Effects of Size and Degree of Abdomen Inflation of the Mother Fish on Ovulation Induction of Red Spotted Grouper, Epinephelus akaara

  • We determined the morphologic characteristics (body weight and degree of abdomen inflation) of the red spotted grouper, Epinephelus akaara, mother fish producing healthy eggs. Experimental fish were chosen from fish reared in a sea cage. The fish were divided into four size groups by body weight: 400~600, 600~800, 800~1,000, and 1,000~1,200 g and four stages (I~IV) of the degree of abdomen inflation. After hormone treatment, we observed the amount of ovulation-induced eggs, and rates of buoyancy, fertilization, embryonic survival, and hatching. As a result, mother fish with a body weight of 600 g or more spawned, and the fertilization rate, embryonic survival rate, and hatching rate were high in the 800~1,000 g range, thus showing effective ovulation induction. As a result of dividing the degree of abdomen inflation based on the anal fin of the mother fish into I-IV stages and determining hormone treatment time, the GSI was $0.9{\pm}0.2%$ at stage I, $2.3{\pm}0.2%$ at stage II, $5.6{\pm0.2%$ at stage III, and $7.9{\pm}0.9%$ at stage IV. The flotation rate and hatching rate were highest at stage III, and the fertilization rate and embryonic survival rate were highest at stage IV. Therefore, in terms of egg quality, the amount of eggs collected per mother fish, maturation, and histology were different depending on the degree of abdomen inflation. At stage III, where the abdomen inflation degree of the mother fish was based on the basal part of the dorsal fin relative to the height of the anal fin was 1, the egg quality was highest.

Effects of temperature and salinity on egg development and larval settlement of an invasive ascidian species, Herdmania momus (Savigny, 1816)

  • The effects of temperature and salinity on egg development and settlement of the ascidian Herdmania momus were investigated. Adult specimens were collected from the Dodu Yacht facility in Jeju Island, Korea(33°30'30.54"N, 126°27'55.46"E) in August 2018. Egg development and larval settlement were observed and recorded at 8 h intervals using a stereomicroscope, under nine temperature (10, 13, 16, 19, 22, 25, 28, 31, and 34℃), and four salinity regimens (28, 30, 32, and 34 psu). The highest hatching rate (82.8±7%) was observed at 32 psu and 25℃ and the lowest hatching rate (1.0±2%) was at 34 psu and 13℃. The developmental rate (0.222±0.0994) was highest at 28 psu and 28℃, and lowest (0.016±0.008) at 30 psu and 13℃. The highest settlement success rate (77.1±5%) was at 32psu and 25℃ and the lowest(0.1±1.0%) was at 30psu, and 13℃. The rate of settlement(0.080±0.000) was highest at 28psu and 28℃, and lowest(0.013±0.000) at 30psu and 13℃. Both hatching and settlement success rates increased as temperature increased and tended to decrease beyond an optimal temperature range. Herdmania momus preferred 30-34 psu salinity and 22-25℃ temperature. This study provides baseline information about the life history of H. momus, and important data to control the damage caused by the increase in number and distribution of this invasive ascidian.

Profiling of skeletal muscle tissue for long non-coding RNAs related to muscle metabolism in the QingYu pig at the growth inflection point

  • Objective: Investigation of muscle growth at different developmental stages is an appropriate strategy for studying the mechanisms underlying muscle development and differences in phenotypes. In particular, the muscle development mechanisms and the difference between the fastest and slowest growth rates. Methods: In this study, we used a growth curve model to fit the growth inflection point (IP) of QingYu pigs and compared differences in the long non-coding RNA (lncRNA) transcriptome of muscle both at the growth IP and plateau phase (PP). Results: The growth curve of the QingYu pig had a good fit (R2 = 0.974) relative to a typical S-curve and reached the IP at day 177.96. At the PP, marbling, intramuscular fat, and monounsaturated fatty acids had increased significantly and the percentage of lean muscle and polyunsaturated fatty acids had decreased. A total of 1,199 mRNAs and 62 lncRNAs were differentially expressed at the IP compared with the PP. Additional to gene ontology and Kyoto encyclopedia of genes and genomes pathway analyses, these differentially expressed protein coding genes were principally related to muscle growth and lipid metabolism. Conclusion: Our results suggest that the identified differentially expressed lncRNAs, could play roles in muscle growth, fat deposition and regulation of fatty acid composition at the IP and PP.

