Qualitative Study on Consumer Experience of Digestive Enzymes Containing Medicinal Herbs (한약재 함유 소화효소제품 복용에 대한 소비자들의 체험에 관한 질적 연구)

  • Objectives: The aim of this study was to analyze consumer experience with Digestive Enzymes, a digestive enzyme product that includes medicinal herbs (Multi-Zyme®). The goal was to provide basic data on improvements and marketability of future digestive enzyme products containing medicinal herbs. Methods: Qualitative research was conducted to explore the current participants' experience. In-depth interviews with a semi-structured interview guide were conducted to investigate the experience and perception of the participants. The participants were recruited using snowball sampling and purposive sampling, and a qualitative content analysis method was adopted. Credibility was ensured by adopting a member check, triangulation, and peer debriefing method. Results: In-depth interviews were completed with a total of 8 participants. The collected data were classified into 16 codes and then further divided into the following seven categories: direct buying, indirect buying, effects, questions, recommendations, positive perception, and negative perception. The categories were grouped into three themes: 'Experience of purchasing existing health foods', 'Experience of taking Multi-Zyme', and 'Opinions on selling Multi-Zyme at Korean medicine clinics'. Conclusion: The participants experienced effect of the Multi-Zyme® and were willing to recommend it around. However, the Korean medicine doctors need to inform consumers about the dose and duration when taking Multi-Zyme® to prevent abuse. Some consumers may trust Multi-Zyme® sales from Korean medicine clinics, but some held opinions that those sales were not trustworthy, so promotion and improvement are needed.

Seroprevalence of Antigens to Bovine Viral Diarrhea Virus in Korean Calves of the Shown Healthy, Digestive and Respiratory Symptom (한우송아지에서 ELISA를 이용한 소 바이러스성 설사병 바이러스 항원 검출)

  • The aim of this study was to investigate the prevalence of bovine viral diarrhea virus (BVDV) infection in Chonbuk province. Blood samples were taken from 92 korean calves to determined their serological status against BVDV, Capture enzyme-linked immunosorbent assay (ELISA) was used to test for antigen. The number of seropositive calves ranged from 3.3% to 12.9%. Antigens against BVDV were detected in 3.3% of healthy calves, 6.4% of digestive symptom calves, 12.9% of respiratory symptom calves, respectively. Sex and age of calves had no significant differences on the prevalence of BVDV. The results indicate that transmission of BVDV may have become exposed as a result of contact with acute infected or persistently infected cattle.

Effects of first feed intake time on growth performance, nutrient apparent metabolic rate and intestinal digestive enzyme activities in broilers

  • Objective: This experiment studied the effects of first feed intake time post-hatch on growth performance, nutrient apparent metabolic rate and intestinal digestive enzyme activities in broilers. Methods: Two thousand five hundred and twenty LingNan Yellow broilers were randomly allotted to seven treatments with six replicates of 60 each. The only experimental factor was the first feed intake time which was 18, 24, 30, 36, 42, 48, and 54 hours after hatching. The whole experiment lasted for 21 days. Results: During the whole period, the 30 h treatment had the best body weight and average daily gain (p<0.05), followed by the 24 h group performance optimization. Also, the 30 h group was observed to have the best apparent metabolic rate for ether extract (p<0.05) and crude protein (p<0.05) and the highest activities of amylase, lipase and trypsin in small intestine. And the 24 h group was second only to the 30 h group in terms of the above two measures. Conclusion: These results indicated that the appropriate first feeding time of LingNan Yellow broilers was 24 to 30 hours after hatching.

Intestinal Development and Function of Broiler Chickens on Diets Supplemented with Clinoptilolite

  • The purpose of this study was to evaluate the effect of natural clinoptilolite (NCLI) and modified clinoptilolite (MCLI) on broiler performance, gut morphology, intestinal length and weight, and gut digestive enzyme activity. A total of 240 d-old male chicks were randomly assigned to 3 treatments, each of which comprised 8 pens of 10 chicks per pen. Birds in the control group were fed the basal diet, while those in the experimental groups were fed diets supplemented with NCLI at 2% (NCLI group), or MCLI at 2% (MCLI group), respectively, for 42 d. Compared with the control, supplementation with NCLI or MCLI had no significant (p>0.05) effects on productive parameters from d 1 to 42. Supplementation with NCLI or MCLI had no influence on the relative length and weight of small intestine at d 1 to 21. But supplementation with NCLI or MCLI significantly reduced the relative weight of duodenum. Supplementation with MCLI and NCLI was associated with greater (p<0.05) villus height in the jejunal and ileal mucosa compared with those areas in the controls from d 1 to 42. However, supplementation with NCLI and MCLI had no significant (p>0.05) influence on the crypt depth in the jejunal and ileal mucosa compared with those in the controls. The addition of either NCLI or MCLI to the diet improved the activities of total protease, and amylase in the small intestinal contents. In conclusion, supplementation with NCLI or MCLI in diets improved intestinal morphology, increased the intestinal length and weigh and gut digestive enzyme activity.

