of Broussometia kazinoki Rhizodermatis has been traditionally used for geopoong, diuresis, hwalhyeol. In the present study, the apoptotic effect of methanol extract of Broussometia kazinoki (MBK) were investigated. Cell viability of A549 cells was measured by MTT assay. Apoptosis-related protein and MAPK protein levels were measured by Western blot. Chromatin condensation of A549 cells was stained with DAPI. MBK inhibited cell proliferation of A549 cell. Based on DAPI staining, MBK-treated cells manifested nuclear shrinkage, condensation and fragmentation. Treatment of A549 cells with MBK resulted in activation of the caspase-3, -8, -9 and cleavage of poly ADP-ribose polymerase (PARP). In the upstream, MBK increased the expressions Bax and Bak, decreased the expression of Bcl-2, and augmented the Bax/Bcl-2 ratio. MBK-induced apoptosis was accompanied by sustained phosphorylation of JNK, p38 MAPK and apoptosis signal-regulating kinase (ASK)-1. These results suggest that MBK induced apoptosis in A549 cells through Bcl-2 family protein-mediated mitochondria/caspase-3 dependent pathway. In addition, MBK increased the activation of ASK-1, which are critical upsteam signals for JNK/p38 MAPK activation in A549 cancer cells.

Analysis of Flower Teas for their Oriental Medicinal Efficacy through Literature (꽃차용 꽃의 문헌을 통한 한의학적 효능 분석)

  • Research materials used were 55 kinds of flower teas proven in medicinal efficacy based on publications in oriental medicinal science. Efficacy in oriental medical science of the selected plants was classified into 11 groups, exterior-relieving herbs, removing dampness by promoting diuresis herbs, heatclearing herbs, purgative herbs, tonic herbs, qi-activating herbs, herbs as blood-tonics, herbs warming the intestine, qi-regulating herbs, herbs for removing dampness by means of aromatics, and herbs activating blood. Moreover, they were assorted by fever (hot, warm, slightly warm, common, cool, and cold), flavor (sweet, bitter, spicy, sour, and salty), and toxicity (toxic or nontoxic). Generally, flower teas have been known to be drinkable by all people. However, it is important to select proper flower teas considering physical constitution and health condition of the individual. One should be cautious in drinking flower teas containing toxic ingredients and weak people should be prohibited drinking them. Further clinical study on efficacy of flower teas will contribute to commercialization of these teas.

Identification of G Protein Coupled Receptors Expressed in Fat Body of Plutella Xylostella in Different Temperature Conditions (온도 차이에 따른 배추좀나방 유충 지방체에서 발현되는 G 단백질 연관 수용체의 동정)

  • BACKGROUND: G protein-coupled receptors (GPCRs) are widely distributed in various organisms. Insect GPCRs shown as in vertebrate GPCRs are membrane receptors that coordinate or involve in various physiological processes such as learning/memory, development, locomotion, circadian rhythm, reproduction, etc. This study aimed to identify GPCRs expressed in fat body and compare the expression pattern of GPCRs in different temperature conditions. METHODS AND RESULTS: To identify GPCRs genes and compare their expression in different temperature conditions, total RNAs of fat body in Plutella xylostella larva were extracted and the transcriptomes have been analyzed via next generation sequencing method. From the fat body transcriptomes, genes that belong to GPCR Family A, B, and F were identified such as opsin, gonadotropin-releasing hormone receptor, neuropeptide F (NPF) receptor, muthuselah (Mth), diuretic hormone receptor, frizzled, etc. Under low temperature, expressions of GPCRs such as C-C chemokine receptor (CCR), opsin, prolactin-releasing peptide receptor, substance K receptor, Mth-like receptor, diuretic hormone receptor, frizzled and stan were higher than those at 25℃. They are involved in immunity, feeding, movement, odorant recognition, diuresis, and development. In contrast to the control (25℃), at high temperature GPCRs including CCR, gonadotropin-releasing hormone receptor, moody, NPF receptor, neuropeptide B1 receptor, frizzled and stan revealed higher expression whose biological functions are related to immunity, blood-brain barrier formation, feeding, learning, and reproduction. CONCLUSION: Transcriptome of fat body can provide understanding the pools of GPCRs. Identifications of fat body GPCRs may contribute to develop new targets for the control of insect pests.

