In order to study auricular acupuncture and treadmill exercise involved in high fat diet-induced obesity rats. Thirty Sprague-Dawley rats which were divided into five group. Group I : Normal diet-induced group, Group II : high fat diet-induced group, Group III: high fat diet-induced group and auricular acupuncture, Group IV : high fat diet-induced group and treadmill exercise, Group V : high fat diet-induced group and auricular acupuncture and treadmill exercise. In each group, body weight, serum lipid, histologic obserations, and fat tissue were observed and analyzed. The following results were obtained. Body weights were significantly increase in group II than group I. The level of total cholesterol, LDL-C and TG were significantly lower in experimental groups than group I. Only auricular acupuncture was tended to body weight change but not significantly different between experimental group. The above result suggest that auricular acupuncture and treadmill exercise effectively repressed in obesity. It would be considered that auricular acupuncture and treadmill exercise have an effects on relieving obesity.

The purpose of this study was to evaluate the obesity index and effect of dietary zinc and iron levels on serum trace minerals status in the high fat diet-induced obese rats. Male Sprague-Dawley rats were randomly assigned to control and high fat diet groups. Ten weeks later, the control and high fat diet group were rearranged into six groups by zinc and iron levels. After 16 wk serum zinc, iron, copper and manganese was analyzed. Obesity index was significantly higher in the group fed high fat diet (20% lard) than that of control group (5% corn oil). Body fat content was 12.10$\pm$4.51g/100g BW in high fat diet group and 7.64$\pm$4.18g/100g BW in control group. So, the obese rats were successfully induced by high fat diet. The trace mineral concentration of obese rats in serum were affected by zinc levels. Serum zinc concentration was increased by dietary zinc overload, whereas the iron, copper and manganese were decreased. Specially the manganese concentration was significantly affected by zinc levels. In both groups, serum trace mineral concentration was not changed significantly by the dietary iron levels. There were positive correlations between zinc, iron and manganese concentration according to dietary zinc and iron levels.
Objectives : Obesity is a public health concern associated with chronic diseases including hyperlipidemia, diabetes, fatty liver, atherosclerosis and cancer. As several anti-obesity drugs have been limited owing to their side effects, the development of new anti-obesity drugs through herbal medicines has been increasing. Cynanchum Wilfordii Radix (CW) traditionally is consumed for various health benefits including immune enhancing, anti-inflammation and anti-tumor activities. The aim of the present study is to evaluate the effects of CW on High fat diet (HFD)-induced obese mice. Methods : The mice were randomly divided into four groups (n=7). The mice were respectively fed a normal diet (ND), a high-fat diet (HFD), HFD plus CW (50 mg/kg/day), HFD plus CW (100 mg/kg/day). All groups were assayed for body weights, food efficiency ratio, blood biochemistry parameters, and organic tissue weights. Results : HFD-fed mice showed an increase in the body weight and serum biochemistry parameters levels (total cholesterol and triglycerides) as well as organic tissue weights. However, the administration of CW to obese mice induced a reduction in their body weight, food efficiency ratio, blood biochemistry parameters and weight of liver and fat compared with the HFD fed mice. Additionally, we observed that CW inhibited the lipid accumulation in liver and serum lipid parameter induced by HFD. Conclusions : Taken together, the findings of this study suggest that CW may be a potential agent for use in the treatment of obesity and obesity-related metabolic diseases.

The present study investigated the anti-obesity effects of Mycoleptodonoides aitchisonii (MA) in mice fed a high-fat (HF) diet. Two groups were fed either a normal control diet or an HF (45% kcal fat) diet for 12 weeks and three groups were fed an HF diet supplemented with powdered MA (MAP, 1%, 3%, and 5%) for 12 weeks. The anti-obesity effects of MAP supplementation on body weight, fat mass development, and lipid-related markers were assessed. Consumption of an HF diet resulted in increased body weight, serum lipids, relative adipose tissues weight, and liver fat accumulation. However, administration of MAP significantly decreased body weight gain, food intake, food efficiency ratio, hepatic cholesterol level, and adipose tissue weight in a dose-dependent manner. In addition, treatment with MAP significantly reduced the occurrence of fatty liver deposits and steatosis, and inhibited an HF diet-induced increase in adipocyte size. These results suggest that dietary supplementation with MAP exerts anti-obesity effects and indicate that MAP could be used as a functional food to control obesity.
