7S globulin has been isolate from the defatted soybean meal(glycine max,. merill) by Sepharose-6B column chromatography and CM-Sephadex column chromatography. Coagulum of 7S globulin formed at a temperature of $65^{\circ}C$ in microbial enzyme treatment. In order to characterize the structure of the coagulum, three kinds of coagulum (enzyme-, calcium-and acid-induced coagulum)were compared througth the Scanning Electron Microscope(SEM). The network structure was found to be of two levels. First, there was an appearance on the molecular level in the form of strands. Second, there was a denser network with a fine structure.
Soybeans contain the phytoestrogens genistein and daidzein, their glucosides genistin and daidzin and coumesterol. These isoflavonoid compounds are capable of producing an estrogenic response in a number of diverse species. This study determined optimum conditions for the extraction of the main isoflavones(daidzin, genistin, daidzein, genistein) in defatted soybean meal using high-performance liquid chromatography. The best optimum extraction was achieved at 75% ethanol, $80^{circ}C$, pH4 and a three hour contact time. In addition, isoflavones with high purity were separated by adding up to 4%(w/v) of calcium chloride dihydrate. Most soybean extracts were composed of $beta$-glucosidic conjugate(daidzin, genistin) which is difficult to adsorb in body. Therefore, $beta$-glucosidase was used to convert as conjugate to aglycone form (daidzein, genistein) which is easy to adsorb. The optimal conditions of enzyme reaction involved to be 8.4 units of enzyme concentration, pH5.0, $40^{circ}C$ and 40 minutes.
In our previous report (Korean J. Food Sci. Technol., 9, 123. (1977), functional properties of soyprotein isolates prepared from defatted soybean meal were studied. Using those properties soyprotein fibers, which may be acceptable as meat analogs, were prepared with protein spinning apparatus. Soyprotein can be converted into the suitable form for the spinning by denaturation with alkali (0.6%) and continuous fibers were spun by extruding spinning solution into an 20% NaCl-1 N acetic acid coagulating bath. The process for producing soyprotein fibers on a bench scale was described and break strength, break elongation and textural parameters of the fibers formed were evaluated. The possible scheme of formation of soyprotein fibers was discussed.

These isflavonoid compounds are capable of producing an estrogenic response in a number of diverse species. This study determined optimum conditions for extraction of isoflavones in defatted soybean meal. Extraction of isoflavones was conducted at various conditions such as difference concentrations of extraction solvent, temperature, time and pH to extact. The most optimum extraction conditions was achieved at 75% ethanol, $80^{\circ}C$, pH4 and extract for three hours. In addition. isoflavones with high purity were separated by adding up to 4%(w/v) of calcium chloride dihydrate.
1) Higher yield of red color was observed by the isolated strain (Monascus D-7) than the type cultures in steamed rice medium. 2) In a case of Monascus purbigerus IAM 8004, best yield of color was obtained at Lin's submerged culture medium containing 1% wheat bran, 2% starch and 3% corn meal instead of rice powder as carbon source. However, in a case of isolated strain (M. D-7), good result was shown at 1% rice bran and 2% starch as a source of carbon in Lin's medium. 3) Good yields were obtained from both strains in Nishikawa's medium which was added with 3% defatted soybean flour. 4) There were no significant differences in pigment extractability among solvents. Extracted pigment was stable in wide range of pH and heat, whereas relatively unstable in sunlight. 5) Toxicological study of extracted pigment determined $LD_{50}$ at 0.2539g/20g, when injected in mouse. When injected in to mouse in 25% ethanol solution: considering the toxicity of ethanol, the toxicity of pigment itself is believed to be none.