Development of Porcine Somatic Cell Nuclear Transfer Embryos Following Treatment Time of Endoplasmic Reticulum Stress Inhibitor

  • We examine the effect of endoplasmic reticulum (ER) stress inhibitor treatment time on the in vitro development of porcine somatic cell nuclear transfer (SCNT) embryos. Porcine SCNT embryos were classified by four groups following treatment time of ER stress inhibitor, tauroursodeoxycholic acid (TUDCA; 100 µM); 1) non-treatment group (control), 2) treatment during micromanipulation process and for 3 h after fusion (NT+3 h group), 3) treatment only during in vitro culture after fusion (IVC group), and 4) treatment during micromanipulation process and in vitro culture (NT+IVC group). SCNT embryos were cultured for six days to examine the X-box binding protein 1 (Xbp1) splicing levels, the expression levels of ER stress-associated genes, oxidative stress-related genes, and apoptosis-related genes in blastocysts, and in vitro development. There was no significant difference in Xbp1 splicing level among all groups. Reduced expression of some ER stress-associated genes was observed in the treatment groups. The oxidative stress and apoptosis-related genes were significantly lower in all treatment groups than control (p<0.05). Although blastocyst development rates were not different among all groups (17.5% to 21.7%), the average cell number in blastocysts increased significantly in NT+3 h (48.5±2.3) and NT+IVC (47.7±2.4) groups compared to those of control and IVC groups (p<0.05). The result of this study suggests that the treatment of ER stress inhibitor on SCNT embryos from the micromanipulation process can improve the reprogramming efficiency of SCNT embryos by inhibiting the ER and oxidative stresses that may occur early in the SCNT process.

Effects of Mito-TEMPO on the survival of vitrified bovine blastocysts in vitro

  • Vitrification methods are commonly used for mammalian reproduction through the long-term storage of blastocyst produced in vitro. However, the survival and quality of embryos following vitrification are significantly low compared with blastocyst from in vitro production (IVP). This study evaluates that the survival of frozen-thawed bovine embryos was relevant to mitochondrial superoxide derived mitochondrial activity. Here we present supplementation of the cryopreservation medium with Mito-TEMPO (0.1 µM) induced a significant (p < 0.001; non-treated group: 56.8 ± 8.7%, reexpanded at 24 h vs Mito-TEMPO treated group: 77.5 ± 8.9%, re-expanded at 24 h) improvement in survival rate of cryopreserved-thawed bovine blastocyst. To confirm the quality of vitrified blastocyst after thawing, DNA fragmentation of survived embryos was examined by TUNEL assay. As a result, TUNEL positive cells rates of frozen-thawed embryos were lower in the Mito-TEMPO treated group (4.2 ± 1.4%) than the non-treated group (7.1 ± 3.5%). In addition, we investigated the intracellular ROS and mitochondrial specific superoxide production using DCF-DA and Mito-SOX staining in survived bovine embryos following vitrification depending on Mito-TEMPO treatment. As expected, intracellular ROS levels and superoxide production of vitrified blastocysts after cryopreservation were significantly reduced (p < 0.05) according to Mito-TEMPO supplement in freezing medium. Also, mitochondrial activity measured by MitoTracker Orange staining increased in the frozen-thawed embryos with Mito-TEMPO compared with non-treated group. These results indicate that the treatment of Mito-TEMPO during cryopreservation might induce reduction in DNA fragmentation and apoptosis-related ROS production, consequently increasing mitochondrial activation for developmental capacity of frozen-thawed embryos.

