Dehydrogenase Activity and Physico-chemical Characteristics of Park Soils in Seoul (서울 공원 토양의 탈수소효소 활성과 물리화학적 특성)

  • The relationships between microbial activity and disturbance level of soil were investigated from 15 parks in Seoul and undisturbed area. The physico-chemical characteristics of soil and dehydrogenase activity(DHA) as an index of soil microbial activity were analysed. There were ranges of 3.84~7.37 in pH, 9.63~40.33% in moisture content, 3.41~21.49% in organic matter, 0.36~0.79g/g in water holding capacity and 0.03~0.53% in total nitrogen investigated sites. DHA values of soil were 8.64~$146.76{\mu}g/g$ in park soil and 545.14~$1, 198.80{\mu}g/g$ in undisurbed area. DHA of park soil with high traffic density and contamination source from human activities was much lower than that of undisturbed area. DHA was positively correlated with moisture content, organic matter, water holding capacity and total nitrogen.

  • The relationships between microbial activity and physico-KDICical characteristics of soils were investigated in three golf courses of Kwanak, Gold and Korea Country Clubs, with different open years. The soil samples were collected in tee, fairway and rough. There were ranges of 4.80-5.55 in pH, $25.55-98.50{\mu}S$ / cm in conductivity, 10.96-16.73% in moisture content, 0.18-0.36g / g in water holding capacity, 3.68-5.39% in organic matter, and 0.10-0.25% in total nitrogen. Dehydrogenase activity(DHA) as an index of soil microbial activity was determined. DHA values of soil were $69.83-314.43{\mu}$g / g in three courses and showed the order of Kwanak>Gold>Korea Country Club with open year. This indicates that DHA was affected by several fertilizer treatments rather than herbicide and pesticide treatments. DHA was significantly different with golf clubs as well as golf courses and positively correlated with water holding capacity and total nitrogen.

  • The relationship between microbial activity and physico-chemical characteristics of golf course soils were investigated in Kyonggi Province with different open years. The increasing tendency of soil PH with depth seemed to the result of mineral leaching. Exchangeable $Ca^2$+ , $Mg^2$+ and K+ showed relatively higher concentration in lower horizon indicating the leaching of minerals. The minerals were heavily leached in older golf course. There was a significant positive correlation between pH and $Ca^2$+.The contents of organic matters, total nitrogen and phosphorus were high in higher horizon.Dehydrogenase activity(DHA) and the content of ATP were determined as an index of soil microbial activity. DHA value was significantly different with golf club as well as golf courses ac-cording todifferent open year. This indicates that DHA was affected by several fertilizertreatments rather than herbicide pesticide treatments. DHA was positively correlated with water holding capacity and total nitrogen. The content of ATP was high in fairway and in golf course with older open year, showing the similar tendency with the results of DHA.

  • The relationships among the contents of Pb, Zn, Cd, Cu and microbial biomass and activity of soil were investigated in Kyongbu, Yongdong, and Chungbu highways. The heavy metal concentrations were the highest in Kyongbu highway with high traffic density. The levels of these metals in soil in three highway sites were much higher than Mt. Kwank as control site. The highest concentrations of heavy metals were found in the upper layer of soil adjacent to the roadside. Dehydrogenase activity (DHA) and adenosine tri-phosphate (ATP) contents were generally lowest at distance of 0.5 m from the roadside. Simple regression analysis indicated that DHA and ATP contents were highly negatively correlated with Pb and Zn concentrations.

