Effects of Wastewater Treatment Plants (WWTPs) on Downstream Water Quality and Their Comparisons with Upstream Water Quality in Major Korean Watersheds

  • The purpose of the study was to evaluate spatial and temporal effects of wastewater treatment plants (WWTPs) on the water quality of downstreams (Tan Stream, TS; Daemyeong Stream, DS; Gwangju Stream, GS, and Kap Stream, KS) located in four major watersheds along with impact analysis of nutrient enrichments on the WWTPs during 2004~2008. In the four streams, seasonal means of BOD, COD, TN, and TP were significantly (p<0.01) greater in the downstreams ($D_s$) than the upstreams ($U_s$). The removal effect of nutrients (nitrogen, and phosphorus) from the WWTPs was much less than the BOD, indicating a greater nutrient impact on the downstreams. Seasonal dilution of organic matter, based on BOD, during the summer monsoon of July~September was most pronounced in the downstreams of all four watersheds. However, mean TN in the downstreams during the monsoon varied little in all four streams. Regression analysis of TN in the downstreams against TN from the WWTPs showed that in the TS, and DS regression slopes in the upstreams were similar to the slopes of downstream but there was a significant difference in the GS (p<0.001) and KS (p<0.01). Tan-Stream WWTP showed low removal efficiency of BOD and COD concentrations, compared to the nutrients, whereas, two WWTPs of Gwangju and Kap Stream had low removal effects in TN and TP. Regression analysis of TN and BOD in the downstreams showed that they was closely related (p<0.01) with stream water volume only in the GS. Our data analysis suggests that greater treatment efficiencies of phosphorus and nitrogen from the WWTPs may improve the downstream water quality.

The Acute and Chronic Toxicity Effects of Triorganotins on the Growth of Microalgae, Skeletonema costatum (미세조류, Skeletonema costatum의 성장에 미치는 유기주석 화합물들의 급성 및 만성 독성효과)

  • The use of triorganotins as insecticides, bacteriocides, and antifouling agents has increased dramatically over the past 30 years, In this paper, to evaluate the acute and chronic toxicity effects of various triorganotins (TBTO, TBTE, CTPT, TBTCI) on the growth of microalgae, Skeletonema costatum, we carried out the short term and long term experiments, respectively. In the short-term experiments, the growth of S. costatum was inhibited by the various triorganotins in the dose dependent manners, but recovered by the time laps. The growth inhibition of triorganotins were decreased in the groups with higher initial cell density. These data were resulted from the dilution effect and the biodegradation of triorganotins by the microalgae. In the long-term experiments, S. costatum died completely at the 1 $\mu$g/L of TBTP and CTPT, but didn't in short-term experiments. The facts suggest that the toxicity of the chronic exposure of triorganotins should be increased in the natural sea water with low phytoplankton density.

Antimicrobial and Anti-Biofilm Activities of the Methanol Extracts of Medicinal Plants against Dental Pathogens Streptococcus mutans and Candida albicans

  • Several medicinal plants are ethnomedically used in Korea as agents for treating infection, anti-inflammation, and pain relief. However, beyond typical inhibitory effects on cell growth, little is known about the potential anti-biofilm activity of these herbs, which may help to prevent cavities and maintain good oral health. This study aimed to investigate the antimicrobial and anti-biofilm activities of the methanol extracts of 37 Korean medicinal plants against dental pathogens Streptococcus mutans and Candida albicans, which synergize their virulence so as to induce the formation of plaque biofilms in the oral cavity. The antimicrobial activities were investigated by broth dilution and disk diffusion assay. The anti-biofilm and antioxidant activities were evaluated based on the inhibitory effect against glucosyltransferase (GTase) and the DPPH assay, respectively. Among 37 herbs, eight plant extracts presented growth and biofilm inhibitory activities against both etiologic bacteria. Among them, the methanol extracts (1.0 mg/ml) from Camellia japonica and Thuja orientalis significantly inhibited the growth of both bacteria by over 76% and over 83% in liquid media, respectively. Minimum inhibitory concentration (MIC) values of these methanol extracts were determined to be 0.5 mg/ml using a disk diffusion assay on solid agar media. Biofilm formation was inhibited by more than 92.4% and 98.0%, respectively, using the same concentration of each extract. The present results demonstrate that the medicinal plants C. japonica and T. orientalis are potentially useful as antimicrobial and anti-biofilm agents in preventing dental diseases.