Cell-Surface Loss of Constitutive Activating and Inactivating Mutants of Eel Luteinizing Hormone Receptors

  • The present study aimed to investigate the mechanism of cell surface receptor loss by two constitutively activating mutants (designated L469R, and D590Y) and two inactivating mutants (D417N and Y558F) of the luteinizing hormone receptor (LHR) in the Japanese eel Anguilla japonica, known to naturally occur in human LHR transmembrane domains. We investigated cell surface receptor loss using an enzyme-linked immunosorbent assay in HEK 293 cells. The expression level of wild-type eel LHR was considered to be 100%, and the expression levels of L469R and D417N were 97% and 101%, respectively, whereas the expression levels of D590Y and Y558F slightly increased to approximately 110% and 106%, respectively. The constitutively activating mutants L469R and D590Y exhibited a decrease in cell surface loss in a manner similar to that of wild-type eel LHR. The rates of loss of cell surface agonist-receptor complexes were observed to be very rapid (2.6-6.2 min) in both the wild-type eel LHR and activating mutants. However, cell surface receptor loss in the cells expressing inactivating mutants D417N and Y558F was slightly observed in the cells expressing inactivating mutants D417N and Y558F, despite treatment with a high concentration of agonist. These results provide important information on LHR function in fish and the regulation of mutations of highly conserved amino acids in glycoprotein hormone receptors.

Nonfatal injuries in Korean children and adolescents, 2007-2018

  • Background: Injury is the leading cause of death or disability in children and adolescents. Rates of deaths from injuries have recently declined, but studies of the occurrence of nonfatal injuries are lacking. Purpose: This study aimed to investigate nonfatal injuries in children and adolescents younger than 20 years based on data from the Korean National Health and Nutrition Survey, 2007-2018. Methods: A questionnaire survey was conducted to determine whether children and adolescents had experienced an injury requiring a hospital visit in the previous year. We investigated each injury's risk factors and characteristics. Results: Of a total of 21,598 children and adolescents, 1,748 (weighted percentage, 8.1%) experienced one or more injuries in the previous year. There was no yearly difference in the proportion of injuries experienced. Among the male subjects, 10.0% had an injury experience; among the female participants, 6.1% had an injury experience (P<0.001). The highest rate was 9.0% in children aged 1-4 years. In multivariate logistic regression analysis, male sex; having an urban residence; having restricted activity due to visual, hearing, or developmental impairment; and attention deficit/hyperactivity disorder were significant risk factors for injury experience. The characteristics of up to 3 injuries per patient were investigated, and 1,951 injuries were analyzed. Falls and slips accounted for 34.9%, collisions for 34.1%, and motor vehicle accidents for 11.3% of the total injuries. Ninety-six percent of injuries were unintentional, 20% caused school absences, and 10% required hospitalization. Conclusion: Among Korean children and adolescents, 8.1% experienced injuries at least once a year with no significant differences in incidence over the past 12 years. Greater attention and effort to prevent injuries are needed.

Bioconversion and growth performance of Hermetia illucens in single fruit by-products

  • As agricultural production increases due to population growth, agricultural by-products that are generated at all production stages increase commensurately. Black soldier fly (BSF) (Hermetia illucens) treatment has potential as an environmentally friendly process to combat the environmental pollution caused by agricultural by-products. This study examined the utilization of BSF larvae in the decomposition of the by-products of apples and mandarins, fruits commonly produced in South Korea. The BSF test larvae were fed apple pomace or mandarin waste, and the control larvae were fed calf feed. Larval weight and size were measured at 4-day intervals until larvae reached the pre-pupal stage. Larval development time, survival rate, and BSF fecundity rate were calculated for all three substrates. Waste reduction and bioconversion ratios were also calculated. The developmental time of larvae fed with apple pomace and mandarin waste was greater than that of the control larvae. The average weight of larvae fed with the fruit by-products was less than that of the control. There was no significant difference in the survival rate of BSF larvae or the fecundity rate of BSF between the substrates used in this study. BSF larvae decomposed 48.0% and 61.5% of apple pomace and mandarin waste, respectively. The bioconversion efficiency rates of BSF larvae fed with apple pomace and mandarin waste were 9.1% and 12.1%, respectively. These results indicated that decomposition of single fruit by-products by BSF larvae is an environmentally friendly and effective bioconversion process.