Hyperuricemic effects of Alpiniae Oxyphyllae Fructus extracts (고요산동물에서의 익지인의 요산저하 효과)

  • Objective : Hyperuricemia is a metabolic disease characterized by elevated blood uric acid levels, and its prevalence is rapidly increasing worldwide. Alpiniae Oxyphyllae Fructus (AO) belonging to Zingiberaceae is one of well-known traditional medicines in China and Korea, and has been used to treat intestinal disorders, urosis, diuresis, and chronic glomerulonephritis traditionally. However, the effect of AO has not been studied. In this study we investigated the anti-hyperuricemic effect of AO, and the mechanisms underlying the effect in potassium oxonate (PO)-induced hyperuricemic rats. Methods : To examine the anti-hyperuricemic effects of the AO extract, serum uric acid levels were analyzed in normal and PO-induced hyperuricemic rats. The mechanism underlying the effects of the AO extract on uric acid levels was studied through xanthine oxidase (XOD) activity test and uric acid uptake assay in vitro. The chemical finger printing of the AO extract was analyzed using HPLC-DAD. Results : The AO extract significantly reduced serum uric acid levels in normal as well as PO-induced hyperuricemic rats. It also significantly inhibited the uptake of uric acid in oocytyes and human embryonic kidney cells (HEK293) expressing urate transporter (URAT)1, but not XOD activity in vitro. The chemical finger printing analysis of the AO extract showed nootkatone as a main component. Conclusion : The AO extract exhibits anti-hyperuricemic effects, and these effect were accompanied by increasing excretion of uric acid in kidney. Therefore, the AO extract could be used for prevention or treatment of hyperuicemia and gout.

A Herbological Study on the Plants of Aspidiaceae in Korea (한국산 면마과(綿馬科) 식물에 관한 본초학적 연구)

  • Objectives : This study was aimed to developing Korean herbalogy of the plants belonging to Aspidiaceae in Korea. Methods : The herbological books and papers published at home and abroad were researched. The total list of Aspidiaceae was made and Medicinal plants in Aspidiaceae was classified. The medicinal properties, action and applications of medicinal plants were investigated. Results : 1. There are totaled to 17 genera and 130 species in Aspidiaceae in Korea and among them medicinal plants are 11 genera, 31 species, some 24% in total. 2. Athyrium genus is main kind enough that it has 32 species among 130 species in Aspidiaceae, and medicinal plants of Dryopteris is 4 species. 3. The rhizome is the main medicinal parts if medicinal plants in the Aspidiaceae, which is used in 24 species. 4. According to nature and flavor of medicinal plants in the Aspidiaceae, they were classified into cool 15 and cold 8 species ; bitter taste 19, and little bitter taste 15 in the order. 5. According to meridian propism of medicinal plants in the Aspidiaceae, they were classified into liver meridian 5 species, stomach meridian 3 species. 6. According to the properties and principal curative action, they were classified into drugs for clearing away heat 25 species, drugs for detoxification 22 species, drugs for promoting diuresis 16 species in the order. 7. The number of toxic species in the Aspidiaceae was examined to be 5 species. Conclusions : There are totaled to 17 genera and 130 species in Aspidiaceae in Korea and among them medicinal plants are 11 genera, 31 species, some 24% in total. Influence of Intracerebroventricular Yohimbine on the Renal Function of the Rabbit (가토 신장기능에 미치는 측뇌실내 Yohimbine의 영향)

