Expressions of Tumor-Related Proteins and $TGF-{\beta}1$ in Colon Cancer

  • This study was designed to investigate the correlation between the expression rate of p53 and p21 proteins, c-erbB-2 oncoprotein and $TGF-{\beta}1$ and tumor prognostic factors in colon cancer including the tumor size, histological differentiation and Dukes' stage. The expression rate of p53 protein was 11.4% (4 cases) at well differentiation, 48.6% (17 cases) at moderately differentiation, and 17.1% (6 cases) at poorly differentiation. In other words, the poorer differentiation, the higher the expression rate of p53 protein (P<0.05). The expression rate of p21 protein was 17.1% (6 cases) at well differentiation, 40.0% (14 cases) at moderately differentiation, and 8.6% (3 cases) at poorly differentiation, indicating that, as the histological malignant degeneration progressed, the expression rate of p21 protein decreased distinctively (P<0.05). However, the correlation of the above mentioned proteins with tumor size and Dukes' stage was not recognized. The expression rate of c-cerbB-2 oncoprotein was 11.4% (4 cases) at well differentiation, 54.3% (19 cases) at moderately differentiation, and 17.1% (6 cases) at poorly differentiation, indicating that the poorer differentiation, the higher expression rate of c-erbB-2 oncoprotein (P<0.05). The expression rate of $TGF-{\beta}1$ was 17.1% (6 cases) at well differentiation, 48.6% (17 cases) at moderately differentiation, and 11.4% (4 cases) at poorly differentiation. As Dukes' stage progressed, the expression rate of $TGF-{\beta}1$ was 8.6% (3 cases) in stage A, 20.0% (7 cases) in stage B, 37.1 % (13 cases) in stage C, and 11.4% (4 cases) in stage D. There was a difference in expression rates between Dukes' stages (P<0.05). In 10 cases, p53 protein was positive while p21 protein was negative, and in 6 cases, p53 protein was negative whereas p21 was positive (P<0.05). Therefore, a statistically significant inverse correlation between the expression rate of p53 protein and that of p21 protein was observed. In conclusion, since there was a signigicant correlation between histological differentiation of colon cancer and the expressions of p53 and p21 proteins and c-erbB-2 oncoprotein, and between Dukes' stage and the expression of $TGF-{\beta}1$, it was conformed that the overexpression of p53 and p21 proteins, c-erbB-2 oncoprotein, and $TGF-{\beta}1$ is closely associated with the occurrence of colon cancer and its progress. Accordingly, this study may be greatly beneficial to the presumption of diagnosis, treatment and prognosis of colon cancer patients.

대학생의 자아분화, 자아존중감과 정신건강간의 관계 - 우울, 불안을 중심으로 (A Study on Relationships among University Students' Self-differentiation, Self-esteem and Mental Health : Focused on Depression and Anxiety)

  • The purpose of this study is to examine relationships among university students' self-differentiation, self-esteem and mental health. The subjects were 400 students of four universities in Busan and Gyungnam area. A questionnaire survey was done. The results of this study are as follows : First, the levels of university students' self-differentiation and self-esteem were high. Second, university students' self-differentiation and self-esteem were correlated positively. Third, the levels of university students' depression and anxiety were relatively low. It showed that the subjects' mental health of this study were not bad. Fourth, depression had no significant difference by gender. However, female students had higher anxiety than male students had. Fifth, university students' self-differentiation and self-esteem had negative correlation with depression and anxiety, and self-esteem played a role of mediating variable between self-differentiation and mental health. Sixth, family projection, family regression and self integration of self-differentiation had indirect influence upon the students' mental health through self-esteem, while cognitive-emotional function had direct influence upon mental health and had indirect influence upon mental health through self-esteem as well. In this study, university students' self-differentiation and self-esteem were found to be important variables having influence upon mental health, and self-differentiation had indirect influence upon mental health through self-esteem. Counselling intervention strategies should be established considering self-differentiation and self-esteem of the students who complained about their maladjusted emotion and human relation problems at schools. Also, programs enhancing self-differentiation and self-esteem of university students should be developed and the execution of these programs will be needed to help the university students who experienced mental health problems such as depression, anxiety and so on.