Effects of Different Levels of Supplementary Alpha-amylase on Digestive Enzyme Activities and Pancreatic Amylase mRNA Expression of Young Broilers

  • Four hundred and forty 1-day-old Arbor Acre broilers were fed commercial starter diets with 0, 250, 750 and 2,250 mg/kg of an alpha-amylase preparation from 1 to 21 days of age to investigate the effects of an exogenous enzyme on growth performance, activities of digestive enzymes in the pancreas and anterior intestinal contents and pancreatic amylase mRNA expression. Body weight gain (BWG) and average daily gain (ADG) increased linearly (p<0.01) with increasing levels of supplementary amylase but feed conversion ratio (FCR) was not affected. There was a negative quadratic change of protease and amylase in the small intestinal contents with the increase of supplementary amylase level. The activity of intestinal trypsin was also increased (p<0.05). Lipase was unaffected by amylase supplementation (p>0.05). The pancreatic protease, trypsin, and lipase were not affected by exogenous amylase levels. Consistent with the tendency for a linear depression of amylase activity, pancreatic ${\alpha}$-amylase mRNA was down-regulated by dietary amylase supplementation. The present study suggested that oral administration of exogenous amylase affected activities of intestinal enzymes and the production of pancreatic digestive enzymes in a dose-dependent manner.

Studies on the Antibacterial Activity of Enzymatic Hydrolyzates of Lactoferrin Derived from Bovine Colostrum (유우 Lactoferrin 효소가수분해물 항균 활성에 관한 연구)

  • The investigative research on the mammalian milk purely consisted of the physiological quality of lactoferrin was conducted to reveal the antimicrobial ativity of specifically functional foods with antibiotic characteristics as a basic data in food manufacturing. Bovine lactoferrin were isolated from raw milk samples, and was digested with pepsin, trypsin and chymotrypsin. It was necessary then to separate and purify lactoferrin from bovine raw milk, and in order to analyze the antimicrobial activity of the enzyme-treated bovine lactoferrin in their required quantitative fraction. Afterwards Escherichia coli and Staphylococcus aureus was incubated in it. It was that investigated to enzyme-treated fractions molecular weight and the peptide fragment with antimicrobial effect. 1. The purity of enzyme-treated bovine lactoferrin(BLF) was tested by SDS-PAGE. As a results of 12% SDS-PAGE assay, pepsin-treated LF did not exhibited band until if reaches 14 KDa, while trypsin and chymotrypsin treated LF, known to contain the non-digestive lactoferrin exhibited band at a molecular weight of 33 KDa. 2. Bovine lactoferrin was sucessfully purified through the use of Sephadex G-50 Column. In order to assay LF through the Sephadex G-50 column chromatography, the digestive bovine lactoferrin (BLFs) was eluted with a linear gradient of 0.05% Tris-HCl. When the gel-filtration analysis, pepsin, trypsin and chymotrypsin treatments of BLF fragments was showed 2, 3, and 2 peak, respectively. The results of the HPLC analysis confirmed that had a non-digestive lactoferrin receptor, and trypsin and chymotrypsin treated BLFs has an antimicrobial effect. 3. To measure the strength of the antimicrobial effect of enzyme treated lactoferrin it was compared to the antimicrobial activity taking place at the incubated Escherichia coli and Staphylococcus aureus. This might explain the resistance of the microorganisms for peptide fragment. The pepsin-treated of bovine lactoferrin was markedly reduced by incubation of the cells. Trypsin-treated of BLF was similar to chymotrypsin-treated of BLF. However, trypsin and chymotrypsin treatments of BLFs were showed the antimicrobial effect until eight hours incubation for native bovine lactoferrin. Therefore the enzyme-treated lactoferrin have an antimicrobial effect even non-digestive lactoferrin. 4. The digestive bovine lactoferrin fragments assay was carried out by the use of Sephadex G-50 column chromatography and SDS-PAGE. The pepsin and chymotrypsin-treated fragments has a low molecular weight and trypsin-treated lactoferrin was only showed a band. It was described that characteristics of digestive protein. It appeared that there may be a relation between virulence and resistance to enzyme-treated BLF.

