Radioprotective effects of dimethyl sulfoxide in the artificial skin reconstructed with cultured human cells (배양된 사람 세포를 이용해 제작한 인공피부에서 방사선조사에 대한 dimethyl sulfoxide의 보호 효과)

  • Purpose: To evaluate cultured human artificial skin as an experimental model for studying radiation effects in vitro. Materials and Methods: The skin was constructed by culturing keratinocytes over collagen lattice which made by culturing fibroblasts. Two groups were irradiated to gamma rays at single dose of 25 Gy with or without 3.5% of DMSO. Ultrastructures were investigated by electron microscopy after irradiation. The number of epidermal layers and expression of cytokeratin (CK) 14 & 10 were also seem by light microscopy. Results: At 2 days after irradiation in experimental group without DMSO, necrotic cells were rarely found in the spinosal layer and undercornified cells were visible in the homey layer. Similar findings were also found in experimental group with DMSO but in mild form. The number of epidermal layers in experimental group without DMSO were significantly fewer than other group. CK 14 expressed in all the layer excluding homey layer but CK 10 expressed over 3∼4 basal layers. Such patterns of CK expression were similar to all groups. It is suggested that structures of the keratinocytes and epidermal formation could be disturbed by irradiation in artificial skin and that DMSO can protect these damages. Conclusion : Therefore this work could be used as an organotypic experimental model in vitro using human cells for studying radiation effect in skin. Furthermore structural findings provided in this study could be used as useful basic data in further study using this model.

Effects of Dimethyl-sulfoxide on Sperm Cryopreservation of Grass Carp (Ctenopharyngodon idellus)

  • This study was conducted to investigate the effects of three different concentrations (6%, 8% and 10% final volume) of dimethyl sulfoxide (DMSO) on cryopreserved sperm of grass carp (Ctenopharyngodon idellus). Grass carp sperm was suspended in Kurokura extender #2 and equilibrated at $4^{\circ}C$ for 10 min. French straws (0.25 ml) of sperm were frozen from $4^{\circ}C\;to\;-4^{\circ}C$ at a rate of $4^{\circ}C\;min^{-1}$ and then ken $-4^{\circ}C\;to\;-80^{\circ}C$ at a rate of $11^{\circ}C\;min^{-1}$. The straws were kept at $-80^{\circ}C$ for 10 min and finally stored in liquid nitrogen $(-196^{\circ}C)$. The cryopreserved sperm was thawed in a water bath at $40^{\circ}C$ for 30 sec and fertilization, hatching rate and larval malformation were compared with fresh sperm (control). The fertilization rate of post-thawed sperm was comparable (from 88.21% to 94.30%) to that of fresh sperm. However, hatching rate of all frozen sperm were significantly lower (P<0.05) than that of control. Additionally, the larval abnormality rate of frozen sperm was significantly higher than that of fresh sperm. The results indicate that DMSO could affect the quality of cryopreserved sperm of grass carp, and a freezing program and a proper extender composition should be further studied.

A Study on Desorption Efficiency of PAHs according to Desorption Solvents by HPLC with Sonication Extraction (탈착용매에 따른 PAHs 흡착매체의 탈착효율 평가 연구)

  • Objectives: In general, NIOSH method 5506 is most widely used for the occupational exposure measurement of PAHs, but 2-4 ring PAHs have poor desorption efficiency, especially for a filter. The purpose of this study was to determine a method to increase the desorption efficiency of 16-PAHs using an ultrasonic extraction procedure. Methods: Test samples prepared spiked XAD-2 tubes and PTFE filters in the range of $0.01-1.0{\mu}g/mL$ for desorption efficiency study. Four different extraction solvents, acetonitrile, acetone, tetrahydrofuran and dichloromethane, were tested in order to select the most suitable solvent for the extraction of the 16 PAHs. The addition of dimethyl sulfoxide and sonication time were considered in order to determine the method with the highest extraction efficiency. All samples were made in three sets and analysis was replicated seven times by HPLC. Results: Acetonitrile and acetone were the optimized as an extraction solvent and desorption efficiency of 2-ring PAHs such as naphthalene, acenaphthylene were increased 3~19% with dimethyl sulfoxide for XAD-2. Acetone was the best extraction solvent for PTFE filter and the desorption efficiency was increased 3~13% for 2- to 4-ring PAHs. The optimum sonication time was 60 minutes and desorption efficiency increased with extraction time. Conclusions: As a result, the best extraction solvent was acetone with dimethyl sulfoxide for ultrasonic extraction procedure and the desorption efficiency of this method was better than NIOSH 5506's. This study could be applied as a method for occupational exposure measurement of PAHs.

Selection of Cryoprotectants for Cryopreservation of Pearl Oyster, Pinctada fucata martensii Trochophore (진주조개, Pinctada fucata martensii 담륜자의 냉동보존을 위한 동해방지제의 선택)

  • To find out the desirable cryoprotectant for cryopreservation of bivalve trochophores, four types of cryoprotectant were tested with trochophores of the pearl oyster (pinctada fucata martensii) generally used in the pearl production in Korea. Each cryoprotectant (dimethyl sulfoxide, ethylene glycol, glycerol and 1,2-propanediol) was mixed with 0.2 M sucrose to make final concentrations of 0.01, 0.1, 1.0 and 2.0 M. The trochophores were immersed in each preparation, waiting for 10 minutes to reach equilibration and cryopreserved in the liquid nitrogen (-l96$^{\circ}C$). Survival rate of trochophores thawed after cryopreservation increased as the media concentration increase. However, a few number of the trochophores seemed to be damaged with the efflux of cell inclusions. Our study rsults indicate that desirable cryoprotectants for cryopreservation of pearl oyster trochophores are 1.0~2.0 M dimethyl sulfoxide and ethylene glycol : 82.8~97.4% of the trochophores cryopreserved with these media survived after thawing.

