Comparison of Soyasapogenol A, B Concentrations in Soybean Seeds and Sprouts

  • Soybean seeds contain many biologically active secondary metabolites, such as proteins, saponins, isoflavones, phytic acids, trypsin inhibitors and phytosterols. Among them, saponins in soybeans have attracted considerable interest because of their health benefits. Soyasaponin A and B are the most abundant types of saponins found in soybeans along with soyasapogenol (aglycone), which is a precursor of soyasaponin. The main purpose of this experiment was to determine the concentration of soyasapogenol in soybean seeds and sprouts as a function of seed size, usage, seed coat color and seed cotyledon color. The 79 Korean soybean varieties were cultivated at Yesan of Chungnam in 2006 for the analysis of soyasapogenol using HPLC with Evaporative Light Scattering Detection (ELSD). The total average concentration of soyasapogenol was $1313.52{\mu}g\;g^{-1}$ in soybean seeds and $1377.22{\mu}g\;g^{-1}$ in soybean sprouts. Soybean sprouts were about 5% higher than soybean seeds in average total soyasapogenol concentration. In the process of sprouting, the average soyasapogenol A content decreased by approximately 1.6%, but soyasapogenol B and total soyasapogenol increased by 8.31% and 4.88%, based on the content of soybean seeds. When classified according to the size of seeds, the total soyasapogenol concentration of soybean seeds were not significantly different (p<0.05) On average, small soybean seeds were increased by as much as $103.14{\mu}g\;g^{-1}$ in sprouting process. As a function of the use of the seeds, The total soyasapogenol in soybean seeds were significantly different (p<0.05). While, the soybean sprouts were not significant different (p<0.05). Altogether, sprout soybean seeds show the greatest change in content during the germination process. When seeds with different coat colors were compared, the total soyasapogenol concentration of soybean with yellow seed coats ($1357.30\mu g\;g^{1}$) was slightly higher than that of soybean with black ($1260.30{\mu}g\;g^{-1}$) or brown ($1263.62{\mu}g\;g^{-1}$) seed coats. For the color of the cotyledon, the total soyasapogenol concentration was significantly increased in green cotyledon during the germination and seedling process. The results of this study suggest the functional characteristics of soybeans through quantitative analysis of soyasapogenol. In addition, the concentration of soyasapogenol exhibited a change during the germination process, which was evaluated by the nutritional value of the soybean sprouts.

콩 Lipoxygenase 결핍계통의 가공적성 (Processing Characteristics of Soybean Genotypes Lacking Lipoxygenase)

  • 콩 비린내에 관여하는 lipoxygenase가 결핍된 계통들의 가공적성 및 제품개발의 타당성을 알아보고자 두부와 두유제조시 일반콩과의 특성을 비교 검토한 바 그 결과는 다음과 같다. 1. 두부 제조시 lipoxygenase가 일부 결핍된 수원169호는 일반콩인 황금콩에 비해 수율 및 색도는 비슷하였으나 관능검사는 높게 평가되었다. 2. 두부의 일반조성도 공시재료간에는 비슷하였으나 수원169호가 황금콩에 비해 총당 함량은 높게 나타났다. 3. 실험실 규모에서 두유 제조후 관능검사를 실시한 결과 lipoxygenase 결핍계통이 향기 및 맛에서 우수한 것으로 평가되었다. 4. 공장 규모의 두유제조에서도 수원169호가 황금콩에 비해 두유 가공적성이 우수하였다.

콩에 대한 Calonectria ilicicola 균주의 병원성 변이 (Variability in Virulence of Calonectria ilicicola Isolates on Soybean)

  • Variability in virulence of Calonectria ilicicola isolates form different hosts and geographic origin provides important information for breeding cultivars resistant to red crown rot. A wide range in virulence for 25 isolates of C. ilicicola from soybean and peanut was observed on six soybean cultivars. Soybean isolates were pathogenic on soybean although some were less virulent. Virulence of isolates was not affected greatly by cultivar and relatively consistent regardless of virulence level. Soybean isolates were more virulent on soybean than were peanut isolates. When virulence of two soybean and tow peanut isolates was compared between laboratory and greenhouse tests, it was stable across a range of cultivars. Mycelial growth of isolates from either soybean or peanut was reduced significantly on potato dextrose chlorate showed significant reduction of fungal growth than isolates from peanut did although their growth on potato dextrose agar was not significantly different. Evidence for physiologic specialization was not recognized in this system. However, the findings that soybean isolates of C. ilicicola were more virulent on soybean and reduction of fungal growth on potato dextrose chlorate than were peanut isolates suggest that host specialization may exist in this fugus.

