Molecular Interaction of Dimethylsulfoxide with Water and Alkanols : A Vapor Pressure Osmometry Study (디메틸술폭시드와 물 및 알칸올과의 분자 상호작용 : 증기압 삼투법에 의한 연구)

  • Studies on the molecular interactions of dimethylsulfoxide (DMSO) with water and/or some alkanols were carried out by vapor pressure osmometry at 40$^{\circ}C$. Negative deviation from Raoult's law was observed for the DMSO-water, methanol, ethanol, 1-propanol, 2-propanol, and 2-methyl-1-propanol systems, whereas positive deviation from Raoult's law was observed for the DMSO-1-butanol and 1-pentanol systems. The results were interpreted in terms of molecular interactions between unlike molecules, and of self-association of DMSO molecules, respectively. Measured chemical shift of hydroxyl proton of the solvents also supported the results.

  • Solvolyses of p-substituted benzyl bromides have been studied in dimethylsulfoxide-water and N,N-dimethylformamide-water mixtures by kinetic method. To determine the ionizing power, Y and the nucleophilicity, $N_{BS}$, the solvolyses of 1-adamantyl halides, t-butyl halides, and methyl tosylate in the same solvent mixtures have been investigated. The solvatochromic parameters for each dimethylsulfoxide-water mixtures have been determined by substituting into the Taft's linear solvatochromic energy relationships with measured $ν_{max}$. The solvolyses of p-substituted benzyl bromides have been found to proceed by borderline mechanism in which bond formation is more advanced than bond cleavage in the transition state based on the m, l values and ${\beta},{\rho}_s$, values.

Establishment of Analytical Method for Chlorophyll Using the N,N-Dimethylformamide and Dimethylsulfoxide in Citrus Leaves (N,N-Dimethylformamide와 Dimethylsulfoxide를 이용한 감귤 잎의 엽록소 측정방법 확립)

  • BACKGROUND: Purpose of the study was to establish the extinction coefficients of chlorophyll a and b in N,N-dimethylformamide(DMF) and Dimethylsulfoxide(DMSO) solvents and to find out the conditions of optimal extraction temperature and time in citrus leaves. METHODS AND RESULTS: Chlorophyll a and b standards were dissolved in DMF and DMSO. Extinction coefficients of chlorophyll pigments were determined and their contents were quantified using spectrophotometer. Chlorophyllous pigments of citrus(Citrus unshiu Marc. cv. Okitsu wase) leaves were extracted at 25, 40, 60 and $80^{\circ}C$ for 4, 6, 8, 24 and 48 hours to determine the optimal extraction condition. CONCLUSION: The extinction coefficients of chlorophyll a(Chl a) and chlorophyll b(Chl b) of DMF extracts for high extinction wavelength were 663.8 and 647.2 nm. Similarly, the high extinction wavelength of DMSO extracts were 665.8 and 649.0 nm for chl a and b respectively. Chl a, Chl b and total chlorophyll content of DMF extracts were Chl a = $12.10A_{663.8}-2.74A_{647.2}$, Chl b = $21.94A_{647.2}-5.06A_{663.8}$ and total $Chl=19.193A_{647.2}+7.04A_{663.8}$. Similarly, Chl a, Chl b and total Chl of DMSO extracts were Chl a = $14.53A_{665.8}-5.40A_{649.0}$, Chl b = $26.98A_{649.0}-7.11A_{665.8}$ and total $Chl=21.58A_{649.0}+7.43A_{665.8}$. The chlorophyll extracts of DMF and DMSO were very stable in dark. High chlorophyll contents of citrus leaves were found at $80^{\circ}C$ extraction for 6 hours in DMF and at $80^{\circ}C$ extraction for 24 hours in DMSO. However, the chlorophyll content was decreased significantly after 8 hours in DMF extraction while it was remained up to 30 hours in DMSO extraction.

  • Polarographic behavior of cadmium(II) and copper (II) complexes of 1,5-diphenylcarbohydrazide in dimethylsulfoxide have been investigated by the DC polarography. The reduction processes are estimated as follows; Cd(II)${\cdot}$DPH Complex$\frac{e^-}{(E_{\frac{1}{2}}=-0.12V)}$${\to}$Cd(I)${\cdot}$DPH Complex. Cd(I)${\cdot}$DPH Complex$\frac{e^-}{(E_{\frac{1}{2}}=-0.74V)}$${\to}$Cd(Hg) + nDPH. Cu(II)${\cdot}$DPH Complex$\frac{e^-}{(E_{\frac{1}{2}}=-0.44V)}$${\to}$Cu(I)${\cdot}$DPH Complex. Cu(I)${\cdot}$DPH Complex$\frac{e^-}{(E_{\frac{1}{2}}=-0.84V)}$${\to}$Cu(Hg) + nDPH. The limiting currents of all reduction wave are irreversible. The number of ligand and the dissociation constant for Cu(I)${\cdot}$1.5-diphenylcarbohydrazide complex were found to be 2 and 5.12 ${\times}10^{-8}$, respectively. All reduction waves of complexes are irreversible. Based on the experimental results, the polarographic reductions of complexes in dimethylsulfoxide solution occurred in two one-electron steps.

