Objectives: The purpose of this ex vivo study was to compare the antifungal activity of a synthetic peptide consisting of 15 amino acids at the C-terminus of human ${\beta}$-defensin 3 (HBD3-C15) with calcium hydroxide (CH) and Nystatin (Nys) against Candida albicans (C. albicans) biofilm. Materials and Methods: C. albicans were grown on cover glass bottom dishes or human dentin disks for 48 hr, and then treated with HBD3-C15 (0, 12.5, 25, 50, 100, 150, 200, and $300{\mu}g/mL$), CH ($100{\mu}g/mL$), and Nys ($20{\mu}g/mL$) for 7 days at $37^{\circ}C$. On cover glass, live and dead cells in the biomass were measured by the FilmTracer Biofilm viability assay, and observed by confocal laser scanning microscopy (CLSM). On dentin, normal, diminished and ruptured cells were observed by field-emission scanning electron microscopy (FE-SEM). The results were subjected to a two-tailed t-test, a one way analysis variance and a post hoc test at a significance level of p = 0.05. Results: C. albicans survival on dentin was inhibited by HBD3-C15 in a dose-dependent manner. There were fewer aggregations of C. albicans in the groups of Nys and HBD3-C15 (${\geq}100{\mu}g/mL$). CLSM showed C. albicans survival was reduced by HBD3-C15 in a dose dependent manner. Nys and HBD3-C15 (${\geq}100{\mu}g/mL$) showed significant fungicidal activity compared to CH group (p < 0.05). Conclusions: Synthetic HBD3-C15 peptide (${\geq}100{\mu}g/mL$) and Nys exhibited significantly higher antifungal activity than CH against C. albicans by inhibiting cell survival and biofilm.
The purpose of this study was to evaluate the marginal adaptability of the amalgam restorations in applying the cavity varnish (Copalite$^{(R)}$) and dentin bonding agent (Scotchbond 2$^{(R)}$) under the scanning electron microscope. For this study, eighteen sound extracted human molars were selected. Class I cavities in 12 teeth and class V cavities in 6 teeth were prepared using an air turbine with No. 701 tungsten carbide bur and finished using a low speed handpiece with No. 557 fissure bur. The prepared specimens were then divided into three groups including 4 class I cavities and 2 class V cavities in each group and restored as follows ; Group I. All the prepared cavities were restored with amalgam only (Control). Group II. Two layers of Copalite$^{(R)}$ cavity varnish were applied to the cavities with a gentle stream of air after each application and cavities were restored with amalgam. Group III. The enamel cavity margins were etched with 37% phosphoric acid gel for 60 sec., rinsed for 30 sec. and dried. One layer of visible lightcured Scotchbond Dental Adhesive$^{(R)}$ was applied and immediately cured for 20 seconds with visible light-cure unit and cavities were restored with amalgam. All the specimens were cut at the neck of the teeth and the occlusal halves of specimens were sectioned buccolingually in the longitudinal axis centering the amalgam restorations, using the disk. The cut specimens were ground with sandpapers (400, 600, 800, 1000 grit), and cleaned for 5 minutes in the ultrasonic cleaner (Brason Co. U.S.A.). In the cut surfaces, the amalgam - tooth interfaces were examined under the scanning electron microscope (JSM, 35C type, JEOL). The obtained results were as follows ; 1. The amalgam-tooth interfaces were reduced more significantly in the Copalite$^{(R)}$ and Scotchbond 2$^{(R)}$ application group than in the control group. 2. In the class I cavities, the Scotchbond 2$^{(R)}$ application group showed the findings similar to the Copalite$^{(R)}$ application group in the cavity floor, and the marginal adaptability was better in the side wall than in the cavity floor. 3. In the class I cavities, the Scotchbond 2$^{(R)}$ application group showed better marginal adaptability in the occlusal margin than in the gingival margin. 4. The marginal adaptability was in the order of the Scothbond 2$^{(R)}$ application group, the Copalite$^{(R)}$ application group and the control group.

