Preparation and Characterization of the L-prolino Co(III) Complexes with the Tetradentate $N_2O_2$-type Ligands

  • Complexes of the type[Co(T)(L-pro)], where T is the quadridentate $N_2O_2$-type ligand, N,N'-dimethylethylenediamine-N,N'-diacetate or N,N'-dimethylethylenediamine-N,N'-di-$\alpha$-butyrate, have been prepared. The complexes were separated into the two stereoisomers, $\Delta$-s-cis-[Co(T)(L-pro)] and $\Lambda$-s-cis-[Co(T)(L-pro)]. They were characterized by their proton magnetic resonance, absorption and circular dichroism spectra, elemental analyses.

Assessment of Mycobacterial Viability by Fluorospectrophotometry (형광분광측정법에 의한 항산균의 생명력 평가)

  • Viable potential of Mycobacterium smegmatis, a slow grower in vitro cultivation and of M. leprae, an obligate intracellular parasitic bacterium, which can not be cultured yet in vitro was assessed by fluorospectrophotometry. Bacterial cells in different numbers and under various physiological status were incubater with fluorescein diacetate(FDA). After an incubation of the bacterial preparations with FDA at specified conditions, amount of fluorescein inside bacteria was measured by a fluorospectrophotometer at 470nm and 510nm of excitation and emission wavelengths, respectively. Fluorounit given by such bacteria showed a correlation with assessment of viability of the same preparations made by other methods, such as optical density and colony forming units of M. smegmatis and intracellular ATP content of M. leprae. The possible use of fluorospectrophotometry in assessing viability or physiological potential of bacteria, particularly intracellular parasites and fastidious organisms to culture in vitro is discussed in relation to other methods.

Catalase Induced by All-Trans Retinoic Acid Is Involved in Antiproliferation of 36B10 Cells (레티노인산에 의한 카타라제의 유도가 36B10세포의 증식억제에 미치는 효과)

  • Purpose: All-trans retinoic acid (ATRA) has anti proliferative effects against brain tumor cells. Recently, ATRA has been reported to induce catalase. We investigated whether catalase induction by ATRA is associated with its anti proliferative effects. Materials and Methods: 36B10 cells were exposed to 0~50${\mu}M$ ATRA for 24 or 48 hours and mRNA, protein, and activity of catalase were measured. Reactive oxygen species (ROS) were measured using 2',7'-dichlorofluorescin diacetate. A clonogenic assay was used to confirm the cytotoxic effect. Results: The mRNA, protein, and activity of catalase were found to increase in a concentration- and incubationtime-dependent manner. The increase in catalase activity induced by ATRA was decreased by the addition of 3-amino-1,2,4-triazole (ATZ). ROS was also increased with ATRA and decreased by the addition of ATZ. The decrease in cell survival induced by ATRA was partly rescued by ATZ. Conclusion: Catalase induction by ATRA is involved in ROS overproduction and thus inhibits the proliferation of 36B10 cells.

Characteristics of Transparent and Conducting Tin Oxide Film (투명전도성 Tin Oxide Film의 특성)

  • Some characteristics of $SnO_2$ film which was deposited on a slide glass substrate, using dibutyl tin diacetate and oxygen, by the chemical vapor deposition were observed. The optimum condition for the preparation of the film was found to be at 420$^{\circ}C$ of substrate temperature for 20 min of deposition. Important optical, electrical, and structural features of the film were examined. It was found that the typical $SnO_2$ film on the untreated substrate was 4000${\AA}$ in thickness, transmitted 90% of the visible liglit, and provided 5800 ohms/${\square}$ of the sheet resistance. It was also found that the surface treatments of the slide glass by acid leaching were beneficial. The film structure was found to be a mixture of polycrystalline tetragonal stannic oxide confirmed by the X-ray diffraction and to be spherical fine grains concluded by the scanning electron microscopy.

Effects of Diesel Oil on the Population and Activity of Soil Microbial Community (토양미생물군집의 개체수와 활성도에 미치는 경유의 영향)

  • The effects of diesel oil on the microbial community in sandy loam soil were investigated, and the effects of bioremediation which was performed to enhance the removal of diesel oil from soil were also measured. The residual percentage of diesel oil was about 50% after 16 week incubation period. The bioremediation treatment increased the removal rate at 60~95%. When the soil was contaminated with diesel oil, the direct bacterial count, length of fungal hyphae, aerobic heterotroph and hydrocarbon degrader were increased by 2~3 orders of magnitude. The bioremediation further increased these numbers 10 to 100-fold. There were no difinite patterns of change in fluorescein diacetate hydrolysis activity in bioremediation-untreated soil, but about 10 times of increase of activity was observed in bioremediation-treated soil. Similar change was occurred in soil dehydrogenase activity.

