Purification and Characterization of Two Alkaline Proteases Produced by Pseudomonas sp. BK7

  • Pseudomonas sp. BK7, an alkalophile, displayed the highest growth and protease activity when grown in a fermenter which was controlled at a pH level of 9.0, and the enzyme production was significantly enganced by the increase of agitation speed. Two formas of alkaline proteases (BK7-1 and BK7-2) were fractionated and purified to near homogeneity. Protease BK7-1 was purified through CM-Sepharose CL-6B and Sephadex G-75 column chromatographies, and Protease BK7-2 was purified through CM-Sepharose CL-6B and Sephadex G-75 column chromatographies, and Protease BK7-2 was purified through CM-Sepharose CL-6B, DEAE-Sepharose, and Sephadex G-75 column chromatographies. The molecular weights of proteases BK7-1 and BK7-2 determined by gel filtration chromatography were 20,700 and 40,800, respectively. The $K_m$ value, isoelectric point, and optimum pH of protease BK7-1 were 2.55 mg/ml, 11.0 and 11.0, respectively, whereas those of protease BK7-2 were 1.57 mg/ml, 7.2, and 10.0, respectively. Both protease were practically stable in the pH range of 5-11. The optimum temperatures for the activities of both protease BK7-1 and BK7-2 were 50℃ and 45℃, respectively. About 56% of the original protease BK7-2 activity remained after being treated at 50℃ for 30 min but protease BK7-1 was rapidly inactivated at above 25℃. Both proteases were completely inhibited by phenylmethane sulfonyl fluoride, a serine protease inhibitor. Protease BK7-2 was stable against EDTA, EGTA, STP, and detergents such as SDS and LAS, whereas protease BK7-1 was found to be unstable.

A Novel Esterase from Paenibacillus sp. PBS-2 Is a New Member of the ${\beta}$-Lactamase Belonging to the Family VIII Lipases/Esterases

  • Screening of a gene library from Paenibacillus sp. PBS-2 generated in Escherichia coli led to the identification of a clone with lipolytic activity. Sequence analysis showed an open reading frame encoding a polypeptide of 378 amino acid residues with a predicted molecular mass of 42 kDa. The esterase displayed 69% and 42% identity with the putative ${\beta}$-lactamases from Paenibacillus sp. JDR-2 and Clostridium sp. BNL1100, respectively. The esterase contained a Ser-x-x-Lys motif that is conserved among all ${\beta}$-lactamases found to date. The protein PBS-2 was produced in both soluble and insoluble forms when E. coli cells harboring the gene were cultured at $18^{\circ}C$. The enzyme is a serine protein and was active against p-nitrophenyl esters of $C_2$, $C_4$, $C_8$, and $C_{10}$. The optimum pH and temperature for enzyme activity were pH 9.0 and $30^{\circ}C$, respectively. Relative activity of 55% remained at up to $5^{\circ}C$ with an activation energy of 5.84 kcal/mol, which indicates that the enzyme is cold-adapted. Enzyme activity was inhibited by $Cd^{2+}$, $Cu^{2+}$, and $Hg^{2+}$ ions. As expected for a serine esterase, activity was inhibited by phenylmethylsulfonyl fluoride. The enzyme was remarkably active and stable in the presence of commercial detergents and organic solvents. This cold-adapted esterase has potential as a biocatalyst and detergent additive for use at low temperatures.

Screening and Characterization of a Novel Cellulase Gene from the Gut Microflora of Hermetia illucens Using Metagenomic Library

  • A metagenomic fosmid library was constructed using genomic DNA isolated from the gut microflora of Hermetia illucens, a black soldier fly. A cellulase-positive clone, with the CS10 gene, was identified by extensive Congo-red overlay screenings for cellulase activity from the fosmid library of 92,000 clones. The CS10 gene was composed of a 996 bp DNA sequence encoding the mature protein of 331 amino acids. The deduced amino acids of CS10 showed 72% sequence identity with the glycosyl hydrolase family 5 gene of Dysgonomonas mossii, displaying no significant sequence homology to already known cellulases. The purified CS10 protein presented a single band of cellulase activity with a molecular mass of approximately 40 kDa on the SDS-PAGE gel and zymogram. The purified CS10 protein exhibited optimal activity at $50^{\circ}C$ and pH 7.0, and the thermostability and pH stability of CS10 were preserved at the ranges of $20{\sim}50^{\circ}C$ and pH 4.0~10.0. CS10 exhibited little loss of cellulase activity against various chemical reagents such as 10% polar organic solvents, 1% non-ionic detergents, and 0.5 M denaturing agents. Moreover, the substrate specificity and the product patterns by thin-layer chromatography suggested that CS10 is an endo-${\beta}$-1,4-glucanase. From these biochemical properties of CS10, it is expected that the enzyme has the potential for application in industrial processes.