The objective of this study was to evaluate the toothbrush abrasion characteristics of class V restorations. Thirty extracted human premolars, which were collected from oral surgery clinics were used. We mounted five teeth in a metal ring mold of 50 mm in diameter and 15 mm in height using chemically cured acrylic resin. Class V cavities were prepared in lingual cervical root surfaces and restored using one of following restorative materials : Dentin Conditioner/Fuji II LC (Group FL), All Bond II/Z-250 (Group ZT), One-up Bond F/Palfigue Estelite (Group PE), F2000 Primer/Adhesive (Group FT), and Prime & Bond 2.1/Dyract AP (Group DR). They were stored under distilled water at $37^{\circ}C$ for seven days. The toothbrush abrasion test was conducted using a wear testing machine of pin-on disk type under a load of 1.5 N for 100,000 cycles. We have examined the bonded interfaces, the changes of surface roughness and color of abraded surfaces. From this experiment, the following results were obtained. 1. The change of surface roughness showed high degree: RMGIC>compomer>composite resin (p<0.05). 2. Because of the protrusion and missing of filler particles, SEM observation of abraded surfaces of RMGIC and compomers revealed the increase of surface roughness due to the selective removal of matrix resin. 3. The color change by toothbrush abrasion was affected in large part by the change of $L^*$ and $b^*$ of resin composites (p<0.05). 4. The color change by toothbrush abrasion was so small to detect by human eyes. 5. SEM observation of abraded surfaces revealed the interface bonding was the best in the FT group.
The purpose of this study was to evaluate the color stability of resin cements with accelerated test. Four dual curing resin cements: Panavia-F (KURARAY). Duolink (BISCO), Variolink-II (Ivoclar Vivadent), and RelyX Unicem (3M ESPE) and 1 self curing resin cement: Resiment CE (j. l. Blosser) were used in this study. In control group, Gradia Anterior (GC) composite resin and Tescera Dentin (Bisco) indirect composite were used. Ten disk shape specimens were made from each resin cement. The specimens were subjected to an accelerated aging process in a refrigerated bath circulator at 60$^{\circ}C$ for 15 and 30 days. Spectrophotometric analyses were made before and after 15 days and 30 days of accelerated aging time. The color characteristics ($L^*,\;a^*,\;b^*$) and the color difference (${\Delta}E^*$) of the specimens before and after immersion were measured and computed. Regardless of type of the resin cements, $L^*$ value was decreased and $a^*$ value was increased, but there were no significant difference. But $b^*$ value was increased significantly (p < 0.05). Tescera inlay showed least color change (p < 0.05), but Gradia showed notable color change after 15 days. After 30 days on accelerated aging, ${\Delta}E^*$ value was increased (Panavia-F < Variolink-II < Resiment CE < Duolink < Unicem) (p < 0.05). but there were no significant difference among Panavia-F, Variolink-II, and Resiment CE groups. After 30 days of accelerated aging, ${\Delta}E^*$ value of all resin cements were greater than 3.0 and could be perceived by the human eye.
Purpose: This study is to evaluate the effect of modeling liquid on the shear-bond strength between zirconia core and veneering ceramic. Methods: Disk-shaped (diameter: 12.0mm; height: 3.0mm) zirconia were randomly divided into six groups according to the surface conditioning method and whether modeling liquid is used or not to be applied (N=60, n=10 per group): group 1-control group with distilled water(ZD); group 2-control group with modeling liquid(ZM); group 3-airborne particle abrasion with $110-{\mu}m$$Al_2O_3$(AD) with distilled water; group 4-airborne particle abrasion with $110-{\mu}m$$Al_2O_3$ with modeling liquid(AM); group 5-liner with distilled water(LD); group $6{\pounds}{\neq}liner$ with modeling liquid(LM). Contact angles were determined by the sessile drop method at room temperature using a contact angle measurement apparatus. The specimens were prepared using dentin veneering ceramics, veneered, 3mm high and 2.8mm in diameter, over the cores. The shear bond strength test was performed in a Shear bond test machine. Load was applied at a cross-head speed of 0.50mm/min until failure. The fractured zirconia surfaces were evaluated by using stereomicroscope (${\times}30$). Collected data were analyzed using SPSS(Statistical Package for Social Sciences) Win 12.0 statistics program. Results: ZD showed the highest contact angle($50.6{\pm}5.4^{\circ}$) and LD showed the lowest value($6.7{\pm}1.3^{\circ}$). Control groups and zirconia liner groups were significantly higher contact angle than liner groups(p<0.05). LD was the highest shear bond strength($43.9{\pm}3.8MPa$) and ZD was the lowest shear bond strength($24.8{\pm}4.9MPa$). Shear bond strengths of control groups and contact angle of liner groups were not significantly different((p>0.05). Liner groups presented adhesive failures. The others groups showed cohesive and adhesive failures. Conclusion: Modeling liquid groups showed lower contact angles and lower shear bond strength compared to those of distilled water groups.

