Surveillance on the Vivax Malaria in Endemic Areas in the Republic of Korea Based on Molecular and Serological Analyses

  • Plasmodium vivax reemerged in 1993. It has been sustained for more than 25 years and become one of the important indigenous parasitic diseases in northern and western parts of the Republic of Korea near the demilitarized zone. In particular, relapse is a significant concern for the control of malaria, as short- and long-term incubation periods vary among those infected in Korea. In this study, the prevalence of asymptomatic carriers was examined among residents of high endemic areas of vivax malaria during nonseasonal transmission of mosquitoes. Blood samples from 3 endemic regions in northwestern Korea were evaluated by microscopic examination, rapid diagnostic testing, and nested PCR to identify asymptomatic patients carrying malaria parasites in the community. However, no positive malaria case among residents of endemic areas was detected. Additionally, serological analysis was carried out to measure antibodies against 3 antigenic recombinant proteins of P. vivax, merozoite surface protein 1-19, circumsporozoite surface protein-VK210, and liver-stage antigen (PvLSA-N), by the protein array method. Interestingly, seropositivity of sera between previous exposure and samples without exposure to malaria was significantly higher using the PvLSA-N antigen than the other antigens, suggesting that PvLSA-N can be used as a serological marker to analyze the degree of exposure for malaria transmission in endemic areas. This indicates a very low asymptomatic carrier prevalence during the nonmalaria season in the endemic areas of Korea.

  • Newcastle disease is a serious infectious disease in the poultry industry. The commercial vaccines can only offer limited protection and some of them are expensive and need adjuvants. At present, DNA vaccines are widely used. However, the immune responses induced by DNA vaccines are too slow and low. Here, we constructed the transfer vectors with a different number of C3d as molecular adjuvants (n = 1, 2, 4, or 6), and the vectors were cloned into the optimal eukaryotic expression plasmid (pVAXI-optiF) that expressed the F gene of Newcastle disease virus (NDV), and named pVAXI-F(o)-C3d1, pVAXI -F(o)-C3d2, pVAXI-F(o)-C3d4, and pVAXI-F(o)-C3d6, respectively. Cell transfection test indicated that pVAXI-F(o)-C3d6 showed the highest expression. In vivo immunization showed that the chickens immunized with pVAXI-F(o)-C3d6 intramuscularly induced better immune responses than the chickens immunized with the other plasmids. The protective efficacy of pVAXI-F(o)-C3d6 was 80% after challenge with the highly virulent NDV strain F48E9. The results in this study showed that C3d6 could be used as a molecular adjuvant to quickly induce an effective immune response to control NDV.

Relative Abundance of Vectors of Scrub Typhus in Jeonnam Province, Korea

  • Studies on the relative abundance of potential vectors of scrub typhus was conducted from April to October 2009 at the Jangseong of Jeonnam Province, Korea. Among the total 62 field rodents collected by the Sherman collapsible traps, 56 rodents were $Apodemus$ $agrarius$ (90.3%), 4 were $Crocidura$ $lasiura$ (6.4%) and 2 were $Cricetulus$ $triton$ $nester$ (3.2%). Out of the 62 field rodents, 39 were parasitized by chiggers, showing 62.9% of the infestation rate and 80.0 of the chigger index. Chigger index of $A.$ $agrarius$ and $C.$ $triton$ $nester$ were 79.9, 82.0, respectively. But chigger mite was not collected from $C.$ $lasiura$. From the trapped field rodents, 3,120 chiggers were collected and identified with 6 species of 2 genera. $Leptotrombidium$ $scutellare$, the vector species of tsutsugamushi disease, was the dominant species, showing 2,276 chiggers (72.9%). $L.$ $pallidum$ was the second dominant species showing 346 chiggers (11.0%). The distribution of chigger mites was showed the different dominant species according to seasons. $L.$ $pallidum$ was the predominant chigger collected in April (34.8%), May (77.8%) and Jun (39.3%), whereas $L.$ $scutellare$ was the predominant chigger collected in September (78.78%) and October (76.8%).

  • DNA vaccines use eukaryote expression vectors to produce immunizing proteins in the vaccinated host and it represents a novel approach to vaccine and immuno-therapeutic development. We constructed a 2.9 kb compact plasmid vector (pVAC) which contains CMV promoter, polycloning site, BGH poly A terminator, ampicillin resistance gene and PBR322 origin. Enriched unmathlyated CpG motifs have introduced into pVAC-ISS1 and pVAC-ISS2 which are derived from pVAC for enhancing Thl responses. These plasmid DNAs rapidly induced interleukin 6 secretion in vivo. It is expected that these vectors will contribute to the DNA inoculation against infectious disease and various cancers without adjuvant.

