Estimation of Water Quality Variation in Sewer Network using MOUSE TRAP Model (MOUSE TRAP 모델을 이용한 하수관거내 수질변화 예측)

  • One of the major problems associated with operation of domestic sewer lines involves hydraulic problems such as insufficient conveyance capacity, exceeding maximum velocity, and deficiency of minimum velocity. It has also been pointed out that influent concentration lower than design concentration of pollutants, which is mainly caused by unidentified inflow and infiltration, degrades the operational efficiency of many sewage treatment plants (STPs). A computer-added analysis method supporting a coupled simulation of sewage quality and quantity is essentially required to evaluate the status of existing STPs and to improve their efficiency by a proper sewer rehabilitation work. In this study, dynamic water quality simulations were conducted using MOUSE TRAP to investigate the principal parameters that governs the changes of BOD, ${NH_4}^+$, and ${PO_4}^{3-}$3- concentrations within the sewer networks based on data acquired through on-site and laboratory measurements. The BOD, ${NH_4}^+$ and ${PO_4}^{3-}$3- concentrations estimated by MOUSE TRAP was lower than theoretical pollution loads because of sedimentation and decomposition in the sewer. The results revealed that sedimentation is a most important factor than other biological reactions in decreasing pollutant load in the sewers of C-city. The sensitivity analysis of parameters pertaining to water quality changes indicated that the effect of the BOD decay rate, the initial DO concentration, the half-saturation coefficient of dissolved BOD, and the initial sediment depth is marginal. However, the influence of settling rate and temperature is relatively high because sedimentation and precipitation, rather than biological degradation, are dominant processes that affect water quality in the study sewer systems.

Biokinetics of Protein Degrading Clostridium cadaveris and Clostridium sporogenes in Batch and Continuous Mode of Operations

  • A quantitative real-time polymerase chain reaction (QPCR) was applied to estimate biokinetic coefficients of Clostridium cadaveris and Clostridium sporogenes, which utilize protein as carbon source. Experimental data on changes in peptone concentration and 16S rRNA gene copy numbers of C. cadaveris and C. sporogenes were fitted to model. The fourth-order Runge-Kutta approximation with non-linear least squares analysis was employed to solve the ordinary differential equations to estimate biokinetic coefficients. The maximum specific growth rate (μmax), half-saturation concentration (Ks), growth yield (Y), and decay coefficient (Kd) of C. cadaveris and C.sporogenes were 0.73 ± 0.05 and 1.35 ± 0.32 h-1, 6.07 ± 1.52 and 5.67 ± 1.53 g/l, 2.25 ± 0.75 × 1010 and 7.92 ± 3.71 × 109 copies/g, 0.002 ± 0.003 and 0.002 ± 0.001 h-1, respectively. The theoretical specific growth rate of C. sporogenes always exceeded that of C. cadaveris at peptone concentration higher than 3.62 g/l. When the influent peptone concentration was 5.0 g/l, the concentration of C.cadaveris gradually decreased to the steady value of 2.9 × 1010 copies/ml at 4 h Hydraulic retention time (HRT), which indicates a 67.1% reduction of the initial population, but the wash out occurred at HRTs of 1.9 and 3.2 h. The 16S rRNA gene copy numbers of C. sporogenes gradually decreased to steady values ranging from 1.1 × 1010 to 2.9 × 1010 copies/ml. C. sporogenes species was predicted to wash out at an HRT of 1.6 h.

