서울 천호동지역 정호의 대장균오염조사 (A Study on the Contamination of Wells in Chonhodong Area)

  • A study was conducted to find out the biological contamination of wells in Chon Ho Dong areas, Seoul, Korea from May 4, 1971 through May 18 1971. Coliform groups were detected by membrane filter method and general sanitary conditions were checked. The results obtained were summarized as follows: 1) 61 wells (81.3%) out of 75 wells showed the evidence of coliform group. In urban areas it is recommended to use disinfectants for wells pretty frequently. 2) The wells in native villages were less contaminated than those in new villages established by poor people from slum areas of Seoul. 9 out of 15 wells at Song Pa Dong, which is However, at Bang E Dong's new village, 19 out of 20 wells were contaminatd by coliform groups. 3) Coliform groups were positive at 57 out of 61 open wells, while only 4 out of 14 pumping wells showed the evidence of coliform groups. 4) 38 out of 40 poorly drained wells were contaminated, however, 13 out of 25 well drained wells were coliform organisms positive.

식도협착과 급성신부전이 합병된 개미산 중독 1례 (Esophageal Stricture and Acute Renal Failure after Formic Acid Poisoning: - A Case Report -)

  • Formic acid or formate is a common industrial compound used in the production of ensilage, disinfectants, decalcifying agents and mainly as a precursor in industrial chemical synthesis. It is also a well-known toxic metabolite produced in methanol poisoning. Thus, formate is a potential source of both accidental and deliberate poisoning. Very few reports have been published thus far, on the toxicology of direct formic acid poisoning. Here, we report a case of a 74-year-old man without a history of depression, who ingested about 30 gm of formic acid. The patient presented with profound high anion gap metabolic acidosis, acute renal failure and esophageal stricture. The patient was successfully treated with hemodialysis and supportive measures. But permanent esophageal stricture was complicated by formic acid burns in the gastrointestinal tract. We discuss the pathophysiology and treatment of this case.

유리잔류염소와 모노클로라민에서의 생물막의 CLPP (CLPP of Biofilm in Free Chlorine Residual and Monochloramine)

  • The disinfection effect of free residual chlorine and monochloramine on biofilm communities were investigated by CLPP (community level physiology profile) using Biolog GN plates. Low concentration of disinfectant, $0.5\;mg/\iota$ free chlorine and $1.0\;mg/\iota$ monochloramine, stimulated the growth of bacteria rather than disinfection. Bacterial concentrations were decreased at more than $1.0\;mg/\iota$ of disinfectants. CLPP was different with the type and concentration of disinfectant and sampling time. Common and different carbon sources were actively used with similar bacterial concentration in free chlorine and monochloramine. This represents the differences of bacterial communities with tap water contact times and disinfectant.

수종방부제의 살균효과에 관한 실험적 연구 (EXPERIMENTAL STUDIES ON ANTIBACTERIAL EFFECT OF DISINFECTANTS)

  • 1. The purpose of this study is to determine the antibacterial effect of the remedies commonly used in operative dentistry. 2. Experiment were performed against staphylococcus albus which were isolated from different root canals. 3. Isolated bacteria were inoculated on nutrient agar and disk which has been saturated antiseptics were placed on it, then this plate was incubated for 48 hours at 37℃. 4. Inhibition zone were measured around antiseptic disks. 5. The results of 45 cases of experiments showed as follows. Average diameter of inhibition zone around phenol disk was 2.99 mm, camphorated phenol, 0.74 mm, formocresol, 13.46mm, formocresol (Korea), 12.7 mm, eugenol, 4.74mm, thymol, 16.46 mm.

Principles and Applications of Non-Thermal Technologies for Meat Decontamination

  • Meat contains high-value protein compounds that might degrade as a result of oxidation and microbial contamination. Additionally, various pathogenic and spoilage microorganisms can grow in meat. Moreover, contamination with pathogenic microorganisms above the infectious dose has caused foodborne illness outbreaks. To decrease the microbial population, traditional meat preservation methods such as thermal treatment and chemical disinfectants are used, but it may have limitations for the maintenance of meat quality or the consumers acceptance. Thus, non-thermal technologies (e.g., high-pressure processing, pulsed electric field, non-thermal plasma, pulsed light, supercritical carbon dioxide technology, ozone, irradiation, ultraviolet light, and ultrasound) have emerged to improve the shelf life and meat safety. Non-thermal technologies are becoming increasingly important because of their advantages in maintaining low temperature, meat nutrition, and short processing time. Especially, pulsed light and pulsed electric field treatment induce few sensory and physiological changes in high fat and protein meat products, making them suitable for the application. Many research results showed that these non-thermal technologies may keep meat fresh and maintain heat-sensitive elements in meat products.