Effect of Lactic Acid Bacteria (Lactobacillus acidophilus, Streptococcus thermophilus, Bsfidobacterium bifidum) on the Enhancement of the Production of Nitric Oxide and TNF-$\alpha$ in RAW 264.7 Macrophage Cell (RAW 264.7 대식세포에서의 유산균에 의한 Nitric Oxide와 $TNF-{\alpha}$의 생성 증가 효과)

  • The immune reinforcement of the probiotic lactic acid bacteria Lactobacillus acidophilus, Streptococcus thermophilus and Bifidobacterium bifidum was studied in RAW 264.7 cell line treated with diluted solution (dilution to $2^{5}$) of the supernatnats of lactic acid bacteria. RAW 264.7 cell line was used as a macrophage model to assess the effects of lactic acid bacteria on the production of nitric oxide (NO) and cytokine tumor necrosis factor (TNF)-$\alpha$ and cell morphological changes. The production of NO and TNF-$\alpha$ were largely affected by lactic acid bacteria in dose-dependent manner in 24 or 48 hr cultures and cell morphological changes were also largely affected by lactic acid bacteria. Especially Bifidobacterium bifidum differentially stimulated the production of NO and TNF-$\alpha$. NO production was increased by Bifidobacterium bifidum 25 $\mu$l/ml more than LPS (20 ng/ml) control, and TW-$\alpha$ by Bifidobacterium bifidum 6.25 $\mu$l/ml more than LPS (10 ng/ml) control. The in vitro approaches employed here should be useful in further characterization of the effects of lactic acid bacteria on systemic immunity.

  • X-ray Fluorescence Spectrographic method has been applied for the rapid determination of main components, such as $SiO_2$, $Al_2O_3$, $Fe_2O_3$, CaO, MgO and $K_2O$ in Silicate Minerals. In this method, Boric Acid was used as a binder after fusion with Lithium Tetraborate in the briquet-making process. The Lithium Flubride, Ammonium di-Hydrogen Phosphate and Ethylene Diamine d-Tartrate crystals were used with Scintillation counter and Gas Flow counter as the detectors. Several influences on this method were discussed, including the particle size of samples and reducing of the matrix effects by dilution with Boric Acid and addition of Lanthanum Oxide with the diluent. In order to test the reproducibility of this method described above, the determination of the same kind of samples were carried out repeatedly, and the results obtained were presented in the table. Calibration curves for each element were presented, and the application of the method was tested with International Rock Standard T-Ⅰ. All the results obtained by X-Ray Fluorescence Spectrographic method were compared with the results by conventional chemical method.

Alkaline Phosphatase Activity in Two Geologically Different Streams in Alabama, U.S.A. (미국 알라바마에서 지질학적으로 다른 두 하천의 Alkaline Phosphatase 활성도)

  • Alakline phosphatase activity (AP A) as a phosphorus deficiency measurement in flowing waters and of microhabitats (rocks, wood, leaves, and sediments) was measured and its relationship to flux of nutrients and response to rainfall events were determined for two geologically different streams in west Alabama from August to November. Results indicated water column AP A in both streams had a low correlation with levels of orthophosphate, total organic phosphorus, nitrate, ammonia, dissolved organic carbon, and discharge (r=0.075-0.583; n=g-IU. Communities on rock surfaces showed a higher AP A level than those on wood and leaves. Sediment passed through a $106{\mu}m$ sieve showed 2-9 times higher AP A level than material passed through $425{\mu}m$ sieve. The first storm after drought at Yellow Creek introduced substantial quantities of DOC (2.5 times baseflow concentrations) and $N0_3-N$ (5.8 times baseflow concentrations) which did not affect AP A significantly. The second storm at Little Schultz Creek caused minor changes in nutrient cocentrations; however $N0_3-N$ levels and AP A were drastically lower due to the dilution effect. Retention of stream water AP A at Yellow Creek and Little Schultz Creek on $0.45{\mu}m$ filter (54 and 43%, respectively) and $0.22{\mu}m$ (83 and 77% of total APA. respectively) indicated more free dissolved portion of the enzyme was present at Little Schultz Creek. Little Schultz Creek (with carbonate and with a higher productivity and biomass) showed a consistantly greater AP A activity $(132{\pm}54\;{\mu}M{\cdot}1^{-1}{\cdot}min^{-I};\;n=g)$ than Yellow Creek $(41{\pm}23\;{\mu}M{\cdot}1^{-I}{\cdot}min^{-I}$, with a sandstone substrate; n=l1, $p{\leq}O.OO1)$. Overall, a greater APA on all microhabitats and the presence of more dissolved enzyme in Little Schultz Creek during the study period may indicates it is more P deficient than Yellow Creek.