Purpose: Surface damage and bonding strength difference after micromechanical treatment of zirconia surface are to be studied yet. The aim of this study was to evaluate the difference of fracture resistance and bonding strength between more surface-damaged group from higher air-blasting particle size and pressure, and less damaged group. Materials and Methods: Disk shape zirconia ($LAVA^{TM}$) was sintered and air-blasted with $30{\mu}m$ particle size (Cojet), under 2.8 bar for 15 seconds, $110{\mu}m$ particle size (Rocatec), under 2.8 bar for 15 seconds, and $110{\mu}m$ particle size (Rocatec), under 3.8 bar for 30 seconds respectively. Biaxial flexure test and bonding failure load test were performed serially (n = 10 per group). For bonding test, specimens were bonded on the base material having similar modulus of elasticity of dentin with $200{\mu}m$-thick resin cement for tension of surface damage. Failure load of bonding was detected with acoustic emission (AE) sensor. Results: There were no significant differences both in the biaxial flexure test and bonding failure load test between groups (P > 0.05). Sub-surface cracks were all radial cracks except for two specimens. Conclusion: Within the limitations of no aging under monotonic load test, surface damage from higher air-blasting particle size and pressure was not significant. Evaluations of failure load with bonded zirconia disks was clinically relevant modality for surface damage and bonding strength, simultaneously.
The purpose of this study was to compare the ability of three resin surface sealants to prevent microleakage in Class V composite resin restorations. Forty Class V cavities with the occlusal margin in enamel and gingival margin in dentin were prepared on the buccal surfaces of sound extracted molars, and restored with composite resin. Restorations were randomly assigned into one of four equal groups (n = 10): a control group, without resin sealing, and three experimental groups in which margins were sealed with Fortify Plus, Biscover and Permaseal, respectively. Specimens were thermocycled, immersed in a 2% methylene blue solution for 4 hours, sectioned longitudinally, and observed the leakage at the occlusal and gingival margins. The result was analyzed using Kruskal-Wallis test, Mann-Whitney test and Wilcoxon signed rank test. In conclusion, the ability to reduce microleakage at occlusal margins was similar in all of three sealants. However at gingival margin, it depended on the type of used resin surface sealant. At gingival margin. control and Fortify Plus group showed statistically higher microleakage than PermaSeal group. and Fortify Plus group also showed higher microleakage than BisCover group (p < 0.05).
The purpose of this study was to investigate the distribution of calcitonin gene-related peptide containing nerve fibers in rat pulp after dentinl injury by means of immunohistochemistry and confocal laser scanning microscope. The Spague-Dawley rats weighing about 250-300gm were used. The animals were devided into normal control and experimental groups. Experimental animals were sacrified 1, 2, 4, 7, 10, 21days after dentinal injury (dentin cutting, and then acid etching with 35% phosphoric acid) on the maxillary molar teeth. The maxillary teeth and alveolar bone were removed and immersed in the 4% paraformaldehyde in 0.1M phosphate buffer (pH 7.4), then were decalcified with 15% formic acid for 10 days. Serial frozen $50{\mu}m$ thick sections were cut on a cryostat. The rabbit CGRP antibody was used as a primary antibody with a dilution of 1:2000 in 0.01M PB. The sections were incubated for 48 hours at $4^{\circ}C$, and placed into biotinylated antirabbit Ig G as a secondary anti body with dilution of 1:200 in 0.01M PB and incubated in ABC(avidin-biotin complex). The peroxidase reaction was visualized by incubating the sections in 0.05% 3,3 diaminobenzidine tetrahydrochloride containing 0.02% $H_2O_2$. For the confocal laser scanning microscopic examination, Primary antibody reaction was same as immunoperoxidase stainning, but fluorescein isothiocyanate(FITC)-conjugate antirabbit IgG as a secondary antibody was used. The confocal laser scanning microscope was used for the examination. A series of images of optical sections was collected with a 20x objective at $3{\mu}m$ intervals throughout the depth of specimen. FITC fluerescence was registrated through a 488nm and 568nm excitation filter, and images were saved on optical disk. The stereoscopic images and three dimentionnal images were reconstructed by computer software, and then were analyzed. The results were as follows : 1. In normal control group, CGRP containing nerve fibers were coursed through the root with very little branching, and then formed a dense network of terminals in coronal pulp. 2. A slight increase in CGRP containing nerve fibers at 1 and 2day postinjury was noted subjacent to the injury site. In the 4day group, there were an extensive increase in the number of reactive fibers, followed by a partial return toward normal levels at 7~10 day postinjury, and return by 21days. 3. The sprouting of the CGRP containing nerve fibers was evident within 2day after dentinal injury, and by 4days there was a maximal increased, but was decreased at 7days and returned to normal 10~21 day postinjury. 4. In confocal laser scanning microscopic exammination, the distinct distribution pattern and sprouting reaction of CGRP containing nerve fibers were observed in stereoscopic images and three dimentional images. These results suggest that CGRP containing nerve fiber can be important role in the response to dental injury and pain regulation.