Detergency of Natural Surfactant for the Cleaning of Excavated Cotton Fabrics (출토 면직물 습식세척을 위한 천연계면활성제의 세척성 연구)

  • The purpose of this study was to investigate the characteristics and detergency of natural surfactants for the cleaning of excavated fabrics. For this purpose, SDS, a synthetic surfactant, was selected as the control, and five types of natural surfactants, namely, LES, apple wash, tea saponin, cornacopa, and coco betaine were selected. The structures of the surfactants were confirmed by FT-IR spectroscopy analysis, and the characteristics of the surfactants were determined by measuring the pH and surface tension. In addition, detergency testing was carried out on four artificially soiled fabrics and fragments of excavated fabrics. From the results, apple wash, tea saponin, and cornacopa were found to be as good as SDS in terms of detergency in the cleaning of artificially soiled fabrics, and the detergency of tea saponin and coco betaine was found to be good for cleaning excavated fabrics. Therefore, considering the safety and detergency of detergents, among natural surfactants, tea saponin is found to be most suitable for the cleaning of excavated fabrics.

Proteomic Analysis of Fructophilic Properties of Osmotolerant Candida magnoliae

  • Candida magnoliae, an osmotolerant and erythritol producing yeast, prefers D-fructose to D-glucose as carbon sources. For the investigation of the fructophilic characteristics with respect to sugar transportation, a sequential extraction method using various detergents and ultracentrifugation was developed to isolate cellular membrane proteins in C. magnoliae. Immunoblot analysis with the Pma1 antibody and two-dimensional electrophoresis analysis coupled with MS showed that the fraction II was enriched with membrane proteins. Eighteen proteins out of 36 spots were identified as membrane or membrane-associated proteins involved in sugar uptake, stress response, carbon metabolism, and so on. Among them, three proteins were significantly upregulated under the fructose supplying conditions. The hexose transporter was highly homologous to Ght6p in Schizosaccharomyces pombe, which was known as a predominant transporter for the fructose uptake of S. pombe because it exhibited higher affinity to D-fructose than D-glucose. The physicochemical properties of the ATP-binding cassette transporter and inorganic transporter explained their direct or indirect associations with the fructophilic behavior of C. magnoliae. The identification and characterization of membrane proteins involved in sugar uptake might contribute to the elucidation of the selective utilization of fructose to glucose by C. magnoliae at a molecular level.

Study on occurrence and management of organic micropollutants in sewer systems (국내·외 하수도시설 미량유기물질의 발생 특성 및 관리체계에 대한 이해)

  • As the modern society is rapidly developing and people become affluent in materials, many new chemical compounds in different forms of products (e.g., antibiotics, pesticides, detergents, personal care products and plastic goods) are produced, used, and disposed of to the environments. Some of them are persistently having a harmful impact on the environment and mimicking endocrine properties; in general they are present in the environment at low concentrations, so they are called organic pollutants. These organic micropollutants flow to sewage treatment plants via different routes. In this study, the generation characteristics, exposure pathways, detection levels, and environmental impacts of organic micropollutants were critically reviewed. In addition, currently available risk assessment methods and management systems for the compounds were reviewed. The United States Environmental Protection Agency (US EPA), for example, has monitored organic micropollutants and set the monitoring and management of some of the compounds as a priority. To effectively manage organic micropollutants in sewer systems, therefore, we should first monitor organic micropollutants of potential concern and then make a watch list of specific substances systematically, as described in guidelines on listing water pollutants in industrial wastewater.

Chemical Components of Seed Oil of Sapium japonicum Pax. et Hoffm. (사람주나무 종실유의 화학적 조성)

  • Physico-chemical characteristics and chemical composition of seed oil of Sapium japonicum were determined by various analysis methods. Physio-chemical characteristics of the extracted oil from fresh seed were specific gravity (0.928), refraction index (1.477), acid value (2.30), saponification number (190.0), and iodine value (126.0). Neutral lipid (93%) in seed oil was identified as major components, followed by glycolipid (4.9%) and phospholipid (1.3%). In GC analysis, nine fatty acids were presented in the seed oil. Among fatty acids, predominant fatty acids were oleic acid (45.8%) and linoleic acid (35.6%). The contents of fatty acids of seed oil somewhat varied with their storage period. Composition of fatty acids from lipid fractions which were isolated on silica open column was investigated. Major fatty acids in three lipid fractions were linoleic acid and linolenic acid. The fatty acid contents of fresh seed oil derived from each lipid fraction were generally high comapred to storage seed oil. From the above results, seed oil of Sapium Japonicum could be useful in cosmetics, detergents and a few pharmaceuticals.