  • The renal function is under regulatory influence of the central nervous system, mainly through activation of sympathetic nerve to the kidney, and it was recently reported that clonidine, an agonist to ${\alpha}_2$-adrenoceptors, induces diuresis and natriuresis when injected directly into a lateral ventricle of the rabbit brain (i.c.v.). This study was undertaken, therefore, to obtain further information as to the role of the central ${\alpha}_2$-adrenoceptors in regulating renal function, by observing the effects of i.c.v. yohimbine, a specific antagonist of adrenoceptors of ${\alpha}_2$-type, on the rabbit renal function, and to elucidate the mechanism involved in it. With 10 ${\mu}g/kg$ i.c.v. of yohimbine sodium excretion transiently increased along with increasing tendency of urine flow, renal plasma flow and glomerular filtration rate. These responses decreased with increasing doses. With 100 and 300 ${\mu}g/kg$ i.c.v. marked antidiuresis and antinatriuresis as well as profound decreases of renal perfusion and glomerular filtration were noted. Systemic blood pressure transiently increased. In reserpinized rabbits, 100 ${\mu}g/kg$ yohimbine i.c.v. did not produce any significant changes in urine flow, sodium excretion as well as in renal hemodynamics. The pressor response was also abolished. In preparations in which one kidney was denervated and the other left intact as control, i.c.v. yohimbine elicited typical antidiuretic antinatriuretic response in the innervated control kidney, whereas the denervated experimental kidney responded with marked diuresis and increases in excretory rates of sodium and potassium and in osmolar clearance in spite of absence of increased filtration and perfusion . Systemic blood pressure responded as in the normal rabbits. These observations indicate that i.c.v. yohimbine affects renal function in dual ways in opposite directions, the first being the antidiuretic antinatriuretic effects which results from decreased renal perfusion and glomerular filtration due to sympathetic activation and which is predominantly expressed in the normal rabbits, and the second less apparent effect being the diuretic and natriuretic action which is not mediated by nerve pathway but brought about by some humoral mechanism and which is effected by decreased sodium reabsorption in the tubules, possibly of the proximal portion.

Comparative Expression of the Aedes aegypti 5-Hydroxytryptamine7 Receptor in Drosophila Schneider2 and Chinese Hamster Ovary-K1 Cells (초파리 Schneider2 세포와 Chinese hamster ovary-K1 세포에서 Aedes aegypti 5-Hydroxytryptamine7 수용체의 발현비교)

  • Serotonin receptor binds to serotonin (5-HT) and activates effector proteins such as adenylyl cyclase, phospholipase C, cyclic GMP phosphodiesterase or ion channel through G protein on the cell membrane, resulting in various physiological responses like diuresis, memory and development. To examine the comparative expression of the 5-HT$\_$7/ receptor of Aedes aegypti, the Aedes 5-HT$\_$7/ receptor gene was transfected into Drosophila Schneider2 (S2) cells and mammalian Chinese hamster ovary (CHO)-Kl cells. The expression of the Aedes 5-HT$\_$7/ receptor gene in selected cell lines, Tr-CHO and Tr-S2, was confirmed with reverse transcription-PCR, Western blot and immunocytochemistry. Compared with the induced intracellular cAMP level of Tr-S2 cell line to 5-HT, the induced cAMP in the Tr-CHO cell line was over 9 times higher and was dose-dependent. These results suggest that the functionality of Aedes 5-HT$\_$7/ receptor is much more effective in mammalian CHO-K 1 cells and that the Tr-CHO cell line expressing Aedes 5-HT$\_$7/ receptor can be used for synthetic agonist or antagonist candidate screening.

Analysis for Compatibility of Daehwangmokdan-tang and Its Pharmacological Activities (대황목란탕(大黃牧丹湯)의 배오(配伍)분석과 그 약리활성)

  • Objectives : I analysed daehwangmokdan-tang's compatibility principle by the system of chief, deputy, assistant, and envoy and investigated pharmacological activities by categorizing with chemical components, molecular level, cellular level, animal level and human level based on Korean and Chinese studies for this formula. Methods : Daehwangmokdan-tang's compatibiltity principle was examined by the system of chief, deputy, assistant, and envoy. I looked into studies that presented in Korea from 1956 to 2016 about daehwangmokdan-tang through KOREA INSTITUTE OF ORIENTAL MEDICINE, Korean medicine information system (OASIS) and in Chinese for 20 years about daehwangmokdan-tang through China National Knowledge Infrastructure, CNKI. Then classify into chemical components, molecular level, cellular level, animal level and human level to analyse. Results : According to the system of chief, deputy, assistant, and envoy, chief herb is Rhei Radix et Rhizoma and Moutan Cortex, deputy herb is Natrii Sulfas and Persicae Semen, assistant and envoy herbs are Trichosanthis Semen. The amount of extraction of paeonol, total anthraquinone, and conjugated anthraquinone from daehwangmokdan-tang with the formulation of the system of chief, deputy, assistant, and envoy was the highest, and in the formulation of chief herb and deputy herb, the extraction amount of paeonol and conjugated anthraquinone was the lowest. With other formulations, the amount of extraction of total anthraquinone and conjugated anthraquinone was improved, although the degree was different. In particular, when it is blended with Persicae Semen as a deputy herb, the extraction amount of total anthraquinone and conjugated anthraquinone of Rhei Radix et Rhizoma as a chief herb is greatly increased, and the extraction amount of paeonol is rather different, but it is lowered. It was found that the amount of Mg Ca K Na in daehwangmokdan-tang was the highest. Pharmacological activities can be detected in inflammatory mediators and enzymes for molecular level. For cellular level, it can be determined in lipopolysaccharide (LPS)-stimulated RAW 264.7 cell line. In mouse and rats for animal level and human level, in inflammatory diseases (acute appendicitis, acute pancreatitis, acute cholecystitis, acute abdominal disease, ect.), pharmacological activities was caught. Conclusions : From the above results, daehwangmokdan-tang is composed in line with the system of chief, deputy, assistant, and envoy, suggesting that there is certain rationality and scientific. Pharmacological activities of daehwangmokdan-tang are effective to anti-inflammation, improvement of sepsis, analgesic, muscle relaxation, and improvement of intestinal flora and its metabolites. Daehwangmokdan-tang is consistent with the action of diuresis and anti-inflammation in terms of the content of elements, suggesting that there is action of purging fire and removing blood stasis, defecation detoxification, cooling blood and clearing heat and activating blood and dispersing stasis.

