농도별(濃度別) 산양산삼(山養山蔘) 증류약침(蒸溜藥鐵)의 Apoptosis에 관(關)한 실험적(實驗的) 연구(硏究) (An Experimental Study on Apoptosis of cultivated Wild Ginseng Distilled Herbal Acupuncture by Concentration Level)

  • Objective : In order to measure the efficacy of cultivated wild ginseng distilled herbal acupuncture by concentration level, we've treated A549 human lung cancer lines with different concentrations of cultivated wild ginseng distilled herbal acupuncture and examined mRNA and proteins which take parts in apoptosis. Methods : A549 human lung cancer lines were treated with various concentration levels of cultivated wild ginseng distilled herbal acupuncture and cell toxicity was carefully examined. From the analysis of DNA fragmentation, RT-PCR and Western blot, manifestation of mRNA and proteins which are associated with apoptosis were inspected. Results : The following results were obtained on apoptosis of A549 human lung cancer lines after administering various concentration levels of cultivated wild ginseng distilled herbal acupuncture. 1. Measuring cell toxicity of lung cancer cells, strong cell toxicity was detected at high concentration level(1000ul, 1200ul), but no consistent concentration dependent reliance was detected. 2. Through DNA fragmentation, we were able to confirm cell destruction in all groups. 3. Experiment groups treated with cultivated wild ginseng distilled herbal acupuncture showed inhibition of Bcl-2 and COX-2 at mRNA and Protein level, whileas increase of Bax was shown. 4. Manifestation of p21, p53, Cyclin E, and Cyclin Dl were confirmed in all groups. 5. Extrication of Cytochrome C was detected at all groups, as well as increased activity of the enzyme caspase-3 and caspase-9, and PARP fragmentation were confirmed. Conclusion : According to the results, we can carefully deduce cell destruction of A549 human lung cancer lines were induced by Apoptosis. At the fixed level, cultivated wild ginseng distilled herbal acupuncture showed decrease of Bcl-2 and COX-2, as well as increase of Bax. Since cultivated wild ginseng distilled herbal acupuncture increases manifestation of p21, p53, Cyclin E, and Cyclin Dl, it affects cellular cycle and through these phenomena, we can consider extrication of Cytochrome C, increase of caspase, and PARP fragmentation are the results.

국 또는 개량 누룩으로 제조한 술덧을 감압 또는 상압으로 증류한 소주의 휘발성 성분 (Volatile Compounds in Liquor Distilled from Mash Produced Using Koji or Nuruk under Reduced or Atmospheric Pressure)

  • 본 연구는 국 또는 개량 누룩을 사용하여 제조한 술덧을 감압 또는 상압에서 증류하여 제조한 증류식 소주의 휘발성 성분을 분석하여 적절한 술덧 제조 원료와 증류방법을 검토하기 위함이다. Acetaldehyde는 상압 증류하였을 때에 감압 증류하였을 때보다 그 함량이 2~3배 많았다. Furfural은 상압 증류법으로 제조한 소주에서만 검출되었다. Ester류는 전체적으로 감압 증류하였을 때에 상압 증류하였을 때보다 더 많이 검출되었으며, 특히 ethyl pelargonate는 상압 증류하였을 때에 감압 증류하였을 때보다 2배 이상 높았다. 대부분의 alcohol류는 감압 증류하였을 때에 상압 증류하였을 때보다 약간 더 많았으며, methanol은 국을 사용한 경우보다 개량 누룩을 사용한 경우 더 많이 검출되었다. 고급 알코올류인 isoamyl alcohol과 2-phenyl ethanol만이 증류 방법에 의한 차이가 있었다. 특히 2-phenyl ethanol은 상압 증류하였을 때에 감압 증류하였을 때보다 2배 이상 많이 검출되었다. Methanol은 국을 사용한 술덧이 개량 누룩을 사용한 술덧에 비해 적게 검출되었다. 본 연구 결과 소주에 부정적 영향을 미치는 휘발성 성분을 줄이기 위해서는 국을 사용한 술덧을 감압 증류함으로써 증류식 소주의 품질을 향상시킬 수 있을 것으로 판단된다.

