조피볼락의 생식주기 (Reproductive Cycle of a Rockfish, Sebastes schlegeli)

  • 본 실험은 조피볼락 (Sebastes schlegeli)의 생식년주기와 생식주기에 따른 sex steroid hormone들의 계절적인 변화를 밝히기 위하여 GSI와 HSI의 연간 변동과 생식소 조직학적인 연간 변화와 sex steroid hormone ($estradiol-l7{\beta}, 17{\alpha}, 20{\beta}-dihydroxy-4-pregnen-3-one, testosterone$, 그리고 $11-ketotestosterone$)의 혈중 수준의 연간 변화를 조사하였다. 암컷의 생식주기는 다음과 같이 5단계로 구분할 수 있었다: 1) 회복기 (6월${\~}$9월): 혈중 $estradiol-l7{\beta}$의 수준이 점차 증가 하였다. 2) 난황형성기 (9월~2월): 난모세포의 난황 형성이 관찰되며, GSI 값과 혈중 $estradiol-l7{\beta}$수준이 증가하였다; 3) 임신기 (2월${\~}$4월): 난소내에 발생중인 자어가 관찰되며, 2월에서 3월에 이르러 혈중 $17{\alpha}, 20{\beta}-dihydroxy-4-pregnen-3-one$와 testosterone의 수준이 증가하였다: 4) 출산기 ($4{\~}5$월): GSI 값이 급격히 감소하면서 난소내에는 자어들의 출산된 흔적이 보였다: 5) 휴지기 ($5월$${\~}$7월) GSI값과 혈중 steroid hormone의 수준이 낮은 값을 유지하였다. 수컷의 생식년주기는 6단계로 나눌 수 있었다: 1) 정자형성 초기(4월${\~}$6월): GSI의 값과 혈중 steroid hormone의 수준이 점진적으로 증가하였으며, 정원세포의 포낭이 수가 증가하고 일부의 정모세포의 포낭들도 관찰되었다; 2) 정자형성 중기 (6월${\~}$9월): GSI의 값과 혈중 steroid hormone의 수준이 증가하고 많은 포낭내의 생식세포들이 활발하게 정자형성을 하고 있었다: 3) 정자형성기 후기 (9월${\~}$11월): GSI와 steroid hormone의 수준이 높은 값을 유지하고 있었으며, 정자들이 정세관 내강으로 방출되었다; 4) 정자 방출기 (11월${\~}$12월): 전세관의 내강이 확장되고 이 내강내에는 성숙된 정자들로 가득차 있었다: 5) 퇴화기 (12월${\~}$2월): GSI의 값이 점차로 감소하면서 정모세포의 포낭수도 점차로 감소하였다: 6) 휴지기 (2월${\~}$4월): 정소조직의 변화는 보이지 않았으며, GSI와 steroid hormone의 수준이 낮은 값을 유지하였다. 11월에 정소들의 정세관 내강에 성숙된 정자들로 가득차 있었고 난소내강에 정자괴들이 관찰되는 것으로 보아 이종의 교미시기는 11월과 12월사이라고 할 수 있다.

Effects of Steroid Hormones on Collagen Biosynthesis in Rat Aorta and Uterus

  • Effects of steroid hormones on the collagen biosynthesis in aorta and uterus were studied with ovariectomized Sprague-Dawley rats. Effects of administration of hormones, such as estrogen, testosterone and prednisolone, to the ovariectomized animals were studied, comparing with the control. Each group was injected with ³H-proline and sacrificed, followed by removals of aorta and uterus. Separations and quantitative analyses of proline and hydroxyproline were performed by means of thin layer chromatography; and radioactivities of the separated amino acids were assayed by liquid scintillation counter. Normally the incorporation of ³H-proline into hydroxyproline was greater in uterus than in aorta, and collagen turnover rate of uterus was observed rapid as well than that of aorta. In the two tissues from ovariectomized rats, the incorporation rate of ³H-proline into hydroxypoline was markedly decreased than that of the former. Changes in the turnover rate of collagen in these tissues were not observed. Decrease in ³H-proline incorporation into collagen in ovariectomized rats was markedly antagonized by estrogen, but not influenced by prednisolone in the tissues tested.