Blockade of Kv1.5 by paroxetine, an antidepressant drug

  • Paroxetine, a selective serotonin reuptake inhibitor (SSRI), has been reported to have an effect on several ion channels including human ether-a-go-go-related gene in a SSRI-independent manner. These results suggest that paroxetine may cause side effects on cardiac system. In this study, we investigated the effect of paroxetine on Kv1.5, which is one of cardiac ion channels. The action of paroxetine on the cloned neuronal rat Kv1.5 channels stably expressed in Chinese hamster ovary cells was investigated using the whole-cell patch-clamp technique. Paroxetine reduced Kv1.5 whole-cell currents in a reversible concentration-dependent manner, with an $IC_{50}$ value and a Hill coefficient of $4.11{\mu}M$ and 0.98, respectively. Paroxetine accelerated the decay rate of inactivation of Kv1.5 currents without modifying the kinetics of current activation. The inhibition increased steeply between -30 and 0 mV, which corresponded with the voltage range for channel opening. In the voltage range positive to 0 mV, inhibition displayed a weak voltage dependence, consistent with an electrical distance ${\delta}$ of 0.32. The binding ($k_{+1}$) and unbinding ($k_{-1}$) rate constants for paroxetine-induced block of Kv1.5 were $4.9{\mu}M^{-1}s^{-1}$ and $16.1s^{-1}$, respectively. The theoretical $K_D$ value derived by $k_{-1}/k_{+1}$ yielded $3.3{\mu}M$. Paroxetine slowed the deactivation time course, resulting in a tail crossover phenomenon when the tail currents, recorded in the presence and absence of paroxetine, were superimposed. Inhibition of Kv1.5 by paroxetine was use-dependent. The present results suggest that paroxetine acts on Kv1.5 currents as an open-channel blocker.

Bio-kinetic and Design Analysis for Box-mill Wastewater Treatment Using Anoxic Activated Sludge Process (무산소 활성오니공정을 이용한 판지공장 폐수처리의 동력학적 해석 및 설계분석)

  • The anoxic activated sludge process was applied to the treatment of industrial box-mill wastewater, which exhibited the high removal efficiencies of $90{\sim}94%$$ TCOD_{Mn}$ and $58{\sim}81%$ Color. For the design of industrial anoxic activated sludge process, Monod bio-kinetic coefficients of box-mill wastewater were estimated as follows: $K_{max}$(maximum specific substrate removal rate)=0.52 $day^{-1}$, $K_s$(half saturation constant)=314 mg/L, $K_d$(decay coefficient)=0.274 $day^{-1}$, y(microbial yield coefficient)=0.908 mg/mg, and ${\mu}_{max}$(maximum specific growth rate)=0.472 $day^{-1}$. Space loading factors for the design analysis were practically determined as the values of F/M ratio=$0.043{\sim}0.07$ kg-$TCOD_{Mn}$/kg-SS-day, BOD space loading=$0.18{\sim}0.3$ kg-$TCOD_{Mn}/m^3-day$, and ${\theta}_x=6.8{\sim}26.4$ day when considering the relationship of these loading factors with growth dynamics of microorganisms, the F/M ratio that is inversely proportional to ${\theta}_x$ should be equivalent to ${\mu}_{max}$ in units, but exhibited the significant difference between theses two values. Therefore, it is considered that high safety factors are requested in the design of anoxic activated sludge process that is based on Monod bio-kinetics of microorganism.

Blockade of Kv1.5 channels by the antidepressant drug sertraline

  • Sertraline, a selective serotonin reuptake inhibitor (SSRI), has been reported to lead to cardiac toxicity even at therapeutic doses including sudden cardiac death and ventricular arrhythmia. And in a SSRI-independent manner, sertraline has been known to inhibit various voltage-dependent channels, which play an important role in regulation of cardiovascular system. In the present study, we investigated the action of sertraline on Kv1.5, which is one of cardiac ion channels. The effect of sertraline on the cloned neuronal rat Kv1.5 channels stably expressed in Chinese hamster ovary cells was investigated using the whole-cell patch-clamp technique. Sertraline reduced Kv1.5 whole-cell currents in a reversible concentration-dependent manner, with an $IC_{50}$ value and a Hill coefficient of $0.71{\mu}M$ and 1.29, respectively. Sertraline accelerated the decay rate of inactivation of Kv1.5 currents without modifying the kinetics of current activation. The inhibition increased steeply between -20 and 0 mV, which corresponded with the voltage range for channel opening. In the voltage range positive to +10 mV, inhibition displayed a weak voltage dependence, consistent with an electrical distance ${\delta}$ of 0.16. Antidepressant drug paroxetine blocks the open pore of Kv3.1 potassium channel

