S-(N,N-Diallyldithiocarbamoyl)-N-acetylcysteine: Synthesis and Biochemical Properties Associated with Chemoprevention (S-(N,N-Diallyldithiocarbamoyl)-N-acetylcysteine의 합성 및 발암억제와 관련된 생화학적 특성)

  • Dithiocarbamate and mixed disulfide containing allyl functions were designed and synthesized as putative chemopreventive agents, i.e. N,N-diallyldithiocarbamate (DATC) and S-(N,N-diallyldithiocarbamoyl)-N-acetylcysteine (AC-DATC). DATC and AC-DATC were administered and the activities of cytosolic glutathione S-transferase (GST), glutathione reductase (GR) and microsomal N-nitrosodiethylamine (NDEA) deethylase were assayed in order to test the effects of these organosulfur com-pounds on the detoxification and metabolic activation system of NDEA. The amounts of hepatic glutathione (GSH and GSSG) was also determined. The administration of DATC to rats led to an increase in the activity of GR and to an inhibition of CYP2E1-mediated NDEA deethylation. AC-DATC induced the activity of GR and GST, increased the hepatic GSH content and inhibited the rate of NDEA deethylation. The level of GSSG was decreased as a consequence of the increased activity of GR. These effects may contribute to possible antimutagenic and anticarcinogenic action of the dithiocarbamates investigated.

  • In the present work, we studied thiram on silver surface by SERS. Investigations of disulfides with SERS revealed that the molecules undergo a surface reaction on silver, namely easy cleavage of the S-S bond. We believe that the two S atoms of resonance formed from the thiram may be chemisorbed strongly on Ag sol. This resonance form adheres perpendicularly to the Ag surface via the two S atoms, since the ${\delta}(CH3)$ and n (CN) mode perpendicular to the surface showed strong enhancement. The important roles of halide anion adsorption have been discussed and the pH effects of thiram on Ag sol in acidic, neutral, and alkaline conditions were examined.

  • Nickel sulfide (NiS) has been utilized in optoelectronic applications, such as transformation-toughening agent for materials used in semiconductor applications, catalysts, and cathodic materials in rechargeable lithium batteries. Recently, high quality nickel sulfide thin films have been explored using ALD/CVD technique. Suitable precursors are needed to deposit thin films of inorganic materials. However, nickel sulfide precursors available for ALD/CVD process are very limited to nickel complexes with dithiocarbamate and alkanethiolate ligands. Therefore, it is essential to prepare novel nickel sulfide suitable for ALD/CVD precesses. Herein we report on the synthesis and characterization of new nickel sulfide complex with designed aminothiolate ligand. Furthermore thin films of NiS have been prepared on silicon oxide substrates by spin coating nickel precursor 10 wt% in THF. The novel complex has been characterized by means of 1H-NMR, elemental analysis, thermogravimetric analysis (TGA), X-ray Diffraction (XRD) and scanning electron microscope (SEM).

Effects of Seongpung-tang on the NO Production of Primary Microglial Cell (성풍탕이 미세아교세포의 NO 생성에 미치는 영향)

  • The water extract of Seongpungtang(SPT) has commonly been used for treatment of ischemic brain damage in Oriental traditional medicine. However, little is known about the mechanism by which the water extract of SPT rescues brain cells from ischemic damage. To elucidate the protective mechanism of ischemic induced cytotoxicity, the regulation of Lipopolysaccharide (LPS) and PMA (phobol-12-myristate-13-acetate) induced iNOS expression in microglial cells was investigated. LPS and PMA treatment for 48 hr in microglial cells markedly induced nitric oxide (NO), but treatment of the cells with the water extract of SPT decreased nitrite formation. In addition, LPS and PMA treatment for 48 hr induced severe cell death in microglial cells. However treatment of the cells with the water extract of SPT did not induce significant changes compared to the control cells. Furthermore, NO production was markedly decreased by treatment of nuclear factor kappa B(NF-kB) inhibitor, pyrrolidine dithiocarbamate(PDTC). According to the above results, it is suggested that the protective effects of the water extract of SPT against ischemic brain damage may be mediated by regulation of iNOS during ischemic condition.

  • The analytical conditions for the fluorescence X-ray determination of small amount of cupper in solution were established. Copper in solution was extracted by diethyldithiocarbamate ethyl acetate and adsorbed by zinc powder. After drying, it was pressed to tablet under the pressure of $250Kg/cm^2.$$K_{\alpha}2(2{\theta}_{LiF} = 45.08^{\circ})$. The limit of detection was estimated to be about $10{\mu}g$ by the method of standard addition with an error of 10 %.