Effects of crocin and metformin on methylglyoxal-induced reproductive system dysfunction in diabetic male mice

  • Objective: This study investigated the effect of crocin in methylglyoxal (MGO)-induced diabetic male mice. Methods: Seventy 1-month-old male NMRI mice weighing 20-25 g were divided into seven groups (n=10): sham, MGO (600 mg/kg/day), MGO+crocin (15, 30, and 60 mg/kg/day), MGO+metformin (150 mg/kg/day), and crocin (60 mg/kg/day). MGO was administered orally for 30 days. Starting on day 14, after confirming hyperglycemia, metformin and crocin were administered orally. On day 31, plasma and tissue samples were prepared for experimental assessments. Results: Blood glucose and insulin levels in the MGO group were higher than those in the sham group (p<0.001), and decreased in response to metformin (p<0.001) and crocin treatment (not at all doses). Testis width and volume decreased in the MGO mice and improved in the crocin-treated mice (p<0.05), but not in the metformin group. Superoxide dismutase levels decreased in diabetic mice (p<0.05) and malondialdehyde levels increased (p<0.001). Crocin and metformin improved malondialdehyde and superoxide dismutase. Testosterone (p<0.001) and sperm count (p<0.05) decreased in the diabetic mice, and treatment with metformin and crocin recovered these variables. Luteinizing hormone levels increased in diabetic mice (p<0.001) and crocin treatment (but not metformin) attenuated this increase. Seminiferous diameter and height decreased in the diabetic mice and increased in the treatment groups. Vacuoles and ruptures were seen in diabetic testicular tissue, and crocin improved testicular morphology (p<0.01). Conclusion: MGO increased oxidative stress, reduced sex hormones, and induced histological problems in male reproductive organs. Crocin and metformin improved the reproductive damage caused by MGO-induced diabetes.

The Effects of Flutamide and Diethylstilbestrol Exposure on the Reproductive Organs and Thyroid of Male Rats by the Rodent 20-day Thyroid/Pubertal Assay

  • To establish a test protocol for the rodent 20-day thyroid/pubertal assay, flutamide and diethylstilbestrol (DES) were administered to intact male Sprague-Dawley rats from postnatal day 33 for 20 days. Flutamide (1, 5, and 25 mg/kg/day) or DES (10, 20, and 40 ug/kg/day) was given once daily by oral gavage to immature male rats. Prepuce separation was significantly delayed in flutamide group and in DES group. One day after the last dose, the rats were killed and pituitary, thyroid, and reproductive organs were removed and weighed. Flutamide treatment resulted in a significant reduction in the weights of epididymides, ventral prostate, seminal vesicles plus coagulating glands and fluid (SVCGF), levator ani. bulbocarvenus muscles (LABC), Cowper's glands, and glans penis. The weight of adrenal glands decreased at % mg/kg/day, while testes and any other organ weights were unaffected. No microscopic changes were observed in the thyroid glands. Serum levels of testosterone wert significantly increased in the flutamide-treated groups and serum levels of estradiol were also increased. A significant reduction in the weights of testes, epididymides, ventral prostate, SVCGF, LABC, Cowpers glands, and glans penis of DES treated group. Serum testosterone and LH decreased significantly in DES group. Decrease of estradiol was observed, but not significant. These results indicate that flutamide and DES delay puberty in the male rat and its mode of action appears to be via altered secretion of steroids, which subsequently affect the development of the reproductive tract. 흰쥐의 내분비선 및 혈장성분에 미치는 뇌하수체척출의 영향과 이에 대한 성 Hormone의 효과 (The Effect of Hypophysectomy and Subsequent Administration of Sex Hormone on Several Endocrine Glands and Plasma Components in Rats)

  • The present experiments were carried out to elucidate the effects of hypophysectomy and subsequent administration of sex hormone on thyroid, adrenal gland, gonads and blood plasma components in the rat. The jresults obtained were summarized as follows: The weight of the thyroid gland of both male and female hypophysectiomzed rats decreased markedly from 7 days up to 56 days after the hypophysectomy as compared to the control group. The administration of sex hormone (6 mg of testosterone propionate to male and 6 mg of hexestrol to female) to the hypophysectomized rat gave on effect on the change in the weight of the thyroid gland. The hopophysectomy decreased the uptake of radioactive iodine in the thyroid gland in both male and female rats with time. Subsequent administration of the sex hormone caused no effect. With regard to the histological changes of the thyroid gland, the hypophysectomy caused significant changes in the gland showing a remarkable degeneration. The function of the gland seemed to disa, pp.ar almost completely on 56th day after the hypophysectomy. Upon the administration of sex hormone after the hypophysectomy, however. the epithelia of the follicle which has changed to flat from has partly returned to its functional cubicfrom and nuclei recovered as nearly as normal. These recovery were more remarkable in the female than in the male. The hypophysectomy kept causing a significant decrease in the weight of the adrenal gland in male and female rats during the period of observation (up to 56 days) as in the case of thyroid gland. The administration of sex hormone has on effect in this respect either. The hypophysectomy also caused a marked morphological change in the gland: zona fasciculata and zona reticularis were dicreased in size quichly after the hypophysectomy. The administraton of the sex hormone to the hypophysectomized rat resulted in clear distinction among the three layers of the adrenal cortex which otherwise very diffused. In the male, this phenomenum was more remarkable than in the female and the pattern of the cell arrangements and the thickening of each layer became similar to those of normal rats. The gonads of both sexes have also kept decreasing in the weight and degenerated in morpohology after the hypophysectomy. However, the degenerate follicle became enlarged after the administration of hexestrol in the female. Furthermore, the vacuoles found in interstitial cells of hypophysectomized rat disa, pp.ared after the administration of testosterone in the male and the formation of spermatocytes seemed to be recovered. Hypophysectomy also caused a gradual increase in the contents of total protein, non-protein nitrogen, total lipid, cholesterol and calcium in the blood plasma with time. The concentrations of sodium, potassium and chloride in the blood did not change after the hypophysectomy. Sex hormone caused practically no change in above tendency.