This study was designed to determine possible weight loss effects of Hangbisan, an oriental medicine, on Sprague-Dawley (SD) rats. SD rats that were fed a high-fat diet for 6 weeks to induce obesity and subsequently fed with a basic diet containing 10%(w/w) Hangbisan or 10%(w/w) cellulose for 8 weeks. The Hangbisan fed rats demonstrated a significantly (p<0.05) reduced weight gain compared to the cellulose fed rats as well as a reduced level of plasma cholesterol and triglycerides and an increased level of (HDL)-cholesterol. These results suggest that dietary Hangbisan has an anti-obesity effect in the high-fat diet-induced obesity (DIG) rat and therefore a potential use as an anti-obesity agent in the application of oriental medicine compounds.
Objectives: The prevalence of obesity continues rise and obesity and metabolic syndrome is a major problem in global health care. Animal models are used in the drug discovery of novel treatment for obesity. One of common models of obesity is a high fat diet induced obesity in a C5BL/6 mouse, and the development of obesity and glucose tolerance in mouse model is different according to period of diet. Therefore, this study was performed to observe the development of obesity and glucose tolerance during a high fat diet (HFD). Methods: Male C57BL/6 mice, 5 weeks of age, were fed on a standard chow diet as a normal diet (18 kcal% fat) or a HFD (60 kcal% fat) for up to 16 weeks. The various factors related with obesity and insulin resistance were measured at 8, 12, and 16 weeks. Results: The weights of body and epididymal fat were gradually increased for 8~16 weeks, however the change of hyperglycaemia and glucose tolerance have shown different with that of body weight. Blood glucose, oral glucose tolerance and insulin tolerance were increased more clearly at week 12 and 16 than week 8. Lipid accumulation of liver and body temperature were also significantly increased at week 16, compared with normal group. Conclusions: The developments of obesity and related factors were different by a HFD period in a C57BL/6 obese mice. This result suggests that the development of obesity with glucose tolerance and liver lipid may induce clearly by a HFD for 16 weeks.
Objectives: This study investigated the effects of water extract of Aconitum carmichaeli Debx (ACD) on obesity and glucose tolerance in high fat diet induced obese mice. Methods: Five-week-old C5BL/6 mice were fed a high fat diet (HFD) containing or not containing ACD (100 or 300 mg/kg) for 16 weeks. Body weight, food intake, fasting blood glucose, and body temperature were checked every week and then organs, blood serums were collected after 16-week treatment. Furthermore oral glucose tolerance test (OGTT) was carried out after treatment. Results: ACD treated mice showed no significant decreases in body weight and adipose tissue weight as compared with HFD mice. Lipid accumulations in liver and serum lipid levels were not different between ACD treated and HFD mice. However, ACD extract administration significantly and dose-dependently reduced fasted plasma glucose and glucose tolerance as determined by OGTT. Conclusions: The present study demonstrates that ACD might prevent diet-induced glucose tolerance in mouse models of obesity.