  • Soil microorganism activity in an agricultural field is affected by various factors including climate conditions, soil chemical properties, and crop cultivation. In this study, we elucidate the correlation between microorganism activity and agricultural environment factors using the dehydrogenase activity (DHA) value, which is one of the indicators of soil microbial activity. As a result, the various factors noted above were related to the DHA value. Annual rainfall, soil Mg2+, bacterial and fungal diversities, types of crops, developmental stages, seasons, and cultivation status were highly correlated with the DHA value. Furthermore, next-generation sequencing (NGS) analysis was used to identify that the type of crop affected soil microbial compositions of both bacteria and fungi. Soil used for soybean cultivation showed the highest relative abundance for Verrucomicrobia, Planctomycetes, and Acidobacteria but Actinobacteria and Firmicutes had the lowest relative abundance. In the case of soil used for potato cultivation, Actinobacteria had the highest relative abundance but Proteobacteria had the lowest relative abundance. Armatimonadetes showed the highest relative abundance in soil used for cabbage cultivation. Among the fungal communities, Mortierellomycota had the highest relative abundance for soybean cultivation but the lowest relative abundance for cabbage cultivation; further, Rozellomycota, Chytridiomycota, and Cercozoa had the highest relative abundance for cabbage cultivation. Basidiomycota had the highest relative abundance for potato cultivation but the lowest relative abundance for soybean cultivation.

Characterization of Glycerol Dehydrogenase from Thermoanaerobacterium thermosaccharolyticum DSM 571 and GGG Motif Identification

  • Glycerol dehydrogenases (GlyDHs) are essential for glycerol metabolism in vivo, catalyzing its reversible reduction to 1,3-dihydroxypropranone (DHA). The gldA gene encoding a putative GlyDH was cloned from Thermoanaerobacterium thermosaccharolyticum DSM 571 (TtGlyDH) and expressed in Escherichia coli. The presence of Mn2+ enhanced its enzymatic activity by 79.5%. Three highly conserved residues (Asp171, His254, and His271) in TtGlyDH were associated with metal ion binding. Based on an investigation of glycerol oxidation and DHA reduction, TtGlyDH showed maximum activity towards glycerol at 60℃ and pH 8.0 and towards DHA at 60℃ and pH 6.0. DHA reduction was the dominant reaction, with a lower Km(DHA) of 1.08 ± 0.13 mM and Vmax of 0.0053 ± 0.0001 mM/s, compared with glycerol oxidation, with a Km(glycerol) of 30.29 ± 3.42 mM and Vmax of 0.042 ± 0.002 mM/s. TtGlyDH had an apparent activation energy of 312.94 kJ/mol. The recombinant TtGlyDH was thermostable, maintaining 65% of its activity after a 2-h incubation at 60℃. Molecular modeling and site-directed mutagenesis analyses demonstrated that TtGlyDH had an atypical dinucleotide binding motif (GGG motif) and a basic residue Arg43, both related to dinucleotide binding.

Measurement of Ammonia Inhibition of Activated Sludge by DHA-INT (DHA-INT를 이용한 활성슬러지의 암모니아 저해도)

  • It is a time consuming work to identify the inhibition of unknown chemicals or industrial wastewater. Thus it is needed to establish a fast assay tool for finding a toxicant source. Biomass activity and ammonia inhibition were measured by DHAINT method. Ammonia inhibition tests were comprised of total ammonia inhibition and free ammonia inhibition. Those inhibitions were carried out by nitrifier and heterotroph each other with nitrifier inhibitor. The ammonia inhibition was proportional to an amount of total ammonia and pH increase. It meaned that a free ammonia played a key role for ammonia inhibition. however both total ammonia and free ammonia should be considered for an accurate assay of the ammonia inhibition. Nitrifier was more sensitive than heterotroph when the ammonia concentration above 3.000mg/L.

