Dyeing Properties of Microbial Violacein on Mutifiber Fabrics (미생물 violacein 색소의 다섬교직포에서의 염색성)

  • Dyeability of microbial violacein produced from Chromobacterum violaceum CV107 on to multifiber fabrics has been studied. The bluish-purple colourants were produced by cultivation of Chromobacterum violaceum using LB liquid medium for 2 days. The colourant was extracted with 80% acetone and identified as violacein by LC/MS analysis. The violacein could be dyed on not only natural fibers such as Cotton, Silk and Wool but also synthetic fibers such as Diacetate, Triacetate, Creslan 61 and Nylon 66. Maximum K/S values were shown at 540-580 nm according to different fiber with color appearance of purple or blue. An optimum pH and temperature under dyeing condition were 10 and $70^{\circ}C$, respectively. Any mordants were not improved colour density and quality on various fabrics. From this studies, pigments produced microbe have a high potentials for natural dyeing on fabrics. Finally, development of new colourants from microbe has made a possible change for new dyeing field in respects of eco-friend and repeatability of natural dyeing for apparels.

Protective Effects of Prunus persica Flesh Extract (PPFE) on UV-Induced Oxidative Stress and Matrix Metalloproteinases Expression in Human Skin Cells

  • In our continuous efforts to procure the active materials from natural products in the protective effects of oxidative stress or UV damage to skin cells we found the Prunus persica flesh extract (PPFE) is considerable to meet the demand to protect the skin damage. PPFE attenuated cell damage induced by hypoxanthine-xanthine oxidase in cultured human keratinocytes, indicating that PPFE has the potential of the scavenging effect of reactive oxygen species (ROS) in human skin cell. Moreover, PPFE significantly suppressed UVA-induced ROS production determined by the oxidation of 2,7-dichlorodihydrofluorescein diacetate (DCFH) using FACS analysis. Additional study revealed that UVA irradiation of HaCaT human keratinocytes increased the gelatinolytic activities of matrix metalloproteinase-2, and -9 (MMP-2, -9) and mRNA expression of MMP-9 analyzing by a real-time reverse transcriptase-polymerase chain reaction (RT-PCR), and these events were significantly suppressed by the treatment with PPFE. These results suggest that PPFE might be applicable as natural ingredients for skin antiaging agents via UV-induced ROS scavenging activity and suppression of MMP expression in the skin cells.

Antioxidant Efficacy of Extracts from a Variety of Seaweeds in a Cellular System

  • As a part of an ongoing search for antioxidants from marine sources, antioxidant activities of 24 kinds of seaweeds (4 green algae, 8 brown algae, and 12 red algae) were investigated. The seaweeds were extracted by acetone/dichloromethane and methanol, respectively. The antioxidant properties of both extracts were evaluated using four different activity tests, including degree of occurrence of intracellular reactive oxygen species (ROS), NO, lipid peroxidation, and GSH (glutathione) in mouse macrophage Raw 264.7 cells. The levels of intracellular reactive oxygen species (ROS) and GSH were measured using 2',7'-dichlorofluorescin diacetate (DCFDA) and monobromobimane as fluorescence probe, respectively. Moreover, the generation of NO and lipid peroxidation products were determined by each method based on the Griess reaction and TBARS assay. Solvent extracts from seaweeds such as Scytosiphon lomentaria, Prionitis cornea, Laruencia okamurae, Callophyllis japonica, Sargassum horneri, Dictyopteris divaricata, Lomentaria catenata, Corallina confuse, Ishige okamurae, and Ahnfeltiopsis flabelliformi exhibited high antioxidant activities in cellular oxidizing systems.

Peroxynitrite Scavenging Activity of Sabohwanin Lipopolysaccharide-Induced Oxidatively-Stressed Mice (Lipopolysaccharide로 산화 스트레스를 유도한 Mouse에서 사보환(四補丸)의 Peroxynitrite 억제 효과)