Prevalence and Intensity of Clonorchis sinensis Metacercariae in Freshwater Fish from Wicheon Stream in Gunwi-gun, Gyeongsangbuk-do, Korea

  • The infection status of Clonorchis sinensis metacercariae (CsMc) was examined in freshwater fish from a highly prevalent site, Wicheon (a branch of Nakdong-gang), which is located in Gunwi-gun, Gyeongsangbuk-do, the Republic of Korea. Total 1,162 fish in 32 species were examined by the artificial digestion method through 6 years. CsMc were detected in 720 (67.5%) out of 1,067 fish (26 spp.) and their density was 610 per fish infected. In the susceptible gobioninid fish group, i.e., Pungtungia herzi, Squalidus gracilis majimae, Squalidus japonicus coreanus, Sarcocheilichthys variegatus wakiyae and Pseudorasbora parva, all of 323 fish were infected with an average of 1,310 CsMc. Total 23 (95.8%) gobioninid fish, i.e., Pseudogobio esocinus, Abbottina springeri, Hemibarbus longirostris, Microphysogobio koreensis, and Microphysogobio jeoni, were infected with 127 CsMc in average. In the acheilognathinid fish (bitterlings) group, the prevalence was 77.0%, and the density was 50 CsMc per fish infected. In the rasborinid fish (chubs) group, i.e., Zacco platypus, Zacco temminckii, Zacco koreanus, and Opsariichthys uncirostris amurensis, 147 (36.5%) out of 403 fish examined were infected with 15 CsMc in average. The susceptibility indices of CsMc were 412 in the overall positive fish group, 1,310 in the gobioninid group-1, 122 in the gobioninid group-2, 38.5 in the acheilognathinid group, and 5.5 in the rasborinid fish group. Conclusively, it was confirmed that CsMc are highly prevalent in fish from Wicheon, and their infection tendency varied according to the subfamily groups in Cyprinidae fish hosts.

Cutaneous Leishmaniasis Situation and Predicting the Distribution of Phlebotomus papatasi and P. sergenti as Vectors of Leishmaniasis in Ardabil Province, Iran

  • Cutaneous leishmaniosis (CL) is the most common form of leishmaniasis.CL caused by L. major and L. tropica is endemic in 17 provinces of Iran. This study was carried out to elucidate situation of CL in Ardabil province and to predict distribution of Phlebotomus papatasi and Phlebotomus sergenti (Diptera: Psychodidae) as vectors of CL in the region. In this cross-sectional study, data on CL patients were collected from local health centers of Ardabil province, Iran during 2006-2018 to establish a geodatabase using ArcGIS10.3. A total of 20 CL cases were selected randomly and skin samples were collected and analyzed by PCR method. MaxEnt 3.3.3 model was used to determine ecologically suitable niches for the main vectors. A total, 309 CL human cases were reported and the highest incidence rate of disease was occurred in Bilasavar (37/100,000) and Germi (35/100,000). A total of 2,794 sand flies were collected during May to October 2018. The environmentally suitable habitats for P. papatasi and P. sergenti were predicted to be present in northern and central areas of Ardabil province. The most variable that contributed ratio in the modeling were Isothermality and slope factors. Ardabil province is possibly an endemic are for CL. The presence of P. papatasi and P. sergenti justifies local transmission while the vectors of CL are existing in the northern and central areas of the province.

Patterns of Plasma Fatty Acids in Rat Models with Adenovirus Infection

  • Adenoviral vectors are among the most promising vectors available for human gene therapy. However, the use of recombinant adenoviral vectors, including replicationcompetent adenovirus (RCA), raises a variety of safety concerns in relation to the development of new therapies based on gene therapy. To examine how organic compounds change in rat plasma following the injection of adenovirus, $\beta$-galactosidase expressing recombinant adenovirus (designated rAdLacZ) or RCA, we investigated the content of fatty acids (FAs), which are important biochemical indicators in pathological conditions. Pattern recognition analysis on the level of FAs in rat plasma is described for the visual discrimination of adenovirus infection groups from normal controls. Plasma FAs from four control rats (normal group), and from four rats with rAdLacZ infection and six rats with RCA infection (the two abnormal groups), were examined by gas chromatography-mass spectrometry in selected ion monitoring modes as their tert-butyldimethylsilyl derivatives. In total, 20 FAs were positively detected and quantified. The results of the Student's t-test on the normal mean of two abnormal groups, the levels of three FAs (p<0.05) from rAdLacZ group and eleven FAs (p<0.05) from RCA group were significantly different. When star symbol plotting was applied to the group mean values of 20 FAs after normalization to the corresponding normal mean values, the resulting eicosagonal star patterns of the two infected groups were distorted into similar shapes, but were distinguishable from each other. Thus, these approaches will be useful for screening and monitoring of diagnostic markers for the effects of infection following the use of adenoviral vectors in gene therapy.