콩 Saponin의 생리활성 기능과 함량변이 (Biological Activities of Soyasaponins and Their Genetic and Environmental Variations in Soybean)

  • There is much evidence suggesting that compounds present in soybean can prevent cancer in many different organ systems. Especially, soybean is one of the most important source of dietary saponins, which have been considered as possible anticarcinogens to inhibit tumor development and major active components contributing to the cholesterol-towering effect. Also they were reported to inhibit of the infectivity of the AIDS virus (HIV) and the Epstein-Barr virus. The biological activity of saponins depend on their specific chemical structures. Various types of triterpenoid saponins are present in soy-bean seeds. Among them, group B soyasaponis were found as the primary soyasaponins present in soybean, and th e 2, 3-dihydro-2, 5-dihydroxy-6- methyl-4H-pyran-4-one(DDMP)-conjugated soyasaponin $\alpha\textrm{g}$, $\beta\textrm{g}$, and $\beta$ a were the genuine group B saponins, which have health benefits. On the other hand, group A saponins are responsible for the undesirable bitter and astringent taste in soybean. The variation of saponin composition in soybean seeds is explained by different combinations of 9 alleles of 4 gene loci that control the utilization of soyasapogenol glycosides as substrates. The mode of inheritance of saponin types is explained by a combination of co-dominant, dominant and recessive acting genes. The funtion of theses genes is variety-specific and organ specific. Therefore distribution of various saponins types was different according to seed tissues. Soyasaponin $\beta\textrm{g}$ was detected in both parts whereas $\alpha\textrm{g}$ and $\beta$ a was detected only in hypocotyls and cotyledons, respectively. Soyasaponins ${\gamma}$g and $\gamma\textrm{g}$ were minor saponin constituents in soybean. In case group A saponins were mostly detected in hypocotyls. Also, the total soyasaponin contents varied among different soy-bean varieties and concentrations in the cultivated soy-beans were 2-fold lower than in the wild soybeans. But the contents of soyasaponin were not so influenced by environmental effects. The composition and concentration of soyasaponins were different among the soy products (soybean flour, soycurd, tempeh, soymilk, etc.) depending on the processing conditions.

Evapotranspiration and Grain Yield in Responses to Different Soil Water Conditions in Soybean

  • This experiment was conducted to evaluate the effect on evapotranspiration and yield of soybean according to different soil water conditions, and to find the optimum time and amount for irrigation in soybean cultivation. The difference between potential evapotranspiration (PET) and maximum evapotranspiration (MET) during growing season of soybean planted in lysimeter was higher during reproductive stage than during vegetative one. The maximum crop coefficient was obtained at beginning seed stage of soybean. Soil water coefficient of irrigation treatment was higher than that of non-irrigation treatment during soybean growth stage in field experiment. Grain yield was highest in lysimeter due to its high water use efficiency and evapotranspiration rate.

Quality Characteristics of Colored Soybean Curd Containing Paprika (Capsicum annuum L.)

  • This study was conducted to examine the quality characteristics of soybean curd containing red paprika juice (RPJ) and green paprika juice (GPJ). The proximate compositions of RPJ showed higher levels of ash, carbohydrate, and vitamin C than GPJ. The yield of soybean curd was not significantly different with the level of RPJ and GPJ. However there was a significant decrease in the pH and an increase in the acidity with the addition of RPJ and GPJ. The L, a, and b values of soybean curd containing RPJ and the L value of soybean curd containing GPJ were significantly different. The hardness and chewiness of soybean curd containing RPJ and GPJ increased significantly with the level of juice.

Evaluation of Soybean Oil as a Lipid Source for Pig Diets

  • An experiment was conducted to determine the effects of soybean oil supplementation replacing tallow in pig diets at different stages of growth. One hundred and twenty crossbred (Landrace${\times}$Yorkshire${\times}$Duroc) pigs weighing 18 kg on average were selected. Pigs were randomly allotted to 12 pens of 10 pigs (5 pigs of each sex) each. Three pens were assigned to each of the four treatments: TA; tallow diet, TA-SO-80; switched from tallow to soybean oil diet at 80 kg average body weight, TA-SO-45; switched from tallow to soybean oil diet at 45 kg average body weight, and SO; soybean oil diet. Treatment SO was significantly lower in ADG than tallow diets (TA, TA-SO-80 and TA-SO-45) during the grower period (18 to 45 kg). However, treatment SO showed greatest compensation in ADFI and ADG during the finisher-2 period (after 80 kg body weight). ADFI and ADG and Gain/Feed for the total period were not significantly different among treatments. Loin area, back fat thickness, firmness and melting point of back fat were not significantly different. The levels of total cholesterol and low density lipoprotein+very low density lipoprotein cholesterol in serum were significantly lower in treatment SO than in treatments TA-SO-45, TA-SO-80 and TA. The level of serum triglyceride linearly increased as the length of the tallow feeding period increased. Serum immunoglobulin-G (IgG) level was significantly higher in the soybean oiltreatment than in other treatments. Major fatty acid composition of short rib muscle and back fat were significantly influenced by treatments. Contents of ${\alpha}$-linolenic acid (C18:3) and docosahexaenoic acid (DHA, C22:6) linearly increased as the soybean oil feeding period increased. In conclusion, soybean oil can be supplemented to the diet of pigs without significant effects on growth performance and carcass characteristics. The level of polyunsaturated fatty acids (PUFA), especially $\omega-3$ fatty acids in the carcass was increased by soybean oil supplementation.

