Aortic Root Remodeling Procedure in Marfan Syndrome associated with Aortic Dissection: Yacoub-David Technique (말판증후군과 동반된 해리성 대동맥류에 대한 대동맥 근부 개형술(改型術): Yacoub-David 수술법)

  • We operated on a Marfan patient who had Stanford type A acute aortic dissection, aortic root aneurysm, and aortic regurgitation. The Yacoub-David aortic root remodeling procedure which preserves native aortic valve and replaces all three aortic sinuses and ascending aorta by a Dacron graft, was applied for this patient. A 24mm Hemashield graft was designed to three tongues at the aortic root end to meet the shape of the Valsalva sinuses. The patient recovered from the procedure uneventfully and there was no aortic regurgitation posto-peratively.

Co-Expression of a Chimeric Protease Inhibitor Secreted by a Tumor-Targeted Salmonella Protects Therapeutic Proteins from Proteolytic Degradation

  • Sunflower trypsin inhibitor (SFTI) is a 14-amino-acid bicyclic peptide that contains a single internal disulfide bond. We initially constructed chimeras of SFTI with N-terminal secretion signals from the Escherichia coli OmpA and Pseudomonas aeruginosa ToxA, but only detected small amounts of protease inhibition resulting from these constructs. A substantially higher degree of protease inhibition was detected from a C-terminal SFTI fusion with E. coli YebF, which radiated more than a centimeter from an individual colony of E. coli using a culture-based inhibitor assay. Inhibitory activity was further improved in YebF-SFTI fusions by the addition of a trypsin cleavage signal immediately upstream of SFTI, and resulted in production of a 14-amino-acid, disulfide-bonded SFTI free in the culture supernatant. To assess the potential of the secreted SFTI to protect the ability of a cytotoxic protein to kill tumor cells, we utilized a tumor-selective form of the Pseudomonas ToxA (OTG-PE38K) alone and expressed as a polycistronic construct with YebF-SFTI in the tumor-targeted Salmonella VNP20009. When we assessed the ability of toxin-containing culture supernatants to kill MDA-MB-468 breast cancer cells, the untreated OTG-PE38K was able to eliminate all detectable tumor cells, while pretreatment with trypsin resulted in the complete loss of anticancer cytotoxicity. However, when OTG-PE38K was co-expressed with YebF-SFTI, cytotoxicity was completely retained in the presence of trypsin. These data demonstrate SFTI chimeras are secreted in a functional form and that co-expression of protease inhibitors with therapeutic proteins by tumor-targeted bacteria has the potential to enhance the activity of therapeutic proteins by suppressing their degradation within a proteolytic environment.

  • Art works reflect the spirit and aesthetic information of the times. More specially clothing shown in paintings indicates the phases of the times and contains the aesthetic values of the era. The purpose of this study is to examine the characteristics of German Romantic Era on male and female clothing style represented in Caspar David Friedrich's paintings. The study aims to highlight the following: First, German romanticism ironically gives conceptual importance but high value to freedom of inner self and imaginative sensitivity of human beings. Second, the paintings of Friedrich show the consistency with nature through human beings' inner self and transcendency ultimately expressing religious ideals paradoxically. Third, the characteristics of male and female clothing style shown in Friedrich's paintings are considered religious, contemplative, paradoxical, transcendental, conceptual, political, grass-roots oriented, and female liberating. Female clothing reflects simple empire-style, while male clothing shows romantic style of people's leaders, reflecting the political features in those days.

  • Aesthetic values reflecting the phases of the time are well represented in the clothing styles in paintings. Especially, modern German romantic artists' ideologies on the essence of nature and life are reflected in the figures of paintings. This paper aims at finding out how the sublime beauty of German romantic clothing styles is represented in the paintings of Caspar David Friedrich. The research is summarized as follows. First, the German romanticism led the modern romanticism and was especially enthusiastic about the sublime beauty of arts. Second, the sublime means the exaltation of passionately soaring souls and has the mysterious transcendence that unpleasant feeling is elevated to pleasant feeling inside men who are so feeble compared with vast nature. Third, the male and female clothing styles in his paintings represent the religious, contemplative, paradoxical, transcendental, liberating sublime beauties of the German romanticism. Accordingly, a study on the sublime beauty of the German romantic clothing styles will renew the interpretation and aesthetic values of early modern clothing.

Imaging Cancer Metabolism

  • It is widely accepted that altered metabolism contributes to cancer growth and has been described as a hallmark of cancer. Our view and understanding of cancer metabolism has expanded at a rapid pace, however, there remains a need to study metabolic dependencies of human cancer in vivo. Recent studies have sought to utilize multi-modality imaging (MMI) techniques in order to build a more detailed and comprehensive understanding of cancer metabolism. MMI combines several in vivo techniques that can provide complementary information related to cancer metabolism. We describe several non-invasive imaging techniques that provide both anatomical and functional information related to tumor metabolism. These imaging modalities include: positron emission tomography (PET), computed tomography (CT), magnetic resonance imaging (MRI), magnetic resonance spectroscopy (MRS) that uses hyperpolarized probes and optical imaging utilizing bioluminescence and quantification of light emitted. We describe how these imaging modalities can be combined with mass spectrometry and quantitative immunochemistry to obtain more complete picture of cancer metabolism. In vivo studies of tumor metabolism are emerging in the field and represent an important component to our understanding of how metabolism shapes and defines cancer initiation, progression and response to treatment. In this review we describe in vivo based studies of cancer metabolism that have taken advantage of MMI in both pre-clinical and clinical studies. MMI promises to advance our understanding of cancer metabolism in both basic research and clinical settings with the ultimate goal of improving detection, diagnosis and treatment of cancer patients.