Study on the Performances and Microbial Community in the Biofilm Process for Treating Nonpoint Source Pollutants (비점오염물질 처리를 위한 생물막 공정의 운전 및 미생물 군집의 특성)

  • In this study, biofilm process was introduced for treating nonpoint source pollutants. The ceramic media were provided for biofilm growth in the reactors. The packing ratio of ceramic media was 5% and 15(v/v)%, respectively. Thereafter, the reactors were operated intermittently with the different interevent periods such as 0, 5, 10 and 15 days, respectively. The removal efficiencies of COD and NH$_4{^+}$-N were investigated at the different operating conditions such as media packing ratio, temperature, and interevent period. Additionally, Polymerase chain reaction(PCR)-denaturing gel gradient electrophoresis(DGGE) and INT-dehydrogenase activity(DHA) test were conducted to observe the microbial community and activity in the biofilm. Consequently, the interevent period seemed to have no significant influence on the COD removal efficiency. COD was removed within 6$\sim$8 hours at 25$^{\circ}C$ and about 15 hours at 10$^{\circ}C$. DGGE profiles showed that the initial species of microorganisms were changed from seeded activated sludge into the microorganisms detected in sediments. INT-DHA test also showed that the activities of microorgnaisms were not decreased even in the 15 days of interevent period.

Effects of ${\alpha}-linolenic$, eicosapentaenoic and docosahexaenoic acids administration on lowering of triacylglycerol level in the hepatic and serum of rats (n-3계 지방산 투여가 성장기 흰쥐의 간장 및 혈청 Triacylglycerol 농도에 미치는 영향)

  • We studied the difference effects of dietary ${\alpha}-linolenic\;acid\;({\alpha}-LA,\;18:3\;n-3)$, eicosapentaenoic acid (EPA, 20:5 n-3) and docosahexaenoic acid (DHA, 22:6 n-3) on the lowering of triacylglycerol in the liver and serum on lipid metabolism in rats. Rats were fed semipurified diets containing 10% fat with constant polyunsaturated/monounsaturated/saturated fatty acids (1:1:1) and n-6/n-3 ratio (1:2). EPA (98%) and DHA (98%) were added in diets as the ethyl esters. The concentration of liver triacylglycerol was significantly lower in rats fed both EPA and DHA than in those fed ${\alpha}-LA$. The concentration of liver phospholipid was significantly higher in rats fed DHA than in those fed ${\alpha}-LA$ and EPA. Both EPA and DHA reduced serum triacylglycerol concentration compared with ${\alpha}-LA$, but this effect was more pronounced in the EPA diet. The activity of phophatidate phosphohydrolase in the liver microsome was significantly lower in rats fed both EPA and DHA than in those fed ${\alpha}-LA$. but, there was no significant difference on the activities of diacylglycerol acyltransferase among the three groups. The concentration of liver triacylglycerol were correlated with changes in the microsomal phosphatidate phosphohydrolase activity (r=0.84). Hepatic NADPH generating enzyme, glucose-6-phosphate dehydrogenase was more effective to reduce the activity in rats fed both EPA and DHA than in those fed ${\alpha}-LA$. In conclusion, EPA or DHA reduced the hepatic triacylglycerol concentration by inhibiting microsomal phosphatidate phosphohydrolase, thereby inhibiting synthesis of triacylglycerol in the liver.

Enzyme Activities in the Soil of Quercus mongolica Forests (신갈나무 산림토양에서의 효소활성도)

  • The present paper describes partial results of the study on the activities of microbes in the soil of Quercus mongolica forest from July, 1994 to April, 1995. To determine the relationship between structure and function of soil microbial ecosystem, the author investigated the seasonal change of physical environmental factors, microbial population and soil enzyme activities. The changes of pH was not significant and the temperature of surface soil was 2℃ higher than lower soil through out the year. Moisture contents (%) of soil samples ranged from 7.64% to 42.11%. However, soils of site 3 at Mt. Komdan in which vegetation is successional have higher moisture content than the others. The bacterial population increased in summer, but continuously decreased in autumn and winter, and then reincreased again in spring. Bacterial population of surface soil was higher than those of 30 cm depth all the year round. Dehydrogenase activity (DHA) was about two-fold higher throughout in surface soil compared to those of lower soil. And the correlation coefficient between DHA and bacterial population size was 0,713, It was suggested that DHA could be used as a primary index of soil microbial population and activity in soil ecosystem.
