Accumulation of pathogenic amyloid-β disrupts the tight junction of retinal pigment epithelium (RPE), one of its senescence-like structural alterations. In the clearance of amyloid-β, the autophagy-lysosome pathway plays the crucial role. In this context, mammalian target of rapamycin (mTOR) inhibits the process of autophagy and lysosomal degradation, acting as a potential therapeutic target for age-associated disorders. However, efficacy of targeting mTOR to treat age-related macular degeneration remains largely elusive. Here, we validated the therapeutic efficacy of the mTOR inhibitors, Torin and PP242, in clearing amyloid-β by inducing the autophagy-lysosome pathway in a mouse model with pathogenic amyloid-β with tight junction disruption of RPE, which is evident in dry age-related macular degeneration. High concentration of amyloid-β oligomers induced autophagy-lysosome pathway impairment accompanied by the accumulation of p62 and decreased lysosomal activity in RPE cells. However, Torin and PP242 treatment restored the lysosomal activity via activation of LAMP2 and facilitated the clearance of amyloid-β in vitro and in vivo. Furthermore, clearance of amyloid-β by Torin and PP242 ameliorated the tight junction disruption of RPE in vivo. Overall, our findings suggest mTOR inhibition as a new therapeutic strategy for the restoration of tight junctions in age-related macular degeneration.
In an infrastructure-based wireless network, an access point is used for all communications among mobile devices. However, when a mobile device moves into a dead Bone, a connectivity disruption between the mobile device and the access point occurs. Such connectivity disruption consequently leads to another connectivity disruption between the mobile device moving toward the dead zone and other wireless-enabled devices located within the area of the infrastructure-based wireless network. To cope with the connectivity disruption in the infrastructure-based wireless network the ad hoc network that dynamically forms a network without any preexisting communication infrastructure needs to be set up to provide seamless connections among mobile devices. In this paper, we propose the DNSQ-MAC (Dynamic Network State aware QoS-Medium Access Control) technique that meets the deadlines of MAC frames forwarded over hop-by-hop multipaths and guarantees the QoS performance of an ad hoc-based wireless network. Mobile devices incorporating the DNSQ-MAC technique are capable of adjusting to the new dynamic network status in order to enhance the QoS performance in the ad hoc-based wireless network. A case study which exploits the Qualnet simulator shows that the proposed DNSQ-MAC technique can guarantee the deadlines of MAC frames forwarded over hop-by-hop multipaths and enhance the QoS performance of various routing protocols and packet schedulers running on the network layer above the MAC layer.
Oriental fruit moth, Grapholita molesta, is a serious pest on apples. To control this pest in an environmentally friendly method, mating disruption strategy using sex pheromone has been developed. Area-wide application of mating disruption has been needed to be effective, with little understanding on how much size of apple cultivating area should be treated in one time application of the mating disruption technique. On this matter, we needed to determine a minimal mating active zone of G. molesta that should be applied with mating disrupters to be effective. Molecular markers to discriminate a specific population should be developed to trace population migration for reproductive behaviors. Here we developed two effective molecular markers using random amplified polymorphic DNA (RAPD) technique. Different field populations of G. molesta, based on locations and seasons, were analyzed with these markers. In a specific location, G. molesta populations varied in genetic composition with different seasons. Different local populations showed differential variation according to their relative distances among apple orchards. In overall, genetic variation among different populations became lessen with progression of seasons.

This study was performed to monitor the seasonal occurrence of Pseudococcus comstocki (Kuwana) by using pheromone traps and to test a mating disruption technique to control this pest in pear orchards. We compared the attractiveness of rubber septa loaded with 1.5 and 3.0 mg of the pheromone, 2,6-dimethyl-1,5-heptadien-3-yl acetate. A total of 1,021 and 1,431 males were caught in traps baited with 1.5 mg and 3 mg of the pheromone, respectively. The numbers of males caught were not affected by trap color, although yellow traps were more attractive than white traps. In 2012, P. comstocki males were captured between June and October, with peaks in late June, early August, and late September. In 2013, the males were trapped between June and October, with peaks in middle June, late July, and late September. In pheromone mating disruption tests, catches were reduced by 17.7, 65.3, and 62.9% in orchards treated with 450, 900, and 1,350 mg per 10 a of the pheromone, respectively. At harvest, 3.4, 2.9, and 4.8% of fruits in orchards treated with 450, 900, and 1,350 mg per 10 a were damaged by P. comstocki, while 9.5% were damaged in the control orchard.
Objective: The purpose of this study was to investigate whether three-dimensional (3D) magnetic resonance imaging could improve diagnostic accuracy for suspected posterior ligamentous complex (PLC) disruption. Materials and Methods: We used 20 freshly harvested goat spine samples with 60 segments and intact surrounding soft tissue. The animals were aged 1-1.5 years and consisted of 8 males and 12 females, which were sexually mature but had not reached adult weights. We created a paraspinal contusion model by percutaneously injecting 10 mL saline into each side of the interspinous ligament (ISL). All segments underwent T2-weighted sagittal and coronal short inversion time inversion recovery (STIR) scans as well as coronal and sagittal 3D proton density-weighted spectrally selective inversion recovery (3D-PDW-SPIR) scans acquired at 1.5T. Following scanning, some ISLs were cut and then the segments were rescanned using the same magnetic resonance (MR) techniques. Two radiologists independently assessed the MR images, and the reliability of ISL tear interpretation was assessed using the kappa coefficient. The chi-square test was used to compare the diagnostic accuracy of images obtained using the different MR techniques. Results: The interobserver reliability for detecting ISL disruption was high for all imaging techniques (0.776-0.949). The sensitivity, specificity, and diagnostic accuracy of the coronal 3D-PDW-SPIR technique for detecting ISL tears were 100, 96.9, and 97.9%, respectively, which were significantly higher than those of the sagittal STIR (p = 0.000), coronal STIR (p = 0.000), and sagittal 3D-PDW-SPIR (p = 0.001) techniques. Conclusion: Compared to other MR methods, coronal 3D-PDW-SPIR provides a more accurate diagnosis of ISL disruption. Adding coronal 3D-PDW-SPIR to a routine MR protocol may help to identify PLC disruptions in cases with nearby contusion.