  • Objectives : Peroxynitrite (ONOO-), superoxide anion radical (?O2-) and nitric oxide (NO) are cytotoxic because they can oxidize several cellular components such as proteins, lipids and DNA. They have been implicated in the aging process, and age-related diseases such as Alzheimer's disease, rheumatoid arthritis, cancer and atherosclerosis. The aim of this study was to investigate Sabohwan's activity for scavenging ONOO- and its precursors. NO and ?02-. Methods : For this study, the fluorescent probes, namely 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), 4.5-diaminofluorescein (DAF-2) and dihydrorhodamine 123 (DHR 123) were used. Results : Sabohwanblocked tert-butylhydroperoxide (t-BHP)-induced cell death in a dose-dependent fashion. It scavenged t-BHP-induced ONOO-, NO and ?O2- in YPEN cells. Sabohwan inhibited the generation of ONOO-, NO and ?O2- in the lipopolysaccharide (LPS)-treated mouse kidney postmitochondria both in vitro and in vivo. The lipid peroxide level increased and glutathione level decreased in the LPS-treated mice, whereas the ones in the Sabohwanadministered group among the LPS-treated mice reversed toward their natural levels. Conclusions : These results suggest that Sabohwanis an effective ONOO-, ?O2- and NO scavenger, and thereby it might have a potential role as a therapy against the aging process and age-related diseases.

Inhibition of COX-2 gene expression via $NF-{\kappa}B$ pathway by Ichungwhan (이정환의 $NF-{\kappa}B$ 활성화 기전을 통한 COX-2 저해 기전)

  • Objectives : The present study was undertaken to investigate the molecular mechanisms of Ichungwhan for inhibition of cyclooxygenase-2 (COX-2) gene expression via suppression of NF-κB (nuclear factor κB) using aged rats. NF-κB is the most important modulator of inflammation and NF-κB regulates the gene expression of several pro-inflammatory cytokines, such as COX-2. Methods : In the experiment, we investigated the scavenging property of Ichungwhan on reactive species (RS) including nitrogen-derived species (RNS), measured by DCF-DA (2,7-dichlorodihydrofluorexcein diacetate) / DHR 123 (dihydrorhodamine 123) assay. Protein expression levels of COX-2, NF-κB, p-ERK and p-p38 were assayed by western blot. Results : We showed that Ichungwhan inhibits RS including RNS and inhibits NF-κB activation by blocking the dissociation of inhibitory IκB-β via suppression of IKK pathway. Also, Ichungwhan inhibits COX-2 gene expression. Conclusions : These findings suggest that Ichungwhan modulates COX-2 gene expression via suppression of the NF-κB pathway.

Metal Effects of Urban Air Particulates on Cytokine Production and DNA Damage

  • Epidemiologic studies have demonstrated an association between short-term exposure to particulate air pollutants and increased mortality. However the biological mechanism underlying these associations have not been fully established and also the chemical and physical characteristics of the pollutant particles are not well understood. The metal constituents of air pollutant particles and their bioavailability are considered to Play an important role as possible mediators of Particle-induced airway injury and inflammation. Sprague-Dawley rat alveolar macrophage cells (NR8383) were exposed to airborne and acid-leached particulate matter (PM). Titanium oxide and nickel subsulfide were used as negative and positive controls. Particle-induced reactive oxygen species formation in cells was detected using the fluorescent probe 2',7'-dichlorofluorescin diacetate. Expression of TNF-$\alpha$ and IL-6 were measured by enzyme-linked immunosorbent assay, and PM-induced DNA double-strand breaks were determined with $\lambda$DNA/Hind III marker. Metals associated with air pollutant particles mediated intracellular oxidant production in alveolar macrophages, and the cytotoxicity and proinflammatory cytokine production induced by PM were associated with oxidative stress. The oxidants produced by air pollutant particles also are likely to induce DNA double-strand breaks. Our findings in alveolar macrophage cells exposed to PM and acid-leached PM support the hypothesis that metal components in urban air pollutants and their bioavailabilities might play an Important role in the induction of the adverse health effects.

  • Reactive oxygen species (ROS) are known to cause oxidative modification of DNA, proteins, lipids and small cellular molecules and are associated with tissue damage and are the contributing factors for inflammation, aging, cancer, arteriosclerosis, hypertension and diabetes. We screened the anti-oxidants in V79-4 hamster lung fibroblast cells induced by hydrogen peroxide with eighteen pure compounds isolated from natural products. Allantoin, brassicasterol, and hypaconitine were found to strongly scavenge intracellular reactive oxygen species, which is measured by dichlorodihydrofluorescin diacetate method (DCHF-DA), and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical.