Effect of retinoic acid and delta-like 1 homologue (DLK1) on differentiation in neuroblastoma

  • The principal objective of this study was to evaluate the chemopreventive and therapeutic effects of a combination of all-trans-retinoic acid (RA) and knockdown of delta-like 1 homologue (Drosophila) (DLK1) on neuroblastoma, the most common malignant disease in children. As unfavorable neuroblastoma is poorly differentiated, neuroblastoma cell was induced differentiation by RA or DLK1 knockdown. Neuroblastoma cells showed elongated neurite growth, a hallmark of neuronal differentiation at various doses of RA, as well as by DLK1 knockdown. In order to determine whether or not a combination of RA and DLK1 knockdown exerts a greater chemotherapeutic effect on neuroblastoma, cells were incubated at 10 nM RA after being transfected with SiRNA-DLK1. Neuronal differentiation was increased more by a combination of RA and DLK1 knockdown than by single treatment. Additionally, in order to assess the signal pathway of neuroblastoma differentiation induced by RA and DLK1 knockdown, treatment with the specific MEK/ERK inhibitors, U0126 and PD 98059, was applied to differentiated neuroblastoma cells. Differentiation induced by RA and DLK1 knockdown increased ERK phosphorylation. The MEK/ERK inhibitor U0126 completely inhibited neuronal differentiation induced by both RA and DLK1 knockdown, whereas PD98059 partially blocked neuronal differentiation. After the withdrawal of inhibitors, cellular differentiation was fully recovered. This study is, to the best of our knowledge, the first to demonstrate that the specific inhibitors of the MEK/ERK pathway, U0126 and PD98059, exert differential effects on the ERK phosphorylation induced by RA or DLK1 knockdown. Based on the observations of this study, it can be concluded that a combination of RA and DLK1 knockdown increases neuronal differentiation for the control of the malignant growth of human neuroblastomas, and also that both MEK1 and MEK2 are required for the differentiation induced by RA and DLK1 knockdown.

Muscle differentiation induced up-regulation of calcium-related gene expression in quail myoblasts

  • Objective: In the poultry industry, the most important economic traits are meat quality and carcass yield. Thus, many studies were conducted to investigate the regulatory pathways during muscle differentiation. To gain insight of muscle differentiation mechanism during growth period, we identified and validated calcium-related genes which were highly expressed during muscle differentiation through mRNA sequencing analysis. Methods: We conducted next-generation-sequencing (NGS) analysis of mRNA from undifferentiated QM7 cells and differentiated QM7 cells (day 1 to day 3 of differentiation periods). Subsequently, we obtained calcium related genes related to muscle differentiation process and examined the expression patterns by quantitative reverse-transcription polymerase chain reaction (qRT-PCR). Results: Through RNA sequencing analysis, we found that the transcription levels of six genes (troponin C1, slow skeletal and cardiac type [TNNC1], myosin light chain 1 [MYL1], MYL3, phospholamban [PLN], caveolin 3 [CAV3], and calsequestrin 2 [CASQ2]) particularly related to calcium regulation were gradually increased according to days of myotube differentiation. Subsequently, we validated the expression patterns of calcium-related genes in quail myoblasts. These results indicated that TNNC1, MYL1, MYL3, PLN, CAV3, CASQ2 responded to differentiation and growth performance in quail muscle. Conclusion: These results indicated that calcium regulation might play a critical role in muscle differentiation. Thus, these findings suggest that further studies would be warranted to investigate the role of calcium ion in muscle differentiation and could provide a useful biomarker for muscle differentiation and growth.

