Comparative Biodegradation of HDPE and LDPE Using an Indigenously Developed Microbial Consortium

  • A variety of bacterial strains were isolated from waste disposal sites of Uttaranchal, India, and some from artificially developed soil beds containing maleic anhydride, glucose, and small pieces of polyethylene. Primary screening of isolates was done based on their ability to utilize high- and low-density polyethylenes (HDPE/LDPE) as a primary carbon source. Thereafter, a consortium was developed using potential strains. Furthermore, a biodegradation assay was carried out in 500-ml flasks containing minimal broth (250ml) and HDPE/LDPE at 5mg/ml concentration. After incubation for two weeks, degraded samples were recovered through filtration and subsequent evaporation. Fourier transform infrared spectroscopy (FTIR) and simultaneous thermogravimetric-differential thermogravimetry-differential thermal analysis (TG-DTG-DTA) were used to analyze these samples. Results showed that consortium-treated HDPE (considered to be more inert relative to LDPE) was degraded to a greater extent (22.41% weight loss) in comparison with LDPE (21.70% weight loss), whereas, in the case of untreated samples, weight loss was more for LDPE than HDPE (4.5% and 2.5%, respectively) at $400^{\circ}C$. Therefore, this study suggests that polyethylene could be degraded by utilizing microbial consortia in an eco-friendly manner.

오일/왁스 구조의 열적 특성과 Sweating과의 상관관계 (The Correlation of Sweating of Oil/wax Structure and Thermal Property)

  • Sweating은 오일 중에 수종의 안료를 분산시키고 이들을 왁스로 고화시킨 계의 표면에 출현하는 특이한 현상으로 립스틱이나 커버메이크업 같은 고형 제품에 흔히 나타나며 원인은 불균형한 처방에 기인한다. 그동안 화장품 분야에서 Sweating 현상을 개선하기 위하여 많은 연구가 진행되어 왔다. DSC는 주로 일정 조건 하에서 온도에 따른 시료의 열적 특성의 변화를 측정하는 분석 방법으로 이 변화를 온도 또는 시간의 함수로 기록하는 기기를 말한다. 오일/왁스구조의 열적 특성(thermal property)을 파악하기 위하여 DSC를 이용하였으며, 이러한 열적 특성과 Sweating 현상이 어떠한 상관관계가 있는지 파악하였다. 오일/왁스 제형 중 온도 변화에 따른 구조 변화가 적은 열적 특성을 나타내고 사용감과 제형 안정성을 갖는 최적융점의 오일/왁스 제형을 연구하였다. 또한, 다양한 온도 범위에서 오일/왁스 제형의 Sweating 현상을 관찰하였으며 그 결과 온도 변화에 따른 구조 변화가 적은 오일/왁스 제형이 Sweating 현상도 최소화하는 조합임을 관찰하였다.

  • 본 연구에서는 DGPS 장비를 장착한 개별 차량으로부터 얻어진 주행궤적자료를 활용하여 VMS영향권에서의 운전자 반응특성을 분석하였다. 실험 대상 구간은 서울도시고속도로(올림픽대로)상의 직선부 도로의 VMS를 대상으로 설정하였다. ANOVA TEST를 통하여 VMS 접근거리에 대한 운전자 반응특성(가속도, 속도)을 분석하였으며, 접근 거리에 따른 가속도의 확률밀도함수를 도출하였다. 본 연구의 분석결과는 VMS 설치로 인해 운전자가 체험하는 작업부하가 교통류패턴에 미치는 영향을 도출하는데 활용 될 수 있으며 나아가 이러한 운전자의 반응행태분석은 보다 현실적인 미시적 교통류 모의실험을 위한 기초자료로도 유용하게 사용될 것으로 기대된다.

Development of Protein Biomarkers for the Authentication of Organic Rice

  • The rice protein profiles of Oryza sativa L (Koshihikari) grown under organic and conventional cultivation regimes were compared on 2-D gels to develop diagnostic marker proteins for organic rice. The selected proteins, differentially expressed between organic and conventional rice, were compared with the differentially expressed proteins of another organic and conventional rice pairing, produced at a different location. In the first comparison among conventional, no-chemical, and organic rice grown in the same region, Korea, 13 proteins exhibiting differential expression in organic and conventionally grown plants were selected. Eight of the 13 proteins were down-regulated, and the 5 remaining proteins were up-regulated from conventional to organic rice. The second comparison pairing from Kyungju, revealed 12 differentially expressed proteins, with 8 down-regulated and 4 up-regulated proteins. Ten of the differentially expressed proteins that overlapped between the two comparison sets could not be clustered into any functional group using a functional annotation clustering tool. Further comparisons using another set of conventional and organic rice, belonging to a different variety of Oryza sativa L and produced in Sanchung, revealed 8 differentially expressed proteins, 5 of which were down-regulated and 3 of which were upregulated in the organic rice. Overall, 3 differentially expressed proteins were commonly found in all three organic rice crops. These 3 proteins, along with other overlapping differentially expressed proteins, can provide a good starting point for the development of signature proteins that can be used for the authentication of organic rice with a follow-up studies with more comparison sets.

