Effects of Antioxidants Treatment on the Cryopreservation of Human Hematopoietic Stem Cells (인간 조혈모 줄기세포의 냉동보존에 미치는 항산화제의 영향)

  • Oxidative damage resulted from reactive oxygen species (ROS) is one of the main causes for the decrease of the viability during in vitro culture and cryopreservation process. This experiment was performed to determine the effects of antioxidants on the human hematopoietic stem cell (HSC) during cryopreservation procedure. HSCs cultured in vitro with or without antioxidants were frozen and then examined for stem cell potential after thawing. The cell viability of thawed HSC was increased in $\alpha$-tocopherol and ascorbic acid treatment group compared to control group ($62.7{\pm}8.0%$) and it was higher in 150 uM $\alpha$-tocopherol treatment group ($70.5{\pm}7.0%$). No significant difference was observed in the membrane integrity in all groups. In auto-differentiation rate, no significant difference was appeared in all groups, but was lower in 150 uM $\alpha$-tocopherol ($7.3{\pm}2.6%$) compared to control group ($10.1{\pm}1.6%$). These results demonstrate that treatment of antioxidants improves the efficiency of cryopreservation for HSC and $\alpha$-tocopherol may be considered effective antioxidant for the protective effect on HSC.

Effects of Nonylphenol and 2,2',4,6,6'-Pentachlorobiphenyl on In Vitro Steroidogenesis in Maturing Oocytes of Ribbed Gunnel, Dictyosoma burgeri (그물베도라치, Dictyosoma burgeri의 성숙기 난모세포 스테로이드 대사에 미치는 Nonylphenol과 2,2',4,6,6'-Pentachlorobiphenyl의 효과)

  • The in vitro effects of nonylphenol (NP) and 2,2',4,6,6'-pentachlorobiphenyl (PCB104) on ovarian steroidogenesis of the ribbed gunnel, Dictyosoma burgeri were investigated. Oocytes taken during maturation stage were incubated with 100 ng/$m{\ell}$ of NP and PCB104 in the presence of exogenous precursor, $[^3H]-17{\alpha}$-hydroxyprogesterone ($[^3H]-17{\alpha}OHP$). Steroids were extracted from the media and the isolated oocytes, and the extracts were separated and identified by thin layer chromatography. The identities of the major metabolites were testosterone (T) and estradiol-$17{\beta}$ (E2). NP treatment inhibited production of E2 metabolite in the oocytes of 1.2, 1.3 and 1.4 mm although NP inhibited production of T metabolite at the oocytes of 1.1, 1.3 and 1.4 mm. PCB104 treatment inhibited production of T metabolite in the oocytes of all groups and E2 metabolite in the oocytes of 1.2, 1.3 and 1.4 mm. In conclusion, these results suggested that NP and PCB104 had an inhibitory effects on conversion of $[^3H]-17{\alpha}OHP$ to T and E2 during the oocyte maturation process of ribbed gunnel.

  • Protein kinase C exists as a family of serine/threonine kinases which are broadly classified into three groups as cPKC nPKC and aPKC depending on their cofactor requirements. Previous studies have shown that the role of PKC in the process of mouse oocyte maturation. For example, phorbol 12-myristate 13-acetate which is known as an activator of cPKC and nPKC inhibits germinal vesicle break down and 1st polar body extrusion in maturing oocytes. In this study, the effect of thymeleatoxin, a specific activator of cPKC not nPKC, was tested comparing with PMA to address the roles of cPKC and nPKC during mouse oocyte maturation. Cumulus-oocyte complex were cultured in M16 medium for 6 or 12 hr with each of these PKC activators to investigate the effect of germinal vesicle breakdown (GVBD) or the extrusion of 1st polar body. IC/sup 50/ of GVBD were at concentrations of 50nM in PMA and 400nM in thymeleatoxin and of 1st polar body extrusion were 20nM in PMA and 200nM in thy- meleatoxin. The results suggest that activation of nPKC is more closely related to the inhibition of GVBD and 1st polar body extrusion than activation of cPKC. Additionally, we found that the oocytes inhibited 1st polar body extrusion with PMA or thymeleatoxin were arrested in metaphase I of first meiosis.