Homeostatic balance of histone acetylation and deconstruction of repressive chromatin marker H3K9me3 during adipocyte differentiation of 3T3-L1 cells

  • Background Adipocyte differentiation is completed by changing gene expression. Chromatin is closely related to gene expression. Therefore, its structure might be changed for adipocyte differentiation. Mouse 3T3-L1 preadipocytes have been used as a cell model to study molecular mechanisms of adipogenesis. Objective To examine changes of chromatin modification and expression of histone modifying enzymes during adipocyte differentiation. Methods Microscopic analysis and Oil Red O staining were performed to determine distinct phenotype of adipocyte differentiation. RT-PCR and Western blot analysis were used to examine expression levels of histone modifying enzymes during adipocyte differentiation. Histone modifications were examined by immunostaining analysis. Results Expression levels of P300 and cbp were increased during adipocyte differentiation. However, acetylation of histones was not quantitatively changed postdifferentiation of 3T3-L1 cells compared to that at pre-differentiation. RT-PCR and Western blot analyses showed that expression levels of hdac2 and hdac3 were increased during adipocyte differentiation, suggesting histone acetylation at chromatin level was homeostatically controlled by increased expression of both HATs and HDACs. Tri-methylation level of H3K9 (H3K9me3), but not that of H3K27me3, was significantly decreased during adipocyte differentiation. Decreased expression of setdb1 was consistent with reduced pattern of H3K9me3. Knock-down of setdb1 induced adipocyte differentiation. This suggests that setdb1 is a key chromatin modifier that modulates repressive chromatin. Conclusion These results suggest that there exist extensive mechanisms of chromatin modifications for homeostatic balance of chromatin acetylation and deconstruction of repressive chromatin during adipocyte differentiation.

대학생의 자아분화 정도가 스트레스 수준 및 대처방식에 미치는 영향에 관한 연구 (The Study of the Effect of Self-Differentiation Degree on Stress Level and Stress Coping Strategies in College Students)

  • The purpose of this study is to study how college students cope with their stress and how the level of their self-differentiation affects degrees of stress and stress-coping strategies. The questionnaires were handed to 497 college students at Y university in Gyeongbuk Province and the data were analyzed in terms of Cronbach's ${\alpha}$-coefficient, frequency and percentage, t-test, and regression analysis by using the SPSS statistical package. The results of this study were as follows: 1. For stress level, degree of self-differentiation and stress coping strategies of the college students, the average scores were 3.56, 2.53 and 3.49, respectively in 5-point Likert Scale, indicating that the students possess the above-average score for the degree of self-differentiation and stress-coping strategies and the below-average score for stress levels. 2. From examination of difference verification of stress levels based on the degree of self-differentiation and effect of degree of self-differentiation on stress levels, it was found that the higher is the degree of self-differentiation the lower is the stress level. 3. From examination of difference verification of stress-coping strategies according to the degree of self-differentiation and effect of degree of self-differentiation levels on stress coping strategies, we found that the students of higher degree of self-differentiation exercise the more active stress coping strategies. These results show that the degree of students' self-differentiation is a significant variable that influences their stress levels and stress control methods. It is thought that students' self-differentiation functions to minimize their stress and to actively cope with their stressful situation and that a high degree of self-differentiation is a significant mediator variable that is beneficial to students' mental health. It follows from the above observations that while college students in this study seem to possess good degrees of self-differentiation and stress coping capabilities and low stress levels they need to seek for the ways to improve their degree of self-differentiation by seeing into themselves and relationships with others around them. Parents, schools and consultation agencies are required to take an active hand to educate and encourage them to cope with their stress positively.

Regulation of Actin Gene Expression During the Differentiation of Naegleria gruberi

  • The regulation of actin gene expression during the differentiation of Naegleria gruberi was examined. Actin mRNA concentration was maximal in amoebae and decreased rapidly after the initiation of differentiation. At 20 min after initiation, the concentration of actin mRNA decreased to 55% of the maximal value. The actin mRNA concentration decreased to the minimum at 80 min (15% of the maximum), and then began to increase slightly at the end of differentiation. This decrease of actin mRNA concentration was regulated by the repression of actin gene transcription based on nuclear run-on transcription experiments. The rates of transcription of actin gene in nuclei prepared at 40 and 80 min after the initiation of differentiation were 50 and 28% of that of nuclei prepared at the beginning of differentiation, respectively. The addition of cycloheximide at the initiation of differentiation inhibited both the rapid decrease in the concentration of actin mRNA and the repression of actin gene transcription. These results suggest that the rapid decrease in the concentration of actin mRNA during the differentiation of N. gruberi is accomplished by the repression of actin gene transcription and this transcriptional regulation requires continuous protein synthesis during the differentiation.