스테인리스 스틸 식품기구 표면에 사용되는 주요 살균소독제의 살균력 평가 및 살균예측모델 개발 (Evaluation of Efficacy and Development of Predictive Model of Sanitizers and Disinfectants on Reduction of Microorganisms on Food Contact Surfaces)

  • 본 연구는 기구 표면에 존재하는 주요 오염균인 E. coli와 S. aureus에 대한 살균소독제의 농도와 처리 시간별 살균력을 평가하여 살균예측모델 개발 및 살균소독지침의 기초자료로 제공하고자 한다. 염소계 살균소독제 100ppm처리 직후, 5분,\0분 후 E. coli( 1.56, 1.49, 1.95 log cfu/25 $cm^2$)와 S. aureus(0.49, 0.88, 1.27 log cfu/25 $cm^2$)에 대해 높은 감소율을 보였고 200ppm처리 시에는 불검출 되었다. 알콜계 살균소독제는 35.5% 처리 직후, 5분, 10분 후 E. coli(0.73, 0.90. 1.55 log cfu/25 $cm^2$), S. aureus(0.37, 1.00, 1.45 log cfu/25 $cm^2$)에 대해 높은 감소율을 보였고, 70% 5분 처리 시 불검출되었다. 과산화수소계 살균소독제는45.5 ppm 처리 직후. 5분, 10분 후 E. coli(0.28, 0.64, l.07 10g cfu/25 $cm^2$), S. aureus(0.53, 0.87, 0.99 log cfu/25 $cm^2$)에 대해 높은 살균력을 보였으나, 91 ppm 5분 처리 시 검출되지 않았다. 4급암모늄계는 100 ppm 처리 직후, 5분, 10분 후 E. coli(0.82, 1.62. 1.71 log cfu/25 $cm^2$), S. aureus(0.46, 0.93, 1.38 log cfu/25 $cm^2$)에 대한 높은 감소율을 보였으나 200 ppm 5분 처리 시 불검출 되었다. 살균예측모델의 경우, $R^2$ 값이 모두 9.4 이상으로 두 균주 모두 높은 적합성을 보였다. 기구 등의 살균소독제의 사용은 적정한 농도와 처리시간을 준수하여 야만 병원성미생물 감소에 효과적이므로 본 연구에서 개발된 4가지 주요 살균소독제의 살균예측모델을 현장에서 활용함으로써 최적의 살균소독제 처리농도와 처리시간을 추가 실험 없이 시뮬레이션으로 도출 가능할 것이다.

Giardia에 의한 환경오염(環境汚染)과 감염(感染)에 관한 역학적(疫學的) 연구(硏究) (Epidemiological Studies on Giardia Infection Associated with environmental Pollution)

