A Study on the Characteristics and Incidences of T. hyodysenteriae on Pigs. (돈적병균의 분포와 특성에 관한 연구)

  • Feces were collected from pigs of various ages on slaughter house and swinery in the southern area of the kangwon-do. Treponema hyodysenteriae were isolated from the feces by the method of Jenkinson. and the effects of temperature on the survival of these organism were investigated. The results abtained are summerized as follows ; 1. T. hyodysenteriae were isolated from 22 fecal samples of 1, 296 samples, and showed higher incidence at 4-8week age groups than other age groups. 2. The organisms were isolated from 17 samples of 329 diarrhea samples and 5 samples of 967 non-diarrhea samples. 3. T hyodysenteriae survived long at $0-10^{\circ}C$ but survived short at $37^{\circ}C$ or above.

CQI Activities for the Reduction of Clostridium difficile Associated Diarrhea in NCU of a University Hospital (일개 대학병원 신경외과중환자실에서 Clostridium difficile 관련 설사 감소를 위한 CQI활동)

  • Background : The Clostridium difficile is the most important identifiable cause of nosocomial infectious diarrhea and colitis, which lengthens hospital stay. Recently incidence of C. difficile has been increasing in an university hospital, and an intervention for prevention and control of C. difficile associated diarrhea (CDAD) was in prompt need. Methods : Subjects were the patients in the neurosurgical intensive care unit(NCU) where C. difficile was most frequently isolated. To increase participation of various departments, we used the CQI method, because management of CDAD requires a wholistic approach including control of antibiotics, barrier precaution and environmental cleaning and disinfection. Duration of the CQI activities was 9 months from April to December 1999. Results : The identified problems were misuse and overuse of antibiotics, lack of consciousness of medical personnels and the possibility of transmission from the contaminated environment and tube feeding. Education for proper use of antibiotics and management of C. difficile infection, use of precaution stickers, supplement of handwashing equipments, emphasis on environmental disinfection, and the change of the process of tube feeding were done. The CDAD rate in NCU was significantly decreased after the CQI program (8.6 case per 1,000 patient days from January to April 1999 vs 4.8 from May to December 1999). The distribution of neurosurgical wards including NCU among the total number of isolated C. difficile from the clinical specimens dropped from 49.4% in January to April to 33,7% in May to December. The average hospital stay of the neurosurgical department changed from 19.6 days to 15.2 days. Also, the effect of the CQI activities for C. difficile may have affected the incidence of vancomycin resistant enterococci (VRE). Duration and dosage of certain antibiotics used in the NS department were decreased. The distribution of neurosurgical department in the number of VRE isolated patients declined from 18.4% to 11.1%. Conclusion : Infection control of resistant organisms such as C. difficile is likely to be successful when management of environmental contamination an collaborative efforts of decreasing the patients' risk factors such as antibiotics management and decreasing the length of hospital stay come simultaneously. For this work, related departments need to actively participate in the entire process under a common target through discussions for identifying problems and bringing up solutions. In this respect, making use of a CQI team is an efficient method of infection control for gathering participation and cooperation of related departments.

Cell adaptation of KPEDV-9 and serological survey on porcine epidemic diarrhea virus(PEDV) infection in Korea (돼지 유행성설사바이러스 국내 분리주(KPED-9) 의 세포증식성 및 혈청학적 역학조사)

  • Korean isolate, porcine epidemic diarrhea virus (KPEDV-9) was adapted through serial passages in vero cell. The viral yield reached up to $10^{5-6}$ $TCID_{50}/ml$ at the passage level of 90th. The cell adapted virus was characterized through genetic and morphological examinations. The RNA extracted from virus infected cell revealed the presence of RNA band with molecular size of >20Kb. The electron microscopic examination on purified virus showed the pleomorphic appearance of enveloped particles with 5-10nm surface projections, which fit with the shape of coronavirus. The etiological survey on swine diarrhea by immunofluorescence test(FA) indicated 17.5% positive rate on the PEDV infection. In addition, the incidence were detected both in piglets within two weeks old as well as fattening pigs. Serological survey by ELISA revealed the overall 45% positive result, thus, indicating the PEDV infection are widespread throughout this country.

Experimental infection of Campylobacter jejuni and Campylobacter coli in chicks (유추(幼雛)에 있어서 Campylobacter jejuni 및 Campylobacter coli의 실험적(實驗的) 감염(感染))

  • The present study was carried out to investigate the incidence of C jejuni and C coli in chicken. Also were examined the pathogenicity of the isolates in chick by experimental inoculation. Thermophilic Campylobacter were isolated from 34(45.9%) of the 74 specimens, and classified as 21.6% C jejuni, and 24.3% C coli. In the biotyping of 16 stranis of C jejuni isolates, 37.5% of the strains were grouped as biotype I, 62.5% as biotype II. In the case of 18 strains of C coli isolates, 49.9% of isolates were grouped as biotype I, 55.6% as biotype II. n oral inoculation with $10^4cfu$ of Campylobacter isolates into infant chicks(1 to 3 days-old), 17 days-old and 34 days-old chicks, 32.5% of the chicks developed diarrhea on day 1, 52.5% on day 3, 70.0% on day 5, and 27.5% on day 7, and the peak incidence of diarrhea was reached on day 5. The organisms were found to be discharged in feces one day afterwards. C jejuni and C coli strains were detected from the feces in 87.5% of the chicks on day 5. The organisms were multiplied from $10^4$ to $10^6cfu/gm$ in feces 5 to 7 days after inoculation. C jejuni and C coli recovered from 100% of the cecum, 64.3% of the duodenum, 50.0% of the spleen, 42.9% of the livers, and from 21.4% of gallbladders 7 days after inoculation.