대학생의 자아분화 및 거부민감성이 대인불안에 미치는 영향 : 성별에 따른 조절된 매개효과 (The Effects of University Students' Self-Differentiation and Rejection Sensitivity on Interpersonal Anxiety : Moderated Mediating by Gender)

  • The purpose of this study was to model the relations between male and female university students' self-differentiation, rejection sensitivity, and interpersonal anxiety. Questionnaires from 502 university students in Seoul were analysed. The findings are as follows. Firstly, self-differentiation, rejection sensitivity, and interpersonal anxiety were significantly different according to gender. Secondly, the level of differentiation from family regression was higher for both male and female students. And the lower the rejection sensitivity experiencing in vertical relations became, and the lower the rejection sensitivity on horizontal relations was, the lower the interpersonal anxiety became. Thirdly, it was confirmed that for male students, differentiation from family regression affected rejection sensitivity on horizontal relations, and for females, differentiation from emotional reactivity affected ejection sensitivity on both horizontal and vertical relations. Finally, rejection sensitivity played a full mediation parameter when self-differentiation affected interpersonal anxiety, and it was demonstrated differences by gender. This study was meaningful in that it confirmed the relations between male and female university students' self-differentiation, rejection sensitivity, and interpersonal anxiety.

환경변인 및 부모-자녀 유대관계가 농촌 고등학생들의 자아분화에 미치는 영향 (Effect of Environment and Parent-Bodning Relationship on rural high School students' Differentiation of Self)

  • The purpose of this study was to investigate the effect of parent-bonding patterns & environmental variables on rural high school students' self-differentiationf. The subjects were 600 rural high school students in the 2nd grade. The rural high school students' self-differentiation, according to parent-bonding style & environmental variables were researched with DOSS(Differentiation of self Scale) PBI(Parent-bonding Instrument) & environmental Scale. The data were analyzed by frequency & Reliability, ANOVA (scheffe'test) & Regression analysis of SPSS PC+ program. The main results were as follows. First, In P-B patterns generally frequencies of mother's care were higher than father's But in overprotection frequencies of father's were higher. Degree of rural highschool students' self-differentiation was on the level of 2.84 Second, According to parent-bonding patterns, the difference of self-differentiation were as follow. In P-B patters, father and mother on each level of P<.001 brought differences in the self-differentiation of the 4 sub-divisions Third, In short the effect on self-differentiation showed that in family environmental variables, parent's scolding, gender, family atmosphere & health conditions are crucial variables in self-differentiation and in P-B patterns, father's overprotection, mother's care, father's care are all influential on self-differentiation.

Dlx3 and Dlx5 Inhibit Adipogenic Differentiation of Human Dental Pulp Stem Cells

  • Dlx3 and Dlx5 are homeobox domain proteins and are well-known regulators of osteoblastic differentiation. Since possible reciprocal relationships between osteogenic and adipogenic differentiation in mesenchymal stem cells exist, we examined the regulatory role of Dlx3 and Dlx5 on adipogenic differentiation using human dental pulp stem cells. Over-expression of Dlx3 and Dlx5 stimulated osteogenic differentiation but inhibited adipogenic differentiation of human dental pulp stem cells. Dlx3 and Dlx5 suppressed the expression of adipogenic marker genes such as $C/EBP{\alpha}$, $PPAR{\gamma}$, aP2 and lipoprotein lipase. Adipogenic stimuli suppressed the mRNA levels of Dlx3 and Dlx5, whereas osteogenic stimuli enhanced the expression of Dlx3 and Dlx5 in 3T3-L1 preadipocytes. These results suggest that Dlx3 and Dlx5 exert a stimulatory effect on osteogenic differentiation of stem cells through the inhibition of adipogenic differentiation as well as direct stimulation.