Objectives: The objective of this study was to explore the anti-obesity effect of Cydonia oblonga Miller fruit extract (COME) and to compare its anti-obesity efficacy with Garcinia cambogia extract (GCE) in diet-induced obese mice. Methods: Five-week-old male C57BL/6 were allocated into four groups: control diet (CD), high-fat diet (HFD), HFD + 400 mg/kg body weight (BW)/day COME (H+C), or HFD + 400 mg/kg BW/day GCE (H+G) groups. COME or GCE was administered once a day by oral gavage for eight weeks. Body weight, body fat percentage, fat weight, and biochemical parameters in serum were measured. The expressions of transcription factors and their target genes in epididymal adipose tissues were analyzed by reverse transcription polymerase chain reaction. Results: COME reduced body weight, weight gain, body fat percentage, total white adipose tissue weight, adipocyte size, and serum levels of insulin and leptin in high-fat diet-induced obese C57BL/6 mice. COME suppressed the mRNA expressions of CCAAT/enhancer binding proteinα, peroxisome proliferator-activated receptorγ, sterol-regulatory element-binding protein-1c, fatty acid synthase, and adipocyte protein 2 and increased carnitine palmitoyl transferase 1 mRNA expression in epidydimal adipose tissues. The anti-obesity efficacy of COME was found to be similar to that of GCE at the same dose. However, COME more effectively decreased adipose tissue weights, epididymal adipocyte size, serum insulin and leptin compared to GCE. Conclusions: These results demonstrated that COME is not toxic and exhibits anti-obesity efficacy at a level similar to that of GCE, suggesting that COME may be applicable as an anti-obesity agent.
Purpose: Ketogenic diets (KDs) have anti-obesity effects that may be related to glucose control and the gut microbiota. This paper hypothesizes that KD reduces body weight and changes the insulin sensitivity and gut microbiota composition in a mouse model of diet-induced obesity. Methods: In this study, C57BL/6 male mice were assigned randomly to 3 groups. The assigned diets were provided to the control and high-fat (HF) diet groups for 14 weeks. The KD group was given a HF diet for 8 weeks to induce obesity, followed by feeding the KD for the next 6 weeks. Results: After the treatment period, the KD group exhibited a 35.82% decrease in body weight gain compared to the HF group. In addition, the KD group demonstrated enhanced glucose control, as shown by the lower levels of serum fasting glucose, serum fasting insulin, and the homeostatic model assessment of insulin resistance, compared to the HF group. An analysis of the gut microbiota using 16S ribosomal RNA sequencing revealed a significant decrease in the proportion of Firmicutes when the KD was administered. In addition, feeding the KD reduced the overall alpha-diversity measures and caused a notable separation of microbial composition compared to the HF diet group. The KD also led to a decrease in the relative abundance of specific species, such as Acetatifactor_muris, Ligilactobacillus_apodemi, and Muribaculum_intestinale, compared with the HF group. These species were positively correlated with the body weight, whereas the abundant species in the KD group (Kineothrix_alysoides and Saccharofermentans_acetigenes) showed a negative correlation with body weight. Conclusion: The current study presents supporting evidence that KD reduced the body weight and altered the insulin sensitivity and gut microbiota composition in a mouse model of diet-induced obesity.
Obesity is an epidemic disease characterized by an increased inflammatory state and chronic oxidative stress with high levels of pro-inflammatory cytokines and lipid peroxidation. Moreover, obesity alters cholesterol metabolism with increases in low-density lipoprotein (LDL) cholesterols and triglycerides and decreases in high-density lipoprotein (HDL) cholesterols. It has been shown that mulberry leaf and fruit ameliorated hyperglycemic and hyperlipidemic conditions in obese and diabetic subjects. We hypothesized that supplementation with mulberry leaf combined with mulberry fruit (MLFE) ameliorate cholesterol transfer proteins accompanied by reduction of oxidative stress in the high fat diet induced obesity. Mice were fed control diet (CON) or high fat diet (HF) for 9 weeks. After obesity was induced, the mice were administered either the HF or the HF with combination of equal amount of mulberry leaf and fruit extract (MLFE) at 500mg/kg/day by gavage for 12 weeks. MLFE treatment ameliorated HF induced oxidative stress demonstrated by 4-hydroxynonenal (4-HNE) and modulated the expression of 2 key proteins involved in cholesterol transfer such as scavenger receptor class B type 1 (SR-B1) and ATP-binding cassette transporter A1 (ABCA1) in the HF treated animals. This effect was mainly noted in liver tissue rather than in cutaneous tissue. Collectively, this study demonstrated that MLFE treatment has beneficial effects on the modulation of high fat diet-induced oxidative stress and on the regulation of cholesterol transporters. These results suggest that MLFE might be a beneficial substance for conventional therapies to treat obesity and its complications.