Experiment on the effect of Artemisia sieversiana extract on hair loss prevention and cell growth

  • Objectives: This study aimed to examine the safety, effects on proliferation of hair papilla cells, and anti-inflammatory and antioxidant mechanisms of Artemisia sieversiana Ehrh. ex Willd. (AS) extract. Methods: Safety tests through purity testing, acute toxicity tests, and repeated toxicity tests were performed using AS extract (ASE) which had been dried for over two years. Cell culture and proliferation tests were conducted; VEGF (vascular endothelial growth factor), bFGF (basic fibroblast growth factor), and EGF (epidermal growth factor) and protein expression analyses were performed for mechanistic evaluation; and inhibitory effects of ASE on the RNA expression of testosterone, 5𝛼-reductase, and aromatase was assessed. The anti-inflammatory and antioxidant efficacy of ASE was confirmed by measuring the levels of nitric oxide, inflammatory mediators (TNF-𝛼 and PGE2), inflammatory cytokines (IL-1𝛽, IL-6, and IL-8), and chemokine MCP-1. Results: The safety of ASE was confirmed. The mechanism of cell proliferation in human hair follicle dermal papilla cells involved the promotion of VEGF, bFGF, and EGF expression. ASE decreased mRNA expression of testosterone, 5𝛼-reductase, and aromatase-1 in a concentration-dependent manner. PGE2 and TNF-𝛼 production by inflammatory mediators was also significantly decreased in a concentration-dependent manner, and inflammatory cytokine and chemokine expression was inhibited. Conclusions: ASE is suggested to promote papillary cell growth at the cellular level, to suppress expression of various enzymes involved in hair cycle and cell death, and to inhibit hair loss through anti-androgen, anti-inflammatory, and antioxidant effects.

  • Recently, there is a worldwide concern that a great number of man-made chemicals have a hormone-like action both in humans and in animals. DECD is developing screening programs using validated test systems to determine whether certain substances may have an effect in humans. In the present study. the establishment oj repeated-dose toxicity test method was tried. Flutamide. an anti-androgenic agent. was administered by gavage to Sprague-Dawley rats for 28 days at dose levels of 0. 0.5. 3 and 18 mg/kg body weight (10-15 rats/sex/group) to examine the effects on general findings. especially reproductive and endocrine parameters. Clinical signs. body weights, food consumption, and sexual cycle were checked and measured. For the gross and microscopic examinations. 10 rats/sex/group were sacrificed at the end of dosing period and the remaining animals of control and high dose groups (5 each) were sacrificed after 14 days recovery. Examinations for hematology and clinical chemistry were carried out at necropsy. There were no treatment-related changes in clinical signs. body weights, food consumption. gross necropsy. hematology and clinical chemistry at all doses of both sexes. The period and regularity of sexual cycle were not adversely affected at all doses by the test agent. At 18 mg/kg. both decreased weights of prostate, seminal vesicle and epididymis in males and increased weights of spleen and thymus in females were observed. In addition, decreased number of spermatids and sperms. increased serum testosterone concentration and increased incidence (100%) of interstitial cell hyperplasia were seen in males. At 18 mg/kg of the recovery group. decreased prostate weight. reduced sperm count and increased incidence (20%) of interstitial cell hyperplasia in males and increased thymus weight in females were observed. At 3 mg/kg. reduced sperm count was found. There were no adverse effects on parameters examined at 0.5 mg/kg of both sexes. The results suggested that the potential target organs of flutamide may be accessory sexual glands including testes for males and spleen and thymus for females. Taken together. this test method was found to be a useful screening test system for endocrine disrupting chemicals.