Effects on Viability of Different Cryoprotectants Treated Mouse Embryos after Quick Freezing (침투성 및 비 침투성 동결보호제를 이용한 생쥐 수정란의 급속동결에 따른 생존성에 관한 연구)

  • In order to improve the cryopreservatory techniques of livestock embryos, the quick freezing method which is directly plunged in liquid nitrogen via prefreezing procedure without freezing machine was carried out for mouse embryos treated with permeable and nonpermeable cryoprotectants. The viability of frozen-thawed embryos were evaluated by FDA vital dye test. The results obtained was summaried as follows: 1. A total of 720 embryos were recovered from frozen embryos for viability test. Evalution of the fluorescein diacetate(FDA) vital dye test with mice embryos were resulted of 2.3 total mean score - evaluted in orderly higher mean grade of P3 453 (63%), P2 133(18%), P1 51(7%) and P0 83(12%). 2. An all-round evalution of these combination, the highest viability was showed in 3M ethylene glycol + 0. 25M trehalose treated with the copper prefreezing. 3. Effects of permeable and nonpermeable cryoprotectants combination were evaluated by means FDA score. 3M ethylene glycol + 0.25M trehalose showed the highest survival rates of 2.8 mean FDA score. 4. Effects of permeable cryoprotectants were evaluated by mean FDA score but the results were not significantly different each other. 5. In evalution of the nonpermeable cryoprotectants, 0. 25M trehalose obtalned higher mean FDA score than of 0.25M sucrose and it was significantly different(P<0.05). 6. There was no significantly difference between copper and stainless-steel in prefreezing procedures.

Inhibitory Phlorotannins from the Edible Brown Alga Ecklonia stolonifera on Total Reactive Oxygen Species (ROS) Generation

  • Reactive oxygen species (ROS) play an important role in the pathogenesis of many human degenerative diseases such as cancer, aging, arteriosclerosis, and rheumatism. Much attention has been focused on the development of safe and effective antioxidants. To discover sources of antioxidative activity in marine algae, extracts from 17 kinds of seaweed were screened for their inhibitory effect on total ROS generation in kidney homogenate using 2',7'-dichlorofluorescein diacetate (DCFH-DA). ROS inhibition was seen in three species: UIva pertusa, Symphyocladia latiuscula, and Ecklonia stolonifera. At a final concentration of 25 $\mu\textrm{g}$/mL, U. pertusa inhibited 85.65$\pm$20.28% of total ROS generation, S. latiscula caused 50.63$\pm$0.09% inhibitory, and the Ecklonia species was 44.30$\pm$7.33% inhibition. E. stolonifera OKAMURA (Lam-inariaceae), which belongs to the brown algae, has been further investigated because it is commonly used as a foodstuff in Korea. Five compounds, phloroglucinol (1), eckstolonol (2), eckol (3), phlorofucofuroeckol A (4), and dieckol (5), isolated from the ethyl acetate soluble fraction of the methanolic extrclct of E. stolonifera inhibited total ROS generation.

The Use of Fluorescein Diacetate (FDA) to Assess Oocyte Viability in the Bovine (FDA를 이용한 소 난포란의 생사 판정)

  • This experiment was carried out to determine the possibility of using FDA on the selection of viable bovine immature oocytes. The results obtained in these experiments were summarized as follows; 1. The rates of nuclear maturation (metaphase II) of bovine oocytes according to FDA concentration were 92.5% (37/40) in the control and those rates in the dilution on FDA to 1:400,000, 1:800,000 and 1:1,600,000 were 74.4% (67/90), 80.3% (38/46) and 71.1% (32/45), respectively. 2. The fertilization rate ( 2-cell) in the control was 72.2% (39/54) and those rates in the dilution of FDA to 1:400,000, 1:800,000 and 1:1,600,000 were 28.8% (23/80), 66.7% (32/48) and 62.2% (28/45), respectively. In these results, a, pp.opriate concentration of FDA to bovine immature oocytes was 1:800,000. 3. Effect of FDA treatment to the blastocyst development of bovine oocytes was indicated that control was 22.2% (18/81) and FDA treatment groups which were classified to strong, partial and weak were 21.4% (36/168), 14.5% (9/62) and 0% (0/15), respectively. This result suggested that in vitro development to the blastocyst was severely reduced except strong group according to the FDA fluorescent level (P<0.05) and that using FDA is possible to select of bovine oocytes.
