Differential Role of Solvents on Human Cytochrome P450 2El Activity in Intact HepG2 Cells (HepG2 세포에서 용매에 의한 차별적인 사람 싸이토크롬 P450 2E1활성 변화)

  • The modification of CYP2El activity is a matter of considerable interest because of its role in the metabolic activation of a variety of environmental toxicants. In the present study, the time-course of changes in human CYP2El activities was determined following treatment with solvents (acetone, dimethylsulphoxide or pyridine) using intact HepG2 cells transfected by human CYP2El. Hydroxylation of chlorzoxazone was used for the measurement of CYP2El activity. CYP2E1 protein level was increased upon cultivation of cells in the presence of the solvents for 24 hr. Determination of CYP2El activities after 24 ht cultivation with the solvents demonstrated that acetone or dimethylsulphoxide increased, whereas pyridine inhibited the activities. This differential effect of the solvents on CYP2El activities persisted to subsequent 24 ht. Competitive inhibition study suggested that pyridine has stronger binding affinity to CYP2E1 than acetone or dimethylsulphoxide. These results demonstrate that different binding affinity of the solvents to CYP2El plays important role in determining real CYP2El activity in intact cells after exposure to the solvents. Present study would be helpful in precise understanding of human CYP2El-mediated toxicity.

  • The dielectric relaxation study for aniline-dimethylsulphoxide (DMSO) and aniline-dim.ethylformamide(DMF) has been carried out using the Time domain reflectometry (TDR) technique, at different temperature and concentrations, in the frequency range of 10 MHz to 10 GHz. The dielectric parameters viz. static permittivity, relaxation time, the Kirkwood correlation factor, excess permittivity, excess inverse relaxation time and thermodynamic parameters have been obtained. The calibration method based on least squares fit method has been used. The dielectric parameters show systematic change with temperature and concentrations.

  • Frequency spectra of the complex permittivity of 2-methoxyethanol (2-ME) with water, ethanol, dimethylsulphoxide (DMSO), N,N-dimethylformamide (DMF) and N,N-dimethylacatamide (DMA) have been determined over the frequency range of 10 MHz to 20 GHz at 25 $^{\circ}C$, using the Time domain reflectometry method, for 11 concentrations for each system. The static dielectric constant, dielectric constant at microwave frequency, relaxation time, excess dielectric parameters, and Kirkwood correlation factor have been determined. The relaxation in these systems within the frequency range can be described by a single relaxation time constant, using the Debye model. The parameters show a systematic change with the concentration.

  • Dielectric relaxation measurements on 3-nitrotoluene (3-NT) mixture of dimethylacetamide (DMA), dimethylformamide (DMF) and dimethysulphoxide (DMSO) have been carried out across the entire concentration range using Time domain reflectometry technique at 15, 25, 35 and $45^{\circ}C$ over the frequency range from 10 MHz to 20 GHz. For all the mixtures, only one dielectric loss peak was observed in this frequency range and the relaxation in these mixtures can be well described by a single relaxation time using Debye model. Bilinear calibration method is used to obtain complex permittivity ${\varepsilon}^{*}({\omega})$ from complex reflection coefficient ${\rho}^{*}({\omega})$ over frequency range 10 MHz to 20 GHz. The excess permittivity, excess inverse relaxation time, Kirkwood correlation factor, molar energy of activation are also calculated for these mixtures to study the solute-solvent interaction.

  • Alcohol consumption and exposure to endocrine disruptors and industrial solvents have been implicated in impaired spermatogenesis, increase in the incidence of malformed sperm and decrease in the percentage of moving sperm. The aim of this study was to determine and compare the direct effects of some organic solvents(bisphenol A; BPA, dibutyl phthalate; DBP, formaldehyde; HCHO, dimethylsulphoxide; DMSO, ethanol) on mucus penetration distance, motility and survival rate of human sperm in vitro. Semen samples from 3 health subjects were prepared using swim-up method and 0.0005-0.5% organic solvents were added to the test medium. BPA, DBP, HCHO and DMSO produced significant decreases in the motility and survival rate with a different potency. The most potent inhibition of mucus penetration distance, motility and survival rate was observed after exposure to HCHO. A concentration of 0.0005% HCHO significantly inhibited sperm motility. When ethanol m.: added directly to sperm, at concentrations equivalent to that in serum after heavy drinking, these damaging effects were lowest compared with other solvents. Present study shows that each compound has different toxic potency to human sperm and we need special caution for the use of HCHO.