Effect of dimethyl sulfoxide on bond durability of fiber posts cemented with etch-and-rinse adhesives

  • PURPOSE. This study was undertaken to investigate whether use of an adhesive penetration enhancer, dimethyl sulfoxide (DMSO), improves bond stability of fiber posts to root dentin using two two-step etch-and-rinse resin cements. MATERIALS AND METHODS. Forty human maxillary central incisor roots were randomly divided into 4 groups after endodontic treatment and post space preparation, based on the fiber post/cement used with and without DMSO pretreatment. Acid-etched root dentin was treated with 5% DMSO aqueous solution for 60 seconds or with distilled water (control) prior to the application of Excite DSC/Variolink II or One-Step Plus/Duolink for post cementation. After micro-slicing the bonded root dentin, push-out bond strength (P-OBS) test was performed immediately or after 1-year of water storage in each group. Data were analyzed using three-way ANOVA and Student's t-test (${\alpha}$=.05). RESULTS. A significant effect of time, DMSO treatment, and treatment${\times}$time interaction were observed (P<.001). DMSO did not affect immediate bonding of the two cements. Aging significantly reduced P-OBS in control groups (P<.001), while in DMSO-treated groups, no difference in P-OBS was observed after aging (P>.05). CONCLUSION. DMSO-wet bonding might be a beneficial method in preserving the stability of resin-dentin bond strength over time when fiber post is cemented with the tested etch-and-rinse adhesive cements.

A New Method for Analysis of Capsaicinoids Content in Microcapsule. (미세캡슐내의 캡사이시노이드의 새로운 분석법)

  • A new method for the quantitative determination of capsaicinoids in microcapsule has been developed. Among seventeen solvents tested for solubilizing wall material (gum arabic and modified starch) of microcapsule, dimethyl sulfoxide (DMSO) was selected as an optimal solvent. The most appropriate mixing ratio of microcapsule to DMSO for solubilizing wall material was 1 to 10(w/v). Appropriate carriersolubilizing temperature and time were $55^{\circ}C$ and 30 min, respectively. Also conditions for extracting oleoresin from the solubilized microcapsule were studied. The mixing ratio of ethanol to DMSO was optimal at 8 to 1(v/v). Optimized vortexing time was 5 min at 40㎐. Pecipitant was obtained by centrifugation at 21000 rpm for 15 min. The precipitant was reextracted with ethanol. The extracted supernatants were combined and adjusted to final volume of 25 ml. Extracted solutions were analyzed for quantitation of total capsaicinoids by employing HPLC and for quantitation of total carotenoids by spectrophotometric method. This method can be used to monitor changes of capsacinoid during manufacturing or storage of red pepper oleoresin microcapsule powder.

Study on Rheological Characterization of Polyacrylonitrile/Dimethyl Sulfoxide Solution with Change of Storage Times and Temperatures (시간 및 온도변화에 따른 폴리아크릴로니트릴/디메틸술폭시드 중합체 용액의 유변학적 특성 연구)

  • In this study, the structural and rheological characterizations of polyacrylonitrile(PAN)/dimethyl sulfoxide(DMSO) solutions for PAN fiber were investigated according to the change of storage times and temperatures. As a result, PAN/DMSO solutions exhibited a very characteristic rheological behavior with variation of temperature. The solutions showed an increase of complex viscosity and a decrease of loss tangent($tan{\delta}$) as temperature was increased over the temperature range of 40 and $70^{\circ}C$ and it could be seen that the viscosity rapidly increased at low frequency. These results indicated that the gel polymer and denser gel structure were formed due to the intermolecular hydrogen bonding of water in the polymer solution depending on the storage time.

Effect of Cryoprotectants on the Cryopreservation of Manila Clam, Ruditapes philippinarum Embryo (바지락 발생배의 냉동보존에 관한 보존액의 효과)

  • The possibility and effectiveness of cryopreservation was determined to assess survival rates and improve stock management of thawed embryos of Manila clam, Ruditapes philippinarum. The ideal freezing rates were designed and tested to allow cryoprotectants to equilibrate across the membrane during freezing. Survival rates ranging from 0 to 64.3% were obtained using a stepwise freezing protocol compared with 82.3% control rates. Embryos of Ruditapes philippinarum were equilibrated in 2 CPAs plus sea water for 10 min at 25℃ and then cooled at -1℃/min from 20℃ to -12℃. Straws containing more than 100 embryos were held at 12℃ for 5 min allowing equilibration after seeding and slowly cooled at 2℃/min. to -35℃ for 30 min for equilibration before quenching in liquid nitrogen. Dimethyl sulfoxide (DMSO) is the best cryoprotectant indicated for embryos of R. philippinarum with a survival rate of 64.3±3.28% in the presence of 2.0 M DMSO.
