Occurrence of bacterial canker of sweet cherry caused by Pseudomonas syringae pv. morsprunorum

  • Bacterial canker of sweet cherry (Prunus cerasus L.) was observed in farmers' orchard in Goesan, Chungbuk in 2003. Typical canker symptom occurred on the branches or twigs of sweet cherry in early spring and bacterial exudates oozed out of the cracked barks of diseased trees. Watersoaked brown symptom appeared on the leaves and severe infection caused thorough defoliation on the branches or twigs of sweet cherry. When cut the severely infected branches or twigs, irregular and rusty-colored symptoms in sapwood and heartwood were clearly found, indicating that they could serve as specific symptoms of bacterial canker of sweet cherry. The gram negative, aerobic bacterium isolated from the lesion produced fluorescent pigments on King's B agar medium but did not grow at 37$^{\circ}C$ The bacterium formed Levan-type colonies, and showed negative reactions in oxidase reaction, arginine dihydrolysis test, and pectolytic activity Based on the biochemical and pathological characteristics, the causal organism was identified as Pseudomonas syringae pv. morsprunorum. This is the first report on bacterial canker of sweet cherry in Korea.

Research of Chemical Properties of Soil and Growth Characteristics of Panax ginseng under Organic and Conventional Cultivation Systems in Korea (국내 유기와 관행 재배 인삼의 토양화학성과 생육 특성 조사)

  • The objective of the present study was to perform a comparative analysis of the chemical properties of the cultivation site soil and growth characteristics of organically and conventionally cultivated ginseng (Panax ginseng C. A. Meyer). Organically and conventionally cultivated ginseng samples (4-, 5-, and 6-year-old) were collected from 52 fields at 14 locations throughout Korea. The samples were collected over three years from 2013 to 2015, with the collection period between October and November of each year. In order to increase the yield of organically cultivated ginseng, the amount of nutrients was increased to match that of the conventional cultivation system, which highlights the need for proper management in accordance with the standards for chemical properties of soil. Growth duration of organic ginseng was ${\geq}60days$ shorter than that of conventional ginseng and its average yield per 1ha was 60% than that of conventional ginseng. Root weight of organically cultivated ginseng was approximately 54% that of conventionally cultivated ginseng. Rhizome diameter and body shape index of organically cultivated ginseng were lower than those of conventionally cultivated ginseng, indicating that organically cultivated ginseng was thinner and longer than conventionally cultivated ginseng. Root length was greater in 5-year-old conventionally cultivated ginseng with a low percentage of paddy-upland rotation fields. The number of rootlets was lower in 5- and 6-year-old organically cultivated ginseng with a high percentage of direct seeding cultivation. Dry weight was distinctly lower in 5- and 6-year-old organically cultivated ginseng with early defoliation than that of conventionally cultivated ginseng. Incidences of notched belly and root rot tended to be higher in conventional cultivation, with the incidence of notched belly being distinctly higher in 4- and 6-year-old roots and root rot being more prevalent in 5- and 6-year-old roots. Red discoloration and eelworm damage, which are highly affected by soil moisture, were most common in the organically cultivated 4-year-old roots. Organically cultivated ginseng showed early defoliation than conventionally cultivated ginseng, as a result, its yield and weight were low, while the incidence of physiological disorders was low. In order to increase the yield of organically cultivated ginseng, studies on cultivation technology that can overcome early defoliation, as well as soil moisture management that can minimize physiological disorders, are required.

Fruit Characteristics Based on Leaf to Fruit Ratio in 'Pione' Grapevine (Vitis vinifera × V. labrusca) during Cultivation with Heating (가온 재배 시 '피오네' 포도(Vitis vinifera × V. labrusca)의 엽과비에 따른 과실 특성)

  • BACKGROUND: Defoliation in grapevine cultivation is practically used to improve light environment within the canopy and thereby fruit quality. Effects of defoliation in five-year-old 'Pione' grapevine during cultivation with heating were investigated to find out optimum ratio of leaf area to fruit cluster weight (L/F). METHODS AND RESULTS: The grapevines were defoliated with berry-thinning 20 days after full bloom to provide various levels of L/F. At harvest, total leaf area values of fruit bearing branches were between 0.23 and $0.60m^2$. With increasing L/F, soluble solids and anthocyanin contents curvilinearly increased ($R^2=0.76^{**}$). At L/F over $0.6m^2/kg$, soluble solids content (SSC) leveled off. With increasing L/F, titratable acidity (TA) linearly decreased ($R^2=0.87^{**}$), but the ratio of SSC to TA linearly increased ($R^2=0.86^{**}$). Anthocyanin content was significantly correlated with SSC and the ratio of SSC to TA ($R^2=0.80^{**}$ and $0.82^{**}$, respectively). When total leaf area per fruit bearing branch was maintained $0.40m^2$, soluble solids and anthocyanin contents linearly decreased ($R^2=0.79^{**}$ and $0.85^{**}$, respectively), but TA linearly increased with increasing fruit cluster weight ($R^2=0.70^{**}$). Fruit was low in quality when the L/F was below $0.6m^2/kg$. CONCLUSION: L/F is recommended to be maintained at least $0.6m^2/kg$ in 'Pione' grapevine during cultivation with heating to produce higher-quality fruits.

