HISTOPATHOLOGY AND PERCUTANEOUS ABSORPTION OF TOPICAL FORMULATION CONTAINING NEW CAPSAICIN ANALOG.

  • A new capsaicin analog modified with 4-hydroxyl and alkyl chain of capsaicin was a very potent antiinflammatory analgesic drug and may be clinically useful for those who have rheumatoid arthritis, diabetic neuropathy and cancer. The purpose of this study was to investigate histopathology after short and long term application of poloxamer-based gels, and percutaneous absorption of various topical formulations. Poloxamer-based gel was prepared by cold method using poloxamer 407. The poloxamer gels was applied to dorsal sites of hairless mouse skin during one week or one month for the evaluation of skin irritation. The applied site was then sectioned for histopathologic examination. The topical formulations were also prepared using CMC, HPMC, MC, carbopol and glycerylmono stearate. Skin variation of poloxamer gels was studied using excised hairless mouse, rat, hamster and human penis skin. Franz-type diffusion cells were used far skin penetration of drug against receptor phase filled with about 10$m\ell$ of 0.9% saline solution kept at 32$^{\circ}C$. The concentration of drug was determined by the reverse phased C18, Symmetry HPLC with fluorometeric detector. No skin erythema was observed after dorsal application of poloxamer-based gels for one week or one month. No histopathologic changes was also examined, suggesting no skin toxicity of poloxamer-based gels. The order of flux rate was HPMC > MC ( CMC > poloxamer >> glycerylmono stearate ( carbopol. There was a skin variation of poloxamer gels. The flux rate of poloxamer gels was highest in case of hairless mouse followed by rat, human and hamster skin. D-Pinitol의 유방암 증식 및 재발 억제 효능 (Inhibitory Effect of D-pinitol on Both Growth and Recurrence of Breast Tumor from MDA-MB-231 Cancer Cells)

  • D-Pinitol, an anti-diabetic substance, is a naturally occurring compound found in legumes. In this study, we investigated the inhibitory effect of D-pinitol on growth and recurrence of breast cancer. When D-pinitol was treated on MDA-MB-231 or MCF-7 breast cancer cells, it was observed that the viability of the two cancer cell lines was reduced in MTT assay. In order to examine the effect on the growth of breast tumor, mouse xenograft assay was carried out. On day 0, nine millions cells of MDA-MB-231 were injected subcutaneously into nude mouse and D-pinitol was administered orally at the dose of 500 mg/kg or 1000 mg/kg body weight for consecutive 45 days. Tumor size was reduced in dose-dependent manner upto 95.4% in 1000 mpk-treated group, compared with the non-treated control group. When D-pinitol was co-administrated with $4{\mu}g$ of doxorubicin, recurrence of breast tumor was delayed by two weeks, compared with the mouse group of doxorubicin monotherapy. Consistent with this data, it was observed that the population of cancer stem cells (CSCs), responsible for recurrence of cancer, within tumor mass was significantly reduced. Taken together, D-pinitol inhibits the growth of breast cancer and relapse of the tumor by suppressing the proliferation of CSCs.

Poly-N-acetyl-glucosamine이 당뇨병 쥐에서 창상치료에 미치는 영향 (Effects of Poly-N-acetyl Glucosamine(pGlcNAc) Patch on Wound Healing in db/db Mouse)

  • Purpose: Poly-N-acetyl glucosamine(PGlcNAc) nanofiber-based materials, produced by a marine microalga, have been characterized as effective hemostatic and angiogenic agents. The similarity between PGlcNAc patch and the natural extracellular matrix allows it to support new healthy tissue growth in an injured area and to encourage fluid absorption. In this study, we hypothesized that a poly-N-acetyl glucosamine fiber patch(PGlcNAc patch) may enhance wound healing in the db/db mouse. Methods: PGlcNAc patches were applied on one square centimeter, full-thickness, skin wounds in the db/db mouse model. Wounds(n=15 per group) were dressed with a PGlcNAc nanofiber patch for 1 hour(1 h), 24 hours(24 h) or left untreated(NT). After the application time, patches were removed and wounds were allowed to heal spontaneously. The rate of wound closure was evaluated by digital analysis of unclosed wound area in course of time. At day 10, wounds(n=7 per group) were harvested and quantified with immunohistochemical markers of proliferation(Ki-67) and vascularization (platelet endothelial cell adhesion molecule, PECAM-1). Results: Wounds dressed with PGlcNAc patches for 1 hour closed faster than control wounds, reaching 90% closure in 16.6 days, nine days faster than untreated wounds. Granulation tissue showed higher levels of proliferation and vascularization following 1 h treatment than the 24 h and NT groups. In addition to its hemostatic properties, the PGlcNAc material also appears to accelerate wound closure in healing-impaired genetically diabetic mice. Conclusion: This material, with its combination of hemostatic and wound healing properties, has the potential to be effective agent for the treatment of complicated wounds.