Effect of Mathematising Learning Using Realistic Context on the Children's Mathematical Thinking (현실적 맥락을 활용한 수학화 학습이 아동의 수학적 사고에 미치는 효과 -초등학교 5학년 도형 영역을 중심으로-)

  • The purpose of this study was to look into whether this mathematising learning utilizing realistic context has an effect on the mathematical thinking. To solve the above problem, two 5th grade classes of D Elementary School in Seoul were selected for performing necessary experiments with one class designated as an experimental group and the other class as a comparative group. Throughout 17 times for six weeks, the comparative group was educated with general mathematics learning by mathematics and "mathematics practices," while the experimental group was taught mainly with mathematising learning using realistic context. As a result, to start with, in case of the experimental group that conducted the mathematising learning utilizing realistic coherence, in the analogical and developmental thoughts which are mathematical thoughts related to the methods of mathematics, in the thinking of expression and the one of basic character which are mathematical thoughts related to the contents of mathematics, and in the thinking of operation, the average points were improved more than the comparative group, also having statistically significant differences. The study suggested that it is necessary to conduct subsequent studies that can verify by expanding to each grade, sex and region, develop teaching methods suitably to the other content domains and purposes of figures, and demonstrate the effects. In addition to those, evaluation tools which can evaluate the mathematical thinking processes of children appropriately and in more diversified methods will have to be developed. Furthermore, in order to maximize mathematising for each group in each mathematising process, it would be necessary to make efforts for further developing realistic problem situations, works and work sheets, which are adequate to the characteristics of the upper and lower groups.

Genistein Prevents Ethanol-Induced Teratogenesis in Mouse Embryos

  • Drinking of excessive ethanol during pregnancy induces a fetal alcohol syndrome. Genistein is one of naturally occurring isoflavones at relatively high levels in soybeans. In this study, we investigated the effects of genistein ($1{\times}10^{-8}$ and $1{\times}10^{-7}\;{\mu}g$/ml) on the ethanol (1 ${\mu}l$/ml)-induced teratogenesis of developing mouse embryos during the critical period (embryonic days 8.5~10.5) of organogenesis using a whole embryo culture system and then morphological scoring analysis. Ethanol-treated embryos exhibited a variety of developmental abnormalities. However, the total morphological scores for ethanol plus genistein groups were significantly higher than those of ethanol alone group (p<0.05). In particular, there were significant increases in the ethanol plus $1{\times}10^{-8}\;{\mu}g$/ml of genistein group on the scores for heart, optic system, branchial bar, mandibular process, and caudal neural tube and further in the ethanol plus $1{\times}10^{-7}\;{\mu}g$/ml of genistein group on the scores for heart, hind-, mid-, and forebrains, optic system, branchial bars, maxillary and mandibular processes, caudal neural tube, forelimb, hindlimb, and somites as compared with those of ethanol alone group (p<0.05). These results indicate that genistein has a preventive effect against ethanol-induced teratogenesis.

  • The paper describes the history and the evolution of the conflict of the Saemangeum reclamation project, focusing on the court trial processes. The Saemangeum project is the world largest coastal reclamation work, regarded as the most controversial environmental issue in the recent history of Korea. Due to the severe pollution found in Lake Sihwa in 1996, the Saemangeum project began to receive a large degree of public concern on the water quality of the proposed artificial freshwater lake. Unlike the Sihwa case, the Korean court system intervened to resolve the heated conflicts between stakeholders in the Saemangeum case. Based on the same set of facts, the Korean courts showed different perspectives on the economic feasibility, value of the ecosystem, land use, and water quality, which represents the limit of legal system to address complicated environmental problems. After the final judgment by the Supreme Court, 'the Special Act for the promotion of the Saemangeum reclamation project', was enacted with strong political support from local leaders and congressmen. A more developmental-oriented land use plan came out in 2009 based on this Act. The Saemangeum project walked along the different pathway from the Sihwa case. The area should be managed in sustainable manners to appropriately consider conservation and development for the prosperity of local residents and future generations.