Population Fluctuation, Developmental Character of Panonychus citri and Damage Degree as its Control Density on Young Yuzu (Citrus junos) (유자에서 귤응애의 발생소장, 온도별 발육특성 및 방제밀도별 피해정도)

  • Experiments were conducted to investigate the population fluctuation, developmental periods, fecundity, hatching rate and demage of Citrus red mite (Panonychus citri M.) on Yuzu trees (citrus junos T.) from 1996 to 1997. Citrus red mite occurred from May to November and made two peaks. The first peak was in July to August and the secondary peak was in October. Density of the second peak was higher (9.5 miteslleaf) than that of the first peak. In the constitution rate of each developmental stage of citrus red mite on Yuzu leaves, egg stage occupied 85%. At the four constant temperature (15, 20, 25, 30 + 1$^{\circ}$C, RH 60 + lo%, 14L- IOD) conditions, the developmental period from egg to adult was 41 .l, 15.5, 11.0 and 9.4 days ; Mean longevity of adult was 23.3, 8.3, 6.3, and 6.1 days; Mean number of egg laid per female per day was 1.6, 3.2, 4.5, 4.0 eggs; Mean hatching rate was 66.6, 85.7, 90.7 and 94.7% at above temperature, respectively. When sprayed acaricide at different density of Citrus red mite, the growth of young Yuzu tree were better at low density. Defoliation rate during winter was 13.5, 20.6, 53.1, 72.6% at 4 control density 1 , 3, 6 mites per leaf and uncontrolled (10 ( ). When every time spray acaricide under 3 mites per leaf, defoliation rate during winter suppressed above 50% compare to uncontrol 72.6%.

Estimation of Leaf Area Using Leaf Length, Leaf width, and Lamina Length in Tomato (엽장, 엽폭, 엽신장을 이용한 토마토의 엽면적 추정)

  • One of the most important factors in predicting tomato growth and yield is the leaf area. Estimating leaf area accurately is the beginning of an effective tomato plant growth assessment model. To this end, this study was conducted to identify the most effective model for estimating plant leaf area through the measurement of tomato plant leaves. Leaf area (LA), leaf length (L), leaf width (W), and lamina length (La) were measured for all leaves of 5 plants at two-week intervals. The correlation between LA and tomato-leaf-independent variables showed a strong positive relationship with the formulas La × W, L × W, La + W, and L + W. For LA estimation, a linear model using the formula LA = a + b (La2 + W2) gave the most accurate estimation (R2 = 0.867, RMSE = 88.76). After examining the positions of upper, middle, and lower leaves from September to December, the coefficient of determination (R2) values for each model were 0.878, 0.726, and 0.794 respectively. The most accurate estimation came from the model that used the upper leaves of the plants. The high accuracy of the upper-leaf-based model is judged by the 50% defoliation performed by farmers after October.

Occurrence and Analysis of Apple Blotch-like Symptoms on Apple Leaves

  • Apple blotch-like symptoms (ABLS) were observed on 'Fuji' apple leaves in Cheongsong, Gunwi and Yeongcheon apple orchards located in Gyeongbuk Province during 2010-2014. Characteristics of ABLS were yellowing, brown spots on leaves, and defoliation, similar to apple blotch diseased (ABD) leaves, which are infected with Marssonina coronaria. It is difficult to differentiate by eye between ABLS and ABD, which has led to misdiagnosis and overuse of fungicides. The present study was conducted to investigate the cause of ABLS using stereomicroscopy, culture isolation, cross-sectional analysis of leaves, and PCR. No acervuli were found on the surface of ABLS leaves and no growth was observed on potato dextrose agar (PDA) plates in culture. Furthermore, cross-sectional analysis revealed similar results, and mycelia were absent in ABLS leaves. By contrast, all these characteristics were present in ABD leaves. Furthermore, no fungi or viruses were detected in ABLS leaves by PCR, suggesting that the disease is not caused by these agents. These findings suggest that ABLS might be a physiological disorder in plants that is distinct from ABD.

