Enhancement of Enzymatic Hydrolysis of Lignocellulosic Biomass by Organosolv Pretreatment with Dilute Acid Solution (효소당화를 위한 목질계 바이오매스의 유기용매 침출 전처리 공정)

  • Organosolv pretreatment is the process to frationation of lignocellulosic feedstocks to enhancement of enzymatic hydrolysis. This process has advantages that organic solvents are always easy to recover by distillation and recycled for pretreatment. The chemical recovery in organosolv pretreatment can isolate lignin as a solid material and carbohydrates as fermentable sugars. For the economic considerations, using of low-molecular-weight alcohols such as ethanol and methanol have been favored. When acid catalysts are added in organic solvent, the rate of delignification could be increased. Mineral acids (hydrochloric acid, sulfuric acid, and phosphoric acid) are good catalysts to accelerate delignification and xylan degradation. In this study, the biomass was pretreated using 40~50 wt% ethanol at $170{\sim}180^{\circ}C$ during 20~60 min. As a results, the enzymatic digestibility of 2-stage pretreatment of rigida using 50 wt% ethanol at $180^{\circ}C$ was 40.6% but that of 1-stage pretreatment was 55.4% on same conditions, therefore it is shown that the pretreatment using mixture of the organosolv and catalyst was effective than using them separately.

  • Recently many scientists have tried to synthesize biodegradable polymers due to durable and non-biodegradable products of conventional synthetic plastics when these were wasted in nature. So to reuse the wastepapers for biodegradable polymer resources, ONP (old newsprint), OCC (old corrugated containerbpard) were carried out by the pretreatment of chlorinite, hypochlorite and oxygen-alkali treatment conditions. For manufacturing of biodegradable polymer with wastepaper, this study performed to investigate change of chemical components and optimal pretreatment condition. The summarized results in this study were as follows: Lignin content in ONP and OCC was was higher than in MOW and ash content was the highest in MOW. More amount of ash components were reduced by wet defiberation than by dry defiberation. Wet defiberation fiber are better than dry defiberated fiber in chemical pretreatment condition for wastepapers, and the best result was obtained in the condition of sodium chlorite at $70^{\circ}C$, because it has high delignification ratio, ${\alpha}$-cellulose contents and degree of polymerization in this treatment condition. Oxygen-alkali treatment condition is the worst method because of low yield, low degree of polymerization in this pretreatments.

Characterization of Pretreatment by NaOH Leaching for Production of Bioethanol from Palm Waste (팜 부산물 활용 바이오 에탄올 생산을 위한 NaOH 전처리 공정의 특성)

  • 본 연구는 팜 부산물로부터 바이오 에탄올을 생산하는 전처리-당화-발효 공정의 첫 번째 단계인 전처리 공정에서 팜 부산물을 NaOH를 이용하여 효율적으로 전처리하고자 하였다. 암모니아 침지법과 NaOH 침출법을 비교한 결과 팜 부산물에 대해서는 암모니아 침지에 의한 탈리그닌 효과가 적으며 NaOH 전처리가 적합한 방법임을 알 수 있었다. 40-100 mesh 크기의 팜 부산물을 이용하여 반응온도(110, 130, $150^{\circ}C$), 반응시간(20, 40, 60분) 및 NaOH 농도(5%, 11%)의 변화에 따른 팜 부산물의 탈리그닌율과 글루코스 및 자일로스 회수율 간의 상호관계를 확인하였다. $150^{\circ}C$까지의 온도 조건에서 온도에 의한 자일로스의 분해는 일어나지 않는 것으로 확인되었다. 팜 부산물의 탈리그닌율은 시간이 증가할수록 증가하였으며, 높은 NaOH 농도에서 더 높은 것으로 나타났다. 그러나 글루코스 및 자일로스의 회수율은 높은 농도에서 낮게 나타났으며, 시간이 지날수록 감소하여 손실이 많은 것으로 나타났다. 따라서 NaOH 농도가 낮을수록 당 회수율은 높게 나타나지만, 탈리그닌율이 낮아 당화 효율이 떨어지므로 효소 당화 후에 최종 당 회수율이 높은 NaOH 농도 조건을 결정하여야 하겠다.

Chemical Characteristics and Ethanol Fermentation of the Cellulose Component in Autohydrolyzed Bagasse

  • The chemical characteristics, enzymatic saccharification, and ethanol fermentation of autohydrolyzed lignocellulosic material that was exposed to steam explosion were investigated using bagasse as the sample. The effects of the steam explosion on the change in pH, organic acids production, degrees of polymerization and crystallinity of the cellulose component, and the amount of extractive components in the autohydrolyzated bagasse were examined. The steam explosion decreased the degree of polymerzation up to about 700 but increased the degree of crystallinity and the micelle width of the cellulose component in the bagasse. The steam explosion, at a pressure of 2.55 MPa for 3 mins, was the most effective for the delignification of bagasse. 40 g/L of glucose and 20 g/L of xylose were produced from 100 g/L of the autohydrolyzed bagasse by the enzymatic saccharification using mixed cellulases, acucelase and meicelase. The maximum ethanol concentration, 20 g/L, was obtained from the enzymatic hydrolyzate of 100 g/L of the autohydrolyzed bagasse by the ethanol fermentation using Pichia stipitis CBS 5773; the ethanol yield from sugars was 0.33 g/g sugars.