  • Three methods for VOCs emissions control in indoor air are reduction at the source, ventilation between indoor and outdoor, and removal. The best alternative should be to replace highly emitting sources with sources having low emissions, but the pertinent information on VOCs is not always available from manufactures. Other ways of improving indoor air quality are needed. It is to increase the outside fresh-air flow to dilute the pollutants, but this method would generally provide only a dilution effect without destruction in residence. An ideal alternative to existing technologies would be a chemical oxidation process able to treat large volumes of slightly contaminated air at normal temperature without additional oxidant such as ozone generator and ion generator. Photocatalytic oxidation(PCO) represents such a process. It is characterized by a surface reaction assisted by light radiation inducing the formation of superoxide, hydroperoxide anions, or hydroxyl radicals, which are powerful oxidants. In comparison with other VOCs removal methods, PCO offers several advantages. The purpose of this study was to explore the possibilities for photocatalytic purification of slightly contaminated indoor air by using visible light such as flurescent visible light(FVL). In this study, a PCO of relatively concentrated benzene using common FVL lamps was investigated as batch type and total volatile organic compounds(TVOCs) using a common FVL lamp and penetrated sun light over window. The results of this study shown the possibility of TiO$_2$ photocatalyst application in the area of indoor air quality control.

Antimicrobial activity of Gynura segetum's leaf extracts and its active fractions

  • $Gynura$ $segetum$ (Lour.) Merr. ($Compositae$) is a well recognized medicinal plant in Indonesia and Malaysia. It is believed to have an anticoagulant effect and is used in treating snake-bites, inflammations and other skin afflictions. This study was carried out to evaluate the antimicrobial effects of $Gynura$ $segetum$ leaves extracts and its fractions. The chemical compositions of the active extracts were also determined. The antimicrobial activities of different solvent extracts of leaves of $Gynura$ $segetum$ were evaluated using the agar well-diffusion method. The Minimum Inhibitory Concentration (MIC) of the active subfractions was determined by the tube dilution method. Gas Chromatography-mass spectrometry (GC-MS) analysis was carried out to identify the chemical compositions of the active extracts. The ethyl acetate fraction and its subfraction E4 performed potent antimicrobial activities and fifteen known chemical constituents were identified by GCMS analysis as 4-vinylphenol, 1-tetradecene, phenol, 2,4-bis(1,1-dimethylethyl), 1-hexadecene, E-15-heptadecenal, hexadecanoic acid, 1,2-benzenedicarboxylic acid, dibutyl ester, 1-docosene, octadecanoic acid, 1-eicosene, cyclotetracosane, 1,2-benzenedicarboxylic acid, bis(2-ethylhexyl)ester, butanedioic acid, monomethyl ester, niacin and 4-hydroxy-benzoic acid. The results of this study suggested a connection between the antimicrobial activities and the chemical structures. The plant may be used as a potential source for antimicrobial agents.

Response of Grain Yield and Milled-Rice Protein Content to Nitrogen Topdress Timing at Panicle Initiation Stage of Rice

  • Response of grain yield and milled-rice protein content to nitrogen topdress (N) timing at panicle initiation stage (PIS) is critical for quantifying real-time N requirement for target grain yield and milled-rice protein content. Two split-split-plot experiments with three replications, one in 2004 and the other in 2005, were conducted in Experimental Farm, Seoul National University, Suwon, Korea. The experiments included three N rates at tillering stage (TS), three N timing treatments at panicle initiation stage (PIS) and two rice cultivars. The N rates at TS, N timing at PIS, and rice cultivars were randomly assigned to main plot, sub plot, and sub-sub plot, respectively. Results showed that the delayed N application at PIS reduced grain yield in 2004 and increased milled-rice protein content in both years significantly at 0.05 probability level. The calculated optimum N timing at PIS from pooled data by N rates and rice cultivars in two years was at 28 days before heading (DBH). However, real-time of N timing at PIS was dependent on plant growth and N status around PIS that in turn was dependent on applied N rates at TS. The optimum N timing at PIS was at 30 DBH for no N treatments at TS while at 27 DBH for 3.6 and 7.2 kg N/10a treatments and at 27 and 29 DBH for Hwaseongbyeo and Daeanbyeo, respectively. In general, earlier applied N at PIS resulted in lower milled-rice protein content but the highest grain yield was expected to be obtained when N topdress at PIS was applied at the time when shoot N concentration started to drop below about 23 mg/g due to dilution effect after transplanting. In conclusion, the results of our experiments imply that the currently recommended N topdress time (24DBH) at PIS in Korea should be reconsidered for the higher grain yield and the better quality of rice.

Dissipation pattern of pesticide residues in/on different varieties of lettuce applied with foliar spraying under greenhouse condition (시설재배에서의 상추품종별 살포농약의 소실양상)

  • The study was carried out to investigate dissipation pattern of pesticide residue in/on different varieties of lettuce applied with foliar spraying under greenhouse. The initial deposited amount of alpha-cypermethrin and imidacloprid in/on the crop was entirely corresponded with shape of the crop. Dissipation of deposited pesticide residue was supposed to be related with degradation by sunlight and wash off by watering rather than dilution effect by biomass increase. The crop grouping in leaf vegetables has to be carefully considered even in lettuce for dissipation pattern of pesticide residue; head type and leafy type.