  • A new and facile protocol for the synthesis of dithiocarabamate in EtOH/$H_2O$ is described. Reaction of aromatic and aliphatic amines with $CS_2$ and 3,4-dihydro-2H-pyran in the presence of N,N,N',N'-tetrabromobenzene-1,3-disulfonamide [TBBDA] and poly(N-bromo-N-ethylbenzene-1,3-disulfonamide) [PBBS] gives the corresponding dithiocarbamates in good to high yields.

Organic Precipitate Flotation of Trace Metallic Elements with Ammonium Pyrrolidinedithiocarbamate (II). Application of Solvent Sublation for Determination of Trace Cd, Co, Cu and Ni in Water Samples

  • A solvent sublation was studied for the determination of trace Cd, Co, Cu and Ni in water samples. Ammonium pyrrolidine dithiocarbamate (APDC) was used as a complexing agent. Experimental conditions such as pH of solution, amounts of APDC, the type and amount of surfactant, the type of solvent, etc. were optimized for the effective sublation of analytes. After metal-PDC complexes were formed in sample solutions of pH 2.5, the precipitate-type complexes were floated in a flotation cell with an aid of sodium lauryl sulfate as a surfactant and by bubbling with nitrogen gas. The precipitates were dissolved and separated into the surface layer of methyl iso-butyl ketone (MIBK). The analytes preconcentrated were determined by a graphite furnace atomic absorption spectrophotometry (GF-AAS). Extractability of each element was 88% for Cd(Ⅱ), 86% for Co(Ⅱ), 95% for Cu(Ⅱ) and 76% for Ni(Ⅱ), respectively. And this procedure was applied to the analysis of real samples. From the recoveries of more than 92%, it was concluded that this method could be simple and applicable for the determination of trace elements in various water samples of a large volume.

Synthesis and Properties of Dicyclohexyltin(IV) Complexes. Structure of $Cy_2Sn(S_2CNMe_2)_2 $

  • Dicyclohexyltin(IV) complexes $Cy_2SnX (X = O,\;S,\;(Me_2NCS_2)_2)$ have been prepared and characterized by means of elemental analysis, cryoscopic data, and IR spectroscopy. X-ray structure determination for $Cy_2Sn(S_2CNMe_2)_2$ (monoclinic; P2/c;a = 8.992(1), b = 6.688(1), c = 19.453(2) ${\AA},\;{\beta}$ = 96.556(7) ; R = 0.041) has shown that the molecule is $C_2$-symmetric with both dithiocarbamate ligands bonding in anisobidentate mode and the local geometry around the hexacoordinate Sn(IV) atom is a distorted octahedral arrangement with the two cyclohexyl groups in trans position. The physical and chemical properties measured suggest polymeric and trimeric structures for $Cy_2SnO\;and\;Cy_2SnS$, respectively.

  • This report describes the results of analysis, synthesis and evaluation of antimony dithiocarbamates (SbDTC). Sb-DTC were synthesized by reacting antimony oxide with dithiocarbamates whish were prepared by reaction of dialkylamines and carbon disulfide. The chemical structures and properties of synthesized Sb-DTCs in lab. were discussed. The frictional properties of SbDTC were tested with Four-Ball Wear Scar Tester and Four-Ball Extreme Tester by ASTM method.

  • Yellowish modified wool, dithiocarbamate(DTC) wool, was synthesized by partial hydrolysis in 0.2 N-NaOH reacting with carbon disulfide to use as ${\alpha}-amylase$ immobilization matrix. ${\alpha}-amylase$ was immobilized reacting with sulfide group of DTC-wool by covalent binding within 1 hour. 0.5 gram of this preparation, $DTC-wool-{\alpha}-amylase$, contained 150 ug of enzyme protein and its specific activity was about 90% of the native one. General properties of $DTC-wool-{\alpha}-amylase$ were a little different from optimum temperature, optimum pH, heat stability, kinetic constants and activation energy. An apparent Michaelis constant and maximum velocity of $DTC-wool-{\alpha}-amylase$ were 5.56 mg/ml and 0.37 mg/ml. $min^{-1}$ respectively, while activation energy was 16.6 kcal/mole.