  • In patients with epilepsy, depression is a common comorbidity but difficult to be treated because many antidepressants cause pro-convulsive effects. Thus, it is important to identify the risk of seizures associated with antidepressants. To determine whether paroxetine, a very potent selective serotonin reuptake inhibitor (SSRI), interacts with ion channels that modulate neuronal excitability, we examined the effects of paroxetine on Kv3.1 potassium channels, which contribute to high-frequency firing of interneurons, using the whole-cell patch-clamp technique. Kv3.1 channels were cloned from rat neurons and expressed in Chinese hamster ovary cells. Paroxetine reversibly reduced the amplitude of Kv3.1 current, with an $IC_{50}$ value of $9.43{\mu}M$ and a Hill coefficient of 1.43, and also accelerated the decay of Kv3.1 current. The paroxetine-induced inhibition of Kv3.1 channels was voltage-dependent even when the channels were fully open. The binding ($k_{+1}$) and unbinding ($k_{-1}$) rate constants for the paroxetine effect were $4.5{\mu}M^{-1}s^{-1}$ and $35.8s^{-1}$, respectively, yielding a calculated $K_D$ value of $7.9{\mu}M$. The analyses of Kv3.1 tail current indicated that paroxetine did not affect ion selectivity and slowed its deactivation time course, resulting in a tail crossover phenomenon. Paroxetine inhibited Kv3.1 channels in a use-dependent manner. Taken together, these results suggest that paroxetine blocks the open state of Kv3.1 channels. Given the role of Kv3.1 in fast spiking of interneurons, our data imply that the blockade of Kv3.1 by paroxetine might elevate epileptic activity of neural networks by interfering with repetitive firing of inhibitory neurons.

  • Soil aquifer treatment is a water reuse technology that secondary or tertiary treated wastewater is infiltrated into the aquifer in which physical and biochemical reactions occur. Major consideration in SAT is the removal and transport of DOC and nitrogen species. In this study, reaction mechanism in SAT was examined considering nitrification, denitrification and organic oxidation. In addition, SAT modeling system was developed as the reaction mechanism was applied to groundwater flow and transport model. In verification of the reaction module by 1-dimensional unsaturated soil column test, the experimental data of all of the species, ammonium, nitrate, DOC and DO, were well matched with the simulation results. In sensitivity analysis, ammonium partition coefficient, dissolved oxygen inhibition constant and biomass decay rate affect ammonium, DOC and DO concentration of effluent, respectively.

Ecological Studies on the Montane Grassland of Mt. Soback in Korea (소백산 산지초원의 생태학적 연구 II. 물질생산과 염류순환)

  • Comparative study of the biomass productioin and the cyclings of nitrogen, phosphorus and potassium was carried out on the east and the northwest facing slope in the montane grassland of Mt. Soback. The maximum productin during the growing season in the east and the northwest slope were 1, 150g/m2.yr, and 755g/m2.yr. in the August, respectively. The positive correlation was appearent between biomass production and A-layer depth of the soil and the correlation coefficient (r=.964) was very significant at 1% level. In the above ground materials, the nutrients contents were high at the early of the growing season but decreased gradually. In roots, however, there are no significant trend throughout the growing season. Total amounts of minerals uptaken by plants during the growing season in the east and the northwest site were 18.20 and 10.66g/m2.yr for N, 0.06 and 0.03g/m2.yr for P, 0.23 and 0.11g/m2.yr for K, respectively. the minerals returned to the soil by litter decomposition and roots decay in the east and the northwest site were 16.19 and 9.62g/m2.yr for N, 0.046 and 0.021g/m2.yr for P, 0.211 and 0.099g/m2.yr for K, respectively. The turnover the rate (absorbed/returned) of the nutrients in the east and the northwest site were 1.13 and 1.14 for N, 1.30 and 1.43 for P, 1.09 and 1.11 for K, and the absorption rates of minerals were 0.39 and 0.29% for N, 3.16 and 1.88% for P, 0.91 and 0.57% for K, respectively.