녹차와 로즈마리 추출물을 첨가한 두유의 항산화 활성 (Antioxidant activities of soymilk added with green tea and rosemary extract)

  • 제주산 녹차, 로즈마리, 레몬 잎, 조릿대 잎을 물과 에탄올을 이용해 다양한 조건에서 추출물을 제조하여 각 추출물에 대한 항산화 활성을 측정하였다. 그 결과, 녹차와 로즈마리 추출물이 레몬과 조릿대 잎 추출물과 비교하여 상대적으로 높은 항산화 활성을 보였다. 항산화 활성이 상대적으로 우수한 녹차와 로즈마리 추출물들을 두유에 첨가한 뒤, 혼합물의 항산화 활성을 측정하였다. 녹차 증류수 가압 가열 추출물과 상온 정치 추출물 10% 첨가한 두유에서 상대적으로 가장 높은 활성을 보였으며, 녹차 증류수 가압 가열 추출물과 상온 정치 추출물의 DPPH 라디칼 소거능은 각각 $85.1{\pm}2.3%$, $84.4{\pm}2.0%$을, ABTS 라디칼 소거능은 각각 $73.4{\pm}2.8%$, $81.3{\pm}4.8%$로 확인되었다. 한편, 로즈마리 증류수 가압 가열 추출물 10% 첨가한 두유에서 무첨가 두유에 비해 더 높은 DPPH 라디칼 소거능이 확인되었으며, ABTS 라디칼 소거능은 무첨가 두유와 유사했다. 따라서 녹차 증류수 가압 가열추출물과 상온 정치 추출물, 그리고 로즈마리 증류수 가압 가열 추출물을 두유에 첨가 시 항산화 활성이 향상된 고부가가치 두유제품 개발 제발이 가능할 것으로 기대된다.

증류주의 품질 최적화를 위한 효모선발 연구 (Yeast Selection for Quality Optimization of Distilled Spirits)

  • 본 연구의 목적은 쌀을 원료로 하여 증류주 제조시 쌀 특성에 적합한 최적의 증류주 효모를 선발하는데 있다. 사용된 5종류의 효모는 전반적으로 정상적인 발효패턴을 보였으나 송천효모(Y1)와 증류주 효모(Y5)가 다른 효모에 비해 우수한 발효력을 보였다. 또한 발효후 술덧을 분석한 결과 고급알코올은 증류주 효모(Y4, Y5)에서, 에스터는 송천효모(Y1)와 증류주효모(Y4, Y5)에서 유의적으로 높게 검출된 반면, 유기산 함량은 증류주 효모(Y3, Y4, Y5)에서 유의적으로 높게 나타났다. 발효술덧을 동증류기를 이용한 상압증류한 증류주 분석 결과 송천효모(Y1)와 증류주 효모(Y5)가 최종 알코올 농도가 높아 제조수율에 유리한 것으로 판단되며, 고급알코올과 에스터 등 향기 성분은 발효술덧 분석 결과와 같이 고급알코올은 증류주 효모(Y4, Y5)에서, 에스터는 송천효모(Y1)와 증류주 효모(Y4, Y5)에서 유의적으로 높게 나타나 발효술덧의 향기 패턴이 증류주에 대부분 전이되는 것으로 판단된다. 효모별 발효특성과 발효술덧 및 증류주의 물리화학적 특성, 관능평가를 종합한 결과 증류주 효모인 siha aktivehefe 6 brennereihefe(Y5)가 쌀을 이용한 증류주 품질 최적화에 가장 적합한 효모로 선발되었다.