Squamous Cell Carcinoma in Pere David Deer (사불상에서 자연발생한 편평상피암)

  • Metastatic squamous cell carcinoma was described in two female Pere David's deer reared at Everland zoo. Both deer with chronic emaciation had the mass on right maxilla(9$\times$11$\times$15cm or 20$\times$17cm) which was composed of miliary tan creamy contents and encapsulated by connective tissue. The undulating contents in the mass was a1so extended in the underlying or adjacent soft palate, maxillary and frontal sinus causing severe bone destruction. In one deer, two fistula were also noted in the right periocular area. Histologically, the neoplastic masses of both deer consist of anastomosing cords or nests of squamous epithelial cells with intercellular bridge or keratin pearl formation. The neoplastic cells invade deep into the subcutis and bony tissue. Mitotic figure was rare. Multifocal areas of necrosis and hemorrhages were also noted in the dermis. Metastasis to maxilla and ethmoid bones and/or to submandibular lymph node was noted in both cases. The diagnosis was based on the results of histopathology.

A Study on the 'Fragmentation-Assemblage' Design Method and Meaning found in the Architecture of David Chipperfield (데이비드 치퍼필드 건축에 나타난 '분절-집합'의 디자인 방법과 의미에 관한 연구)

  • Over the 30 years of practice, most of David Chipperfield's design projects shows that there are several unique design methods and characteristics applied in the design processes. 'Form Matters' exhibition held in London Design Museum in 2009 was crucial opportunity to summarize his architectural philosophy. The main target of this study is to analyze the Chipperfield's consistent design methods and their meaning based on his architectural philosophy. In chapter 2, his architectural background, early projects and the essence of 'Form Matters' exhibition were studied. In chapter 3, the projects built after 2000 were summarized and main design method that is 'Fragmentation & assemblage' were extracted from them. After selecting 6 case projects, each cases were analyzed in detail based on 3 analytical elements: A) Reason to segment the given volume into smaller volumes, B) Relationship between Fragmentation and Program, C) Relationship between Fragmentation-Assemblage and Interior/Exterior Space. In chapter 5, case analysis were summarized and deep meaning of design method were researched. This study founded that Chipperfield has used Fragmentation-Assemblage method to get 'flexible respond to site conditions', 'appropriate composition of program and space', 'various visual-perceptual experience through intermediary space and materiality'. As a conclusion, his unique design method has a role to logically and aesthetically spatialize all the given contextual situations into a specific architectural 'form'.

David Henry Hwang's M. Butterfly: Postmodern Other, (Post-)Imperialist Melancholy and Western Masculinity in Crisis (포스트모던 제국의 우울증-데이빗 헨리 황의 『엠. 버터플라이』)

  • This article discusses David Henry Hwang's M. Butterfly as a suggestive text for examining Western masculinity in crisis in the post-imperialist age, in which territorial imperialism is no longer valid. Previous scholarship on M. Butterfly has centered around the interlocking dynamics of imperialism, racism and sexism. Such critical attentions focus on how Hwang deconstructs racialized significations of the East and the West. In these discussions, the issue of gender is often addressed merely as a trope to represent the power relations between the East and the West. As such, gender as well as sexuality is highlighted as the very source of subversion of the power relations. My discussion departs from a critique of the gendered trope of the East and the West, highlighting a postmodern agent, the allegedly feminized character Song Lining: a Chinese actor who passes for a woman for political purposes in postcolonial China. Remaining an "inappropriate/d other" in the gendered imperialist discourse, Song becomes an emergent subject, who is capable of playing gender ambiguity for reclaiming a devalued identity, that of homosexual Asian man. Discussing how the central character Rene Gallimard's masculine identity is constructed in a cross-cultural space and how it evolves, I also argue that Gallimard's melancholic death signifies a historical unsustainability of imperialist masculinity in the postmodern/postcolonial age since World War II.

Flow Velocity Change of David Glacier, East Antarctica, from 2016 to 2020 Observed by Sentinel-1A SAR Offset Tracking Method

  • This study measures the change of ice flow velocity of David Glacier, one of the fast-moving glaciers in East Antarctica that drains through Drygalski Ice Tongue. In order to effectively observe the rapid flow velocity, we applied the offset tracking technique to Sentinel-1A SAR images obtained from 2016 to 2020 with 36-day temporal baseline. The resulting velocity maps were averaged and the two relatively fast points (A1 and A2) were selected for further time-series analysis. The flow velocity increased during the Antarctic summer (around December to March) over the four years' observation period probably due to the ice surface melting and reduced friction on the ice bottom. Bedmap2 showed that the fast flow velocities at A1 and A2 are associated with a sharp decrease in the ice surface and bottom elevation so that ice volumetric cross-section narrows down and the crevasses are being created on the ice surface. The local maxima in standard deviation of ice velocity, S1 and S2, showed random temporal fluctuation due to the rotational ice swirls causing error in offset tracking method. It is suggested that more robust offset tracking method is necessary to incorporate rotational motion.