Effect of Cisplatin on Glomerular Filltration Rate and Effective Renal Plasma Flow (Cisplatin의 투여 후 사구체여과율 및 신혈류량의 변화)

  • While cisplatin has been widely used in the treatment of a variety of cancers, nephrotoxicity is one of the major problems which frequently limit clinical usefulness of cisplatin. This study has been conducted to investigate nephrotoxicity of cisplatin in terms of changes in glomerular filtration rate (GFR) and effective renal plasma flow (EFPF) measured by the simultaneous use of $^{99m}Tc-DTPA$ and $^{131}I-OIH$, before and after administration of cisplatin, in 12 patients with lung cancer and four patients with esophageal cancer. Cisplatin was administrated at total doses of $75\sim100mg/m^2$ with two hour hydration and diuresis method. GFR determined by the use of $^{99m}Tc-DTPA$ had a good correlation with 24-hour creatinine clearance rate (r=0.77, p<0.001). GFR and filtration fraction decreased immediately after administration of cisplatin, however, they showed a tendency to be in completely recovered four weeks after administration. ERPF was not changed immediately after and four weeks after administration of cisplatin. GFR before and immediately after administration of cisplatin were analyzed with regard to age, sex, performance status, previous adminstration of cisplatin and method of administration. None of these factors had any influence on the rate of decrease in GFR except method of administration. Administration of cisplatin as a single dose lowered GFR more compared with that as divided doses. In this study, we have also demonstrated that the simultaneous use of $^{99m}Tc-DTPA$ and $^{131}I-OIH$ was a useful tool for the measurement of GFR and ERPF respectively.

Cosmetical Properties of Polysccharides from the Root Bark of Ulmus davidiana var. Japonica (유근피로부터 추출한 다당류의 화장료적 특성)

  • The fruit or root of Ulmus davidiana var. Japonica and its extract has traditionally been used as anti inflammatation medicines as well as a remedy of the digestive ulcer for a long time. Ulmus davidiana var. Japonica has also been used to cure palliative and diuresis, because it contains lot of mucus and tannin. In this study, we examined the moisturizing effect and anti-inflammation effect of polysaccharides extracted from Ulmus davidiana var. Japonica to investigated whether it can be used as a cosmetic ingredient or not. The polysaccharides were extracted from Ulmus davidiana var. Japonica. Then the hydrolyzed was obtained through the hydrolysis. It has been ascertained that the polysaccharides we mainly composed of rhamnose, galactose, and glucose through HPLC experiments, and it has 20,000 of the molecular weight and 89.3 dL/g of intrinsic viscosity In a moisturizing test of measuring water loss in a desiccator and moisture content by Comeometer CM820, Ulmus davidiana root extract showed a similar moisturizing effect as hyaluronic acid. And its anti-inflammation effect on carrageenin-induced edema evaluated by measuring the thickness of swelling over paw induced by carrageenin (25 uL of $2\%$ carrageenin saline injection), the $\%$ swelling over paw treated with Ulmus davidiana root extract was much less than that of paw treated with carrageenin only. The anti-inflammatory effect of Ulmus davidiana root extract was almost similar to that of ketoprofen. These results can say that Ulmus davidiana root extract can be effectively used as a cosmetic ingredient.
