Residual solvents in foods are defined as organic volatile chemicals used or produced in manufacturing of extracts or additives, or functional foods. The solvents are not completely eliminated by practical manufacturing techniques and they also may become contaminated by solvents from packing, transportation or storage in warehouses. Because residual solvents have no nutritional value but may be hazardous to human health, there is a need to remove them from the final products or reduce their amounts to below acceptable levels. The purpose of this study was to develop and evaluate an analytical method for the screening of residual solvents in health functional foods. Furthermore, the aim of this study was to constitute a reasonable management system based on the current state of the market and case studies of foreign countries. Eleven volatile solvents such as MeOH, EtOH, trichloroethylene and hexane were separated depending on their column properties, temp. and time using Gas Chromatography (GC). After determining the GC conditions, a sample preparation method using HSS (Head Space Sampling) was developed. From the results, a method for analyzing residual solvents in health functional foods was developed considering matrix effect and interference from the sample obtained from the solution of solvents-free health functional foods spiked with 11 standards solutions. Validation test using the developed GC/HSS/MS (Mass Spectrometry) method was followed by tests for precision, accuracy, recovery, linearity and adequate sensitivity. Finally, examination of 104 samples grouped in suits was performed by the developed HSS/GC/MS for screening the solvents. The 11 solvents were isolated from health functional foods based on vapor pressure difference, and followed by separation within 15 minutes in a single run. The limt of detection (LOD), limit of quantification (LOQ), recovery and coefficient of variation (C.V.) of these compounds determined by the HSS/GC/MS were found to be 0.1 pg/mL, 0.1-125 pg/g, 51.0-104.6%, and less than 15%, respectively. Using the developed HSS/GC/MS method, residual solvent from 16 out of 104 health functional products were detected as a EtOH. This method therefore seems t o be a valuable extension ofanalytical method for the identification of residual solvents in health functional food.
세균내독소는 lipopolysaccharide (LPS)라고 불리우며, 그람음성 세균의 세포벽에 존재하는 물질로 어느 생체재료나 액체에 존재할 수 있다. 세균내독소가 혈액으로 침투되면 발열 및 염증을 유발시킨다. 본 연구에서는 장내 그람음성세균인 Escherichia coli O157:H7, Klebsiella oxytoca, Salmonella Typhi, Shigella sonnei, Morganella morganii의 LPS 양을 비교하려 하였다. 각 세균은 배양한 후 $1.5{\times}10^8CFU/mL$ 농도로 희석하여 동일한 농도로 하였으며 실험의 정당성을 위하여 LAL시약의 표시감도확인시험을 진행하였다. 10 fold dilution 후 1차 시험을 하여 각 균의 반응 end point를 확인하고 바로 윗단계 희석배수 사이에서 2 fold dilution하여 균의 최종 반응 end point를 확인하였다. 결과적으로 E. coli O157은 75~37.5 CFU/mL, K. oxytoca는 37.5~18.75 CFU/mL, M. morganii와 S. Typhi는 3.75~1.875 CFU/mL, S. sonnei는 7.5~3.75 CFU/mL에서 0.015 EU/mL 이상의 endotoxin이 검출되었다. 최종적으로 억제인자에 대한 확인시험을 진행하여 결과값을 보증하였다. 본 연구를 통하여 세균내독소에 대한 추가적인 연구가 지속되기를 기원한다.