Protective Effect of Taurine on TNBS-induced Inflammatory Bowel Disease in Rats

  • We had previously reported that the protective effect of taurine against indomethacin-induced gastric mucosal injury was due to its antioxidant effects, which inhibited lipid peroxidation and neutrophil activation. In this study, we examined the effect of taurine on reducing the inflammatory parameters of trintrobenzene sulfonic acid (TNBS)-induced inflammatory bowel disease (IBD) in rats. In order to induce IBD, ethanolic TNBS was given to rats intracolonically. Then they received 500 mg/kg.day of taurine orally and were sacrificed one week after IBD induction. While ulceration and inflammation of distal colon with formation of granuloma in the vehicle-treated IBD rats two days after administration of TNBS were observed, treatment with taurine ameliorated colonic damage and decreased the incidence of diarrhea and adhesion. also, colon weight as an index of tissue edema, which was mardedly increased in the IBD rats, became significantly lower after administration of TNBS were observed, treatment with taurine ameliorated colonic damage and decreased the incidence of diarrhea and adhesion. Also colon weitht as an index of tissue edema, which was markedly increased in the IBD rats, became significantly lower after taruine treatment. Myeloperoxidase (MPO) activity in the vehicle-treated IBD rats was substantially increased, compared with that of normal control. the taurine-treated animals significantly reduced MPO activity (35% lower) when compared with that of the vehicle-treated animals. Taurine treatment decreased both basal and formyl-methionyl leucyl phenylalanine-stimulated reactive oxygen generation from colonic tissue in the IBD rats. These results suggest that the administration of taurine reduce the inflammatory parameters in this IBD rat model by increasing defending capacity against oxidative damage.

Effects of soybean meal fermented by Bacillus coagulans NRR1207 and kefir on the feeding characteristics of weaned HANWOO calves and Holstein cows

  • This study was conducted to evaluation the effects of dietary soybean meal (SBM) and fermented soybean meal (FSBM) on the growth performance of Korean native cattle as Hanwoo calves (Bos taurus coreanae) and Holstein cows. In total, 16 calves three to four months old (Control group, SBM: 5 males and 3 females, average weight 105.7 kg; Experimental group, FSBM: 5 males and 3 females, average weight 103.7 kg) were fed 5% of the feed amount for 53 days. In terms of growth performance outcomes, the average gain body weight was significantly higher in the FSBM group than in the SBM group in the final fattening period. The average daily gain in body weight (ADGBW) for the FSBM group was higher than that of the SBM group in the final fattening period. The average gain body weight in four months for FSBM was higher than that at three months in the final fattening period. Diarrhea incidence for FSBM was significantly decreased compared to that in the SBM group in the six-week period after weaning. These results indicate that dietary FSBM can improve the growth rate and health condition during the calving period. Holstein cows fed fermented soybean meal had higher milk urea nitrogen levels and decreased somatic cell counts compared to those fed SBM. These results may be closely related with the increased average daily gain body weight associated with dietary FSBM.

Nationwide Multicenter Study of Eosinophilic Esophagitis in Korean Children

  • Purpose: In East Asian countries, there are only a few epidemiologic studies of eosinophilic esophagitis (EoE) and no studies in children. We investigated the incidence and compared the clinical characteristics of EoE and eosinophilic gastroenteritis involving the esophagus (EGEIE) in Korean children. Methods: A total of 910 children, who had symptoms of esophageal dysfunction, from 10 hospitals in Korea were included. EoE was diagnosed according to diagnostic guidelines and EGEIE was diagnosed when there were >15 eosinophils in the esophagus per high power field (HPF) and >20 eosinophils per HPF deposited in the stomach and duodenum with abnormal endoscopic findings. Results: Of the 910 subjects, 14 (1.5%) were diagnosed with EoE and 12 (1.3%) were diagnosed with EGEIE. Vomiting was the most common symptom in 57.1% and 66.7% of patients with EoE and EGEIE, respectively. Only diarrhea was significantly different between EoE and EGEIE (p=0.033). In total, 61.5% of patients had allergic diseases. Exudates were the most common endoscopic findings in EoE and there were no esophageal strictures in both groups. The median age of patients with normal endoscopic findings was significantly younger at 3.2 years, compared to the median age of 11.1 years in those with abnormal endoscopic findings (p=0.004). Conclusion: The incidence of EoE in Korean children was lower than that of Western countries, while the incidence of EGEIE was similar to EoE. There were no clinical differences except for diarrhea and no differences in endoscopic findings between EoE and EGEIE.