Osteosarcopenic Obesity in Elderly: The Cascade of Bone, Muscle, and Fat in Inflammatory Process

  • Conditions related to body composition and aging, such as osteopenic obesity, sarcopenia/ sarcopenic obesity, and the newly termed osteosarcopenic obesity(triad of bone, muscle and adipose tissue impairment), are beginning to gain recognition. Currently, it has begun to attract the attention of scholars from all over the world, however, for this disease, it still needs a more clear understanding and perception. Therefore, this article considered the osteoporosis, muscle depression, and obesity, these diseases as a gate to study the relationship among muscle, bone, and fat. In addition, in the aging process, the formation of IGF-cortisol, testosterone, and estrogen is sensitive. These hormones can not only absorb muscle protein metabolism, but also affect alienation. The decrease in IGF-cortisol in the elderly resulted in increased visceral fat, decreased muscle mass and bone mineral density, and then affected decreased skeletal muscle atrophy and decreased quality. The reduction of skeletal muscle quality and strength and increase body fat affected the adipose tissue to produce inflammatory cytokines, thereby reduced skeletal muscle, promoted cardiovascular disease, metabolic syndrome and insulin resistance in chronic diseases. Almost all chronic inflammatory diseases were associated with bone, muscle and fat. These mechanisms were complex and interrelated. Inflammation reduces bone formation, increases fat and reduces muscle mass. And thus not only had a significant impact on the motor system, but also made the incidence increase of fracture, osteoporosis, fragile syndrome, fall, osteomalacia and other bone disease. This article aimed to start from the interaction between the muscles and bones of the elderly, extended to obesity, muscle deficiency, osteoporosis and other diseases, finally, from a nutritional point of view, to discuss how to treat osteoporosis obesity.

Changes in Plasma Sex Steroid and Cortisol Levels during Annual Reproductive Cycle of Ribbed Gunnel, Dictyosoma burgeri

  • We investigated the changes in plasma sex steroid hormones, testosterone (T), estradiol-$17{\beta}$ ($E_2$), 17,$20{\beta}$-dihydroxy-4-pregnen-3-one ($17{\alpha}20{\beta}P$), 11-ketotestosterone (11KT) and cortisol levels from ribbed gunnel, Dictyosoma burgeri in associated with annual reproductive cycle. The gonadosomatic index (GSI) of females increased from November, peaked in February and decreased rapidly from March. The GSI of males also increased from November, peaked in January and then decreased gradually. In females, $E_2$ levels increased and remained high from December to February. The levels of T showed a similar tendency and correlated ($r_s$=0.898, p<0.01) with $E_2$ levels. The levels of $17{\alpha}20{\beta}P$ increased rapidly in February ($4.78{\pm}1.01ng/ml$) and peaked in July ($5.08{\pm}0.65ng/ml$). Cortisol level was peaked in March and correlated with $17{\alpha}20{\beta}P$ levels ($r_s$=0.696, p<0.01). In males, the levels of T was peaked in January and then decreased rapidly. The levels of 11KT were remained high from October to January. On the other hand, the levels of $17{\alpha}20{\beta}P$ fluctuated during reproductive cycle. These results suggest that plasma sex steroids in ribbed gunnels have annual periodicity, and that cortisol may involve in maturation of females.

양식산 수컷 뱀장어 Anguilla japonica의 인위적 성숙유도에 따른 혈중 성호르몬 변동과 정소 발달 (Plasma Sex Steroid Hormone Profiles and Testicular Development in Artificially Maturing Cultured Mille Eel, Anguilla japonica)

  • We investigated the changes in body weight (BW), plasma sex steroid hormone profiles, and testicular development of cultured male eel Anguilla japonica during an artificial maturation process. Eels that received weekly intraperitoneal injections of eel's ringer solution containing human chronic gonadotropin (HCG) were examined. In the ringer-treated control, BW changes decreased slowly during the experimental period. Plasma testosterone (T), 11-ketotestosterone (11-KT) and $17{\alpha},\;20{\beta}$-dihydroxy-4-pregnen-3-one (DHP) levels In the control remained low and did not show significant changes. Moreover, all germ cells in the testes of the control were spermatogonia. In the HCG-treated male eels, however, BW changes increased gradually from the fifth week and then decreased slowly. The plasma T level increased rapidly (p<0.05) in the second week and then decreased slowly. The plasma 11-KT level increased dramatically (p<0.05) in the second week and was maintained until the end of the experiment. The plasma DHP level increased progressively from the second week and peaked in the eighth week (p<0.05). The testes of HCG-treated male eels were more developed than those of the control; most were at the spermatozoa and spermatid stages and showed active spermiation. Thus, spermatogenesis and spermiation in the cultured eel can be induced by repeated injections of HCG.