본 연구에서는 V. parahaemolyticus를 신속정확하게 고감도로 검출하기 위해 단클론성 항체를 개발하고 이를 이용하여 일반적인 효소면역분석법(ELISA)과 효소면역분석법의 낮은 민감도를 보완할 수 있는 면역선택여과법(ISF)을 개발하여 민감도 및 효율성을 비교분석 하였다. 먼저 heat killed V. parahaemolyticus (HKVP)를 준비하여 7주령 BALB/c mouse에 면역한 후 세포융합 및 클로닝을 통해 HKVP 4H9-9번 및 16번 2종의 hybridoma cell을 확보하였다. Western blot을 통해 보다 특이성이 높은 것으로 확인된 HKVP 4H9-9번 항체를 대량 생산정제하여 분석법을 확립한 결과 검출한계가 ID-ELISA법은 $10^6cell/mL$, ISF 법은 $5{\times}10^1cell/mL$로 나타나 ISF법의 민감도가 매우 높은 것으로 확인되었다. 분석효율성은 ID-ELISA법의 경우 분석을 위해 9단계의 과정을 거쳐 총 16시간의 분석시간이 소요된 반면, ISF법의 경우 3단계의 과정을 거쳐 1시간 이내에 분석을 완료할 수 있는 것으로 확인되었다. 교차반응성의 경우 두 분석법 모두 일부 Vibrio spp. (IDELISA법: V. alginolyticus, ISF법: V. vulnificus)과 S. aureus에 반응성이 있는 것으로 확인되었지만 V. parahaemolyticus에 보다 강한 반응성을 나타내었기 때문에 V. parahaemolyticus에 특이적인 분석법인 것으로 확인되었다. 특히 ISF법은 ELISA법 보다 민감도가 높고 분석에 소요되는 시간도 짧아 V. parahaemolyticus를 신속하게 분석하는데 활용이 가능할 것으로 판단된다.
Objectives : Dementia has emerged as a leading public health problem in elderly persons, and its early detection is important for the treatment of curable cases, and in the educational support for other family members. Although dementia screening tests are available, they have not gained widespread use in community or primary care settings. Our goal was to validate the Tine and Change (T&C) Test, -including its validity and reliability in patients, and to assess it as a simple, standardized method for the screening of dementia in the rural elderly. Methods : The participants in this study comprised of 59 patients from an urban hospital and 405 persons from a rural community aged 65 years or older. The time test evaluated the understanding of clock hands indicating 11:10, and the change test the ability to make 1,000 Won from a group of coins, consisting of one 500, seven 100, and seven 50 Won coins. The T&C ratings were validated against a reference standard based on the physician's diagnosis of the patients. The convergent validity in relation to other cognitive measure, test-retest agreement, and inter-observer reliability were assessed. To assess the relationship between the Korean Mini-Mental State Exam (K-MMSE) and the T&C Test, the mean K-MMSE scores were compared with the results of the T&C Test in the elderly from a rural community. Results The T&C Test had a sensitivity and specificity of 73.0, and 90.9%, and positive and negative predictive values of 93.1, and 66.7%, respectively. The test-retest and inter-observer agreement rates were both 95%. The K-MMSE scores and T&C Test were significantly related in the elderly from a rural community (p<0.01), The T&C Test was not influenced by the educational status. The Time and Change Tests took a mean of 6.3 and 12.7 seconds, respectively, to complete Conclusion : The T&C Test is a simple, accurate and reliable, performance-based tool in the screening for dementia. Because it is quick, and easy-to-use, it is hoped the T&C Test will be used for the widespread cognitive screening of aging populations.
In this study, the features of canine lymphoma on fluorine-18 fluorodeoxyglucose (FDG) positron emission tomography/computed tomography (PET/CT) were evaluated in three small breed dogs. In case 1, ultrasonography and CT indicated neoplastic involvement of the sternal, right axillary, submandibular, lower cervical, tracheobronchial, mesenteric, and sublumbar lymph nodes; spleen; and liver. However, intense FDG uptake on PET/CT images was detected only for the lymph nodes and spleen. No FDG uptake by the liver was detected for case 1 despite the confirmation of lymphoma by cytology. In case 2, ultrasonography and CT indicated neoplastic involvement of the axillary, mesenteric, and sublumbar lymph nodes and the spleen, while intense FDG uptake on PET/CT images was detected for the axillary and a few mesenteric lymph nodes, and the spleen. FDG uptake was additionally observed from popliteal lymph nodes, however there was no uptake by the sublumbar lymph nodes and some mesenteric lymph nodes. In case 3, neoplastic changes in the splenic, mesenteric, and sublumbar lymph nodes and spleen were suspected on ultrasonography, and lower cervical and popliteal lymph node involvements were additionally detected on PET/CT. Compared to ultrasonography, repeated PET/CT showed increased FDG uptake by the lymph nodes at an earlier stage after chemotherapy in case 3. This study illustrated the features of PET/CT in canine lymphomas and compared those to ultrasonography and CT findings. FDG uptakes were not detected from some lesions which were suspected to be neoplastic involvement in case 1 and 2. We could not clearly explain the reason of this result in the present study because cytological or histological examination was not performed for lesions that showed different results on ultrasonography, CT, and PET/CT. Further studies on the subclassification of canine lymphoma and the sensitivity and specificity of PET/CT for the detection of canine lymphoma are required. PET/CT data can provide useful information for predicting the therapeutic response at an early stage after treatment.