Study on Development Effect on Zebrafish Embryo by Alacholr, Butachlor and Fipronil (농약 alacholr, butachlor 및 fipronil이 제브라피쉬 배아 발생에 미치는 영향)

  • This study is aimed to search the possibility of developmental toxicity test using the zebrafish from the pesticide. We selected herbicides alachlor and butachlor, reported for fish toxicity, and insecticide fipronil reported for the high fish toxicity and the honey bee risk among the pesticides with high usability for the examples of the pesticides in this experiment. In this study, we showed those effects on the zebrafish embryo development by exposing different kinds of pesticide with different concentration and exposed time periods. As a result, the rates of hatching and abnormality of the zebrafish embryo after treatments of alachlor were increased in 24-48 hpf group, and the juvenile fishes in every group exposed to $40{\mu}M$ or more of alachlor displayed sever morphological changes such as bent tails, edema and activity failures. In case of the butachlor, the rates of hatching and the abnormality in 24-48 hpf group were higher than the other groups exposed in different time periods. The fatality before hatching was high in $40{\mu}M$ or more of butachlor treatment, and entire zebrafish embryos in 48 hpf group died before hatching. All the living juvenile fishes showed morphological changes as like as the treatment of alachlor. The rate of hatching and the survival of the zebrafish embryo by the fipronil were higher than other pesticides. However, morphological changes such as bent tails were observed from the most of living juvenile fishes. Therefore, the effects of three different pesticides with different concentrations and exposing time periods on the development of zebrafish embryos showed that all the pesticides effects were proportional to the concentration, and exposing time periods may cause the morphological abnormality.

Apoptosis and Development of Porcine Parthenogenetic Embryos Activated and Cultured in Different Condition (활성화 및 배양조건이 돼지 단위발생란의 발달 및 Apoptosis에 미치는 영향)

  • This study investigated apoptosis and in vitro development of parthenogenetic preimplantation porcine embryos. In vitro matured oocytes for $42{\sim}44h$ were used. Apoptotic cell death was analyzed by using a terminal deoxynucleatidyl transferase mediated deoxyuridine 5-triphosphate nick-end tabling (TUNEL) assay. In experiment 1, oocytes were activated with two electric pulses (CH) of 1.2 kV/cm for $30{\mu}sec$ (E), E + 6-dimethylaminopurine (6-DMAP) or E + cycloheximide (CH) and cultured in PZM-3 under 5% $CO_2$ in air at $38.5^{\circ}C$. In experiment 2, oocytes were activated by E and cultured in PZM-3 or NCSU-23 under a gas atmosphere of 20% $O_2$ ($5%\;CO_2$, in air) or 5% $O_2$ $(5%\;CO_2,\;5%\;O_2\;90%\;N_2)\;at\;38.5^{\circ}C$. Oocytes activated with E+6-DMAP or E+CH showed higher blastocyst rates (36.3% and 32.5%) compared to E alone (27.7%). The frequency of apoptosis according to treatments were 5.3%, 7.7% and 7.1% respectively. Oocytes activated with E alone showed lower (P<0.05) frequency of apoptosis compared to other groups. In experiment 2, parthenotes cultured in PZM-3 showed slightly higher blastocyte rates (28.2% and 29.7%) compared to NCSU-23 (22.6% and 24.4%) regardless of atmosphere. Blastocysts generated in PZM-3 showed lower (P<0.05) apoptosis rate under 20% $O_2$ (9.2% vs 16.9%), whereas those in NCSU-23 had slightly lower apoptosis rate under 5% $O_2$ (14.0% vs 18.4%). This result represents that activation method and culture condition could affect the frequency of apoptosis as well as in vitro developmental rate.