Effect of Different Herbicides on Forage Yield and Nutritive Value in Corn-soybean Mixture Cropping

  • The aim of this study was to evaluate different herbicides in corn-soybean intercropping for optimum growth, yield and nutritive value of forage under Korean environment conditions. Herbicidal treatments were randomly applied over five plots, viz. (1) Control treatment: no herbicide; (2) Alachlor; (3) Simazine; (4) Pendimethalin and (5) Mixture of alachlor and simazine herbicides in RCBD have three replications. Results depicted that treated herbicides exhibited significant effect over control of weeds. Simazine herbicide alone or mixed with alachlor had adverse effects on soybean but enhanced (p<0.05) corn production in terms of survivability, dry matter and digestible nutrients yield. Corn-soybean coupling and total dry matter yield were greater (p<0.05) in pendimethalin. Simazine-alachlor mixture reduced (p<0.05) soybean height. Conclusively, simazine could not be suitable for corn soybean intercropping because of having an adverse effect on soybean component. Pendimethalin could maintain growth and yield of corn and soybean both components. Effect of alachlor was comparable to that of pendimethalin.

유전자변형 콩과 자연 콩의 알레르기 유발원 비교 (Comparison of Allergens in Genetically Modified Soybean with Conventional Soybean)

  • Genetically modified organism (GMO) using recombinant DNA technique has been exponentially increased, however there are still arguments for the safety of GM foods. The objective of this research was to compare the allergens of GM soybean(Roundup Ready$^{TM}$) with conventional soybeans. Each soybean extracts were prepared as crude extracts, heated extracts, and as heated and simulated gastric quid (SGF)-digested samples to characterize the stability of allergens to physicochemical treatment. Positive sera from 20 soybean-sensitive patients and control sera from 5 normal subjects were used to identify the endogenous allergens in soybeans. Specific-IgE binding activities to each soybean preparations were evaluated by ELISA and immunoblot technique. In ELISA result, IgE binding activities of positive sera to soy crude extracts generally showed two fold higher mean value than those of control sera, how-ever there was no significant difference between GM soybean and natural soybean varieties. Extracted proteins form each of the soybean preparations were separated with SDS-PAGE. The band pattern of GM soybean was very similar to those of natural soybean varieties. Immunoblots for the different soybeans revealed no differences in IgE-binding protein patterns, moreover, disclosed five prominent IgE-binding bands (75, 70, 50, 44 and 34 kDa) in crude extracts, four (75, 70, 44 and 34 kDa) in heated preparations, one (50 kDa) in heated and SGF-digested preparations. These IgE binding bands were consistent with previously reported results on the soybean. These results indicate that GM soybean (Roundup Ready$^{TM}$) is no different from natural soybean in terms of its allergen.gen.

건조방법에 따른 비지의 품질변화 (Changes in Quality of Soybean Curd Residue as Affected by Different Drying Methods)

  • This study was carried out to determine the changes in quality during the drying process and the optimum drying condition for utilizing soybean curd residue. The quality criteria for soybean curd residue were acid value, peroxide value, fatty acid composition and microbial concentration. The acid values of soybean curd residue were 7.5, 4.5 and 5.9 KOH mg/g upon 12 hour drying with open-air sun, ambient-air blast and warm-air blast, respectively. The numbers of total aerobic bacteria and molds increased remarkably during drying with open-air sunlight, ambient-air blast and warm-air blast except for hot air blast. Among different drying methods, the hot air blast drying(1kg of sample) was the most effective methods, which completed in three hours. Also, the drying method demonstrated a typical drying curve ; settling down, constant rate drying and falling rate drying period were shown within one hour, from one and three hours and after three hours, respectively. Moreover, there was significant variation in the constant drying period for the quality of soybean curd residue.