  • 각종백혈구(各種白血球) 백분비계산방법(百分比計算方法)에는 거이 백종(百種)에 가까울 정도(程度)인데 학자(學者)에 따라서 각각상이(各各相異)하여 각각상이(各各相異)한 백분비(百分比)가 산정(算定)되어 어느방법(方法)을 택(擇)하여야 할것인가 곤란(困難)하다. 여기에서 가장 널리 사용(使用)하고 있는 5종방법(種方法)과 필자(筆者)가 고안(考案)한 13종방법(種方法)을 적용(適用)하여 각혈액도말표본(各血液塗抹標本)에 있는 백혈구(白血球) 실측수(實測數)와 비교검토(比較檢討)한 결과(結果) 도말표본장축(塗抹標本長軸)의 중안선부(中央線部)와 이 중안선부(中央線部)의 양변연(兩邊緣)과의 양(兩) 중안선부(中央線部) 즉(卽) 삼평행선연(三平行線緣)을 종단부(終端部)에서 초단부(初端部)까지 산정(算定)하는 방법(方法)이 비교적(比較的) 이상적(理想的) 방법(方法)이었다. 끝으로 시종강력(始終强力)히 지도편달(指導鞭撻)하여주신 김용필교수(金容珌敎授)에 심심(甚深)한 사의(謝意)를 표(表)하며 선생님(先生任)의 의도(意圖)를 백분발휘(百分發揮)치 못함에 미안(未安)한 마음 금(禁)치 못하며 선배제현(先輩諸賢)의 냉철(冷徹)한 비판(批判)과 지도편달(指導鞭撻)을 바라는바입니다.

Differential Coupling of G$\alpha$q Family of G-protein to Muscarinic $M_1$ Receptor and Neurokinin-2-Receptor

  • The ligand binding signals to a wide variety of seven transmembrane cell surface receptors are transduced into intracellular signals through heterotrimeric G-proteins. Recently, there have been reports which show diverse coupling patterns of ligand-activated receptors to the members of Gq family $\alpha$ subunits. In order to shed some light on these complex signal processing networks, interactions between G$\alpha$q family of G protein and neurokinin-2 receptor as well as muscarinic M$_{1}$ receptor, which are considered to be new thearpeutic targets in asthma, were studied. Using washed membranes from Cos-7 cells co-transfected with different G.alpha.q and receptor cDNAs, the receptors were stimulated with various concentrations of carbachol and neurokinin A and the agonist-dependent release of [$^3H$]inositol phosphates through phospholipase C beta-1 activation was measured. Differential coupling of Gaq family of G-protein to muscarinic M$_{1}$ receptor and neurokinin-2 receptor was observed. The neurokinin-2 receptor shows a ligand-mediated response in membranes co-transfected with G$\alpha$q, G$\alpha$11 and G$\alpha$14 but not G$\alpha$16 and the ability of the muscarinic $M_1$ receptor to activate phospholipase C through G$\alpha$/11 but not G$\alpha$14 and G$\alpha$16 was demonstrated. Clearly G$\alpha$/11 can couple $\M_1$ and neurokinin-2 receptor to activate phospholipase C. But, there are differences in the relative coupling of the G$\alpha$14 and G$\alpha$16 subunits to these receptors.

초등학교 교사의 교실환경 인식에 관한 연구 (A Study on the Elementary School Teachers' Perceptions of Classroom Environments)

  • This study aimed to identify elementary school teachers' perceptions of various aspects of physical environments of a classroom. For the purpose, a survey questionnaire was administered to 982 classroom teachers(female 82.2%) working at 67 elementary schools(Seoul 55%) located in Seoul and Busan. According to the analyses of the survey data, 50.8% of the respondent teachers suggested 20-25 as an optimum class size. Also, 57% of the teachers were positive about the current size of a classroom($67.5m^2$), but evaluations of a classroom size were divided between the teachers who had more and less than 30 students in class. Furthermore, three factors, labelled as 'uniqueness', 'residential stability' and 'visually pleasing', were extracted from a factor analysis of the Semantic Differential Scale consisting of 24 adjectives evaluating a classroom environment. Teachers from Busan tended to rate higher in the uniqueness domain while female teachers scored higher in the domain of residential stability, respectively, compared to their counterparts. Overall, the teachers perceived their classrooms having no particular characteristics, but rated highly in terms of stability. In addition, it was found out that the teachers' requests for the improvement of a classroom environment were not so much as the increased size of a classroom as a reduced class size as well as educational facilities that are corresponding to a variety of instructional methods. In summary, this study confirmed that elementary school teachers' perceptions of a classroom environment varied according to such factors as sex, teaching career, grade of their concerned class, and class size.

