Effects of Adenoviral Gene Transduction on the Stemness of Human Bone Marrow Mesenchymal Stem Cells

  • Human mesenchymal stem cells (MSCs) are currently being evaluated as a cell-based therapy for tissue injury and degenerative diseases. Recently, several methods have been suggested to further enhance the therapeutic functions of MSCs, including genetic modifications with tissue- and/or diseasespecific genes. The objective of this study was to examine the efficiency and stability of transduction using an adenoviral vector in human MSCs. Additionally, we aimed to assess the effects of transduction on the proliferation and multipotency of MSCs. The results indicate that MSCs can be transduced by adenoviruses in vitro, but high viral titers are necessary to achieve high efficiency. In addition, transduction at a higher multiplicity of infection (MOI) was associated with attenuated proliferation and senescence-like morphology. Furthermore, transduced MSCs showed a diminished capacity for adipogenic differentiation while retaining their potential to differentiate into osteocytes and chondrocytes. This work could contribute significantly to clinical trials of MSCs modified with therapeutic genes.

누에분말이 혈청중의 활성산소 및 제거효소에 미치는 영향 (Effects of Silkworm Powder on Oxyen Radicals and Their Scavenger Enzymes in Serum of Rats)

  • This study was designed to investigate the effects of silkworm powder (SWP) on oxygen radicals and their scavenger enzymes in serum of rats. Sprague Dawley (SD) male rats (160${\pm}$10g) were fed experimental diets (SWP-200and SWP-400 groups)added 200 and 400mg/kg BW/day for 6 weeks. Triglyceride (TG)levels were significantly inhibited (10% and 25%) in SWP-200 and SWP-400 groups, but there were no singificant differences in total, LDL-and HDL-cholesterol levels in both SWP-200 and SWP-400 groups. Hydroxyl radical ($.$OH) formations resulted in a marked decreases (about 20%) in both SWP-200 and SWP-400 groups compared with control group. Superoxide radical (O2.-)and hydrogen peroxide formations resulted in a significant decreases in SWP-400 group compared with control group. Lipid peroxide (LPO)and oxidized protein(>C=O group) productions resulted in a significant decreases (SOD) and catalase (CAT) activiteies were remarkably increased (10∼25% and 40∼50%)in SWP-200 and SWP-400groups. Glutathione peroxidase(GSHPX) activities were significantly increased (about 10%) in SWP-400 groups compared with control group. These results suggest that anti-aging effect of silkworm powder (SWP) may play a pivotal role in attenuation a various chronic degenerative diseases age-related changes.

닥나무 뿌리껍질의 항산화 성분 (Antioxidant Components from Broussonetia kazinoki)

  • From the root bark of Broussonetia kazinoki (Moraceae) two antioxidant components were identified. Their structures were determined as kazinol A (1) and kazinol E (2) (is oprenylated flavan and isoprenylated 1,3-diphenyl propan derivatives) by comparing NMR data with those of the reported compounds from relative plants. The antioxidant activity of 1 and 2 were monitored by the method of DPPH radical scavenging activity, whose $SC_{50}$ values were $41.4{\mu}M\;and\;33.4{\mu}M$, respectively. These compounds also exhibited inhibitory activity against tyrosinase, which is the sole key enzyme for the melanin biosynthesis and play a role in conversion of tyrosine to dopa, and dopa to dopaquinone. The antioxidant effect and the suppression of melanin biosynthesis are useful for anti-aging, increasing vitality in the incidence of major degenerative diseases, and cosmetic products in relation to hyperpigmentation.

미주신경 전기 자극이 노인의 항노화 호르몬과 전신적 염증에 미치는 효과 (Effect of Electrical Vagus Nerve Stimulation to Control Systemic Inflammation and Anti-Aging Hormone on Elder)

  • Purpose : As age increases, a low-level systemic inflammatory state develops and the levels of anti-aging hormones decrease. The vagus nerve activates parasympathetic nerves and promotes sulfation and secretion of neurotransmitter in the brain. Therefore, the purpose of this study was to investigate the effects of electrical vagus nerve stimulation on systemic inflammation (CRP) and anti-aging hormone (DHEA-S) levels in elder people. Methods : A total of 30 healthy elder people participated in this study, randomly divided into two groups of 15 subjects. Electrical vagus nerve stimulation was applied to the experimental group for 4 weeks. CRP and DHEA-S levels were compared with those of the control group. Results : The CRP level was significantly lower in the experimental group than in the control group. In the experimental group, there was a significant decrease in CRP before and after the intervention. However, the DHEA-S level was not significantly different between groups. Conclusion : Electrical vagus nerve stimulation may alleviate the low-level systemic inflammatory state found in elderly people. These results suggest that it may have the effect of reducing the degenerative inflammatory diseases of the elderly and delaying aging.

  • Cell therapeutic agents for treating degenerative brain diseases using neural stem cells are actively being developed. However, few systems have been developed to monitor in real time whether the transplanted neural stem cells are actually differentiated into neurons. Therefore, it is necessary to develop a technology capable of specifically monitoring neuronal differentiation in vivo. In this study, we established a system that expresses cell membrane-targeting red fluorescent protein under control of the Synapsin promoter in order to specifically monitor differentiation from neural stem cells into neurons. In order to overcome the weak expression level of the tissue-specific promoter system, the partial 5' UTR sequence of Creb was added for efficient expression of the cell surface-specific antigen. This system was able to track functional neuronal differentiation of neural stem cells transplanted in vivo, which will help improve stem cell therapies.