  • The effect of cryoprotectants and suspending solutions on the preservation of marine heterotophic bacteria was investigated. Six halotolerant and four halophilic bacterial isolates suspended in either distilled water or artificial seawater were preserved in glycerol and dimethylsulfoxide at -70$\^{C}$, respectively. After one year of preservation, the recovery rates on the appropriate agar plates were estimated. The survival rate was found to be dependent on the strain tested, regardless of the preservation conditions tested.

  • Polyphenylenediamine(PPD) film was prepared with dimethylsulfoxide after the synthesis of PPD by chemical polymerization. The molecular structure of conductive polymer synthesized were discussed by using SEM, FT-IR, NMR. The electrical conductivity measurements were carried out at room temperature. The electrical conductivity which was obtained from electrical instrument was 1.98${\times}$10$\^$-2/ S/cm at ambient temperature.

Effect of Solvent on Some Excited States Processes of Mg- and Zn-Phthalocyanines$^\dag$

  • The solvent coordination effect on the excited state processes of Mg(II)- and Zn(II)-phthalocyanines has been described. The triplet state of these compounds decays with mixed first and second order kinetics or mainly second order kinetics depending on the solvents used. The first order component of the rate constants decrease along with the series, dimethylsulfoxide (5-coordinated), 1-chloronaphthalene (4-coordinated) and piperidine (6-coordinated), while the second order rate constant is dependent on the diffusion rate constant of the solvents. The excited state quenching by methylviologen or p-benzoquinone is discussed. And ion recombination rate constant is given.

Cryopreserved Marine Microalgae Grown Using Different Freezing Methods

  • Marine microalgae are a key diet component in finfish and shellfish aquaculture. Cryopreservation of the microalgae is suggested by many other studies as the best method for long-term storage. To test cryopreservation efficacy, 19 taxas of marine microalgal species were examined. In the first experiment we compared dimethylsulfoxide ($Me_2SO$) and glycerol, which are most widely used as cryoprotectant agents (CPAs). The cryopreservation comprised two freezing procedures. Firstly, the samples containing the CPAs were kept at $4^{\circ}C$ for 10 min before being plunged into liquid nitrogen ($-196^{\circ}C$). Secondly, samples containing CPAs were pre-cooled ($-1^{\circ}C$ $min^{-1}$ to $-80^{\circ}C$ before being plunged into liquid nitrogen. Most of the species were successfully cryopreserved using $Me_2SO$, whereas the Prasinophyceae (T. striata and T. suecica) were successfully cryopreserved using glycerol. In general, the cooling method had no influence on the survival of the microalgae except in the case of the Tetraselmis species. In the second experiment, the cultured solution was divided before cryopreservation into concentrated and non-concentrated groups to identify the effect of cell density during cryopreservation. After 12 months of storage, the samples were again divided into centrifugation and non-centrifugation groups to learn the effect of $Me_2SO$ on the culture. Viability and growth of the microalgae were not influenced by cell density or the centrifugal removal of the $Me_2SO$ after thawing.

Radioiodine Labeling of Insulin Using Dimethylsulfoxide as a Labelling-Aid

  • Using dimethylsulfoxide (DMSO) as a labelling aid, insulin--$^{126}$ I of radioimmunoassay use has been effectively prepared. A small amount of DMSO was added to usual labelling mixture ana the reaction time was controled. The labelled insulin obtained in such a way showed improved bindabilities to the antibody and thus expressed larger dose-gradients in the plots of standard dose-response curves even though the labelling rate was decreased to some extent. However, by extending the reaction time to about 1 min, average labelling yield of 30% could be obtained. The average increase of bindability (B/F) in definite antiserum dilution was 2.5 comparing with 1.5 obtained in the absence of DMSO. Thus, the net bindability increase was 70% of those obtained in tile absence of DMSO. By means of a NMR spectrometry, it has been confirmed that the DMSO in the labelling mixtutre is converted to dimethylsulfone by chloramine-T. The results, generally agreed with the Stags's postulation, were discussed in view of a competitive oxidation of DMSO with disulfide linkages of the insulin molecule by the chloramine-T.