Substitution of Wheat for Corn in Beef Cattle Diets: Digestibility, Digestive Enzyme Activities, Serum Metabolite Contents and Ruminal Fermentation

  • The objective of this study was to evaluate the effect of diets containing different amounts of wheat, as a partial or whole substitute for corn, on digestibility, digestive enzyme activities, serum metabolite contents and ruminal fermentation in beef cattle. Four Limousin${\times}$LuXi crossbred cattle with a body weight ($400{\pm}10kg$), fitted with permanent ruminal, proximal duodenal and terminal ileal cannulas, were used in a $4{\times}4$ Latin square design with four treatments: Control (100% corn), 33% wheat (33% substitution for corn), 67% wheat (67% substitution for corn), and 100% wheat (100% substitution for corn) on a dry matter basis. The results showed that replacing corn with increasing amounts of wheat increased the apparent digestibility values of dry matter, organic matter, and crude protein (p<0.05). While the apparent digestibility of acid detergent fiber and neutral detergent fiber were lower with increasing amounts of wheat. Digestive enzyme activities of lipase, protease and amylase in the duodenum were higher with increasing wheat amounts (p<0.05), and showed similar results to those for the enzymes in the ileum except for amylase. Increased substitution of wheat for corn increased the serum alanine aminotransferase concentration (p<0.05). Ruminal pH was not different between those given only corn and those given 33% wheat. Increasing the substitution of wheat for corn increased the molar proportion of acetate and tended to increase the acetate-to-propionate ratio. Cattle fed 100% wheat tended to have the lowest ruminal $NH_3-N$ concentration compared with control (p<0.05), whereas no differences were observed among the cattle fed 33% and 67% wheat. These findings indicate that wheat can be effectively used to replace corn in moderate amounts to meet the energy and fiber requirements of beef cattle.

Trypsin Activity in the Digestive Organs and Gastric Evacuation Rate of Litopenaeus vannamei at the Different Rearing Water Temperatures (사육수온에 따른 흰다리새우 Litopenaeus vannamei 소화기관의 trypsin 활성과 배설률)

  • Tryptic enzyme activities in the digestive glands (hepatopancreas) and digestive tracts of Pacific white shrimps Litopenaeus vannamei were assayed at three water temperature regimes. At $26^{\circ}C$, trypsin activity in the hepatopancreas was 200% higher than at $23^{\circ}C$ and 300% higher than at $20^{\circ}C$. The highest foregut trypsin activity levels showed no significant difference in the temperature regimes, but the time between peaks in foreguts and midguts shortened at higher temperature. In the midgut, the level of enzyme activity was highest at $26^{\circ}C$ regardless of the amount of ingested feed. The ratio of foregut and/or midgut dry weight to the body wet weight indicated feed movement through the digestive track directly and gave accurate account about the feeding mechanism. Maximum feed ingestion in the foregut was equivalent to 0.6% of the body weight (wet weight) at $23^{\circ}C$, 0.4% of the body weight at $20^{\circ}C$, and 0.5% of the body weight at $26^{\circ}C$. In view of the temperatures chosen for this study, although maximum ingestion was observed at $23^{\circ}C$, the shrimps showed highest enzyme activity, but lowest feed retention time at $26^{\circ}C$ and lowest enzyme activity, but highest retention time at $20^{\circ}C$.

Changes in Antioxidant Enzyme Activities in the Gill And Digestive Glands of the Manila Clam Ruditapes philippinarum exposed to Cu

  • We investigated the effect of Cu exposure on the activities of protective antioxidant enzymes in the gills and digestive glands of the manila clam Ruditapes phillippinarum exposed to subchronic concentrations (0, 20, 40, and $80{\mu}gL^{-1}$) of waterborne Cu. No mortality occurred during the experimental period, and no significant condition index differences were observed in any exposure group compared with the control. No significant differences were observed in the digestive glands and gills of the clams observed during 15 days of exposure, but after 30 days, the SOD activity in the gill showed a significant difference between the $80{\mu}gL^{-1}$ Cu-exposed group and the control. GPx activities in the digestive glands and gills were significantly lower after 30 days of Cu exposure. Gill GR activity in the high-exposure group ($80{\mu}gL^{-1}$) was significantly elevated compared with that in the control group. GST activities in the digestive glands of all groups did not change over 30 days. However, GST activity in the gill at $80{\mu}gL^{-1}$ Cu was significantly higher after 15 and 30 days of exposure. GSH activities in the gill showed patterns similar to those of GST activities during exposure periods. In the digestive glands, GSH activity was higher only at $80{\mu}gL^{-1}$ after 30 days exposure. In digestive glands and gills, the MDA levels of clams exposed to $80{\mu}gL^{-1}$ Cu were significantly higher after 30 days of exposure.