Agonist (P1) Antibody Converts Stem Cells into Migrating Beta-Like Cells in Pancreatic Islets

  • Tissue regeneration is the ultimate treatment for many degenerative diseases, however, repair and regeneration of damaged organs or tissues remains a challenge. Previously, we showed that B1 Ab and H3 Ab induce stem cells to differentiate into microglia and brown adipocyte-like cells, while trafficking to the brain and heart, respectively. Here, we present data showing that another selected agonist antibody, P1 antibody, induces the migration of cells to the pancreatic islets and differentiates human stem cells into beta-like cells. Interestingly, our results suggest the purified P1 Ab induces beta-like cells from fresh, human CD34+ hematopoietic stem cells and mouse bone marrow. In addition, stem cells with P1 Ab bound to expressed periostin (POSTN), an extracellular matrix protein that regulates tissue remodeling, selectively migrate to mouse pancreatic islets. Thus, these results confirm that our in vivo selection system can be used to identify antibodies from our library which are capable of inducing stem cell differentiation and cell migration to select tissues for the purpose of regenerating and remodeling damaged organ systems.

  • Human body ages differently due to environmental, genetic and pathological factors. DNA methylation patterns also differs depending on various factors such as aging and several other diseases. The aging clock model, which uses these differences to predict age, analyzes DNA methylation patterns, recognizes age-specific patterns, predicts age, and grasps the speed and degree of aging. Aging occurs in everyone and causes various problems such as deterioration of physical ability and complications. Alzheimer's disease is a disease associated with aging and the most common brain degenerative disease. This disease causes various cognitive functions disabilities such as dementia and impaired judgment to motor functions, making daily life impossible. It has been reported that the incidence and progression of this disease increase with aging, and that increased phosphorylation of Aβ and tau proteins, which are overexpressed in this disease and accelerates epigenetic aging. It has also been reported that DNA methylation is significantly increased in the hippocampus and entorhinal cortex of Alzheimer's disease patients. Therefore, we calculated the biological age using the Epi clock, a pan-tissue aging clock model, and confirmed that the epigenetic age of patients suffering from Alzheimer's disease is lower than their actual age. Also, it was confirmed to slow down aging.

성인여성(成人女性)의 사상체질(四象體質)과 골밀도(骨密度)의 상관성(相關性)에 대한 연구(硏究) (A Clinical Study on the Relation between Sasang Constitution and Bone Mineral Density(BMD) in Women)

  • Purposes : As the population of old people has increased by the improvement of medical techniques, the concern on degenerative diseases has increased gradually. Especially the researches of osteoporosis have been carried out in view of prevention and treatment. But there wasn't any research related with Sasang Constitution. So we investigated the relation between Sasang Constitution and BMD(bone mineral density). Methods : Authors measured BMD and BMI(body mass index) of healthy women. We devided our subjects into Young age group and Old age group, and analyzed the data of Sasang Constitution subgroup. Results : 1. BMD of Taeumin was higher than those of Soyangin and Soeumin in Lumbar spine and Femoral neck in both Young and Old age group. In Old age group, there were significant differences in Taeumin vs Soeumin, and Soyangin vs Soeumin in BMD of Lumbar spine 2. BMI of Taeumin was higher than those of Soyangin and Soeumin in both Young and Old age group. In Young age group, there were significant differences in Taeumin vs Soyangin and Taeumin vs Soeumin. In Old age group, there were significant differences in Taeumin vs Soyangin, Taeumin vs Soeumin and Soyangin vs Soeumin. Conclusion : BMD and BMI of Taeumin was higher than those of Soyangin and Soeumin.

  • Cellular prion protein (PrPC) encoded at Prnp gene is well-known to form a misfolded isoform, termed scrapie PrP (PrPSC) that cause transmissible degenerative diseases in central nervous system. The physiological role of PrPC has been proposed by many studies, showing that PrPC interacts with various intracellular, membrane, and extracellular molecules including mitochondrial inner membrane as a scaffold. PrPC is expressed in most cell types including reproductive organs. Numerous studies using PrPC knockout rodent models found no obvious phenotypic changes, in particular the clear phenotypes in development and reproduction have not demonstrated in these knockout models. However, various roles of PrPC have been evaluated at the cellular levels. In this review, we summarized the known roles of PrPC in various cell types and tissues with a special emphasis on those involved in reproduction.

감염성 척추염과 감별질환의 병태생리와 MRI 소견 (Pathophysiology and MRI Findings of Infectious Spondylitis and the Differential Diagnosis)

  • MRI에서 추간판의 이상 신호와 위, 아래 척추체 종판의 파괴, 종판 주변의 골수부종 등은 감염성 척추염의 전형적인 소견으로 여겨지나 퇴행성 척추질환, acute Schmorl's node, 척추관절병증, synovitis, acne, pustulosis, hyperostosis, and osteitis (SAPHO)/chronic recurrent multifocal osteomyelitis, 척추신경관절병증, calcium pyrophosphate dehydrate 결절침착질환 등 다양한 비감염성 척추질환에서도 나타날 수 있다. MRI에서 이러한 비감염성 척추질환과 감별되는 감염성 척추염의 영상 소견은 추간판의 고신호와 농양, 척추 연부조직의의 농양, 그리고 T1 강조영상에서 저신호로 보이는 종판의 경계가 불명확해지는 점 등이다. 그러나 이러한 감별점이 항상 적용되는 것은 아니며 감염성, 비감염성 질환의 영상 소견에 유사점이 많기 때문에 정확한 진단을 위해서는 감염성 척추염뿐만 아니라 감염과 감별해야 하는 다양한 질환의 병태생리와 연관된 영상학적 특징을 아는 것이 중요하다.