  • A major problem in the enzymatic hydrolysis of lignocellulosic substrates is the very strong bonding of cellulase to lignin and even cellulose in the hydrolysis residues. This phenomenon inhibits recycle of the cellulase which is a major expense of the enzymatic hydrolysis process. In this paper, autohydrolyzed wood was delignified by two-stage with a 0.3% Na OH extraction and oxygen-alkali bleaching and was subjected to enzymatic hydrolysis with cellulase. Also, an improved almost quantitative recycle process of cellulase enzyme was discussed. In enzyme recovery by affinity method. the first recycling showed relatively high hydrolysis rate of 97.4%. Even at the third recycle. hydrolysis rate was 86.7 percents. In the case of cellulase recovery by ultrafiltration method, first 2 recycling treatments resulted very high hydrolysis rate(97.0-97.7%). Even the third recycling showed about 94.2%. Authoydrolysis of oak wood followed by 2-stage delignification with alkali and oxygen-alkali produced a substrate for enzymatic hydrolysis that allowed almost quantitative recycle of cellulase.

  • Steam-exploded woods were delignified by two-stage with a 0.3% NaOH extraction and oxygen-alkali bleaching and were subjected to the enzymatic hydrolysis with cellulase enzyme. Also, an improved almost quantitative recycle process of cellulase enzyme was discussed. In enzyme recovery by affinity method, The first recycling showed relatively high hydrolysis rate of 96.4%. Even at the third recycle, hydrolysis rate was 87.0 percents. In the case of cellulase recovery by ultrafiltration method, first 2 recycling treatments resulted in very high hydrolysis rates, 96.8% and 95.0%, respectively. Even the third recycling showed about 93.6%. Steam-explosion treatment of oak wood followed by 2-stage delignification with alkali and oxygen-alkali produced a excellant substrate for the enzymatic hydrolysis that allowed almost quantitative recycle of cellulase.

Impact of Residual Extractives and Hexenuronic Acid on Lignin Determination of Kraft pulps

  • The amount of non-lignin components in unbleached and oxygen-delignified kraft pulps and their impact on lignin determinations was investigated. The lignin analyses investigated were kappa number and Klason lignin in conjunction with acid-soluble lignin. The species investigated were loblolly pine, and aspen. The non-lignin components that impacted on lignin determination were residual extractives and hexenuronic acid in unbleached and oxygen-delignified kraft pulps. In the hardwoods, significant amounts of extractives remained after kraft pulping and oxygen delignification. These residual extractives in the hardwood pulps had an impact on the lignin determination, more so on the acid lignin method than kappa number. Hexenuronic acid only impacts on kappa number determination both softwood and hardwood pulps, not on acid lignin. Hexeneuronic acid contributed as lignin content more in aspen than pine pulps, and more in oxygen-delignified than unbleached kraft pulps. Impact of hexenuronic acid on should be corrected both softwood and hardwood pulps for accurate kappa number.

Effect of Molar Ratios of DES on Lignin Contents and Handsheets Properties of Thermomechanical Pulp (TMP의 LB 공융용매 처리 시 젖산/베타인 몰 비율이 리그닌 함량 및 수초지 특성에 미치는 영향)

  • This study was conducted to investigate the effects of deep eutectic solvent (DES) treatment on properties of TMP fibers and handsheets. DES was prepared by mixing of lactic acid and betaine (L and B), and the molar ratio of these two components mixtures was controlled to 2:1 (L:B=2:1) and 5:1 (L:B=5:1). As results, lignin was partly extracted from the TMP fibers. Especially, the delignification of TMP samples was promoted according to the increase of the molar ratio of lactic acid. Except for tear index, both tensile index and burst index of handsheets were increased when higher molar ratio of lactic acid was mixed for DES preparation. However, the LB DES treatment of TMP fibers didn't give any effect on the optical properties of TMP handsheets. The plausibility of TMP fiber was to be enhanced by LB DES treatment.

Enzymatic hydrolysis and micro-structure of ozone treated wood meal (오존 처리에 의한 목재 세포벽의 미세구조변화와 효소가수분해)

  • Pine (Pinus densiflora) and aspen (Populus euramericana) wood meals were treated with ozone at various time schedule in acidic condition. The lignin contents and surface area of the ozone treated wood meals were determined and the enzymatic hydrolysis rate of ozonated wood meals was evaluated. The feasibility of enzymatic hydrolysis of the ozone treated wood meal was obviously influenced with the degree of delignification. After ozone treatment of wood meal for 10min, total pore volume were slightly increased in the surface of wood meal. When wood meals were treated with ozone longer than 10min, few change in the pore volume was observed. However, the area of over $50{\AA}$ of pore size is increased with ozonation time. As a conclusion, the rate of enzymatic hydrolysis of wood is more effective with the pore size distribution than the total pore volume.

  • A series of studies on nitric-acid pulping of municipal recycled waste papers were carried out to substitute the bleached chemical pulp imported for producing printing paper as well as to use its solidified spent-liquor as fertilizer. The first experiment was carried out to find the optimum treatment conditions such as pulp consistency, nitric acid charge and temperature in $HNO_3$-alkali pulping process. The results obtained were as follows: 1. Some selective delignification of OCC pulp was conducted by $HNO_3$-alkali process. The higher the temperature and concentration of nitric acid, the lower the pulp yield and kappa number of treated pulp. while its brightness was increased. 2. The higher consistency required the stronger mixing in case of more than 5% pulp. 3. In the laboratory, the suitable $HNO_3$-treating condition seemed to be less than 6% consistency, lower than 500% $HNO_3$charge on pulp and lower than $100^{\circ}C$ in cooking temperature. 4. The spent liquor with 1.77% N-content seemed to be slow-release nitrogen fertilizer suitable for agriculture.