청혈산(淸血散)이 ob/ob mouse의 혈당, 고지혈증, Polyol Pathway 및 Reactive Oxygen Species에 미치는 영향 (Effects of Cheonghyul-san on Blood Glucose, Hyperlipidemia, Polyol Pathway and Reactive Oxygen Species in ob/ob Mice)

  • Etiological studies of diabetes and its complications showed that oxidative stress might play a major role. Therefore, many efforts have been tried to regulate free oxygen radicals for treating diabetes and its complications. Cheonghyul-san has been known to be effective for the antidiabetic, antihyperlipidemic and antiobesitic prescription, and composed of four crude herbs. In male ob/ob mouse with severe obesity, hyperinsulinemia, hypergiycemia, hyperlipidemia, the acting mechanisms of Cheonghyul-san were examined. Mice were grouped and treated for 5 weeks as follows. Both the lean (C57/BL6J black mice) and diabetic (ob/ob mice) control groups received standard chow. The experimental groups were fed with a diet of chow supplemented with 7.5, 15 and 30 mg Cheonghyul-san per 1 kg of body weight for 14 days. The effects of Cheonghyul-san extract on the ob/ob mice were observed by measuring the serum levels of glucose, insulin, lipid components, and the kidney levels of reactive oxygen species (ROS), MDA+HAE, GSH and also the enzyme activities involved in polyol pathway. Cheonghyul-san lowered the levels of serum glucose and insulin in a dose dependent manner. Total cholesterol, triglyceride and free fatty acid levels were decreased, while the HDL-cholesterol level was increased, in Cheonghyul-san treated groups. Renal aldose reductase and sorbitol dehydrogenase activities were increased in the ob/ob mice, whereas those were inhibited in the Cheonghyul-san-administered groups. Cheonghyul-san inhibited the generation of ROS in the kidney. Finally, MDA+HAE level was increased and the GSH level was decreased in the ob/ob mice, whereas those were improved in the Cheonghyul-san-administered groups. The results suggested that Cheonghyul-san exerted the antidiabetic and antihyperlipidemic activities by regulating the activities of polyol pathway enzymes, scavenging ROS, regulating the MDA+HAE and GSH levels in the ob/ob mice.

Novel Three-Dimensional Knitted Fabric for Pressure Ulcer Prevention: Preliminary Clinical Application and Testing in a Diabetic Mouse Model of Pressure Ulcers

  • Background Population aging has led to an increased incidence of pressure ulcers, resulting in a social burden and economic costs. We developed a three-dimensional knitted fabric (3-DKF) with a pressure-reducing function that can be applied topically in the early stages of pressure ulcers to prevent progression. Methods We evaluated the effects of the 3-DKF in a streptozotocin-induced diabetes mellitus pressure ulcer mouse model, and the fabric was preliminarily applied to patients. Twelve-week-old male C57BL/6 mice were used for the animal experiments. In the pressure ulcer mouse model, an ischemia-reperfusion injury was created using a magnet on the dorsa of the mice. Pressure was measured with BodiTrak before and after applying the 3-DKF to 14 patients at risk of sacral pressure ulcers. Results In the 3-DKF-applied mice group, the ulcers were shallower and smaller than those in the control group. Compared with the mice in the control group, the 3-DKF group had lower platelet-derived growth factor-α and neutrophil elastase expression, as parameters related to inflammation, and increased levels of transforming growth factor (TGF)-β1, TGF-β3, proliferating cell nuclear antigen, and α-smooth muscle actin, which are related to growth factors and proliferation. Additionally, typical normal tissue staining patterns were observed in the 3-DKF group. In the preliminary clinical analysis, the average skin pressure was 26.2 mm Hg before applying the 3-DKF, but it decreased to an average of 23.4 mm Hg after 3-DKF application. Conclusion This study demonstrated that the newly developed 3-DKF was effective in preventing pressure ulcers through testing in a pressure ulcer animal model and preliminary clinical application.