스킨헬스케어를 위한 증류 죽초액 함유 천연화장품의 여드름 피부 임상 적용 연구 (A Clinical Research of Acne Skin through Natural Cosmetics with Distilled Bamboo Vinegar Contents for Skin Health Care)

  • 본 연구는 천연물질인 증류 죽초액이 함유된 천연화장품의 여드름 피부 개선에 대한 효과성을 검증하여 여드름 화장품 소재 개발을 목적으로 하였다. 연구의 대상 10대 남·여 20명을 선정하여 난수표방식에 따라 10명은 천연화장품(폼 클렌징, 토너, 팩)을 사용한 대조군으로 10명은 증류 죽초액이 함유된 천연화장품(폼 클렌징, 토너, 팩) 사용한 실험군으로 배정하였다. 화장품 사용 기간은 12주로 하였고, Mark-·Vu 피부 측정기를 사용하여 사전, 6주 후, 12주 후 피부의 피지, 모공 크기, 붉은기를 측정하였다. 수집된 자료는 SPSS v. 21.0 통계 패키지 프로그램을 활용하여 분석하였으며, 결과는 다음과 같다. 첫째, 천연화장품의 피부 안정성 확인을 위한 인체첩포시험 결과 피부에 안전한 제품으로 확인되었다. 둘째, 대조군과 실험군의 피지, 모공 크기, 붉은기에 대한 동질성 검증 결과 동질성이 확보되었다. 셋째, 증류 죽초액이 함유된 천연화장품의 여드름 피부 개선 효과 검증에서 실험군이 대조군에 비해 이마와 오른 볼 부위의 피지, 모공 크기, 붉은기 변화의 감소율이 더 크게 나타났다. 이에 증류 죽초액이 함유된 천연화장품은 피부에 안전하며, 여드름 피부에서 피부의 피지 감소와 모공 크기 및 붉은기 감소에 효과적인 것으로 확인됨에 따라 화장품 산업에서 증류 죽초액 활용을 기대하는 바이다.

소리쟁이(Rumex crispus) 뿌리로부터 가용성 고형분의 추출특성 (Extraction Characteristics of Soluble Solid from Rumex crispus(Curled Dock) Roots)

  • In this study, the extraction characteristics of soluble solid from Rumex crispus(Curled dock) was studied from the investigation of the effects of experimental conditions on extraction rate; extraction ratio, composition of extractants, extraction time and pH of extractant, etc. The proximate composition of Rumex crispus was 2.58% crude lipid, 5.59% crude protein, 7.39% crude ash, 6.13% moisture and 78.31% carbohydrate, respectively. Turbidity of extract by distilled water was higher and increased with extraction time and extraction temperature, where as the turbidity didn't increase by ethanol and methanol in 20 folds of extraction ratio. Turbidity was inversely proportional to the extraction ratio for the three extractants at 25$^{\circ}C$ and 1 hour extraction. But turbidity of extract was highest by composition of 50% methanol-water extractant than any other compositions of extractants. Eighteen and fifteen free aminoacids were detected in extracts with distilled water, methanol and ethanol extractant, respectively, and it's contents were order of glutamic acid>proline>aminobutyric acid>alanine. The extraction rate of soluble solid from Rumex crispus was order of distilled water>methanol>ethanol within experimental extraction ratio. In extraction with distilled water, the contents of soluble solid was inversely proportional to the pH of extractant.

치자염의 염색과정이 염색견뢰도에 미치는 영향(II) (The effect of color fastness in the Gardenic Jasminoides Ellis Dyeing(II))

  • The purpose of this study is to investigate the effect of mordanting condition in the Gardenic Jasminoides Ellis Dyeing. The results were as follows; 1. The color fastness was compared and analysed that colored sample as $AIK(SO_4)_2,\;CuSO_4{\cdot}5H_2O,\;Ca(OH)_2,\;NaOH,\;HOOC(CHOH)_2COOH,\;K_2\;Cr_2O_7$ group was better than non-colored sample and also a silk was better than a cotton in fastness. Mordant as NaOH, HOOC$(CHOH)_2COOH,\;K_2Cr_2O_2,\;AIK(SO_4)_2$ were better than anything else in fastness and the effective method of the mordant to a silk was pre & aft mordant(4) and to a cotton was aft-mordant(3). 2. The comparison between methanol and distilled water, which used in extracted dyenig solution, the latter was more than one grade in fastness. 3. The color-difference value-test indicated that colored sample groups more redish and bluish color than non-colored sample. 4. The comparison between methanol and distilled water, which used to extract dyeing solutions, the latter were more light, greenish and yellowish colors. 5. When compared with the methanol and distilled water, as used to extract dyeing solutions, showed that the methanol was more clear and light yellow and the distilled water was more orange color and more effective in color fastness.