목 적 : 신경내분비종양 진단에 대한 I-123 MIBG 스캔의 4시간 조기 영상과 SPECT 영상의 임상적 유용성을 평가하고자 시행하였다. 대상과 방법 : CT 또는 MRI를 시행하여 신경내분비종양이 의심되었던 21예를 대상으로 I-123 MIBG 스캔을 시행하였으며 4시간에 전신 평면 영상과 SPECT 영상을 얻고, 17예에서는 12-24시간 사이에 지연 평면 영상도 얻었다. 결 과 : 21예중 12예에서 신경내분비종양으로 확진되었고, 4시간 I-123 MIBG 스캔의 진단적 예민도는 75%, 특이도는 89%였다. SPECT 영상은 CT나 MRI와의 비교 분석에 용이하였다. 지연 영상은 일부 진양성을 소견을 보인 예에서 종양 대 배후 방사능의 비가 증가하여 병변 확인에 도움이 되었으나 4시간 평면 영상과 진단율에 차이는 없었다. 결 론 : I-123 MIBG 스캔은 4시간 조기 영상으로도 신경내분비종양의 진단에 유용했으며, SPECT 영상은 다른 진단매체와의 비교 분석이 용이하였고 향후 종양의 종류에 따라 더 많은 예의 연구가 필요할 것으로 생각하였다.
본 연구에서는 동시 형광분광법과 GC/FID법을 이용하여 폐윤활유 시료 중의 PAHs를 acetonitrile 용매로 추출하여 정량분석 하였다. 동시 형광분광법을 이용하여 7종의 PAHs, 즉 acenaphthene (Ace), anthracene (Anth), benzo(a)pyrene (BaP), chrysene (Chry), phenanthrene (Phen), fluoranthrene (Fl) 및 perlyrene (Per)을 분석하였다. 이들 성분의 검정선은 모두 0.4~166 ppb(상관계수; 0.9985~0.9999)의 범위에서 직선을 보였다. 다른 8종의 PAHs를 GC법으로 분석하기 위한 검량선은 10.0 ppm 표준용액을 사용하였고, split ratio를 10에서부터 100까지 변화시킬 때에 발생되는 peak 면적을 이용하여 작성하였다. 검출감도는 동시 형광분광법이 GC법보다 적어도 100배 이상 우수하였다. 폐윤활유 시료 중의 총 PAHs 함량은 LNG(버스)와 LPG(택시)의 폐윤활유에서 각각 5.5 ng/g과 10.5 ng/g의 수준으로 검출되었으며, 가솔린을 사용하는 일반승용차, 경유를 사용하는 승합차 및 트레일러의 폐윤활에서 각각 92.2 ng/g, 92.6 ng/g 및 130.3 ng/g 등이 측정되어 경유를 사용하는 대형트레일러에서 가장 많은 양이 발생됨을 알 수 있었다.
물 시료에 존재하는 극 미량의 니켈과 코발트를 착물로 형성시켜 이온 교환수지 서스펜션에 흡착시켜 분리 농축하여 정량하는 방법을 연구하였다. 리간드로 APDC (ammonium pyrrolidine dithiocarbamnate)를 사용하여 극 미량 이온들을 착물로 형성시켜 농축한 다음, 전열 원자흡수 분광광도법으로 정량하였다. 이때 착물 형성을 위한 수용액의 pH와 착화제인 APDC의 양, 흡착을 위한 이온교환 수지의 종류 및 저어주는 시간, 역 분산에 사용하는 산의 종류 및 농도, 초음파 진동시간 등의 실험조건들을 최적화 하였다. 시료용액의 pH를 5로 조절하고 APDC의 양을 몰 비로 분석원소 전체의 430배 이상 첨가하여 코발트와 니켈을 정량적으로 착물을 형성시켰다. 이온교환 수지는 음이온 형태의 Dowex 2-X8이 우수하였다. pH를 조절한 시료용액, 리간드 및 수지 서스펜션을 혼합하고 1분간 저어주어 흡착을 완전하게 하였다. 역 분산을 위해서는 0.1 M 염산이 가장 좋았고, 이때 막 필터로 거른 교환 수지를 초음파 진동기에서 7분간 진동하면 충분하였다. 팔라듐을 염산과 함께 사용하면 매트릭스를 개선하여 재현성과 감도가 개선되었다. 바탕흡수 신호표준편차의 세배에 해당하는 검출한계는 Co 0.36 ng/mL, Ni 0.27 ng/mL로 극미량 검출이 가능하였고 시료에 일정량 첨가한 분석원소의 회수율은 각각 99-102%와 100-105% 이었다.