Analysis of Structure and Expression of Grapevine 2-oxoglutarate Oxygenase Genes in Response to Low Temperature

  • 2-Oxoglutarate (2OG) acts as a signaling molecule and plays a critical role in secondary metabolism in a variety of organisms, including plants. Six 2-oxoglutarate (2OG) and Fe(II) oxygenase (2OGO) genes, VlCE2OGO1 [Vitis labruscana 2-oxoglutarate (2OG) and Fe(II) oxygenase 1], VlCE2OGO2, VlCE2OGO3, VlCE2OGO4, VlCE2OGO5, and VlCE2OGO6, which show different expression patterns upon transcriptome analysis of 'Campbell Early' grapevine exposed to low temperature for 4 weeks, were analyzed for their structure and expression. Comparison of the deduced amino acid sequences of the 2OGO genes from the V. labruscana transcripts revealed sequence similarities of 38.6% (VlCE2OGO1 and VlCE2OGO2) to 19.2% (VlCE2OGO2 and VlCE2OGO3). The lengths of these genes ranged from 1053 to 2298 bp, and they encoded 316 to 380 amino acids. The prediction of the secondary structure of the encoded proteins by Self-Optimized Prediction Method with Alignment (SOPMA) indicated that all the genes contained alpha helix (23.95 to 41.71%), extended strand (16 to 22.34%), beta turn (6.65 to 9.22%), and random coil (32.97 to 51.58%) in the analysis. Specific primers from unique regions in each gene obtained by alignment of nucleotide sequences were used in real time PCR for analysis of gene expression. All tested genes showed differential expression in grapevines exposed to low temperature. Of the six transcripts, VlCE2OGO1, VlCE2OGO2, and VlCE2OGO3 were up-regulated and VlCE2OGO4, VlCE2OGO5, and VlCE2OGO6 were down-regulated in response to cold treatments at all tested time points. The 2OG genes can be used for elucidation of mechanisms of tolerance to cold and as valuable molecular genetic resources for selection in breeding programs for cold-hardy grapevines.

Molecular cloning of peroxidase cDNAs from dehydration-treated fibrous roots of sweetpotato and their differential expression in response to stress

  • Three peroxidase (POD) cDNAs were isolated from dehydration-treated fibrous roots of sweetpotato (Ipomoea batatas) plant via the screening of a cDNA library, and their expressions were assessed to characterize functions of each POD in relation to environmental stress. Three PODs were divided into two groups, designated the basic PODs (swpb4, swpb5) and the anionic PODs (swpa7), on the basis of the pI values of mature proteins. Fluorescence microscope analysis indicated that three PODs are secreted into the extracellular space. RT-PCR analysis revealed that POD genes have diverse expression patterns in a variety of plant tissues. Swpb4 was abundantly expressed in stem tissues, whereas the expression levels of swpb5 and swpa7 transcripts were high in fibrous and thick pigmented roots. Swpb4 and swpa7 showed abundant expression levels in suspension cultured cells. Three POD genes responded differently in the leaf and fibrous roots in response to a variety of stresses including dehydration, temperature stress, stress-associated chemicals, and pathogenic bacteria.

다양한 기하학적 형상을 갖는 복합 적층쉘 구조의 유한차분해석 (Finite Difference Analysis of Laminated Composite Shell Structures with Various Geometrical Shapes)

  • 본 논문은 전단변형효과를 고려한 복합신소재 적층 쉘을 해석하기 위하여, 일반 쉘의 지배방정식을 유도하고, 이 방정식을 풀기 위하여 수치해석 기법중 하나인 유한차분법을 수행하였다. 유한차분법을 미분방정식을 지배방정식으로 가지는 구조물 해석시 간편하게 사용될 수 있고, 오차의 범위를 선택적으로 정할수 있는 장점이 있다. 수치해석 결과의 타당성을 검증하기 위하여 수렴도 분석과 범용 구조해석 프로그램인 LUSAS의 해석결과와 비교하였다. 본 논문의 목적은 전단변형 효과를 고려한 일반 쉘의 거동 특징 및 분석, 복합재료로 구성되었을 경우 정확한 거동을 분석하고, 쉘 구조물이 보다 높은 강성을 가질 수 있도록 하는 적절한 화이버의 보강방안과 다양한 조건 변화를 통해서 최적의 쉘 구조물을 제시하는 것이다. 쉘의 곡률의 변화에 따른 거동과 합응력의 변화를 분석하고, 쉘의 높이-너비 비와 화이버 보강각도 변화에 따른 처짐 및 합응력의 변화를 비교, 분석하여 보다 유리한 쉘 구조물을 제시하였다. 또한 다양한 하중을 가하여 쉘의 형상 변화를 비교 분석함으로서 비등방성 재료로 이루어진 일반 쉘의 거동에 대하여 분석하였다.