The follow up study after massive outbreak of Akabane and bovine ephemeral fever viruses in Korea

  • Since a large number of Akabane and bovine ephemeral fever (BEF) infection occurred in the southern part of Korea in 2010, recent information about seroprevalence of Akabane virus (AKAV) and bovine ephemeral fever virus (BEFV) has been required for preventing both diseases. In this study, serological assay against AKAV and BEFV using virus neutralization assay was conducted using 1,743 bovine sera collected from Namwon, Miryang, Yeongju and Uljin which located in Southern part of Korea from March to May in 2012. The overall seropositive rates for AKAV and BEFV were found to be 49.8% and 1.2%, respectively. The regional distribution of seroprevalence for AKAV ranged from 18.1% to 63.7%. Seroprevalences of AKAV were 63.7% in Miryang, 62.3% in Uljin, 50.7% in Namwon, and 18.1% in Yeongju. The seropositive rates for AKAV in southern part of Korea were higher than the annual average at the national level. On the other hand, seropositive rates of BEFV in four regions were from 0.3 to 3.1%. In detail, regional seroprevalences were 3.1% in Miryang, 2.0% in Uljin, and 1.7% in Yeongju, and 0.3% in Namwon. Even only one year after massive outbreaks, overall seropositive rates were very low, similar to the annual average at the nation level. This result indicates that many number of cattle infected with BEFV may be replaced by new born calf or cattle in farm may not be immunized with vaccines. To prevent another epidemic, a national wide warning should be issued and more aggressive control measure must be implied. Recent global warming phenomenon could lead to more vigorous activity of haematophagous vectors and it is possible that arboviral diseases such as AKAV and BEFV are increased. Therefore, continuous sero-monitoring and extensive vaccination combined with control of haematophagous vectors are important to effectively prevent and control diseases caused by AKAV and BEFV.

Monitoring Insecticide Resistance and Target Site Mutations of L1014 Kdr And G119 Ace Alleles in Five Mosquito Populations in Korea

  • Mosquitoes are globally distributed and important vectors for the transmission of many human diseases. Mosquito control is a difficult task and the cost of preventing mosquito-borne diseases is much lower than that for curing the associated diseases. Thus, chemical control remains the most effective tool for mosquito. Due to the long-term intensive use of insecticides to control mosquito vectors, resistance to most chemical insecticides has been reported. This study aimed to investigate the relationship between insecticide resistance and target site mutation of L1014 kdr and G119 ace alleles in 5 species/species group of mosquitoes (Aedes vexans, Ae. albopictus, Anopheles spp., Culex pipiens complex, and Cx. tritaeniorhynchus) obtained from 6 collection sites. For Anopheles spp., the proportion of mosquitoes with mutated alleles in L1014 was 88.4%, homozygous resistant genotypes were observed in 46.7%, and heterozygous resistant genotypes were observed in 41.8%. For the Cx. pipiens complex and Cx. tritaeniorhynchus species, homozygous resistant genotypes were found in 25.9% and 9.8%, respectively. However, target site mutation of L1014 in the Ae. vexans nipponii and Ae. albopictus species was not observed. Anopheles spp., Cx. pipiens complex, and Cx. tritaeniorhynchus mosquitoes were resistant to deltamethrin and chlorpyriphos, whereas Ae. vexans nipponii and Ae. albopictus were clearly susceptible. We also found a correlation between the resistance phenotype and the presence of the L1014 kdr and G119 ace mutations only in the Anopheles spp. population. In this study, we suggest that insecticide resistance poses a growing threat and resistance management must be integrated into all mosquito control programs.

Automatic Disk Disease Recognition based on Feature Vector in T-L Spine Magnetic Resonance Image (척추 자기 공명 영상에서 특징 벡터에 기반 한 디스크 질환의 자동 인식)

  • In anatomical aspects, magnetic resonance image offers more accurate information than other medical images such as X ray ultrasonic and CT images. This paper introduces a method that recognizes disk diseases from spine MR images. In this method, image enhancement, image segmentation and feature extraction for sagittal plane and axial plane images are performed to separate the disk region. And then template matching method is used to extract disease region for axial plane imges. Finally, disease feature vectors are integrated and disease discrimination processes are performed. Experimental results show that the proposed method discriminates between normal and diseased disk with a considerable recognition ratio.