Analysis of Production of Thermostable Alkaline Protease using Thermoactinomyces sp. E79. Jung, Sang Won, Sung-Sik Park, Yong-Cheol Park" Tae Kwang Oh2, and Jin-Ho Seo*, Department of Food Science and Technology, Seoul National University, Suwon 441-744, Korea, 1lnterdisciplinary program [or Biochemical Engineering & Biotechnology, Seoul National Univer5it}~ Seoul 151 "7421 Koreal 2Microbial Enzyme RU, Korea Research Institute of Bioscience & Biotechnology, Po. Box 1151 Yusong, Taejon 305"6001 Korea - This research was undertaken to analyze fermentation properties of Thermoactinomyces sp. E79 for production of a thermostable alkaline protease, which is able to specifically hydrolyze defatted soybean meal (DSM) to amino acids. TIle optimum pH for cell growth and protease production was pH 6.7, Thermoactinomyces sp. E79 did not grow at pHlO Among carbon sources tested, soluble starch was the best for protease production, while glucose repressed protease production. Tryptone was found to be the best nitrogen source for cell growth and soytone was good tor protease production. Oxygen transfer rate played an important role in producing thermostable alkaline protease. Ma'<..imum values of 6.58 glL of dry cell weight and 43.0 UJmL of protease activity were obtained in a batch fermentation using a 2.5 L jar fermentor at 1.93 X 102 hr-l of volumetric oxygen transfer coeff'jcient (kLa). Addition of 200 mgIL humic acid to the growth medium resulted in 1.64 times higher protease activity and 1.77 times higher cell growth than the case without humic acid addition.
The heat generation at the start-up period of an autogeneous single screw extruder was determined with various feed materials, die structure and operational conditions. The highest heat generation rate was observed with defatted soybean meal, while the lowest value was obtained with rice flour, and wheat and barley flour showed the intermediate rate. As the moisture content of the flour decreased and the screw speed increased, the electric power requirement and heat generation rate increased. The temperature at compression section increased with the decrease in the particle size. The same effect was also observed as breaker plate was installed. The optimum operation was established as the temperature profile was maintained in decreasing order of metering section, die and compression section.
In our previous report (Korean J. Food Sci. and Technol., 9, 123, 1977) we have studied the functional properties of soy protein isolates prepared from defatted soybean meal. For the practical application of the data described, beef patties substituted with SPI, and imitatied ice cream were prepared and sensory evaluations were carried out. The sensory analyses indicated that the beef patties substituted with gelated SPI to 30% have no detectable difference to the control at the significant level of 0.01. The imitation of ice cream whose milk-solid-not-fat was substituted with soy protein isolates showed the same texture as the control. Flavor of ice cream, however, has significant difference to the control due to beany-off flavor. Therefore removal of beany-off flavor is required for the substitution in ice cream.
The effects of cultural conditions in the rice straw media for cellulolytic enzymes production by Pleurotus sajor-caju were investigated. The optimum moisture content, pH and temperature for enzymes production were 60%, 7.0 and $35^{\circ}C$ in $C_1-cellulase$, and 60%, 5.0 and $25^{\circ}C$ in $C_x-cellulase$, and 60%, 7.0 and $20^{\circ}C$ in ${\beta}-glucosidase$, respectively. When light was irradiated during the cultivation period, $C_1-cellulase$ and ${\beta}-glucosidase$ production were decreased but $C_x-cellulase$ production increased at $500{\sim}1,000\;lux$. During the cultivation period, $C_1-cellulase$ production was contrary to $C_x-cellulase$ and ${\beta}-glucosidase$. Among the various materials added, rice bran was effective to $C_1-cellulase$ production, cotton seed cake and rice bran to $C_x-cellulase$ production, and defatted soybean and fish meal to ${\beta}-glucosidase$ production. The optimum concentration of rice bran for enzymes production were 20% in $C_{1-}$, $C_x-cellulase$ and 10% in ${\beta}-glucosidase$.

In order to improve the quality of amino acid soysauce, this experiment was carried out as follows; koji added amino acid soysauce(KAAS) was prepared by addition of defatted soybean and meal koji into amino acid soysauce containing $0.5%{\sim}1.5%$ of total nitrogen. Various physicochemical characteristics of KAAS during aging were determined. Total nitrogen, amino-nitrogen, specific gravity, pure extract, total acidity and color were high in KAAS at high total nitrogen. After one month aging, reducing sugar was high in KAAS at high total nitrogen, but pH was high in KAAS at low total nitrogen. Ethyl alcohol was 2.07% in KAAS at 0.5% of total nitrogen after five months aging and 1.4% in KAAS at 1.5% of total nitrogen. Levulinic acid and lactic acid were high in KAAS at low total nitrogen. The results obtained above summarized as the quality of KAAS were highly good at $0.5{\sim}0.7%$ of total nitrogen after two or three months aging.