Inhibition of Oxidative Damage by Phlorotannins from Ecklonia cava in Normal Human Dermal Fibroblasts

  • Phlorotannins which is oligomeric polyphenol of phloroglucinol unit were isolated from solvent fractions of methanolic extract of the brown alga, Ecklonia cava (EC). The inhibitory effects of phlorotannins from EC solvent fractions on oxidative stress were examined in human dermal fibroblasts (HDFs) related to wrinkle formation. Among the solvent fractions, phlorotannins from ethyl acetate fraction exerted the highest scavenging effect on DPPH radical, hydroxyl radical and alkyl radical analyzed by electron spin resonance (ESR) spectroscopy. The levels of intracellular reactive oxygen species (ROS) and lipid peroxidation were measured using 2',7'-dichlorofluorescin diacetate (DCFH-DA) and diphenyl-1-pyrenylphosphine (DPPP), respectively. Their levels were significantly decreased in the presence of phlorotannins from ethyl acetate fraction, compared with other fractions obtained from EC extract (P < 0.01). Furthermore, intracellular glutathione (GSH) level was significantly increased in a time dependent manner by the phlorotannins. Therefore, these results suggest that phlorotannins from EC extract could have a therapeutic potential for prevention and treatment of several diseases such as wrinkle formation related to oxidative stress.

  • The phytochemical investigation of the crude methanolic extracts roots and stem bark of Anthonotha cladantha (Harms) J.Léonard led to the isolation and identification of twelve secondary metabolites: 2,3-dihydroxypropyl hexacosanoate (1), hederagenine (2), cycloeucalenol (3), 2α-hydroxylupeol (4), betulinic acid (5), lupeol (6), heptacosan-2-one (7), triacontanoic acid (8), stigmast-4-en-3-one (9), β-sitosterol (10), stigmasterol (11), and stigmasterol-3-O-β-D-glucopyranoside (12). Their structures were elucidated with the help of their spectroscopic and physical data and by comparison with those reported in the literature. To the best of our knowledge, from all those compounds, 2,3-dihydroxypropyl hexacosanoate (1), hederagenine (2), cycloeucalenol (3), 2α-hydroxylupeol (4), and betulinic acid (5) are being reported for the first time from this genus. In addition, the acetylation of compound 1 afforded a new derivative 3-(hexacosanoyloxy)propane-1,2-diyl diacetate (1a). Compound 1 possessed a moderate α-glucosidase inhibitory activity with an IC50 value of 39.2 ± 0.22 μM; it neither showed antioxidant activity nor inhibition against the enzyme urease. Compound 1a exhibited weak antioxidant activity in the DPPH assay with an IC50 value of 80.3 ± 0.83 μM but was inactive against α-glucosidase and urease. Furthermore, both compounds 1 and 1a were inactive against seven pathogenic bacterial strains.

  • Novel 2'(${\beta}$)-methyl-5'-deoxyapiose purine phosphonic acid analogues were designed and synthesized from 2-propanone-1,3-diacetate. Condensation successfully proceeded from a glycosyl donor 9 under Vorbr${\ddot{u}}$ggen conditions. Condensation of aldehyde 14 with Wittig reagent [(diethoxyphosphinyl)methylidene] triphenylphosphorane gave the desired nucleoside phosphonate analogues 15. Ammonolysis and hydrolysis of phosphonates gave the nucleoside phosphonic acid analogue 17 and 19. The synthesized nucleoside analogues were subjected to antiviral screening against HIV-1. The adenine analogue 19 exhibited weak in vitro activities against HIV-1.

  • Reactive oxygen species (ROS) are known to cause oxidative modification of DNA, proteins, lipids and small cellular molecules and are associated with tissue damage and are the contributing factors for diabetes, inflammation, aging, cancer, arteriosclerosis, and hypertension. We screened the anti-oxidative effect on V79-4 hamster lung fibroblast cells induced by hydrogen peroxide with eleven extracts of combined medicinal plants. Dancheonhwankakambang and Samikangyabtang were found to show the scavenging activities of DPPH radical and intracellular reactive oxygen species, which is measured by dichlorodihydrofluorescin diacetate method (DCHF-DA).

Dyeing and Fastness Properties of Vat Dyes on a Novel Regenerated Cellulosic Fiber

  • enVix is a novel regenerated cellulosic fiber, which is prepared from cellulose diacetate fiber using environmentally friendly manufacturing process. Vat dyeing properties of the enVix were investigated and compared with those ofregular viscose rayon. The enVix exhibited better dyeability than viscose rayon. The colour yields of vat dyes on the enVix were found to be dependent on dyeing temperature as well as the amount of levelling agent and salt. Good build-up and good to excellent fastness properties were obtained on the en Vix fabric.