Triazole 계의 대표적인 농약 중 하나인 amitrole (3-amino-1,2,4-triazole)은 널리 사용되고 있는 제초제로, 환경 중에 유입될 경우 지하수 및 지표수를 오염시킴으로써 궁극적으로 음용수의 오염까지 일으킬 수 있다. 이 물질은 물에 대한 용해도가 높은 반면, 유기 용매에 대한 용해도가 낮고 극성이 강하기 때문에 수질 시료로부터 추출하기 매우 어렵다. 본 연구에서는 수질 시료중의 amitrole을 GC/MS를 이용하여 ppb 수준 이하의 극미량까지 분석하기 위한 방법을 조사하였다. 10 mL의 수질 시료를 진공회전증발기로 완전히 증발시킨 후, isobutyl chloroformate (iso-BCF)를 사용하여 실온에서 15~20분간 반응시켜 유도체화 하였다. 그 결과, N-isobutoxycarbonyl amitrole derivative를 생성함으로써 GC/MS에서의 감도가 향상됨을 확인할 수 있었다. $1.0{\mu}g/L$, $10.0{\mu}g/L$ 및 $100.0{\mu}g/L$ 농도의 수질에서 본 분석방법의 회수율은 각각 98.0%, 94.9% 및 105.3%였고, RSD는 2.4%, 2.8% 및 1.5%였다. 10 mL의 수질 시료로부터 얻어진 검량선의 상관계수는 $0.1{\sim}100.0{\mu}g/L$의 농도 범위에서 0.997로 나타나 직선성이 우수하였으며, 검출한계는 $0.1{\mu}g/L$로 나타났다.
In order to elucidate the influence of intestinal and hepatic first-pass effect on the pharmacokinetics of triflusal, the biotransformation of triflusal in the gastrointestinal tract and liver was designed. Moreover, we tried to establish an HPLC method applicable for bioassay and available to pharmacokinetics, not only with the simultaneous determination of triflusal and its active metabolite, 2-hydroxy-4-trifluoromethyl benzoic acid (HTB), but also with improving sensitivity. After the administration of triflusal (10 mg/kg) and HTB (10 mg/kg) into femoral vein, portal vein (only triflusal) and oral route (only triflusal), pharmacokinetic parameters were investigated from the plasma concentration-time profiles of triflusal and HTB in rats. An HPLC method was developed for the simultaneous determination of triflusal and HTB in rat plasma, urine and bile. The HPLC analysis was carried out using a C18 column and acetonitrile-methanol-water (25:10:65, v/v/v) as the mobile phase and UV detection at 234 nm. Furosemide was used as the internal standard. The calibration curves were linear over the concentration range $0.05-5.0\;{\mu}g/ml$ for triflusal and $0.2-200.0\;{\mu}g/ml$ for HTB with correlation coefficients greater than 0.999 and with intra-day or inter-day coefficients of variation not exceeding 10.0%. This assay procedure was applied to the study of metabolite pharmacokinetics of triflusal and HTB in rats. It was supposed that triflusal was almost metabolized in vivo because urinary and biliary excreted amounts of triflusal could be ignored as it was lower than 1.2% of the administered dose. According to the gastrointestinal and hepatic biotransformation pathways of triflusal, it was found that triflusal was hydrolyzed by about 5% in intestine and metabolized by about 53% in liver, and that the bioavailability of triflusal after oral administration of triflusal was 0.44, and also that the fraction of total elimination rate of triflusal which formed HTB in liver $(F_{mi},\;%)$ was about 98%. These results showed that triflusal was almost metabolized in liver, and the total elimination of triflusal in the body was dependent to the formation rate of HTB from triflusal in liver.

회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
