Probe Diffusion and Viscosity Properties in Dimethyl Sulfoxide Solution of Poly(vinyl alcohol) with High Degree of Hydrolysis (고검화도의 폴리(비닐 알코올)/디메틸설폭사이드 용액에서의 점성도 특성과 탐침입자의 확산)

  • Poly(vinyl alcohol)(PVA) with high degree of hydrolysis of above 98% was dissolved in dimethyl sulfoxide(DMSO), and the shear viscosity was measured up to $C{\simeq}0.14\;g/mL$ in the semi-dilute solution regime. Next, as probe particle, polystyrene(PS) latex was introduced into this matrix system and its delayed diffusion due to polymer concentration was investigated by means of dynamic light scattering. When the solution viscosity of PVA/DMSO was plotted against the reduced concentration $C[{\eta}]$, which is scaled by the intrinsic viscosity, the molecular weight dependence was strongly appeared at C$[{\eta}]$ >2. Some heterogeneties in polymer solution were considered as its source. Contrary, the diffusion of probe particle in the matrix solution was observed as a single mode motion at whole concentration range but its ratio of its diffusion coefficient at solution to that at solvent, D/Do did not show any molecular weight dependence at all. However, the application limit of the stretched exponential function was disclosed at C$[{\eta}]$ >2.5.

  • Reaction of monochloroacetic acid with zinc in presence of toluene, dimethylformamide, dimethyl sulfoxide, tetrahydrofuran, acetonitrile, and dioxane solvents showed the solvent effect in order of dimethylformamide, dioxane, dimethyl sulfoxide, toluene, acetonitrile, and tetrahydrofuran. The increasing reactivity of the reagents was observed in order of the polarity and hydrophilicity of the solvent. The same reaction of ethyl monochloroacetate in the same series of solvents showed also the solvent effect, but the difference was not large as compared to that of the acid. The reaction of the acid, ester, zinc, and carbonyl precursors such as benzaldehyde and 4-heptanone gave the Reformatsky reaction product in the case of the aldehyde, but the reactivity with 4-heptanone was slight. The yield of the product was varied by the method of addition of reagents. The best yield observed in case of the acid reaction was 38.5% of the hydroxy acid and 0.8g of the salt in presence of acetonitrile and in case of the ester reaction 19.3% of ethyl cinnamate and 21.6% of polymer. The variation of the reactivity of reagents due to the reaction temperature was observed in case of carbonyl reaction. The result was discussed in terms of the solvent effect and the procedures were described.

The Effect of Dimethyl-Sulfoxide and Sucrose as a Cryoprotectant on the Adenosine Triphosphate and Ultrastructure of Bovine Oocytes Matured In Vitro

  • The present study was undertaken to assess the influence of dimethyl-sulfoxide plus sucrose solution as a cryoprotectant on the adenosine triphosphate (ATP) content, the ultrastructure and the embryonic development of bovine oocytes matured in vitro. We measured the amount of ATP in cumulus cells enclosed oocytes (CO) or denuded oocytes (DO) equilibrated with or removed from the cryoprotectant (1.5 M DMSO + 0.25 M sucrose + 20% fetal bovine serum in physiological saline). As a result, the ATP contents in both CO and DO, equilibrated with the cryoprotectant, were significantly lower (p<0.05) than that of the each control group. However, ATP content of DO was recovered to the level of the control group ailer removal of the cryoprotectant, but failed to restore for CO. In the observation of the ultrastructure by a transmission electron microscope, all of the mitochondria in the ooplasm of CO and DO equilibrated with the cryoprotectant were swollen with peripherally located cristae following decreased electron density. However, a large proportion of these swollen mitochondria were restored to the normal shape which can be observed usually in the control group after removal from the cryoprotectant. To the contrary, the morphology of many mitochondria of the cumulus cells in CO were not recovered to that of the control group after removal of the cryoprotectant. CO with removed cryoprotectant had significantly lower embryonic development up to the blastocysts stage (p<0.05) after in vitro fertilization compared with that in the control group. These results suggest that the addition and removal of a cryoprotectant has a negative effect for the ATP content of cumulus enclosed oocytes. One of the factor(s) causing the lower embryonic development after removal of cryoprotectant, may be associated with ATP metabolism.