열화된 종이자료의 보존성 개선을 위한 세척처리 특성 (Effect of Washing Treatment of Aged Paper Materials for Better Conservation)

  • Paper materials for long term conservation suffer slowly mechanical and chemical deterioration, the extent of which may depend upon their conservation environment. Those deterioration includes discoloring, low moisture content, acidification, and brittleness. To slow deterioration, washing treatment, deacidification, and polymer reinforcement on paper materials are usually used. One easy and simple method of fixing low moisture content and acidification was an washing method, and we used both distilled and alkali water in washing method in this study. Alkali water is electrolyzed cathode water of high pH, and has no alkali metal ions in it. Experiment showed that washing treatment with both distilled and alkali water gave improvement in raising moisture content, pH, and mechanical strength of paper materials even after severe accelerated aging test. Advantageous effect of alkali water over distilled water on preventing deterioation was also shown clearly.

Saccharomyces와 Hansenula의 혼합배양에 의해 제조한 민속증류수의 향미특성 (Flavor Characteristics of Korean Traditional Distilled Liquors Produced by the Co-culture of Saccharomyces and Hansenula)

  • Andong Soju is a Korean traditional distilled liquor brewed with Nuruk which is cultured with wild microorganisms. To provide useful information for scientific production and systematic quality control of traditional distilled liquor, the effects of mixed culture of the alcoholic yeasts and saccharifying molds isolated from the Nuruk, and mashes on the flavor and sensory characteristics were investigated. Distillate from mashes cocultured with Saccharomyces cerevisiae and Hansenula anomala using Mucor Nuruk was compared with distillate from mashes brewed with Andong Nuruk and with distillate from plant fermented mashes to analyze their flavor characteristics. The volatile flavor compounds in distillates were analyzed by GC and GC-MS using direct injection, solvent extraction, and purge & trap methods. Alcohols such as 3-methyl-1-butanol, 2-methyl-1-propanol, 1-propanol, and 2-phenyl ethanol; aldehydes such as acetaldehyde and 2-furancar-boxaldehyde; esters such as ethyl ester of acetic acid, hexanoic acid, octanoic acid, decanoic acid; alkanes, alkenes, ketone, sulfur, and pyrone compounds were detected. Alcohols were chief components of flavor compounds. No significant difference in overall acceptability test was shown among three experimental groups(p<0.05), but Nuruk-like aroma, Kaoliangchiew-like aroma, sweet taste, and well rounded mouthfeel showed significant differences among them(p<0.05).

섬유강화 복합재료에서 섬유열화에 따른 계면특성의 변화 (Change of Interfacial properties by the Fiber Degradation in the Fiber Reinforced Composites)

  • Single fiber fragmentation technique was used to evaluate the change of interfacial properties by degradation of fiber tensile strength in the fiber reinforced composites. The influences of fiber tensile strength on the interfacial properties have been evaluated by the fragmentation specimens(weak fiber samples) of glass fiber/epoxy resin that was made using the pre-degraded glass fiber in distilled water at $80^{circ}C$ for specified periods. The effects of the immersion time on the interfacial properties in the distilled water at $80^{circ}C$ also have been evaluated by the fragmentation specimens(original fiber samples) of glass fiber/epoxy resin that was made using the received glass fiber. As the result, the tensile strength of glass fiber was decreased with the increasing of the treatment time in the distilled water at $80^{circ}C$ and the interfacial shear strength was independent of the change of the glass fiber strength in the single fiber fragmentation test. But in the durability test using the single fiber fragmentation specimen, interfacial shear strength decreased with the increasing of the immersion time in distilled water ar $80^{circ}C$. And it turned out that the evaluating of interfacial shear strength using original fiber tensile strength was valuable in the durability test for the water environment by the single fiber fragmentation technique.