Enterobacter cloacae, an Emerging Plant-Pathogenic Bacterium Affecting Chili Pepper Seedlings

  • A previously unreported bacterial disease on chili pepper (Capsicum annuum L.) seedlings affecting as many as 4% of seedlings was observed in greenhouses in Chihuahua, Mexico (Delicias and Meoqui counties). Initial lesions appeared as irregular small spots on leaves and brown necrosis at margins tips were observed. Later, the spots became necrotic with a chlorotic halo. Advanced disease was associated with defoliation. A Gram negative, rod-shaped bacterium was isolated from diseased chili pepper seedlings. Three inoculation methods revealed that isolated strains produce foliage symptoms, similar to those observed in naturally infected seedlings. Pathogenic strains that caused symptoms in inoculated seedlings were re-isolated and identified to fulfill koch's postulate. Polyphasic approaches for identification including biochemical assays (API 20E and 50CH), carbon source utilization profiling (Biolog) and 16S rDNA, hsp60 and rpoB sequence analysis were done. Enterobacter cloacae was identified as the causal agent of this outbreak on chili pepper seedlings.

  • Occurrence of cercospora leaf spot by Cercospora capsici is assamed to remarkbly cause the yield reduction through the leaf spot, yellowing of leaves and early defoliation in pepper cultivation. To develop resistant cultivar, optimal screening conditions of pepper against cercospora leaf spot were studied. Optimal screening conditions were 3.4 ${\times}$ $10^5$ conidia/ml conidial suspension were sprayed on to the leaves of 30~40 days old pepper seedlings, incubated at 15~$20^{\circ}C$/20~$25^{\circ}C$(day/night) and examined 20 days after inoculation.

Effectiveness of Silver Thiosulfate Treatment in Reducing Ozone Injury to Tomato Plants (Silver Thiosulfate 처리가 토마토의 오존피해경감에 미치는 효과)

  • This study was conducted to determine the effectiveness of silver thiosulfate(STS) in reducing $O_3$ injury to tomato plants(Lycopersicon esculentm Mill. 'Pink Glory'). Two days prior to $O_3$ fumigation, plants were given a foliar spray of STS solution at concentrations of 0, 0.2, 0.4, 0.6 mM contained with 0.05% Tween-20. STS concentrations below 0.6 mM were significantly effective in providing protection aginst $O_3$ exposure(16 h at 0.3 ppm). STS reduced leaf injury rate, defoliation of cotyledons, ethylene production and degree of epinasty induced by $O_3$ injury. STS slightly increased ethylene production in non-$O_3$-fumigated plants, but changes of chlorophyll content and transpiration rate on a whole plant basis were not observed. In $O_3$-fumigated plants, STS treatment reduced chlorophyll destruction but did not affect transpiration rate. STS treatment seemed not to affect peroxidase(POD) and superoxide dismutase (SOD) activity in non-fumigated plants but reduced increasing activity of POD by $O_3$ fumigation. However, such an effect as above was not found in SOD activity. Even though enzymatic protection effects were not confirmed, the fact that reduction of acute injury rate was attained for 16 h fumigation indicates that the phytoprotective effects of STS are not necessarily related to blocking the action of strees-induced-ethylene as an anti-ethylene agent.

Mass Production of Eleutherococcus senticosus Plants through in vitro Cell Culture (세포 배양을 통한 가시오갈피(Eleutherococcus senticosus) 묘목의 대량 생산 시스템 개발)

  • Immature zygotic embryos of Eleutherococcus senticosus seeds matured rapidly within one month when the seeds comprising zygotic embryos were pieced to small size and cultured on 1/2 MS medium. Frequency of somatic embryos formation was declined rapidly when the zygotic embryos germinated and grew to plantlets. Embryogenic cells were induced by consecutive subculture of somatic embryos on MS medium with 1.0mg/L2,4-D. After heart-shaped somatic embryos were induced by suspension culture, these embryos were plated onto petri dish to support maturation of embryos. Germination of embryos occurred on medium with 5mg/L GA$_3$and transferred to culture bowl to stimulate the further growth. Frequency of soil survival of plantlets was influenced by soil mixture (perlite and peatmoss). The suitable combination of perlite and peatmoss was 1:5, and the soil survival rate was 78% after 4 months. The soil transferred plantlets were over-wintered in field condition after defoliation. New year sprouting of plants was achieved successfully and they grew to adult plants. These results indicate that the systematic procecure of plant production in E. senticosus for micro propagation.