  • The public interest and popularity of animation has already forecasted the value of animation as the leader of pop culture and cultural industry. The trend in animation changed from 2D to 3D animation, a new genre that appeared in the 1990s with the development of computer technology and innovative changes in media, and 3D animation secured its position in the 2000s. Researches on 3D animation are conducted in various ways. Integrating the area it shares with film and the area it shares with design, it is produced and consumed. This study focused on 'formativeness(design)', the basis of character design, and tried to find out how it is recognized by the audience, who actually watch and feel it. Also, it approached basic theory and tried to deduce logic that is easy to understand and can analyze design or formativeness which keeps changing. Lastly, this study regards the parts that viewers are not satisfied with as problems, and tries to find out a developmental direction. Among the animations for theaters that were big hits in 2015, this study focused on the characters of and . It reviewed errors and what was lacking in the previous studies, collected experts' opinions and made adjustment. Using the frame prepared through this process, it conducted a survey on study participants, the viewers, to see how it is perceived by them. Finally, it tried to find out the reasons why the audience felt that way through theoretical reviews and applications.

  • The reactivation for Art & Culture Content Space in Gangwon province need new turning point according to new approach with internal and external change. That should be established in sustainable possibility model with preexisted natural advantage instead of economical depression and global warming of in these days. That is not a choice to get a successful construction of art & culture content space in Gangwon province. Throughout of reactivation of art & culture content space in Gangwon province, this proposal should go on with diversity of culture and concrete art & culture educational program absolutely to get a goal in regional economical success and social good influence. Already Yeongwol region had lots of art & cultural spaces and developmental strategy compared with other regions in decade. In these points, I choose this region and would like to research Yeongwol region in limit. Thus I would examine and study about art & culture content space in Yeongwol region. And I will concretely suggest that this proposal is composed with economical creating employment and incoming. The reactivation for Art &Culture Content Space in Yeongwol will build up social and cultural influences to this region to get a new modern cultural images and improvement of common life of regional people by and large. In the way of the process to globalization, this reactivation project for Art &Culture Content Space in Yeongwol should go on the base of regional culture background as a mean of glocalization paradigm strategically. Ahead of developing model for globalization in macroscopic view point, we have to research the demend of reginal situation of cultural content and build up its own strength points. Because Art & Culture Content Space in Yeongwol is the place of people in Yeongwol directly as itself. And managing plans of reactivation of art & culture content space in Yeongwol should build up with diversity of culture and environmental circumstances.

A Study on the Selection Criteria of Media for the Textbook: Based on the Review of domestic and foreign Media Rating Systems (교과용도서 내 영상물 선정 기준 연구: 국내외 영상물 등급 제도를 중심으로)

  • This study is a basic study to prepare policies related to selecting media to be included in textbooks. Researchers looked at previous studies focusing on how media affect children's and adolescent's emotions according to their developmental stages. Researchers also wanted to identify how the media is related to student emotions, health, and educational effectiveness. The researchers then examined a wide range of domestic media review and rating criteria, and argued that it is necessary to institutionalize the rating system of media at national level policy level. Based on the previous discussion, the researchers made seven suggestions. First, it is necessary to specify media selection criteria in the guidelines for issuing textbooks at the national level. Second, guidelines are needed to help review the political neutrality and human rights aspects of the media for inclusion in textbooks. Third, media selection guidelines in textbooks should be detailed with reference to categories and age based criteria of domestic and foreign media rating system. Fourth, the media rating system should be applied flexibly if there is a clear educational purpose. Fifth, institutional support is needed for setting guidelines for media collection of textbooks. Sixth, media experts should participate in the whole process of textbook development. Seventh, educational programs should be implemented to support teachers to use self directed learning by using educational media in classroom instruction.

  • This study was to investigate the relationship between safety & quality management changes of patient and changes in management activities based on hospital workers in five mental hospitals and five geriatric hospitals which should be required medical certifying authorities. As a result of the research study, participation whether or not of certification service of mental hospital & geriatric hospital workers was positive correlation to improve change of 'Performance level of Safety Activities for the patient' 'Provide the High Quality Medical Service for the patient' 'Respect the Rights and Responsibility of the patient' 'Performance level of Infection Control Activities' out of contents of Patient Safety & Medical Service Quality. Also developmental changes of Safety Activities for the patient Hospital Quality for the patient Rights and Responsibility of the patient out of contents of Patient Safety & Medical Service Quality need to the Capacity Management Activities through Education and training, and Medical System & Evaluation of Management Provide the High Quality Medical Service for the patient out of contents of Patient Safety & Medical Service Quality need to the need to the Customer Orientation Process.