Rheological Measurement of Fiber Spinnability of PVA Solution Dopes in DMSO

  • The effects of molecular weight (MW) and concentration on the rheological properties of poly(vinyl alcohol) (PVA) solutions in dimethyl sulfoxide (DMSO) were investigated at $30^{\circ}C$. Ubbelohde viscometer and rotational rheometer were employed for dilute and concentrated regime, respectively. In the dilute regime, the Mark-Houwink exponent ($\alpha$) of the solutions determined from three different MWs proved 0.73. The critical concentration (C*), in which the entanglement and overlap of polymer molecules began to take place, decreased with increasing the MW of PVA. Huggins constant ($K_H$) values ranged from 0.33 to 0.45 over the MW examined. In the log-log plot of $\eta_{sp}$ versus [$\eta$]C, the PVA with higher degree of polymerization (DP) gave a greater slope exhibiting the inflection point in the vicinity of C*. In the dynamic viscosity ($\eta'$) curve, the PVA solutions of DP 1700 presented Newtonian fluid behavior over most of the frequency range examined. However, the lower Newtonian flow region reduced with increasing the DP. As the PVA concentration increased, $\eta'$ was increased and the onset shear rate for pseudoplasticity was decreased. In the Cole-Cole plot, PVA solutions showed almost a single master curve in a slope of ca. 1.65 regardless of the DP. However, the increase of the concentration from 8 to 12 wt% for PVA solutions of DP 5000 decreased the slope from 1.73 to 1.57. In the tan $\delta$ curve, the onset frequency for sol-gel transition was shifted from 154 to 92 rad/s with increasing the DP from 3300 to 5000 and from 192 to 46 rad/s with increasing the concentration from 8 to 12 wt%. In addition, longer relaxation time ($\lambda$) was observed with increasing the DP and concentration.

Activation of Caspase-3 and -7 on Porcine Bone Marrow Derived Mesenchymal Stem Cells (pBM-MSCs) Cryopreserved with Dimethyl Sulfoxide (DMSO) (동결 보호제(DMSO) 농도에 따른 돼지 중간엽 줄기세포의 Caspase 3과 7 발현)

  • Adult stem cell transplantation has been increased every year, because of the lack of organ donors for regenerative medicine. Therefore, development of reliable and safety cryopreservation and bio-baking method for stem cell therapy is urgently needed. The present study investigated safety of dimethyl sulfoxide (DMSO) such as common cryoprotectant on porcine bone marrow derived mesenchymal stem cells (pBM-MSCs) by evaluating the activation of Caspase-3 and -7, apoptosis related important signal pathway. pBM-MSCs used for the present study were isolated density gradient method by Ficoll-Paque Plus and cultured in A-DMEM supplemented 10% FBS at $38.5^{\circ}C$ in 5% $CO_2$ incubator. pBM-MSCs were cryopreserved in A-DMEM supplemented either with 5%, 10% or 20% DMSO by cooling rate at $-1^{\circ}C$/min in a Kryo 360 (planner 300, Middlesex, UK) and kept into $LN_2$. Survival rate of cells after thawing did not differ between 5% and 10% DMSO but was lowest in 20% DMSO by 0.4% trypan blue exclusion. Activation of Caspase-3 and -7 by Vybrant FAM Caspase-3 and -7 Assay Assay Kit (Molecular probes, Inc.OR, USA) was analyzed with a flow cytometer. Both of cryopreserved and control groups (fresh pBM-MSCs) were observed after the activation of Caspase-3 and -7. The activation did not differ between 5% and 10% DMSO, but was observed highest in 20% DMSO. Therefore 5% DMSO can be possibly used for cell cryopreservation instead of 10% DMSO.

Measurement of Polymer Chain Depletion Layer in the Poly(vinyl alcohol)/Dimethyl Sulfoxide/Polystyrene Latex System by Dynamic Light Scattering (동적 광산란법에 의한 폴리(비닐 알코올)/디메틸설폭사이드/폴리스티렌 라텍스 계에서의 고분자 사슬 배제층 측정)

  • The polymer concentration dependence of depletion layer was investigated by means of dynamic light scattering after the very small amount of polystyrene spherical latex particles was added into the matrix solution of poly(vinyl alcohol)(PVA)/dimethyl sulfoxide. At the dilute regime, the magnitude of depletion layer kept constant at the level of $63{\pm}3%$ of the radius of gyration of the corresponding PVA chain. Next, at the early semi-dilute regime of $1.5{\leq}C[{\eta}]{\leq}3$, polymer concentration dependence of the layer thickness ${\delta}$ was obtained as ${\delta}{\sim}C^{-0.8}$, and this experimental value was very close to theoretical one of -0.75. However it was observed above $C[{\eta}]$ >3 that its thickness decreased abruptly, and this was ascribed to aggregation effect of latex particles which was driven by Oosawa type attractive interaction.

Nucleophilic Substitution Reactions of N-Methyl α-Bromoacetanilides with Benzylamines in Dimethyl Sulfoxide

  • Kinetic studies of the reactions of N-methyl-Y-${\alpha}$-bromoacetanilides with substituted X-benzylamines have been carried out in dimethyl sulfoxide at $25.0^{\circ}C$. The Hammett plots for substituent X variations in the nucleophiles (log $k_N$ vs ${\sigma}_X$) are slightly biphasic concave upwards/downwards, while the Bronsted plots (log $k_N$ vs $pK_a$) are biphasic concave downwards with breakpoints at X = H. The Hammett plots for substituent Y variations in the substrates (log $k_N$ vs ${\sigma}_Y$) are biphasic concave upwards/downwards with breakpoints at Y = H. The cross-interaction constant $\rho_{XY}$ values are all negative: $\rho_{XY}$ = -0.32 for X = Y = electron-donating; -0.22 for X = electron-withdrawing and Y = electron-donating; -1.80 for X = electron-donating and Y = electronwithdrawing; -1.43 for X = Y = electron-withdrawing substituents. Deuterated kinetic isotope effects are primary normal ($k_H/k_D$ > 1) for Y = electron-donating, while secondary inverse ($k_H/k_D$ < 1) for Y = electronwithdrawing substituent. The proposed mechanisms of the benzylaminolyses of N-methyl-Y-${\alpha}$-bromoacetanilides are a concerted mechanism with a five membered ring TS involving hydrogen bonding between hydrogen (deuterium) atom in N-H(D) and oxygen atom in C = O for Y = electron-donating, while a concerted mechanism with an enolate-like TS in which the nucleophile attacks the ${\alpha}$-carbon for Y = electronwithdrawing substituents.