A Study on the Long-Run Equilibrium Between KOSPI 200 Index Spot Market and Futures Market (분수공적분을 이용한 KOSPI200지수의 현.선물 장기균형관계검정)

  • This paper compares long term equilibrium relation of KOSPI 200 which is underling stock and its futures by using general method fractional cointegration instead of existing integer cointegration. Existence of integer cointegration between two price time series gives much wider information about long term equilibrium relation. These details grasp long term equilibrium relation of two price time series as well as reverting velocity to equilibrium by observing difference coefficient of error term when it renounces from equilibrium relation. The result of this study reveals existence of long term equilibrium relation between KOSPI200 and futures which follow fractional cointegration. Difference coefficient, d, of 'two price time series error term' satisfies 0 < d < 1/2 beside bandwidth parameter, m(173). It means two price time series follow stationary long memory process. This also means impulse effects to balance price of two price time series decrease gently within hyperbolic rate decay. It indicates reverting speed of error term is very low when it bolts from equilibrium. It implies to market maker, who is willing to make excess return with arbitrage trading and hedging risk using underling stock, how invest strategy should be changed. It also insinuates that information transition between KOSPI 200 Index market and futures market does not working efficiently.

Gamma-ray Exposure Rate Monitoring by Energy Spectra of NaI(Tl) Scintillation detectors

  • Background: Nuclear facilities in South Korea have generally adopted pressurized ion chambers to measure ambient gamma ray exposure rates for monitoring the impact of radiation on the surrounding environment. The rates assessed with pressurized ion chambers do not distinguish between natural and man-made radiation, so a further step is needed to identify the cause of abnormal variation. In contrast, using NaI(Tl) scintillation detectors to detect gamma energy rates can allow an immediate assessment of the cause of variation through an analysis of the energy spectra. Against this backdrop, this study was conducted to propose a more effective way to monitor ambient gamma exposure rates. Materials and Methods: The following methods were used to analyze gamma energy spectra measured from January to November 2016 with NaI detectors installed at the Korea Atomic Energy Research Institute (KAERI) dormitory and Hanbat University. 1) Correlations of the variation of rates measured at the two locations were determined. 2) The dates, intervals, duration, and weather conditions were identified when rates increased by $5nSv{\cdot}h^{-1}$ or more. 3) Differences in the NaI spectra on normal days and days where rates spiked by $5nSv{\cdot}h^{-1}$ or more were studied. 4) An algorithm was derived for automatically calculating the net variation of the rates. Results and Discussion: The rates measured at KAERI and Hanbat University, located 12 kilometers apart, did not show a strong correlation (coefficient of determination = 0.577). Time gaps between spikes in the rates and rainfall were factors that affected the correlation. The weather conditions on days where rates went up by $5nSv{\cdot}h^{-1}$ or more featured rainfall, snowfall, or overcast, as well as an increase in peaks of the gamma rays emitted from the radon decay products of $^{214}Pb$ and $^{214}Bi$ in the spectrum. This study assumed that $^{214}Pb$ and $^{214}Bi$ exist at a radioactive equilibrium, since both have relatively short half-lives of under 30 minutes. Provided that this assumption is true and that the gamma peaks of the 352 keV and 1,764 keV gamma rays emitted from the radionuclides have proportional count rates, no man-made radiation should be present between the two energy levels. This study proved that this assumption was true by demonstrating a linear correlation between the count rates of these two gamma peaks. In conclusion, if the count rates of these two peaks detected in the gamma energy spectrum at a certain time maintain the ratio measured at a normal time, such variation can be confirmed to be caused by natural radiation. Conclusion: This study confirmed that both $^{214}Pb$ and $^{214}Bi$ have relatively short half-lives of under 30 minutes, thereby existing in a radioactive equilibrium in the atmosphere. If the gamma peaks of the 352 keV and 1,764 keV gamma rays emitted from these radionuclides have proportional count rates, no man-made radiation should exist between the two energy levels.
