Delphinidin Chloride Effects on the Expression of TNF-$\alpha$ Induced Cell Adhesion Molecules (TNF-$\alpha$에 의해 유도된 세포부착분자의 발현에 대한 Delphinidin chloride의 억제 효과)

  • The process of atherosclerosis begins through secretion of inflammatory cytokine or adhesion of leukocyte from damage in blood vessels and transmigration. This study was conducted to investigate the effects of delphinidin chloride (DC) in the initial process of atherosclerosis on the expression of ICAM-1 (Intracellular Adhesion Molecule-1) and VCAM-1 (Vascular Adhesion Molecule-1) related to adhesion of leukocyte at the HUVEC (human umbilical vein endothelial cell line. As a result, cell growth inhibition rate at 50 ${\mu}M$ was respectively 4, 3 and 5% without cell toxicity. As a result of morphological observation monocyte-endothelial cell adhesion assay and optical microscope carried out to measure attachment of mononuclear cells to endothelial cells induced by Tumor necrosis factor-alpha (TNF-$\alpha$) at concentrations without cell toxicity, DC concentration-dependently suppressed attachment. When effects on the expression of VCAM-1 and ICAM-1, cell adhesion molecules induced from endothelial cells by TNF-$\alpha$, were comparatively analyzed using western blot analysis and RT-PCR methods, protein of VCAM-1 and ICAM-1 and expression at the level of mRNA were concentration-dependently reduced. Taken together, the results of this studies provide evidence that DC possess an anti-metastatic activity.

Effects of polyphenol and anthocyanin contents of Punica granatum fruit parts on their antioxidant and anticancer activities (석류 과실 부위별 폴리페놀 및 안토시아닌 함량이 항산화 및 항암 활성에 미치는 영향)

  • The purpose of this study was to determine the content of phenolics and various anthocyanin compounds, and antioxidant and anticancer activities of the extracts from pomergranates, which were produced in two different regions including Goheung, Korea and California, USA. These pomergrantes were divided into juice, pericarp, and seed parts and each part was extracted with 95% methanol. Content of total phenolics [mg chlorogenic acid equivalents/kg DW] was highest in pericarp, followed by juice and seeds from pomergrantes in both regions. The anthocyanins identified in pomegranate fruit were cyanidin-3,5-diglucoside chloride (Cy3,5G), cyanidin-3-O-glucoside chloride (Cy3G), delphini din-3,5-di-O-glucoside chloride (Dp3,5G), delphinidin-3-O-glucoside chloride (Dp3G), pelargonidin-3,5-di-glucoside chloride (Pg3,5G), and pelargonidin-3-glucoside chloride (Pg3G). Among these, cyanidin-3,5-diglucoside chloride (Cy3,5G) was the major anthocyanin in California pomegranate fruit juice and Goheung pomegranate fruit pericarp. DPPH (1,1-diphenyl-2-picryl hydrazyl radical) free radical scavenging activity was dose-dependently increased, and was higher in pericarp part than juice or seed parts from pomergrantes in both regions. By MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay, pericarp extracts from pomergrantes in both regions showed the highest anticancer activity, which was higher on Calu-6 for human pulmonary carcinoma than SNU-601 for human gastric carcinoma. Correlation between polyphenols and anticancer activity on Calu-6 was determined to be in the range of $r^2=0.8904$ to 0.9706.

Effects of Acidic Treatments for Anthocyanin and Proanthocyanidin Extraction on Black Bean (Glycine max Merrill.) (검정콩 안토시아닌 및 프로안토시아니딘 추출에 대한 산 처리의 영향)

  • This study was performed to investigate the effects of various acidic treatment for anthocyanin and proanthocyanidin extraction from black bean. Extracts were prepared with 80% methanol solutions containing HCl, acetic, formic, phosphoric, and citric acids of different concentrations (0.1, 0.2, 0.3, 0.5, and 0.7%). Total anthocyanin content ranged from 0.74 mg/g in 0.7% citric acid to 1.74 mg/g in 0.3% HCl, depending on acid type and concentration. The major anthocyanins were delphinidin-3-glucoside (D3G), cyanidin-3-glucoside (C3G), and petunidin-3-glucoside (Pt3G). The highest C3G content was 1.12 mg/g in 0.3% HCl. Proanthocyanidin content ranged from 2.01 mg/g in 0.5% acetic acid to 5.29 mg/g in 0.3% HCl. Thus, acidic condition is a significant factor affecting anthocyanin and proanthocyanidin extraction from black bean, and the optimum extraction conditions were determined as 80% methanol containing 0.3% HCl.