  • An electrochemical study related to the redox characteristics of Ethyl-3-acetyl-6-methyl-1, 4-diphenyl-4, 3a-dihydro-1, 3, 4-triazolino[3, 4-a] pyrimidine-5-carboxylate ester and its derivatives (1a-f) and (2a-e) in nonaqueous solvents such as 1, 2-dichloroethane (DCE), dichloromethane (DCM), acetonitrile (AN), dimethylsulphoxide (DMSO) and tetrahydrofurane (THF) using $0.1\;mol\;dm^{-3}$ tetrabutylammonium perchlorate (TBAP) as a supporting electrolyte at platinum, glassy carbon and gold electrodes, has been performed using cyclic voltammetry (CV). Controlled potential electrolysis (CPE) is also carried out to elucidate the course of different electrochemical reactions through the separation and identification of the intermediates and final electrolysis products. The redox mechanism is suggested and proved. It was found that all the investigated compounds in all solvents are oxidized in a single irreversible one electron donating process following the well known pattern of the EC-mechanism to give a dimer. On the other hand, these compounds are reduced in a single irreversible one electron step to form the anion radical, which is basic enough to proton from the media forming the radical which undergoes tautomerization and then dimerization processes to give also another bis-compound through N-N linkage formation.

Practical Procedure of Sperm Cryopreservation of the Bar-tailed Flathead Platycephalus indicus (양태, Platycephalus indicus의 정자의 냉동보존)

  • This study was conducted to investigate protocol standardization for cryopreservation spermatozoa of the bar-tailed flathead Platycephalus indicus. The suitability of the cryoprotectants, dimethyl sulphoxide (DMSO), glycerol and methanol were tested against three freezing rates and three thawing temperatures. DMSO and glycerol gave significantly higher motile index and survival rates than methanol. Among the freezing rates, freezing at a height of 2 cm above $LN_2$ surface for $10\;min^{-1}$ gave higher motile index and survival rates. In terms of best thawing temperature, $20^{\circ}C$ obtained the highest motility.

  • A sensitive and accurate method for the detection of astaxanthin was developed. The optimum HPLC system for the extraction and quantification of astaxanthin was based on a reversed phase column, and a quaternate mobile phase contained dichloromethane. The amount of astaxanthin can be accurately quantified in Phaffia rhodozyma cultures. The results of this study suggest that the mixture solution of dimethylsulphoxide and acetone is the optimal extraction solvent for astaxanthin and samples treatment for astaxanthin analysis must control low temperature and absent light operation strictly.

Effect on Survival and Developmental Competence of Vitrified Mouse Embryos Using Various Cryoprotectants and Cooling Speeds (생쥐 배아의 유리화 동결에 동결액의 조성과 냉각속도의 영향)

  • Objective: Vitrification requires a high concentration of cyroprotectant (CPA) and an elevated cooling speed to avoid ice crystal formation. We have evaluated the effect of different combinations of cooling rate and CPA on embryonic integrity (developmental competence) in order to increase the efficiency of vitrification without impairing embryo viabilit. We hypothesized that the combination of CPA or the increase of cooling rates can reduce the concentration of toxic CPA for vitrification. As consequently, we performed experiments to evaluate the effect of various composition of CPA or slush nitrogen ($SN_2$) on the mouse embryonic development following vitrification using low CPA concentration. Methods: Vitrification of mouse embryos was performed with EM grid using liquid nitrogen ($LN_2$) or $SN_2$ and different composition of CPAs, ethylene glycol (EG) and dimethylsulfoxide (DMSO). After vitrification-warming process, their survival and blastocyst formation rates were examined. For analyzing long-term effect, these blastocysts were transferred into the uterus of foster mothers. Results: Survival and blastocyst formation rates of vitrified embryos were higher in EG+DMSO group than those in EG only. Furthermor, the group using $SN_2$ with a lower CPA concentration showed a higher survival of embryos and developmental rates than group using $LN_2$. Conclusion: The combination of EG and DMSO as CPAs may enhance the survival of mouse embryos and further embryonic development after vitrification. $SN_2$ can generate high survival and developmental rate of vitrified/warmed mouse embryos when a lower concentration of CPA was applied. Therefore, these systems may contribute in the improvement of cryopreservation for fertility preservation.