A Case Report of Gastroesophageal Reflux Disease and Dysphagia Caused by Ingestion of Detergent (세정제 음독후 발생한 역류성 식도질환 및 연하곤란 환자 치험 1례)

  • The recent increased interest in hygiene has led to the use of a variety of detergents everyday life. Accordingly, accidents occur frequently, but proper precautions or treatments have not yet been established for these occasions. The patient in this case had accidentally ingested basic detergent, after mistaking it for a drink. This had caused acid reflux swallowing distress. Treatment in a western medical hospital had resulted in improvement in inflammation, as observed with an esophageal endoscopy test, but the patient still found it almost impossible to drink or eat. Following treatment with herbal medicine and acupuncture, the patient’s food intake increased and the pain and dysphagia were diminished. We cannot generalize based only on this one case study, so further studies are needed.

Washing-Related Garment Damage to Children's Clothing and Consumer Complaint Behavior (유.아동 의류 제품의 세탁과 관련된 소비자 피해 사례 및 불평 행동 연구)

  • As the materials being used in children's clothing become diverse, the number of reported problems related to various aspects of washing and wearing is increasing. Many customers experience great dissatisfaction when using these children's garments due to their lack of professional knowledge in washing, fabric, and detergents. Thus, this paper focuses on various cases in which housewives have experienced product damage after washing their children's garments according the prescribed directions. Of particular interest is how the consumers respond to such damage in garments and how their experiences with garment damage affect the sellers of these garments. A total of 292 housewives with children less than 8 years old living in Seoul responded to our survey, out of which 83 reported to have suffered negative consequences. By analyzing the 83 responses, the following results were found: First, the problems reported by the housewives in regards to washing children's garments were mostly related to the durability or color change problems. Secondly, the most favored means of response to garment damage were 'complaining directly to the seller' (16.7%) and 'complaining at the customer service department of the manufacturer or seller' (13.5%). Thirdly, among the actions they requested to the seller/manufacturer, 'ask for an exchange' marked the highest. Lastly, respondents admitted that their experiences with garment damage as well as the attitude or response of the manufacturer/retailer to their complaints would influence their repurchase intention.

Cloning, Expression, and Characterization of a Cold-Adapted Lipase Gene from an Antarctic Deep-Sea Psychrotrophic Bacterium, Psychrobacter sp. 7195

  • A psychrotrophic strain 7195 showing extracellular lipolytic activity towards tributyrin was isolated from deep-sea sediment of Prydz Bay and identified as a Psychrobacter species. By screening a genomic DNA library of Psychrobacter sp. 7195, an open reading frame of 954 bp coding for a lipase gene, lipA1, was identified, cloned, and sequenced. The deduced LipA1 consisted of 317 amino acids with a molecular mass of 35,210 kDa. It had one consensus motif, G-N-S-M-G (GXSXG), containing the putative active-site serine, which was conserved in other cold-adapted lipolytic enzymes. The recombinant LipA1 was purified by column chromatography with DEAE Sepharose CL-4B, and Sephadex G-75, and preparative polyacrylamide gel electrophoresis, in sequence. The purified enzyme showed highest activity at $30^{\circ}C$, and was unstable at temperatures higher than $30^{\circ}C$, indicating that it was a typical cold-adapted enzyme. The optimal pH for activity was 9.0, and the enzyme was stable between pH 7.0-10.0 after 24h incubation at $4^{\circ}C$. The addition of $Ca^{2+}\;and\;Mg^{2+}$ enhanced the enzyme activity of LipA1, whereas the $Cd^{2+},\;Zn^{2+},\;CO^{2+},\;Fe^{3+},\;Hg^{2+},\;Fe^{2+},\;Rb^{2+}$, and EDTA strongly inhibited the activity. The LipA1 was activated by various detergents, such as Triton X-100, Tween 80, Tween 40, Span 60, Span 40, CHAPS, and SDS, and showed better resistance towards them. Substrate specificity analysis showed that there was a preference for trimyristin and p-nitrophenyl myristate $(C_{14}\;acyl\; groups)$.
