An improved tandem time-of-flight (TOF) mass spectrometer for the photodissociation (PD) study of ions generated by matrix-assisted laser desorption ionization (MALDI), MALDI-TOF-PD-TOF, has been designed and constructed. Recording a full spectrum with better than unit mass resolution even in low mass range has been achieved without reflectron voltage stepping which was needed in the previous version. Other aspects of the improvement, such as those in the data system which now allow 10-100 times faster spectral acquisition than with the previous instrument, are described. Rationale for the ideas which have led to the improvements is presented also.

Sorption and desorption studies were conducted to evaluate several media as a potential biobarrier for the remediation of groundwater contaminated with hydrophobic organic compounds (HOCs). Pahokee and Bion peats, Devonian Ohio shale, vermicompost, and 50% HDTMA-montmorillonite were used as model sorbents. Sorption and desorption isotherms were determined using the radiolabeled phenanthrene (Phe). Sorption capacity of Phe on several sorbents was in the order Ohio shale > 50% HDTMA-montmorillonite > vermicompost > Pahokee peat. Mineralization kinetics was investigated for Phe-sorbed sorbents using Pseudomonas putida 17484. Among the tested sorbents, active biodegradation of Phe was observed in shale and vermicompost: degradation in shale exhibited little lag time while that in shale showed a significant lag time. Results of this study indicate that sorbents used in this work can be utilized as permeable reactive biobarrier media for the remediation of HOC-contaminated groundwater.
Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) was used to observe folic acid and its derivatives such as tetrahydrofolate and 5-methyltetrahydrofolate in a vitamin tablet and in foods. Folic acid in a vitamin tablet was determined using angiotensin I as an internal reference. Tetrahydrofolic acid, 5-methyltetrahydrofolic acid, and an oxygenated folate were observed from a human blood sample using graphite plate. The results show that these mass spectrometric methods are useful for quickly obtaining a profile of folates.
A rapid and efficient method for analyzing the nonionic surfactants and silicone polymers, which control the shape and characteristics of cosmetic products and give influence on product quality, has been developed using Matrix-Assisted Laser Desorption Ionization Time of Flight Mass Spectrometry (MALDI- TOF IMS). The MALDI-TOF/MS could easily and effectively determine the molecular weight distribution and monomer units of nonionic surfactants. As a result, creating a library of mass spectrum data of surfactants used in cosmetic products using MALDI-TOF/MS and analyzing surfactants extracted from the products may become a useful method for detailed structural characterization of the surfactants. Furthermore, the MALDI-TOF/MS analysis was effective in obtaining the spectrum of silicone polymers from which the molecular weight distribution could be determined. The repetition units and structural data could also be obtained through molecular mass peaks. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) was used to demonstrate cross-linking of peptides induced by transglutaminase. The presence of ε-( Υ-glutamyl)lysine isopeptide cross-link in the acid hydrolysate of the cross-linking reaction mixture was also demonstrated by MALDI-TOF-MS without prior separation. MALDI-TOF-MS quickly provided peptide mass maps after pronase digestion of the cross-linked peptide adduct, which enabled us to monitor the hydrolytic sequence. Pronase appears to preferentially hydrolyze peptide bonds distant from the cross-link before hydrolyzing peptide bonds around the cross-link. The results suggest that pronase digestion followed by MALDI-TOF-MS could be used for determination of amino acid sequence around a modification site.

Gold nanostructures (Au NSs) are useful and interesting matrices for mass spectrometric analysis of various biomolecules based on organic matrix-free laser desorption/ionization time-of-flight mass spectrometry (LDI-TOF-MS). Au NSs provide high efficiency and versatility in LDI-TOF-MS analysis based on their well-established synthesis and surface functionalization, large surface area, high laser absorption capacity, and photothermal conversion efficiency. Therefore, Au NSs based LDI-TOF-MS can be a facile, functional, and efficient analytical method for important small biomolecules owing to its simple preparation, rapid analysis, salt-tolerance, signal reproducibility, and quantitative analysis. This review chronologically summarizes the important advance of Au NSs-based LDI-TOF-MS platforms in terms of in-depth mechanism, signal enhancement, quantitative analysis, and disease diagnosis.
In many drinking water treatment plants, chlorination process is one of the main techniques used for the disinfection of water. This disinfecting treatment leads to the formation of disinfection by-products (DBPs) such as haloacetonitriles (HANs), trihalomethanes (THMs), haloacetic acids (HAAs). In this study, headspace-solid phase microextraction (HS- SPME) technique was applied for the analysis of HANs in drinking water. The effects of experimental parameters such as selection of SPME fiber, the addition of salts, magnetic stirring, extraction temperature, extraction time and desorption time on the analysis were investigated. Analytical parameters such as linearity, repeatability and detection limits were also evaluated. The $50/30{\mu}m$-divinylbenzene/carboxen/polydimethylsiloxane fiber, extraction time of 30 minutes, extraction temperature of $20^{\circ}C$ and desorption time of 1 minute at $260^{\circ}C$ were the optimal experimental conditions for the analysis of HANs. The correlation coefficients ($r^2$) for HANs was 0.9979~0.9991, respectively. The relative standard deviations (%RSD) for HANs was 2.3~7.6%, respectively. Detection limits (LDs) for HANs was $0.01{\sim}0.5{\mu}g/L$, respectively.
High molecular weight syndiotactic poly(vinyl alcohol)(HMW s-PVA) with number-average degree of polymerization of 10000 and syndiotactic diad content of 61.5%/iodine complex film was prepared. Its adsorption and desorption behaviors of iodine in hot water were investigated. In comparison with atactic PVA film or low molecular weight s-PVA film, the degree of solubility of s-PVA film and the iodine desorption of HMW s-PVA/iodine film in hot water were limited to an extremely lower level. As the soaking time increased, the iodine desorption in hot water was increased. This reason might be explained by the fact that as the soaking time increased, so the iodine adsorption increased not by the stable molecular complex between PVA and iodine but by the simple physical adsorption of iodine.
In the automotive industry, the hole expanding test is widely used to assess the formability of punched holes in sheets. This test provides a good representation of formability within the framework defined by the ISO 16630 standard. During hole expanding tests on galvanized high strength steels, a negative effect was observed when there was a delay between hole punching and expansion, as compared to performing both operations directly. This effect is believed to be caused by hydrogen aging, which occurs when hydrogen diffuses towards highly-work hardened edges. Therefore, the aim of this study is to demonstrate the migration of hydrogen towards work-hardened edges in high strength Zn-coated steel sheets using a novel Thermal Desorption Analyzer (TDA) designed for small samples. This newly-developed TDA setup allows for the quantification of local diffusible hydrogen near cut edges. With its induction heating and ability to analyze Zn-coated samples while reducing artifacts, this setup offers flexible heat cycles. Through this method, a hydrogen gradient is observed over short distances in shear-cut galvanized steel sheets after a certain period of time following punching.