실크단백질 효소 가수분해물이 2형 당뇨 마우스의 혈당 및 혈청지질에 미치는 영향 (Effects of Silk Protein Hydrolysates on Blood Glucose and Serum Lipid in db/db Diabetic Mice)

  • 본 연구는 누에고치를 효소 가수분해하여 얻은 실크단백질 효소 가수분해물이 2형 당뇨병 모델인 C57BL/KsJ db/db mouse에 혈당조절 및 혈장지질 개선에 미치는 영향을 조사하였다. 12주령의 C57BL/KsJ db/db 마우스를 대조군과 실크단백질 효소 가수분해물 섭취군으로 나누어 8주 동안 음수로 섭취시켰다. 8주 동안 실험동물의 체중, 식이섭취량, 음수섭취량을 측정하고 매주 1회씩 4시간 공복 후 혈당변화를 관찰하였으며, 8주 후 모든 동물을 희생시킨 후 장기무게와 혈액을 채취하여 혈중 triglyceride와 cholesterol을 측정하였다. 실험동물의 체중은 실크단백질 효소 가수분해물 섭취군에서 증가하는 경향을 보였다. 장기무게는 간장의 경우 실크단백질 효소 가수분해물 섭취군에서 약간 증가하는 경향을 보였으며, 신장은 당뇨대조군에서 약간 증가하였으나 간장과 신장 모두에서 각 군간의 유의적 차이는 없었다. 혈당은 실크단백질 효소 가수분해물 섭취 1주 후부터 유의적인 차이를 보였으며, 실크단백질 효소 가수분해물 섭취군이 대조군에 비해 유의적으로 혈당상승을 억제하는 효과가 나타났다. 당뇨대조군의 경우 초기 혈당에 비해 8주후 혈당은 약 18% 증가하였고, 실크단백질 효소 가수분해물은 초기혈당과 비교 시 약 5.8%증가하여 실크단백질 효소 가수분해물이 혈당상승을 억제하는 것으로 나타났다. 포도당 부하후 측정한 내당능의 결과에서도 실크단백질 효소 가수분해물 섭취는 혈당 개선에 영향을 미치는 것으로 나타났다. 실크단백질 효소 가수분해물 섭취군의 혈청지질 중 total cholesterol과 non-HDL cholesterol은 당뇨대조군과 비교시 큰 차이가 없었으나, HDL-cholesterol을 유의적으로 증가시키고 triglyceride 함량을 유의적으로 감소시켰다. 실크단백질 효소 가수분해물의 섭취는 혈당상승을 억제하며, 혈청 지질 중 triglyceride를 낮추고 HDL-cholesterol을 높여 혈청지질 개선에 도움을 주는 것으로 사료된다.

  • Effects of Sotosaja hwan on Blood Glucose, Hyperlipidemia, Polyol Pathway and Antioxidative Mechanism in ob/ob Mouse Diabetes is a disease in which the body does not produce or properly use insulin. Etiological studies of diabetes and its complications showed that oxidative stress might play a major role. Therefore, many efforts have been tried to regulate free oxygen radicals for treating diabetes and its complications. Sotosaja-hwan has been known to be effective for the antiaging and composed of four crude herbs. In male ob/ob mouse in severe obesity, hyperinsulinemia and hyperlipidemia, which are features of NIDDM, the hyperglycemic activites and mechanisms of Sotosaja-hwan were examined. Mice were grouped and treated for 5 weeks as follows. Both the lean (C57/BL6J black mice) and diabetic (ob/ob mice) control groups received standard chow. The experimental groups were fed with a diet of chow supplemented with 30 and 90 mg Sotosaja-hwan per 1 kg of body weight for 14 days. The effects of Sotosaja-hwan extract on the ob/ob mice were observed by measuring the serum levels of glucose, insulin, lipid components, and the kidney levels of superoxide anion radical $({\cdot}O_2)$, MDA+HAE, GSH/GSSG ratio, and also the enzyme activities involved in polyol pathway. Sotosaja-hwan lowered the levels of serum glucose and insulin in a dose dependent manner. Total cholesterol, triglyceride and free fatty acid levels were decreased, while the HDL-cholesterol level was increased, in Sotosaja-hwan treated groups. Renal aldose reductase and sorbitol dehydrogenase activities were increased in the ob/ob mice, whereas those were inhibited in the Sotosaja-hwan-administered groups. Sotosaja-hwan inhibited the generation of ${\cdot}O_2$ in the kidney. Antidiabetic Activity and Mechanisms of Acarbose in $KKA^{y}$ Mice