A HISTOLOGIC STUDY OF BONE FORMATION ACCORDING TO DIFFERENT CRYOPROTECTANTS DURING CRYOPRESERVATION OF BONE (동결보호제의 종류에 따른 냉동보관자가골의 골형성능에 대한 연구)

  • Purpose: Several cryoprotectants are in use to help the survival of cells during cryopreservation of bone in maxillofacial region. Among them, $Me_2SO$(dimethyl sulfoxide), EG(ethylene glycol), sucrose were used for experimentally created defects with accompanying cryopreserved bone graft in the rabbit model. The aim of this study is to analyze the effect of above mentioned agents on bone formation using histologic and histomorphometrical methods, thus to provide experimental support for clinical application of these agents. Materials and methods: Nine rabbits were used as experimental animals. Surgical defects were created on the distal femoral heads and mesial tibial heads of each animal using trephine drill(5mm diameter and 5mm length). The harvested bones were cryopreserved in $-80^{\circ}C$ refrigerator for one week. The defects were filled with cryopreserved bone with cryoprotectants as experimental groups and cryopreserved bone without cryoprotectant as control. Then, the animals were sacrificed at 1, 2, and 3 weeks after surgery. With Goldner's modified Masson trichrome staining and semiautomatic image analysis system, we observed the change of the cells and bone formation. Results: After bone graft, bone formation and active remodeling process were examined in all experimental groups and the control. But the intensity of such activities of the control were somewhat weaker than that of the experiments. Especially $Me_2SO$+sucrose group was the best in bone formation and bone remodeling. $Me_2SO$ group was more than that of EG group in bone fomation. Sucrose seems to be helpful in survival of the bone cell. Histologic findings showed superior bony quantity and quality in experimental groups than that in control. Conclusions: The data from this study provides the basis for future studies for evaluating the effect of cryoprotectants in the cryopreservation of bone and clinical study for predictable use of these agents.

Measurement of Phase Behavior for Dextran/DMSO/scCO2 System (Dextran/DMSO/초임계 CO2계의 상거동 측정)

  • Micron-sized dextran particles, which now attract wide attention as a promising drug delivery systems, can be prepared via the supercritical anti-solvent (SAS) process. In SAS process, dextran particles are obtained as a result of recrystallization of dissolved dextran in dimethyl sulfoxide (DMSO) on addition of supercritical $CO_2$ as an anti-solvent. In this work, with an intention to provide information on the feasible operating conditions of the process, the phase behavior of Dexran/DMSO/$CO_2$ is observed by measuring the cloud point in favor of a variable volume cell. From the experimental study, it is concluded that a feasible operating condition of the SAS process for preparation of dextran particles would be 300.15 K~330.15 K and 90 bar~130 bar, respectively, and solute concentration ranges from 5mg/ml to 20 mg/ml.

Effect of sec-O-glucosylhamaudol on mechanical allodynia in a rat model of postoperative pain

  • Background: This study was performed in order to examine the effect of intrathecal sec-O-glucosylhamaudol (SOG), an extract from the root of the Peucedanum japonicum Thunb., on incisional pain in a rat model. Methods: The intrathecal catheter was inserted in male Sprague-Dawley rats (n = 55). The postoperative pain model was made and paw withdrawal thresholds (PWTs) were evaluated. Rats were randomly treated with a vehicle (70% dimethyl sulfoxide) and SOG ($10{\mu}g$, $30{\mu}g$, $100{\mu}g$, and $300{\mu}g$) intrathecally, and PWT was observed for four hours. Dose-responsiveness and ED50 values were calculated. Naloxone was administered 10 min prior to treatment of SOG $300{\mu}g$ in order to assess the involvement of SOG with an opioid receptor. The protein levels of the ${\delta}$-opioid receptor, ${\kappa}$-opioid receptor, and ${\mu}$-opioid receptor (MOR) were analyzed by Western blotting of the spinal cord. Results: Intrathecal SOG significantly increased PWT in a dose-dependent manner. Maximum effects were achieved at a dose of $300{\mu}g$ at 60 min after SOG administration, and the maximal possible effect was 85.35% at that time. The medial effective dose of intrathecal SOG was $191.3{\mu}g$ (95% confidence interval, 102.3-357.8). The antinociceptive effects of SOG ($300{\mu}g$) were significantly reverted until 60 min by naloxone. The protein levels of MOR were decreased by administration of SOG. Conclusions: Intrathecal SOG showed a significant antinociceptive effect on the postoperative pain model and reverted by naloxone. The expression of MOR were changed by SOG. The effects of SOG seem to involve the MOR.