Risk and Protective Factors for Gastrointestinal Symptoms associated with Antibiotic Treatment in Children: A Population Study

  • Purpose: Gastrointestinal symptoms are often related to antibiotic treatment. Their incidence, risk and protective conditions in children are not well defined and represent the aims of this study. Methods: We prospectively enrolled inpatient children submitted to antibiotic treatment. Indication, type, dose and duration of treatment, probiotic supplementation and gastrointestinal symptoms were recorded at recruitment, after two and four weeks. Antibiotic-associated diarrhea (AAD) was defined as the presence of at least 3 loose/liquid stools within 14 days from antibiotic onset. Results: AAD occurred in 59/289 (20.4%) of patients, with increased risk in children younger than 3 years (relative risk [RR]=4.25), in lower respiratory (RR=2.11) and urinary infections (RR=3.67), intravenous administration (RR=1.81) and previous AAD episodes (RR=1.87). Abdominal pain occurred in 27/289 (9.3%), particularly in children >6 years (RR=4.15), with previous abdominal pain (RR=7.2) or constipation (RR=4.06). Constipation was recorded in 23/289 (8.0%), with increased risk in children having surgery (RR=2.56) or previous constipation (RR=7.38). Probiotic supplementation significantly reduced AAD (RR=0.30) and abdominal pain (RR=0.36). Lactobacillus rhamnosus GG (LGG) and L. reuteri significantly reduced AAD (RR=0.37 and 0.35) and abdominal pain (RR=0.37 and 0.24). Conclusion: AAD occurred in 20.4% of children, with increased risk at younger age, lower respiratory and urinary tract infections, intravenous treatment and previous AAD. LGG and L. reuteri reduced both AAD and associated abdominal pain.

Effects of Copper-bearing Montmorillonite (Cu-MMT) on Escherichia coli and Diarrhea on Weanling Pigs

  • Copper-bearing montmorillonite (Cu-MMT) was produced by $Cu^{2+}$ cation exchange reaction. X-ray diffraction analysis showed that that the (001) basal spacing of the MMT crystal lattice increased from 1.544 to 1.588 nm after $Cu^{2+}$ exchange. This indicated that $Cu^{2+}$ entered into interlayer position of MMT as a hydrated cation or composite cation. In vitro results indicated that Cu-MMT had antibacterial activity on Escherichia coli $K_{88}$. Cu-MMT had unbalanced positive charge after cation exchange. Its antibacterial activity resulted from two aspects, one was electrostatic attraction which made E. coli $K_{88}$ being adhered on the montmorillonite surface, the other was the $Cu^{2+}$ slowly released, which could kill bacteria. In an in vivo study, four replicates of eight weanling pigs were assigned to each of two dietary treatments to study the effects of Cu-MMT on diarrhea, E. coli in the lumen of the jejunum and morphology of jejunal mucosa. As compared to the control, supplementation of the diet with 0.2% Cu-MMT improved average daily gain by 12.50% (p<0.05) and decreased F/G by 9.42% (p<0.05). The mean diarrhrea incidence was decreased by 71.80% (p<0.05). The viable counts of Escherichia coli in jejunal contents were significantly reduced (p<0.05). Villus height and the villus height to crypt depth ratio at the jejunal mucosa were increased by 19.09% (p<0.05) and 37.10% (p<0.05), respectively.

Performance of Hanwoo calves fed a commercial colostrum replacer versus natural bovine colostrum

  • Supplying a sufficient amount of high-quality colostrum is a prevailing challenge in Hanwoo calf production systems. This study aimed to describe the efficiency of a lacteal-derived colostrum replacer versus natural bovine colostrum (from Holstein) fed to Hanwoo calves. Forty newborn Hanwoo calves (25 males and 15 females; 30.7 ± 3.15 kg body weight [mean ± standard deviation]) were blocked by sex and birth weight and fed either natural colostrum or a commercial colostrum replacer. Calves displayed a narrow difference in the evolution of body weight and structural body dimensions when they received a commercial versus natural colostrum. Minor differences existed in the starter and total dry matter consumption between calves fed natural or commercial colostrum. Although the colostrum source had no significant effects on days to first diarrhea (average of 7.6 days; p = 0.17), the duration of diarrhea was longer in natural colostrum-fed calves (3.2 vs. 4.5 days; p = 0.04). Consumption of natural colostrum resulted in greater mean serum immunoglobulin G (IgG) on day 2 (14.7 vs. 10.8 ± 0.92 mg/mL) and day 7 (8.21 vs. 5.12 ± 0.82 mg/mL) of calf life, resulting in a greater proportion of commercial colostrum-fed calves (50% vs. 15%; day 2) to experience failure of passive transfer of immunity (serum IgG < cutoff point of 10 mg/mL). Overall, growth rate, body frame development, and incidence of diarrheal disease were not different in both groups. These results suggested that the colostrum replacer product tested in this study could be an alternative to natural colostrum derived from Holstein cow in securing calf growth and health in Hanwoo calf-rearing operations.