  • Giardia lamblia is a pathogenic flagellate causing intestinal disorders such as diarrhea, abdominal pain and malabsorption of nutrients. Giardia is mainly infected by the ingestion of contaminated foods per os. Craun (1979) has recently reported that mass infection of this flagellate through the contaminated water supply systems is one of public health hazards. Also, so-called traveller's diarrhea is sometimes caused by Giardia infection (CDC, U.S.A., 1971). However, a few epidemiological studies figuring out the mode of infection or control measures of Giardia infection has been done so far in Korea. The present study was aimed to know the prevalence of Giardia infection in several Korean populations, detectability of this flagellate in water systems and the viability of the cysts against sewages and disinfectants applying to drinking water. In the present study, 388 stool specimens from orphanage children in Chun-joo, Chung-joo, On-yang and Chun-an areas and 538 stool specimens from inhabitants in Woo-do, In-chon, and Chun-joo were examined by formalin-ether concentration technique to detect out Giardia cysts. On the other hand, water samples from 14 sites of Han River and its tributaries were collected in May through July, 1984. Fifty liter of water sample in each sampling site was then filtered through water filtering system deviced by U.S. Environmental Proutection Agency and the sediments rinsed out from the thread rolls, a part of water filtering system, were examined to detect out the Giardia cysts. In order to observe the viability of Giardia cysts in the sewage samples, the cysts were treated in it at $4^{\circ}C$ or $25^{\circ}C$ for 7 through 28 days. For this purpose, the cysts were also exposed to various concentrations of disinfectants such as chlorine, iodine and ozone gas for proper time intervals. After treatment, the viability test of the Giardia cysts were carried out by method of Rice and Schaefer (1981) with minor modification. The results obtained in this study were as follows : 1) The detection rates of G lamblia cysts in the stool specimens were 18.3% in orphans and 4.3% in general examinees. 2) The prevalences of Giardia Infection were higher in the young age groups than in-adults. The highest positive rate was 18.4% in the age group less than 10. 3) Of 14 water specimens sampled from Han River system and its tributaries around the Seoul area, the Giardia cysts were detected from 4 samples, and no cyst was found in the water supply systems. 4) The cysts treated in the sewage survived for 28 days at $4^{\circ}C$ and for 13 days at $25^{\circ}C$. 5) The cysts were completely destroyed within 60 minutes by exposure to 8 mg/l of residual chlorine at 4g and within 30 minutes by exposure to the same concentration of chlorine at $25^{\circ}C$. 6) The cysts were all dead when exposed to 1 mg/1 of iodine for 60 minutes at $4^{\circ}C$ or $25^{\circ}C$. 7) The cysts were destroyed after 10 minute exposure in 0.15 mg to 0.25mg of residual ozone gas per liter. Summarizing the above results, it is considered that Giardia infection is regarded as water-borne disease and the cysts are able to be controlled by the application with the disinfectants in the water supply systems.

Well-Plate를 사용한 치과용 유니트 수관 바이오필름 모델 확립 (Establishment of a Dental Unit Biofilm Model Using Well-Plate)

  • DUWL에 형성한 바이오필름을 효율적으로 제거할 수 있는 새로운 소독제는 개발되어야 하며, 실험실에서 소독제의 효과를 확인하기 위해 DUWL 바이오필름 시료는 필요하다. 이 연구의 목적은 well plate를 사용하여 간단하고 재현 가능한 DUWL 바이오필름 모델을 개발하는 것이다. 사용중인 4대의 DUWL에서 배출된 1 L의 물을 여과지에 여과시켜 세균을 얻었다. 여과지를 PBS (pH 7.4) 용액 20 ml에 현탁시킨 후, 현탁액을 R2A 액체배지에 접종하고 $25^{\circ}C$에서 10일 동안 배양하였다. 10일 배양한 세균 배양액을 $-70^{\circ}C$에 보관하였고 매 실험에 사용하였다. 세균배양액을 R2A 배지에서 5일 동안 회분 배양하였다. 12-well plate에 회분 배양한 세균 배양액과 멸균한 폴리우레탄 튜빙 조각을 넣고 정체된 상태로 $25^{\circ}C$에서 바이오필름을 형성시켰다. R2A 액체배지는 2일마다 2 ml씩 교체해주었다. 폴리우레탄 내형성된 바이오필름의 축적량을 확인하기 위해 폴리우레탄 튜빙 조각을 내면에서 수집한 바이오필름을 R2A 고체배지에 도말하였다. 도말한 R2A 고체배지는 $25^{\circ}C$에서 7일 배양하고 $CFU/cm^2$를 계산하였다. 그리고 바이오필름의 두께와 구성 세균의 형태 및 분포를 확인하기 위해 CLSM과 SEM을 사용하였다. 4일 동안 배양시킨 바이오필름의 평균 축적량은 $1.15{\times}10^7CFU/cm^2$였다. 바이오필름은 구균, 짧은 길이의 간균, 그리고 중간길이의 간균을 포함하고 있었고 폴리우레탄 튜빙 내면에 넓게 분포하고 있었다. 바이오필름두께는 위치에 따라 차이가 있지만 $2{\mu}m{\sim}7{\mu}m$였다. 이 연구에서 제작된 DUWL 바이오필름 model은 DUWL에서 수집된 모든 세균을 사용하여 세균의 다양성을 확보했고 또한 실험실에서 쉽게 구할 수 있는 well-plate를 사용했기 때문에 비용 효율적이고 재현이 간단하다. 본 연구의 DUWL 바이오필름 모델은 새로운 소독방법의 개발하는 데 유용하게 사용될 수 있다.