  • To elucidate antidiabetic effect and mechanism(s) of acarbose in a polygenic spontaneous hyperglycemic and hyperinsulinemic diabetic animal model, $KKA^y$ mice, acarbose was administered orally for 4 weeks and effects on body weight, plasma glucose and insulin levels, genetic expressions of intestinal sucrase-isomaltase (SI), sodium-glucose cotransporter (sGLT1) and glucose transporter in quadriceps muscle (GLUT4) were examined in this study. Although no differences in body weight were detected between control and acarbose-treated groups, plasma glucose level in acarbose-treated group was markedly reduced as compared to the control. In the mechanism study, acarbose downregulated the SI and SGLT1 gene expressions, and upregulated the GLUT4 mRNA and protein expressions when compared to the control group. In conclusion, the data obtained strongly implicate that acarbose can prevent the hyperglycemia in $KKA^y$ mice possibly through blocking intestinal glucose absorption by downregulations of SI and sGLT1 mRNA expressions, and upregulation of skeletal muscle GLUT4 mRNA and protein expressions.

Effect of Cinnamon and Rhodiola rosea treatment on blood Glucose, Triglyceride, Total cholesterol and Glycohemoglobin in db/db mouse

  • The purpose of this study is to investigate the effect of the samples on the blood levels of glucose, glycohemoglobin(HbAlc), total cholesterol and triglyceride. The samples have been used in the treatment of a type 2 diabetic animal model (C57BlKsj db/db). The samples were administrated orally before each meal for 6 weeks. Cinnamon dose was 50mg/kg/day, 100mg/kg/day, 150mg/kg/day and 200mg/kg/day, respectively. Rhodiola rosea dose was 50mg/kg/day, 100mg/kg/day, 15-mg/kg/day and 200mg/kg/day, respectively. (omitted)

고지방식이급여 마우스에서 잠상산물의 항당뇨 효능 (Anti-Diabetic Effect of Sericultural Product in High Fat Diet-Fed Mice)

  • 본 연구는 고지방식이로 유도된 제2형 당뇨 동물모델에서 잠상산물의 항당뇨 및 지질개선 효과를 규명하기 위하여 C57BL/6N 마우스에 14주간 잠상산물 분말(ML, SK, MF, CT)을 급여한 후 제2형 당뇨의 임상적 지표 및 인슐린 신호 전달체계에 미치는 영향을 분석하였다. HFD군보다 잠상산물을 급여한 군들의 체중증가량이 감소하는 경향이었으며, 혈장 포도당 및 인슐린 농도는 HFD군보다 ML분말을 급여한 군에서 특히 감소하였다. 혈장 T-C 농도는 HFD군에 비하여 ML과 SK 분말 급여 군에서 유의적으로 감소하였으며, 혈장 HDL-C 농도 및 HTR은 HFD군보다 ML군과 SK군에서 현저히 증가하였다. 간 조직 지질 농도는 HFD군보다 잠상산물을 급여한 네 군 모두에서 TG와 T-C 농도의 유의적인 감소가 나타났다. 이상의 결과 잠상산물 중 ML분말이 혈장 포도당, 인슐린, T-C 및 TG 농도를 가장 효과적으로 감소시켰으며, 혈장 HDL-C 농도 및 HTR은 증가시켰다. 또한, 내장지방 조직의 AKT, S6K, 그리고 AMPK의 활성도 증가시킨 것으로 보아 고지방식이에 의한 인슐린 저항성 및 지질 대사를 개선한 것으로 생각된다. 향후 ML의 인슐린 저항성 개선 효과에 대해 더 깊이 있는 기전연구가 수행된다면 제2형 당뇨환자를 위한 건강기능성 식품으로 개발될 수 있을 것으로 생각한다.
